目的 建立大孔树脂富集纯化蜘蛛香总黄酮工艺方法。方法 对D-101、AB-8、HPD-600、D-301 4种不同极性大孔树脂进行吸附率和解吸率筛选,确定选择HPD-600树脂。考察了HPD-600树脂上样量、pH值、上样体积流量、上样液浓度、洗脱液配比、酸液浓度、乙醇体积分数、洗脱剂用量对蜘蛛香总黄酮的影响,优化大孔树脂纯化工艺参数。结果 HPD-600树脂装柱,上样浓度为3.03 mg/mL,上样容量为24.24 mg/g,先0.1 mol/L HCl的10%乙醇溶液洗脱除杂,直至洗液颜色不再变浅、固形物含量不再增加,后用0.1 mol/L HCl的80%乙醇溶液,洗脱体积流量1 mL/min,洗脱5倍柱体积,收集洗脱液减压浓缩至干得提取物。结论 HPD-600树脂适用于蜘蛛香中总黄酮的纯化,可使蜘蛛香总黄酮质量分数达55.25%,收率可达39.01%。  相似文献

The objective of this study was to examine ochratoxin A (OTA) binding by three lactic acid bacteria (LAB) species: Lactobacillus plantarum, L. brevis, and L. sanfranciscensis. Experiments were conducted using MRS medium and PBS buffer contaminated with 1000 ng/mL OTA and inoculated with live or thermally inactivated bacterial biomass at a concentration of 1 or 5 mg dry weight/mL. It was found that, depending on the strain and biomass density, live bacterial cells reduced OTA content by 16.9% to 35% in MRS medium and by 14.8% to 26.4% in PBS after 24 h of contact. OTA binding was higher in the case of thermally inactivated bacterial biomass (46.2% to 59.8%). The process is very rapid: OTA was removed from PBS as early as after 30 min of contact. The binding of the toxin by cells was partially reversible under the treatment by water and 1 M HCl. The results show that OTA is adsorbed to the surface structures of the cell wall, which is promoted not only by the hydrophobic properties of the cell wall, but also by electron donor-acceptor and Lewis acid-base interactions.  相似文献

Introduction

The plasminogen activator inhibitor-1 (PAI-1) 4G/5G polymorphism was considered to be associated with risk of venous thromboembolism (VTE), while evidence remains inadequate. To provide a more accurate estimation of this relationship, we performed an updated meta-analysis of all eligible studies.

Materials and Methods

A systematical search was performed in PubMed, EMBASE, Wanfang, China National Knowledge Infrastructure (CNKI) and Cqvip databases to identify relevant studies published before March 6th 2014. The odds ratios (ORs) with 95% confidence intervals (CIs) were pooled using the fixed/random-effects model using Review Manager 5.1 and STATA 12.0.

Results

A total of 34 studies with 3561 cases and 5693 controls were analyzed. Overall, significant association between the PAI-1 4G/5G variant and VTE risk in total population (dominant model: OR = 1.32, 95%CI: 1.13-1.54) was observed. And this variant was also related to the deep vein thrombosis risk (dominant model: OR = 1.60, 95%CI: 1.24-2.06, P = 0.0003). In the subgroup analyses on ethnicity, significant results were obtained in both Asians (dominant model: OR = 2.08, 95%CI: 1.29-3.35, P = 0.003) and Caucasians (dominant model: OR = 1.31, 95%CI: 1.10-1.56, P = 0.003). However, no significant association was found in patients with provoked VTE. In terms of subgroup analyses on co-existence of other thrombotic risk factors, the PAI-1 4G/5G polymorphism was significantly associated with VTE risk in patients with factor V Leiden mutation (dominant model: OR = 1.72, 95%CI: 1.17-2.53), but not in patients with cancer or surgery.

Conclusion

Our findings demonstrate the role of PAI-1 4G/5G polymorphism being a risk candidate locus for VTE susceptibility, especially in patients with other genetic thrombophilic disorders.  相似文献

目的:观察脊柱平衡手法推拿治疗椎动脉型颈椎病的疗效。方法:选该病患者240例,随机分为两组。治疗组以脊柱平衡手法为主推拿治疗,对照组以颈部推拿为主治疗。两组7天为1疗程,治疗两个疗程。结果:治疗组痊愈66例,好转51例,无效3例,总有效率为97.50%(95%CI=92.38%~99.45%);对照组痊愈42例,好转43例,无效35例,总有效率为70.83%(95%CI=62.70%~78.94%);两组综合疗效比较(u=4.226 2,P=0.000 0),差异有显著性意义。结论:脊柱平衡手法推拿治疗椎动脉型颈椎病疗效优于颈部推拿治疗,其收益为OR=0.06(95%CI=0.02~0.21),NNT=4(95%CI=2.78~5.74)。  相似文献

