目的 建立中药配方颗粒辅料麦芽糊精的检测方法,为中药配方颗粒质量评价提供分析技术支持。方法 建立配方颗粒中果糖、葡萄糖、蔗糖、麦芽糖、乳糖的HPLC定量检测方法,根据样品经糖化酶水解后的葡萄糖增加量换算出麦芽糊精含量。基于中药浸膏与麦芽糊精不同比例混合物的中红外光谱(mid-infrared spectroscopy,MIRS)特征峰差异,建立配方颗粒中麦芽糊精的半定量检测方法。结果 所建立的HPLC方法可以准确定量检测配方颗粒中果糖、葡萄糖、蔗糖、麦芽糖、乳糖。如果中药浸膏自身含有较多可经糖化酶水解产生葡萄糖的成分(淀粉、蔗糖等),HPLC检测配方颗粒中麦芽糊精时存在系统性正误差,可通过中药浸膏酶解后葡萄糖增加量、待测样品酶解后蔗糖减小量等部分修正正误差。如果中药浸膏自身少含或不含淀粉、蔗糖等可经糖化酶水解产生葡萄糖的成分,HPLC检测配方颗粒中麦芽糊精时存在系统性负误差,换算公式偏差、麦芽糊精纯度、样品处理损失等导致根据样品酶解后葡萄糖增加量计算的麦芽糊精“检测含量”预期低于根据生产投料计算的麦芽糊精“名义含量”。麦芽糊精含量越高,配方颗粒MIRS中1200~900 cm-1区域的...  相似文献

探究生脉方(Shengmai formula,SMF)对脓毒症小鼠组织损伤、血清炎症因子和外周血固有免疫细胞比例的作用;考察肠道菌群在SMF治疗脓毒症中的作用。灌胃0.3,0.6,1.2 g/kg或腹腔注射0.6 g/kg SMF 4 d后腹腔注射20 mg/kg脂多糖(LPS)建立脓毒症模型,考察小鼠生存率,通过H&E染色观察小鼠肝、肺、肾组织病理改变,检测血清IL-6、TNF-α、谷丙转氨酶(ALT)、谷草转氨酶(AST)、尿素氮(BUN)和肌酐(Cr)水平;建立LPS和盲肠结扎穿刺(CLP)脓毒症模型,通过流式细胞术检测SMF灌胃或腹腔注射对外周血单核细胞、巨噬细胞和中性粒细胞比例的影响;考察抗生素(ABX)处理对SMF灌胃治疗脓毒症的影响;考察SMF灌胃小鼠的粪菌对脓毒症的治疗作用。结果表明,SMF灌胃显著提高LPS模型小鼠生存率,减轻肝、肺、肾损伤和炎性浸润,降低血清IL-6、ALT、AST、BUN、Cr水平;显著降低LPS模型24 h外周血巨噬细胞比例,下调CLP模型24 h外周血单核、巨噬细胞和中性粒细胞比例。SMF腹腔注射对脓毒症模型上述指标均无显著作用。ABX...  相似文献

目的:观察参芍颗粒对结扎犬冠状动脉前降支造成血瘀症模型的血液流变学的影响。方法:采用结扎犬冠状动脉前降支造成血瘀症模型,于造模前、造模后15,30,60,90min分别股静脉采血测定血液流变学指标、体外血栓长度、血小板粘附率和全血粘度(切变率为5-200/s)。结果:参芍颗粒可缩短血栓长度、降低全血粘度、血小板粘附度,抑帛血栓形成。结论:参芍颗粒有改善实验性犬血瘀症的血液流变学指标的作用。  相似文献

用日本第一化学药品株式会社LDL—C试剂盒在BeckmanCX4型自动生化分析仪上直接测定血清LDL—C含量。方法批内CV 0 37%~ 1 11% ,批间CV为 2 11%~ 3 0 2 % (n =2 0 )。平均回收率为 98 9% ,在 8 10mmol/L浓度范围内呈良好线性关系。TG <5 0 0mmol/L时与聚乙烯硫酸沉淀法 (X1)及Friedewald(X2 )公式计算结果相关良好 ,回归方程分别为Y =0 988X1+0 0 47,r=0 95 1,Y =0 96 1X2 +0 0 5 2 ,r=0 930。LDL—C直接测定法简单、快速 ,用血量少 ,勿需沉淀分离 ,适宜于自动化分析。  相似文献

In 129 term infants at birth and at the age of 4 months, zinc and copper concentrations of plasma and urine were determined by graphite furnace atomic absorption spectrophotometry and the values correlated to other biochemical parameters and somatic data. Of the infants, 49 were exclusively breast-fed, 44 fed with various commercially available cow's milk formula, 35 fed with a hypollergenic formula (cows's milk whey hydrolysate, commercially available, supplemented with zinc and copper). Plasma zinc values declined from birth to the age of 4 months in all three groups (P<0.001). In formula fed children, 4 months old, the values (11.1±1.7 mol Zn/l) were significantly lower than in breast-fed (12.2±1.7 mol Zn/l;P=0.004) or babies on hypo-allergenic formula (12.4±1.6 mol Zn/l;P=0.0015). In accordance with the literature plasma copper and caeruloplasmin values increased significantly within the first 4 months of life, the plasma levels were similar in either feeding group, only urinary copper excretion was higher in male infants on hypoallergenic formula (P<0.03) at the age of 4 months. There were no correlations between zinc or copper values and alkaline phosphatase. In infants on hypo-allergenic formula there was a negative correlation between plasma zinc and weight or height increments. Despite different zinc and copper supply, presumedly different bioavailability, and different plasma zinc values, all infants thrived and weight and length increments were similar in each group.  相似文献

