We recorded activity from neurones in cortical motion-processing areas, middle temporal area (MT) and middle posterior superior temporal sulcus (MST), of anaesthetised and paralysed macaque monkeys in response to moving sinewave gratings modulated in luminance and chrominance. The activity of MT and MST neurones was highly dependent on luminance contrast. In three of four animals isoluminant chromatic modulations failed to activate MT/MST neurones significantly. At low luminance contrast a systematic dependence on chromaticity was revealed, attributable mostly to residual activity of the magnocellular pathway. Additionally, we found indications for a weak S-cone input, but rod intrusion could also have made a contribution. In contrast to the activity of MT and MST neurones, speed judgments and onset amplitude of evoked optokinetic eye movements in human subjects confronted with equivalent visual stimuli were largely independent of luminance modulation. Motion of every grating (including isoluminant) was readily visible for all but one observer. Similarity with the activity of MT/MST cells was found only for motion-nulling equivalent luminance contrast judgments at isoluminance. Our results suggest that areas MT and MST may not be involved in the processing of chromatic motion, but effects of central anaesthesia and/or the existence of intra- and inter-species differences must also be considered.  相似文献

目的探讨汉语阅读障碍(read disorder,RD)儿童视觉编码和词语成分加工的脑功能定位及血氧水平依赖(blood oxygenation level-dependent, BOLD)响应与加工作业之间的关系.方法以19名RD儿童为病例组,以28名正常儿童为对照组,2组儿童年龄分别为(9.81±1.22)岁和(9.77±1.19)岁.运用功能近红外成像技术,观察760 nm和850 nm的漫射光强变化,测定受试者左前额皮层的血流量和血氧的变化.结果 2组儿童左前额叶的BOLD响应强度在词语成分加工时均显著强于视觉编码加工.视觉编码加工时,RD儿童左前额叶的血流量增加,对照组的血流量减少;词语成分加工时,RD儿童和正常儿童左前额叶的血流量均呈现增加趋势,但RD儿童脑区域激活强度和激活范围较对照组明显减弱(P<0.05).结论左前额叶参与RD儿童视觉编码和词语成分加工作业.汉语RD儿童在完成上述2项加工作业时的脑局部血流量的代谢机制和脑功能机制与正常儿童存在明显差异,提示汉语RD儿童神经生理机制的异常可能是其发病的大脑物质基础.  相似文献

Photoreceptors in the vertebrate retina are electrically coupled with one another. Such coupling plays important roles in visual information processing. Physiological properties of rod-rod and rod-cone coupling have been best studied in the salamander retina, yet the cellular and molecular basis of these electrical synapses has not been established. Recently, connexin35/36 (Cx35/36) gap junction proteins were found to be highly expressed in brain and retina, suggesting that it may mediate photoreceptor coupling. To test this idea, we examined the cellular distribution of Cx35/36 in the salamander retina. Western blot analysis showed the expression of Cx35/36 proteins, and confocal microscopy revealed characteristic punctate Cx35/36 immunoreactivity in both synaptic layers. In addition, Cx35/36-positive plaques were detected in the outer nuclear layer (ONL) between neighboring rods, and these plaques outlined the mosaic of the rod network at a level distal to the external limiting membrane. Moreover, although Cx35/36 plaques were detected between some cones and their adjacent rods, the number and size of these plaques was smaller, and their staining intensity was diminished compared with the plaques between adjacent rods. Furthermore, Lucifer yellow injection together with confocal microscopy revealed that Cx35/36-puncta were colocalized with finlike structures of rod cell membrane, with the ultrastructure of gap junctions between paired rod fins having been found by electron microscopy. Therefore, our findings demonstrate that Cx35/36 expression in photoreceptors is primarily located between rods and to a lesser extent between rods and cones, suggesting that Cx35/36 may participate in electrical coupling between rods and between rods and cones in the salamander retina.  相似文献

