The leukocyte recruitment and transmigration across the endothelial barrier into the vessel wall are crucial steps in atherosclerosis. Leukocyte trafficking on the endothelium is elicited by induction of endothelial adhesion molecules, and its transmigration is mediated by degradation of basement membrane proteins through enzymatic activity of matrix metalloproteinases (MMP). The current study investigated whether resveratrol, a polyphenol present in grapes and red wine, was capable of inhibiting leukocyte adhesion to tumor necrosis factor (TNF)-α-activated endothelium. It was found that resveratrol inhibited the TNF-α-activated endothelial expression of vascular cell adhesion molecule-1 in a dose-dependent manner. In addition, resveratrol hampered THP-1 monocyte adhesion to activated endothelial cells. This study further examined whether resveratrol interfered with transendothelial migration of leukocytes. The MMP-2 gelatinolytic activity of endothelial cells was enhanced by TNF-α, which was attenuated by an addition of ≥25 µM resveratrol. In addition, 25 µM resveratrol mitigated the MMP-9 activity of THP-1 cells, followed by a marked inhibition of transendothelial migration. These results demonstrated that resveratrol suppressed monocyte adhesion and migration induced by TNF-α through modulating expression of adhesion molecules and gelatinolytic activity of MMP. These findings suggest that dietary resveratrol may be therapeutic agent for inhibiting leukocyte recruitment into the subendothelium during inflammatory atherosclerosis. 相似文献

目的研究白藜芦醇诱导宫颈癌细胞株Hela的细胞生长和发育的改变,细胞周期的进行和凋亡等。方法MTT法测定白藜芦醇对Hela细胞的作用。3H-TdR掺入法检测白藜芦醇对Hela细胞DNA合成的影响。流式细胞术分析细胞周期。结果白藜芦醇作用后,Hela细胞的增殖速度减缓,DNA合成减少。白藜芦醇在较低浓度时即可诱导较快的可逆的S期停滞。结论合适浓度的白藜芦醇可有效预防早期肿瘤,但不至于引起细胞毒作用和细胞死亡。 相似文献

目的研究顺铂(CDDP)联合白藜芦醇(RES)对人胃癌MGC803细胞株的影响。方法采用MTT法检测两种药物单用和联合应用对细胞的作用,联合应用判断两药舍用效果,流式细胞仪检测MGC803细胞周期分布。结果CDDP可抑制MGC803细胞的生长,且呈荆量及时间依赖性(P〈0.05);CDDP对人胚肺成纤维细胞(HPF)生长起促进作用,且呈剂量依赖性(P〈0.05);CDDP与RES联合应用对MGC803细胞生长的抑制具有协同作用(P〈0.05);CDDP对G0/G1期有阻滞作用,RES有明显的S期阻滞作用,两者可从不同环节抑制细胞增殖。结论CDDP对胃癌MGC803细胞有抑制作用,相同剂量对正常细胞无抑制作用。CDDP与RES对胃癌细胞的抑制具有协同作用,与单用RES相比,CDDP与RES联合应用可减少RES的用药剂量。 相似文献

Four new stilbene C-glucosides, namely trans-3-O-methyl-resveratrol-2-C-β-glucoside (1), cis-3-O-methyl-resveratrol-2-C-β-glucoside (2), trans-3-O-methyl-resveratrol-2-(2-p-coumaric)-C-β-glucoside (cissuside A) (3), and trans-3-O-methyl-resveratrol-2-(3-p-coumaric)-C-β-glucoside (cissuside B) (4), were isolated from the aerial parts of Cissus repens, along with known trans-resveratrol (5), trans-resveratrol-2-C-β-glucoside (6) and cis-resveratrol-2-C-β-glucoside (7). Their structures were established by spectroscopic methods. Stilbene C-glucosides were found in the genus Cissus for the first time. 相似文献

目的研究白藜芦醇对D-半乳糖所致衰老小鼠氧化损伤的缓解作用.方法用D-半乳糖皮下注射制备衰老小鼠模型,同时分别给予不同剂量白藜芦醇(15、30 mg/kg)灌胃,观察其对衰老模型小鼠血清和脑组织抗氧化系统的影响.结果衰老模型小鼠脑组织脂褐质(LF)升高,血清和脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性下降,一氧化氮(NO)和丙二醛(MDA)含量增加.分别给予15,30 mg/kg白藜芦醇灌胃后能使衰老模型小鼠脑组织和血清中NO、MDA不同程度下降,SOD及GSH-Px活性不同程度升高,脑组织中LF降低.结论白藜芦醇可通过清除氧自由基及抗脂质过氧化而抗衰老. 相似文献

In the present study, the effects of resveratrol on 6-hydroxydopamine (6-OHDA)-induced p-aminohippurate (PAH) transport impairment were investigated in vitro using rat renal cortical slices. Cisplatin and cephaloridine (CPH), known nephrotoxins, were used as positive controls. In one series of experiments, renal cortical slices were incubated in a cisplatin-containing medium or a cisplatin-free medium. In another series of experiments, renal cortical slices were incubated in a CPH-containing medium, in a CPH- and probenecid-containing medium, or in a CPH-free medium. Subsequently, for each series of experiments kidney slices were incubated in a media containing PAH or tetraethylammonium. In a further series of experiments, renal cortical slices were incubated in a 6-OHDA-containing medium and in a 6-OHDA-free medium. In another series of experiments, renal cortical slices were incubated in a medium containing 50 µM 6-OHDA, in a 6-OHDA- and resveratrol-containing medium or in a 6-OHDA- and resveratrol-free medium. Subsequently, for each series of experiments kidney slices were incubated in media containing PAH. The results of this study in which slices were incubated in 6-OHDA-containing media indicate that 6-OHDA induced a time- and concentration-dependent decrease in PAH accumulation by renal cortical slices. Resveratrol inhibited the 6-OHDA-induced time-dependent decrease of PAH accumulation in a concentration-dependent manner. Therefore, 6-OHDA causes functional injuries of renal proximal tubule cell membrane, thus leading to impairment of transport processes across the cell membrane and to nephrotoxicity. Resveratrol is proposed to account in part for the protective effect of red wine on the cardiovascular system. Angiotensin II (Ang II) is a potent hypertrophic stimulus in cardiomyocytes. In this study, we determined the effect of resveratrol on Ang II-induced cardiomyocyte hypertrophy. Cultured neonatal rat cardiomyocytes were stimulated with Ang II, and [3H]leucine incorporation and -myosin heavy chain (-MyHC) promoter activity were examined. Intracellular reactive oxygen species (ROS) were measured by a redox-sensitive fluorescent dye, 2 7-dichlorofluorescin diacetate, and the extracellular signal-regulated kinase (ERK) phosphorylation was examined by Western blotting. Resveratrol inhibited Ang II-increased intracellular ROS levels. Furthermore, resveratrol, as well as the antioxidant N-acetyl-cysteine, decreased Ang II- or H2O2-increased protein synthesis, -MyHC promoter activity, and ERK phosphorylation. In summary, we demonstrate for the first time that resveratrol inhibits Ang II-induced cardiomyocyte hypertrophy via attenuation of ROS generation.Abbreviations Ang II
