目的 观察不同剂量抗血管内皮生长因子单克隆抗体Bevacizumab兔眼玻璃体腔注射的视网膜毒性作用。 方法 16只新西兰无色素兔的32只眼随机分为药物注射组和对照组,药物注射组又根据玻璃体腔注射药物剂量不同分为A、B、C组,玻璃体腔注射Bevacizumab剂量分别为0.05、0.10、0.25 ml,分别含Bevacizumab 1.25、2.50、6.25 mg。对照组玻璃体腔注射0.9%生理盐水0.10 ml。注药后1、2、4周行视网膜电图(ERG)检查。另外 ,在兔眼玻璃体腔注射Bevacizumab后1、2、4周,每组各摘除2只兔眼,行视网膜组织形态及超 微结构的光学显微镜和透射电子显微镜观察。 结果 兔眼玻璃体腔注射 Bevacizumab后1、2、4周,兔眼ERG各项反应波形均正常,振幅均未出现异常改变(P>0.05)。光学显微镜下观察 ,药物注射组和对照组视网膜各层组织形态在各时间点均未见异常。透射电子显微镜观察, A、B组与对照组无明显差异;C组视网膜光感受器细胞出现部分线粒体损伤,发生肿胀和积 水变,4周时病变无缓解。 结论 单次兔眼玻璃体腔注射Bevacizumab 1.25 mg或2.50 mg是安全的。 (中华眼底病杂志,2008,24:193-196)  相似文献

We have investigated the properties of the newly synthesized proton-pump inhibitor, 3-butyryl-8-methoxy-4-[(2-thiophenyl)amino]quinoline (YJA20379–6), on gastric mucosal proton-pump (H+/K+-ATPase) activity, gastric acid secretion and gastroduodenal lesions in experimental rats. YJA20379–6 markedly inhibited H+/K+-ATPase activity in rabbit isolated gastric mucosal microsomes, confirming its classification as a proton-pump inhibitor. The inhibitory efficacy of YJA20379-6 on the proton pump was approximately 14-times higher than that of omeprazole at pH 7.4. YJA20379–6 given intraduodenally had a potent inhibitory effect on gastric secretion in pylorus-ligated rats (ED50 22.9 mg kg?1) but was less active than omeprazole. Pretreatment of rats with YJA20379-6 dose-dependently protected the gastric mucosa from damage induced by water-immersion stress, indomethacin and absolute ethanol, and the duodenal mucosa from damage induced by mepirizole. Repeated administration of YJA20379-6 also dose-dependently accelerated the spontaneous healing of acetic acid-induced gastric ulcers. These results suggest that YJA20379-6 has potent anti-secretory and anti-ulcer effects which are exerted by suppression of H+/K+-ATPase activity in gastric parietal cells. YJA20379–6 might be useful for the clinical treatment of peptic ulcer diseases.  相似文献

[目的]探讨针刺加推拿治疗神经根型颈椎病的疗效。[方法]选择100例颈椎病的患者,采用针刺配合推拿治疗。针刺主穴选取颈夹脊3~7穴、风池、后溪。推拿采用颈项部理筋、端提旋扳法。[结果]治愈77例(77%),好转20例(20%),未愈3例(3%)。总有效率为97%。[结论]针刺合推拿治疗颈椎病疗效确切。  相似文献

Aim: A low‐protein diet (LPD) is a conservative treatment in patients with chronic kidney disease (CKD) to improve uremic symptoms and slow the progression of renal dysfunction. However, the deleterious effects of protein restriction on nutritional status have raised concern. We investigated whether ketoanalogs supplementation in CKD patients who had training on LPD retards the progression of CKD and maintains nutritional status. Methods: Data were collected retrospectively from 120 consecutive patients in the CKD stages III and IV. Firstly all patients were restricted to LPD alone for 6 months (LPD alone), and then ketoanalogs of essential amino acids (KA) were supplemented for 6 months. Results: The adequate LPD had not achieved in both periods. The declining slopes of glomerular filtration rate (GFR) during the LPD + KA period were significantly lower than those during the LPD alone period. This improvement in GFR was apparent in both subjects with diabetics and non‐diabetic patients. Mean serum total cholesterol levels decreased in LPD + KA compared with LPD alone period. However, serum albumin levels did not change. Responders showed a higher prevalence of diabetes and higher serum albumin levels during the LPD alone period. Multivariate analysis revealed that responsiveness to LPD + KA was independently related to diabetes (p = 0.006) and high serum albumin levels (p = 0.011) in the LPD alone period. Conclusion: KA supplementation on over LPD delayed the progression of CKD without deteriorating nutritional status, and initial serum albumin levels could be an independent factor.  相似文献

The introduction of many new packable composites suggests that these products are rapidly gaining popularity. The purpose of this study was to evaluate the in vitro properties of a variety of packable composites and to determine if significant enhancements in physical and mechanical properties have been achieved for these materials compared with two popular nonpackable posterior composites. For the five packable and two regular composites tested (ALERT, Pyramid-Dentin, Pyramid-Enamel, Solitaire, SureFil, Heliomolar, and Z100), the values for fracture toughness, flexure strength, flexure modulus, hardness, and volumetric polymerization shrinkage were determined. In general, although the packable composites were of heavier consistency, they had mechanical properties that were intermediate to (ALERT, Pyramid, and SureFil) or lower than (Solitaire) those of the nonpackable materials. These results could have been predicted based on the similar meth-acrylate resin chemistry and filler volumes of the various composites. No composite had adequate depth-of-cure when tested in increments greater than 2 mm thick. Polymerization contraction of the packable composites was similar to or higher than that of the nonpackable composites. In addition, the radiopacity of at least one material, Solitaire, was not considered to be adequate (less than 2 mm of aluminum). The results of this study suggest that these packable composites are unlikely to offer improved clinical performance over well-placed nonpackable composites.
CLINICAL SIGNIFICANCE
Packable composites have physical and mechanical properties that are similar to those of nonpackable posterior composites and are, therefore, expected to provide equivalent clinical performance.  相似文献

目的:应用蛋白质指纹图谱技术筛选子宫腺肌病患者血清中特异表达蛋白,并建立多标记物诊断模型。方法:应用表面增强激光解吸电离飞行质谱(SELDI-TOF-MS)技术检测子宫腺肌病患者和健康人(对照组)血清中的差异表达蛋白,并绘制蛋白指纹图谱,利用多标记物诊断模型对建模组及盲筛组分别进行检测。结果:在子宫腺肌病患者和对照组之间共找到12个差异蛋白峰(P<0.05), 利用质核比为 2 748 和5 759 的蛋白质峰建立子宫腺肌病的诊断模型,其敏感度为100%,特异度为80%,经盲法验证,该模型诊断的敏感度和特异度分别为86%和100%。结论:利用SELDI-TOF-MS技术筛选并建立的多蛋白标记物的诊断模型具有较高的敏感度和特异度,为初步诊断子宫腺肌病提供了方向。  相似文献