AIM:To compare conventional slow equilibrium cooling and directional freezing (DF) by gauze package for cryopreservation of human umbilical vein endothelial cells (HUVECs).METHODS: HUVECs were randomly assigned to conventional freezing (CF) and DF by gauze package group. The two groups of HUVECs were incubated with a freezing liquid consisting of 10% dimethylsulfoxide (DMSO), 60% fetal bovine serum(FBS) and 30% Dulbecco’s modified Eagle’s medium(DMEM) and then put into cryopreserved tubes. CF group, slow equilibrium cooling was performed with the following program:precool in 4℃ for 30min, -20℃ for 1h, and then immersion in -80℃ refrigerator. DF group, the tubes were packaged with gauze and then directional freezing in -80℃ refrigerator straightly. One month later, the vitality of HUVECs were calculated between two groups.RESULTS: There was no significant difference in the survival rate and growth curve between CF and DF groups. The DF group was significantly better than CF group in adherent rates, morphological changes and proliferative ability.CONCLUSION:In the conventional cryopreserved method, cells are slow equilibrium cooling by steps (4℃, -20℃ and finally -80℃), which is a complicated and time-consuming process. But the improved DF by gauze package method is better than conventional method, for which is convenient and easy to operate.  相似文献

目的比较静电吸附法和生物素-亲和素桥联法制备前列腺特异性膜抗原(PSMA)抗体靶向纳米微泡的物理特性和靶向黏附率。方法分别应用静电吸附法和生物素一亲和素桥联法制备PSMA抗体靶向的纳米微泡,检测其物理性状;光镜下观察两种微泡与前列腺癌细胞LNCaP和c4—2的靶向能力,并与胃癌细胞MKN45对照;对比分析两种靶向纳米微泡的粒径、浓度及与前列腺癌细胞结合能力。结果两种靶向纳米微泡均呈圆形,表面光滑,大小均匀,分散度好,无聚集,粒径均〈1000nm。静电吸附法制备的靶向纳米微泡平均粒径为(702.96±66.65)nm,平均浓度为(3.46±0.30)×10^8/ml,与前列腺癌细胞LNCaP黏附率(77.2±3.19)%,与C4-2细胞黏附率(61.00±4.47)%;生物素-亲和素桥联法制备的靶向纳米微泡平均粒径为(609.90±37.40)nm,平均浓度约(4.52±0.19)×10^8/ml,与前列腺癌细胞LNCaP黏附率(96.60±1.14)%,与C4-2细胞黏附率(94.00±1.58)%。两种靶向纳米微泡的粒径、浓度及与前列腺癌细胞黏附率比较,差异均有统计学意义(均P〈0.05);与胃癌细胞MKN45均无黏附。结论生物素-亲和素桥联法是一种高效、灵敏、稳定、特异性强的PSMA抗体靶向微泡制备方法,较静电吸附法具有更大的优势。  相似文献

Purpose. To assess postural control in individuals with Down syndrome.

Method. Sixty young adults with Down syndrome were assessed and compared to 10 non-handicapped young. The subjects were asked to stay on a force platform for 30 seconds. Postural control was evaluated in two conditions: open and closed eyes. The kinetic data carried out from the force platform (i.e., ground reaction forces and Center of Pression (COP) displacements) were both evaluated in time domain and in frequency domain.

Results. Patients with Down syndrome are characterized by instable postural control. In particular the data evaluation in frequency domain underlined for Down syndrome subjects versus control group an increase in frequency oscillation both in anterior-posterior and in medio-lateral direction, that are confirmed in time domain analysis only for medio-lateral direction. In DS no changes are evident between eyes open and eyes closed condition.

Conclusion. This study finds that subjects with Down syndrome included in this research demonstrate that deficits in postural control system that may provide a partial explanation for function balance problems that are common in these subjects.  相似文献

依据浸提效果、吸附率和洗脱率确定最佳工艺参数。结果:最佳工艺条件体积分数为40
%乙醇浸提,采用FL-3大孔树脂,以pH值为 4、20%乙醇条件下静态吸附,再以pH 值为7、60%乙醇溶液
洗脱。按此工艺制备的样品中黄酮的含量为70.6%,收率为70.2%。结论:乙醇浸提结合FL-3大
孔树脂静态吸附方法可分离纯化啤酒花黄酮;该工艺操作简便,样品处理量大,适合规模
化生产。

目的 筛选红景天苷的大孔树脂纯化工艺。方法 以红景天苷质量浓度为指标,考察多种型号大孔吸附树脂纯化红景天苷的吸附及洗脱条件。结果 SP-825型大孔吸附树脂为分离纯化红景天苷的最佳大孔吸附树脂,最佳工艺:上样红景天苷质量浓度为1.713~2.570 g/mL,洗脱剂为10%乙醇,洗脱剂用量为30 BV,体积流量为2 BV/h。结论 SP-825型大孔吸附树脂能显著提高样品液红景天苷的纯度,具有吸附量大、洗脱率高、经济环保等优点,适合于规模化生产。  相似文献
