目的 探讨mi RNA-135a(mi R-135a)在肝癌组织中的表达及其临床意义。方法 采用实时荧光定量PCR检测20例肝癌组织及其癌旁组织中mi R-135a的表达水平,并通过脂质体介导的方法将mi R-135a模拟物(mi R-135a mimics)转染肝癌细胞株Hep G2,采用实时荧光定量PCR测定转染后细胞中mi R-135a的表达水平;通过平板克隆实验和MTT法分别检测转染后细胞克隆形成率和存活率的变化,并对mi R-135a调控相关靶细胞的基因表达水平进行检测,观察mi R-135a在肝癌细胞维持正常功能中的作用。结果 通过对20例肝癌组织及其癌旁组织中mi R-135a的表达水平进行检测,结果 mi RNA-135a的表达在正常组织中相对更高,且两组之间存在显著性差异(P0.05);与阴性对照组比较,转染mi R-135a mimics后Hep G2细胞中mi R-135a表达水平显著提高(P0.05),而形成单克隆能力和细胞存活率均显著下降(P0.05),同时转染组细胞中与mi R-135a细胞调控相关靶基因FOS、PI3、Jak2、Stat3的m RNA表达量相较于NC组均显著降低(P0.05),表明mi R-135a表达提高后抑制细胞形成单克隆能力并降低存活率。结论 mi R-135a在正常组织和肝癌组织中差异表达,同时当mi R-135a表达提高时抑制肝癌细胞形成单克隆能力和降低细胞存活率,表明mi R-135a可能通过一些相关靶基因直接或间接调控Hep G2肝癌细胞的存活,在肝癌的发生过程中可能起到作用,mi R-135a可能成为临床治疗肝癌和预后的重要靶点。 相似文献

Background
In an experimental animal model of islet transplantation, stable induction of insulin-dependent diabetes mellitus (IDDM) and islet isolation from donor pancreas are essential. Total pancreatectomy for IDDM induction and islet procurement in nonhuman primates leads to unwanted loss of exocrine function and may lead to morbidities associated with IDDM.Methods
IDDM induction with streptozotocin (STZ) is associated with drug toxicity of STZ and necessitates the killing of another animal for islet procurement. In this study, we performed a subtotal pancreatectomy combined with reduced STZ injection to induce IDDM and procure islets in a nonhuman primate model.Results
Twelve cynomolgus monkeys received low-dose STZ injections (60 mg/kg) simultaneously with subtotal pancreatectomy. All monkeys recovered from the procedure without complications. IDDM was induced in the animals. 57,691 ± 16,050 islets were isolated from the resected pancreas and transplanted into other monkeys.Conclusions
Simultaneous subtotal pancreatectomy and low-dose STZ injection represent an effective and safe method to create an animal model of insulin dependence diabetes, while at the same time providing sufficient amounts of fresh islet cells for allotransplantation without requiring killing of additional animals. 相似文献54.

ObjectiveSubconjunctival hemorrhage (SCH) is an important minor side effect that might affect patient compliance to antivascular endothelial growth factor (anti-VEGF) intravitreal injection treatment (IVI). We sought to compare SCH incidence and pain score responses after topical oxymetazoline in naïve patients undergoing a single IVI of ranibizumab for diabetic macular edema.MethodsProspective, randomized, double-blinded, single centre study. One hundred two patients naïve to anti-VEGF were assigned to receive either topical oxymetazoline or placebo 30 minutes before IVI. SCH incidence and area were measured by slit lamp 24 hours after, and pain was evaluated 5 minutes and 24 hours after.ResultsSCH incidence was reported on 72% in control group versus 51% in oxymetazoline group (p = 0.037). Mean size of SCH was 16.82 mm2 in control group versus 12.55 mm2 in oxymetazoline group (p = 0.394). Prevalence of local pain in the overall study population was 60%. No significant statistical difference was achieved between groups 5 minutes or 24 hours after IVI in either pain scale evaluation.ConclusionAdministration of topical oxymetazoline 30 minutes before IVI is a single, harmless, cost-effective intervention that decreases the incidence of subconjunctival hemorrhage. ObjectiveThe alternations of mtDNA may play an important role in the molecular pathogenesis and process of Pelvic Organ Prolapse (POP) formation in both pre-menopausal and post-menopausal women. The aim of the present study is to analyze the association between the mitochondrial biogenesis gene and development of POP in the uterosacral ligaments (UL) of pre-menopausal women.Materials and methodsSeventy one pre-menopausal women, all below 52 years of age, were enrolled in this study. UL biopsies were obtained from uterine specimens taken from 33 women with POP (n = 33, study group) and 38 myoma patients without POP (n = 38, control group). Quantitative Real-Time PCR was performed to measure mitochondrial DNA (mtDNA) copy number and mtDNA4977. Western blotting and immunohistochemistry were used to assess the protein expression of PGC-1α, TFAM, NRF-1 and NRF-2. Statistical analysis was performed using SPSS statistical software and the Mann–Whitney U test, and the continuous variables were analyzed using the Student's t-test in demographic data.ResultsThere were no significant differences in the patient demographics between the two groups (p > 0.05). The mtDNA copy number in the UL of pre-menopausal patients with prolapse was significantly higher than that in the no prolapse group (p = 0.008). There were no significant differences between the mtDNA4977 of the POP and non-POP groups, but a significantly higher expression of PGC-1α in the POP group compared to the non-POP group (1.59 ± 1.30 v.s. 0.66 ± 0.53; p = 0.036). The expression of TFAM in the POP group was higher than in the non-POP group). There was no significant difference in the TFAM(p = 0.377), NRF-1 and NRF-2 expression between the POP and non-POP groups (p = 0.647; p = 0.682).ConclusionsChanges in the PGC-1α and mtDNA copy number may play a role in the development of Pelvic Organ Prolapse in pre-menopausal patients. 相似文献

目的了解安徽省合肥市健康人群Y群和W135群流脑抗体水平,预测流脑疫情。方法2008年9-10月,在合肥市采用分层随机抽样方法调查健康人群1184人,并采集血清,采用酶联免疫吸附试验(ELISA)检测Y群和W135群流脑抗体水平。结果调查1184人中,Y群流脑抗体阳性率为61.8%,几何平均浓度(GMC)为2.33μg/mL;W135群流脑抗体GMC为1.32μg/mL,阳性率为20.9%;抗体几何平均浓度农村和城市、性别间比较,差异均无统计学意义,各年龄段抗体水平差异有统计学意义。结论合肥市健康人群流脑Y群和W135群抗体水平总体不高,有计划地在人群中接种含有W135群和Y群的流脑疫苗是必要的。 相似文献
