目的 评价碳纤维复合材料人工颅骨板在急性期的生物相容性。方法 制作碳纤维复合材料、含重组人骨形态发生蛋白-2(rhBMP-2)的碳纤维复合材料、钛网、MEM培养基、含有0.64%苯酚的MEM培养基的浸出液,用于测试。采用L929小鼠成纤维细胞进行细胞毒性试验,采用健康ICR小鼠进行急性毒性试验,采用新西兰大白兔耳静脉血进行急性溶血试验,采用白化豚鼠进行皮肤致敏试验。结果 碳纤维复合材料、含rhBMP-2的碳纤维复合材料、钛网浸提液对L929细胞的相对增殖率均在75%以上,细胞毒性1级,无明显细胞毒性;小鼠腹腔注射相应材料的浸提液后,均活动良好,均无不良反应,均无明显急性毒性;碳纤维复合材料、含rhBMP-2的碳纤维复合材料、钛网浸提液溶血率均小于5%,均不引起溶血反应;碳纤维复合材料、含rhBMP-2的碳纤维复合材料、钛网浸提液不引起致敏性。结论 碳纤维复合人工颅骨板急性期生物相容性良好。  相似文献

Nanostructured neural interfaces, comprising nanotubes or nanowires, have the potential to overcome the present hurdles of achieving stable communication with neuronal networks for long periods of time. This would have a strong impact on brain research. However, little information is available on the brain response to implanted high-aspect-ratio nanoparticles, which share morphological similarities with asbestos fibres. Here, we investigated the glial response and neuronal loss in the rat brain after implantation of biostable and structurally controlled nanowires of different lengths for a period up to one year post-surgery. Our results show that, as for lung and abdominal tissue, the brain is subject to a sustained, local inflammation when biostable and high-aspect-ratio nanoparticles of 5 μm or longer are present in the brain tissue. In addition, a significant loss of neurons was observed adjacent to the 10 μm nanowires after one year. Notably, the inflammatory response was restricted to a narrow zone around the nanowires and did not escalate between 12 weeks and one year. Furthermore, 2 μm nanowires did not cause significant inflammatory response nor significant loss of neurons nearby. The present results provide key information for the design of future neural implants based on nanomaterials.  相似文献

Background: The ability of urinary biomarkers to predict residual renal function (RRF) decline in peritoneal dialysis (PD) patients has not been defined. The present study aimed to explore the utility of established biomarkers from kidney injury models for predicting loss of RRF in incident PD patients, and to evaluate the impact on RRF of using neutral-pH PD solution low in glucose degradation products.♦ Methods: The study included 50 randomly selected participants from the balANZ trial who had completed 24 months of follow-up. A change in glomerular filtration rate (GFR) was used as the primary clinical outcome measure. In a mixed-effects general linear model, baseline measurements of 18 novel urinary biomarkers and albumin were used to predict GFR change. The model was further used to evaluate the impact of biocompatible PD solution on RRF, adjusted for each biomarker.♦ Results: Baseline albuminuria was not a useful predictor of change in RRF in PD patients (p = 0.84). Only clusterin was a significant predictor of GFR decline in the whole population (p = 0.04, adjusted for baseline GFR and albuminuria). However, the relationship was no longer apparent when albuminuria was removed from the model (p = 0.31). When the effect of the administered PD solutions was examined using a model adjusted for PD solution type, baseline albuminuria, and GFR, higher baseline urinary concentrations of trefoil factor 3 (TFF3, p = 0.02), kidney injury molecule 1 (KIM-1, p = 0.04), and interferon γ-induced protein 10 (IP-10, p = 0.03) were associated with more rapid decline of RRF in patients receiving conventional PD solution compared with biocompatible PD solution.♦ Conclusions: Higher urinary levels of kidney injury biomarkers (TFF3, KIM-1, IP-10) at baseline predicted significantly slower RRF decline in patients receiving biocompatible PD solutions. Findings from the present investigation should help to guide future studies to validate the utility of urinary biomarkers as tools to predict RRF decline in PD patients.  相似文献

Mesoporous materials are promising candidates for improving dissolution rate of poorly water-soluble drugs in vitro and their bioavailability in vivo. In the present study, sixteen batches of celecoxib-loaded PSi particles with pore sizes ranging from 17 to 58 nm and celecoxib content from 5 to 36 w-% were prepared and a detailed physicochemical characterization of the drug was performed by several methods. Interaction between co-culture of Caco-2/HT29-MTX cells and unloaded PSi particles was tested in toxicity assays, and increased toxicity for particles with large pore size was observed. Dissolution rate of celecoxib was improved in vitro by lowering the drug loading degree which hindered the recrystallization of celecoxib on the external surface of the particles. The fastest permeation of loaded celecoxib through the co-culture monolayer as well as the highest bioavailability in rats was observed with the particles with small pore size and low loading degree. New insights were obtained on how various parameters of the mesoporous delivery system affect the state of the drug inside the pores and its release in vitro and in vivo.  相似文献