Genetic and protein studies have indicated abnormalities in alpha-synuclein in neurodegenerative diseases. However, the developmental localization and cellular role of synuclein isoforms is contentious. We investigated the cellular localization of alpha-, beta-, and gamma-synuclein in developing cultured rat neurons and following axonal transection of relatively mature neurons, a model that disrupts the axonal cytoskeleton and results in regenerative sprouting. Cortical neurons were grown up to 21 days in vitro (DIV). Axon bundles at 21 DIV were transected and cellular changes examined at 4 and 24 h post-injury. Immunohistochemistry demonstrated that alpha- and beta-synuclein were localized to cellular cytosol and growth cones at 3DIV, with accumulating puncta-like labeling within axons and growth cones by 10-21DIV. In contrast, gamma-synuclein immunoreactivity was limited at all time points. By 21DIV, alpha- and beta-synuclein were present in the same neurons but largely in separate subregions, only 26% of puncta contained both alpha- and beta-synuclein immunoreactivity. Less than 20% of alpha-, beta-, and pan-synuclein immunoreactive puncta directly colocalized to synaptophysin profiles at 10DIV, decreasing to 10% at 21DIV. Both alpha- and beta-synuclein accumulated substantially within damaged axons at 21DIV and were localized to cytoskeletal abnormalities. At latter time points post-injury, alpha- and beta-synuclein immunoreactive puncta were localized to growth cone-like structures in regenerating neurites. This study shows that alpha- and beta-synuclein have a precise localization within cortical neurons and are generally nonoverlapping in their distribution within individual neurons. In addition, synuclein proteins accumulate rapidly in damaged axons and may have a role in regenerative sprouting.  相似文献

In an attempt to clarify its possible physiological role, we studied the eye of the Zambian mole rat Cryptomys anselli by light, electron and confocal microscopy using conventional staining as well as immunolabelling with rod and cone cell markers. The small eyes of Cryptomys are located superficially and display all features typical of sighted animals: iris, pupil and well-developed lens, separating the anterior chamber and the vitreous. The retina shows a well stratified organization and the folds described in blind subterranean or nocturnal mammals were not observed. The major population of the photoreceptor cells in the Cryptomys retina consists of rod cells, again with a morphology quite similar to that found in sighted animals. The relatively short outer segments contain numerous well-stacked disks and show a strong rod-opsin as well as transducin immunoreaction. Synapses were evident in the spherules, the round basal processes of the rod cell, but they lacked the precise organization reported for sighted mammals. Cone cells were present as well, as indicated by peanut lectin staining, but no immunolabelling with polyclonal M/L-opsin antisera was detectable. The presence of cone cells was also suggested by some basal processes at the outer plexiform layer which displayed several synaptic active sites and irregular contours. While the other retinal layers also showed an organization typical of sighted mammals, there were signs of less tightly preserved morphology as well. Displaced rods and amacrine and/or ganglion cells were observed, and some sparse rod spherules penetrated into the inner nuclear layer. A major reduction was observed in the number of ganglion cells, estimated from the number of axons in the optic nerve, that was very low (approximately 1000 per retina on average) relative to sighted mammals. The data we have suggest a slow, ongoing loss of cells with ageing. Apoptotic nuclei, mainly corresponding to photoreceptor cells and ganglion cells, were detected in young individuals, and an overall reduction in the thickness of the retina was observed in older animals. The morphological data presented here allow some first speculations on the physiological role of the Cryptomys eye and will hopefully trigger detailed studies on the chronobiology and the anatomy of the retinal projections and of the visual cortex of this remarkable species.  相似文献

The kinetics of rod outer segment production and turnover were studied in Irish setter dogs with a recessively inherited photoreceptor degeneration (rod-cone dysplasia). Affected and littermate control (heterozygote) dogs, 16–50 days of age, were injected intravitreally with [3H]leucine after electroretinographic studies. Normal age-matched mongrel dogs were also used as controls. Between 6 hr and 8 days following intravitreal injection, eyes were removed and processed for microscopic and radioautographic studies.In affected dogs, the rod components of the electroretinogram were not recordable. Cone components were recordable but failed to develop normally. Morphologically, rod inner segments were diminutive and the outer segments short and contained disorganized and disoriented disc material. Cone inner segments became broad and club-shaped. Radio-autography showed label accumulation within inner segments, but only sporadic and randomly distributed label in the narrow outer segment layer. These results suggest that in Irish setters with rod-cone dysplasia, there is a defect in the morphogenesis of the rod outer segments during postnatal retinal differentiation.  相似文献