Summary Partially purified chromaffin granules (granular fraction), crude mitochondria (mitochondrial fraction) and a microsomal fraction were prepared from bovine adrenals by differential centrifugation and characterized by their catecholamine content, succinate dehydrogenase and glucose-6-phosphatase activity. During isotonic incubation with 0.1 mM 45Ca2+ all fractions showed an uptake of 45Ca2+, which was stimulated by ATP. In addition, after incubation the granular fraction was further fractionated by sucrose density gradient centrifugation (2.0–1.3 M sucrose; 170,000g·60 min). In some of these experiments the granular fraction was incubated simultaneously with 45Ca2+ and [3H](-)noradrenaline to compare the uptake of both. The rate of uptake of 45Ca2+ into the 2.0 M sucrose fraction (characterized by the highest catecholamine content and the lowest succinate dehydrogenase activity) was doubled by ATP. The ATP-stimulated uptake of 45Ca2+ into the 2.0 M sucrose fraction of chromaffin granules was inhibited by N-ethylmaleimide (NEM) (0.1 mM), 2,4-dinitrophenol (DNP) (0.1 mM), azide (1 mM), carbonyl cyanide mchlorophenylhydrazone (CCCP) (20M), atractyloside (50 M), ruthenium red (40 M) and amobarbital (1 mM). This inhibition pattern was different from that of the ATP-stimulated uptake of 45Ca2+ into the microsomal fraction, but it was similar to that of the ATP-stimulated uptake of 45Ca2+ into the mitochondrial fraction. However, the following differences are incompatible with the view that a mitochondrial contamination, with a highly active uptake, is responsible for the ATP-stimulated uptake of 45Ca2+ into the 2.0 M sucrose fraction of chromaffin granules: a) The uptake of 45Ca2+ into the mitochondrial fraction was insensitive to 1 mM amobarbital, whereas this agent inhibited the uptake of 45Ca2+ into the 2.0 M sucrose fraction of chromaffin granules. b) Replacement of ATP by succinate stimulated the uptake of 45Ca2+ into the mitochondrial fraction only. c) The dependence of the ATP-stimulated uptake of 45Ca2+ on the concentration of ATP (0.1–5 mM) was determined: while uptake into the 2.0 M sucrose fraction of chromaffin granules exhibited saturation kinetics, that into the mitochondrial fraction was linearly related to the concentration of ATP. Interestingly, uptake of 45Ca2+ into those fractions of chromaffin granules that are known to be contaminated with mitochondria (1.6 M sucrose fraction) exhibited bot a saturable and a nonsaturable component. d) The uptake of 45Ca2+ into the mitochondrial fraction was more sensitive to 0.1 mM DNP than the uptake of 45Ca2+ into the 2.0 M sucrose fraction of chromaffin granules.The comparison of the ATP-stimulated uptake of 45Ca2+ with that of [3H](-)noradrenaline into the 2.0 M sucrose fraction of chromaffin granules revealed that amobarbital, N,N-dicyclohexylcarbodiimide (DCCD), DNP and increasing concentrations of ATP had the same influence on both uptake processes. From our results we conclude that an inherent, ATP-stimulated uptake of 45Ca2+ exists in chromaffin granules. A 61-year old man presented with a five-year history of a swelling initially developing in his right lower lid that progressed to involve the lateral canthal skin and eventually the upper lid and anterior orbit. He was discovered to have an infiltrating, poorly differentiated, mucin-producing carcinoma. Systemic work-up failed to disclose a visceral malignancy, and it was concluded that his tumor was primary in the lids, arising from an adnexal sweat gland. Three other reports in the literature also share almost identical clinical and pathologic features, in that all of the earlier reports dealt with middle-aged or elderly men who had diffusely indurated lids. Histopathologically, the tumor cells grow diffusely in a sclerotic stroma, and resemble the "histiocytoid" variant of metastatic breast carcinoma to the lids in women. Ultrastructural studies in our case point toward an apocrine origin, although earlier authors have favored an eccrine origin. Despite its indolent clinical course, the tumor is capable of producing regional and distant metastases on long-term follow-up. Complete local excision, possibly necessitating radical surgery, is probably the preferred method of treatment, but local radiotherapy may have a beneficial effect in retarding spread of the disease.  相似文献

The Merkel cell is a distinctive nondendritic, nonkeratinocytic, epithelial clear cell believed to migrate from the neural crest to the epidermis and dermis, which is usually located in or near the basal layer of the epidermis and associated with nerve terminations. Merkel first described these cells in 1875 as "Tastzellen" occurring in the snout of a mole. They are believed to function as slowly adapting mechanoreceptors that mediate the sense of touch. Tumors arising from Merkel cells have been reported to occur on the head and neck area, the trunk, arms, and legs, and resemble a primary cutaneous lymphoma or cutaneous metastasis of a lymphoma or a carcinoma. Electron microscopy, to locate the characteristic membrane-bound, dense core neurosecretory granules, is needed for accurate diagnosis. These tumors must be treated aggressively to minimize the chance of local recurrence and nodal or visceral metastases. The authors present a case of Merkel cell tumor occurring on the eyelid. The clinical history, light and electron microscopic findings are shown.  相似文献