As the official journal of the International Federation for Artificial Organs, the International Faculty for Artificial Organs, the International Society for Rotary Blood Pumps, the International Society for Pediatric Mechanical Cardiopulmonary Support, and the Vienna International Workshop on Functional Electrical Stimulation, Artificial Organs continues in the original mission of its founders “to foster communications in the field of artificial organs on an international level.” Artificial Organs continues to publish developments and clinical applications of artificial organ technologies in this broad and expanding field of organ Replacement, Recovery, and Regeneration from all over the world. We take this time also to express our gratitude to our authors for offering their work to this journal. 目的:研究国内2种新研制骨植入钛合金Ti1、Ti2对成骨细胞生物学行为的影响。方法:采用SD乳鼠体外原代分离培养的成骨细胞,将细胞分别接种于新型钛合金表面建立体外共同培养。采用MTT比色试验检测第3天成骨细胞的增殖百分率,采用扫描电镜(SEM)观察细胞形态学变化,并且对培养第5天时细胞碱性磷酸酶(ALP)功能活性进行检测。结果:成骨细胞在新合金表面增殖百分率、碱性磷酸酶活性检测值均高于对照组,细胞增殖百分率及碱性磷酸酶吸光度值与对照组间统计学分析无显著性差异(P>0.05)。扫描电镜观察细胞在新合金表面伸展状况良好、黏附牢固,并具有成骨细胞典型形态特征。结论:2种新型钛合金对成骨细胞学行为无不良影响。  相似文献

目的 比较3D打印钛网与成品钛网的机械力学性能差异,并评价3D打印钛网对细胞生长及分化的影响。方法 采用激光打印技术制备个体化3D打印钛网,用静态拉伸压缩载荷实验检测机械力学性能,评估机械力学性能。选择4周龄雄性SD大鼠骨髓间充质干细胞,与不同孔径大小的3D打印钛网共培养。采用CCK-8法检测各组细胞增殖情况;对各组细胞进行成骨诱导分化后,检测碱性磷酸酶(ALP)活性改变,运用实时荧光定量PCR检测相关成骨基因的表达;通过扫描电镜及活/死细胞染色,观察细胞在3D打印钛网上的黏附和生长情况。采用SPSS 22.0软件包对数据进行统计学分析。结果 3D打印钛网的拉伸压缩负载实验结果显示其具有优异的力学性能;相比对照组,不同孔径钛网对细胞增殖的影响不大,细胞在钛网上显现出良好的黏附和生长状态。3D打印钛网对成骨分化具有一定的促进作用。结论 3D打印钛网的机械力学性能优异,且具有良好的生物相容性。  相似文献

Introduction

The aim of this study was to evaluate the rat alveolar bone response after the implantation of experimental light-cured mineral trioxide aggregate (MTA) or Angelus MTA (Angelus, Londrina, Paraná, Brazil) by histological and fluorescence analysis.

Methods

Thirty Wistar Albino rats were divided into three groups. In the control group, empty polyethylene tubes were inserted into the rat alveolar sockets immediately after extraction. In the other groups, the tubes were filled with light-cured MTA or Angelus MTA. Five animals from each group were injected with calcein on day 7, alizarin on day 14, and oxytetracycline on day 21. On day 30, these animals were killed, and the right hemimaxillas were removed and histologically processed. Half of the maxillas were processed and stained with hematoxylin and eosin. The remaining maxillas were processed for fluorescence analysis and stained with Stevenel blue and alizarin red. New bone was histomorphometrically evaluated using a Merz grid.

Results

The light-cured MTA presented a similar response when compared with Angelus MTA; it was characterized by a mild inflammatory response and complete bone healing. In the light-cured MTA group, the fluorescence areas were more evident at 21 days, showing an increase in bone formation. However, dystrophic mineralization was observed only with Angelus MTA.

Conclusions

It was concluded that both materials present a similar inflammatory response and bone healing, but dystrophic mineralization was observed only with Angelus MTA.  相似文献

目的 :对表面微弧氧化 (microarcoxidation ,MAO)处理后的纯钛材料进行成骨细胞生物相容性检测 ,评价改进的MAO工艺应用于钛植入材料表面处理的可能性。方法 :纯钛材料经过 2种MAO处理后 (MAO 1和MAO 2工艺 ) ,采用MC 3T3细胞系对不同时间点成骨细胞在材料表面的附着率、生长增殖情况以及ALP活性进行检测 ,以未经处理光滑纯钛表面作为对照 ,以SPSS对实验结果进行统计分析。结果 :早期 (0 .5h、1h)细胞附着率差异有统计学意义 ,MAO 1组 >MAO 2组 >纯钛组 ;2h后 ,MAO 1组与MAO 2组无差异 ,2组细胞附着率都显著高于纯钛组。细胞增殖及ALP活性测试中 ,MAO 1处理组在各时间点都显著高于另外 2组。结论 :MAO处理后的纯钛对成骨细胞的生物相容性优于未处理组 ,改进的MAO 1处理工艺较一般工艺可以更有效提高成骨细胞的早期粘附、增殖及ALP活性。  相似文献

目的:了解类骨质羟基磷灰石(OHA)的生物相容性,初步探讨其作为骨替代材料的可行性。方法:12只新西兰白兔随机分为4组,在双侧股骨内侧髁形成9mm×4mm×5mm大小的缺损,分别植入OHA和对照材料,术后2、4、8、12周取标本,进行大体标本、X线、组织学切片、扫描电镜观察及成骨面积的定量分析。结果:实验组成骨细胞易于粘附、生长、增殖,骨小梁形成钙化早,新骨面积百分比高于对照组,差异有统计学意义。结论:类骨质羟基磷灰石是一种良好的骨替代材料,具有广阔的应用前景。  相似文献
