目的 筛选最佳的以黄柏为君药的复方溃疡乐的提取工艺。方法 以小檗碱、靛玉红为工艺筛选指标性成分 ,采用正交设计方法筛选最佳提取工艺及其条件。结果 最佳提取工艺为 80 %乙醇回流 4次 ,每次均为 4倍量 ,第一次回流 1.5 h,第二次回流 1h,第三次和第四次回流 0 .5 h,小檗碱、靛玉红转移率分别为81.6 %、87.0 %。结论 该提取工艺小檗碱、靛玉红转移率高并经验证 ,合理可行。  相似文献

The vasculature of diarthroidal joints has been well documented; however, the volume of vessels supplying different articular tissues is unknown. Angiogenesis, the formation of new vessels from preexisting ones, is difficult to quantify in joints due to the unavailability of a suitable technique. Although angiogenesis is known to occur in rheumatoid arthritis, the development of new vessels following joint injury has not been ascertained. A vascular casting technique was developed using carmine red dye to measure the vascular volume of the medial collateral ligament (MCL), lateral collateral ligament (LCL), menisci, medial capsule, and infrapatellar fat pad of the rabbit knee joint. Vascular volume determinations were repeated at 4 weeks in a group of anterior cruciate ligament (ACL)-transected animals and in a sham-operated control group. The volume of vessels supplying the MCL was estimated to be 0.22 ± 0.07 μl (mean ± S.E.M.), the LCL volume was 0.25 ± 0.05 μl, the medial meniscus volume was 0.19 ± 0.03 μl, the lateral meniscus volume was 0.40 ± 0.08 μl, the medial capsule volume was 0.14 ± 0.05 μl, and the infrapatellar fat pad volume was 1.90 ± 0.62 μl. Following ACL transection, angiogenesis was found to occur in the MCL only. All other tissue vascularities were not significantly different from sham-operated controls. A quantifiable method for measuring vascular volume of knee joint tissues has been described. Joint instability stimulates angiogenesis in the ipsilateral MCL; however, the absence of angiogenic activity in other articular tissues might help explain the lack of posttraumatic healing associated with these joints. 目的体外观察复方青黛饮大鼠含药血清对人永生化角质形成细胞株HaCaT细胞增殖、凋亡和细胞周期的影响,并探讨其作用机制。方法根据中药血清药理学方法,用SD大鼠制备实验血清,采用MTT法、流式细胞术、免疫荧光Hoechst染色,观察含药血清对细胞生长抑制率、凋亡率、细胞周期分布及细胞形态学的影响。结果体外培养HaCaT细胞24h和48h后,复方青黛饮高、中、低三个剂量不同浓度的含药血清组,对HaCaT细胞的生长均有不同程度的抑制,与空白血清组相比,差异有统计学意义(P<0.05),其中高剂量组抑制作用最明显。复方青黛饮含药血清组细胞凋亡率和G0/G1比例明显增加,与空白血清组相比,差异有统计学意义(P<0.05),在镜下呈现凋亡的特征性形态改变。结论复方青黛饮大鼠含药血清对体外培养的角质形成细胞株HaCaT细胞有生长抑制和诱导凋亡作用,并对细胞周期有一定的影响。  相似文献

目的建立青黛中靛玉红含量的测定方法,为2005年版《中国药典》提供含量测定控制方法。方法采用薄层扫描法对青黛中靛玉红进行含量测定,以苯-氯仿-丙酮(5∶4∶1)为展开剂,λS=540nm,λR=700nm;同时,采用薄层色谱法对青黛中的靛蓝和靛玉红进行定性鉴别。结果靛玉红的回归方程为y=11.915x 1052.4,r=0.9967;平均回收率为99.40%,(n=8),RSD为3.50%。测得7批青黛样品,含量在0.161%~0.225%之间。结论该方法具有简便、迅速、灵敏、重现性好等特点,可用于控制青黛药材的质量。  相似文献

目的:考察不同采收期板蓝根的质量。方法:对1a、2a生板蓝根药材进行性状、显微鉴别,用高效液相色谱法测定药材中腺苷、靛蓝及靛玉红的含量。结果:1a生者韧皮部宽广,薄壁细胞排列疏松;2a生者韧皮部窄,薄壁细胞排列紧密。采挖后未及时晒干或贮藏不善易致其断面变色,靛蓝含量增高,腺苷、靛玉红含量显著降低;生长期过短及2a生板蓝根中的腺苷、靛蓝及靛玉红含量较低。结论:需进行板蓝根规范化种植,并进行质量监控,避免因产地、种植采收时间、初加工及贮藏方法等不同而引起的质量差异过大。  相似文献

目的:青黛难溶,与甘草配伍以增加其在水中的溶解度,提高药液中青黛有效成分的含量及生物利用度。 方法:将甘草与青黛、甘草酸分别与靛蓝和靛玉红、柠檬酸与青黛进行配伍,均以HPLC法分别测定各实验中靛蓝、靛玉红的含量。 结果:青黛与生、炙甘草配伍,溶液中靛蓝和靛玉红的含量均明显增多,但炙甘草较生甘草的低约30%左右;将甘草酸单铵盐直接与靛蓝、靛玉红分别按比例混合,测得两者含量均显著增多,约增溶4~8倍左右,且2:1为最佳比例;青黛配伍不同比例柠檬酸,溶液中靛蓝和靛玉红含量无明显变化。 结论:甘草配伍青黛到达了增溶的目的;甘草酸起主要作用,是以其表面活性进行增溶;同时为中药药对配伍应用找到了一定的理论依据。  相似文献

A rapid, selective, and sensitive LC-APCI-MS method is developed in this study for detecting and analyzing tryptanthrin, indigo, and indirubin in daqingye and banlangen, which are, respectively, the leaves and roots of Isatis indigotica and Strobilanthes cusia in traditional Chinese medicine. The detection of the three active components is linear in concentrations ranging from 100 to 1500 ng/mL, the squared correlation coefficient is higher than 0.996, the precision as measured by the relative standard deviation is no larger than 9.5%, and the recovery is greater than 86.6%. The analysis of the 21 banlangen samples led to considerably different conclusions on the contents of tryptanthrin, indigo, and indirubin in fresh leaves versus those in dried leaves. These results should shed some light on future plant selection and breeding. Compared with the traditional TLC and HPLC-UV methods, the new LC-APCI-MS approach has proven to be an optimal tool for detecting and analyzing the three marker compounds in the Chinese herbal medicines of daqingye and banlangen.  相似文献

Introduction and importanceAn unicornuate uterus with a non-communicating rudimentary horn (UUNCRH) is a very rare uterine malformation which is difficult to diagnose and to decide the surgical plan. We aim to describe the case of pediatric UUNCRH patient and our operative technique of laparoscopic intra-uterus indigo carmine injection (LIUICI) to confirm that the rudimentary horn (RH) is non-communicating before the resection and review the relevant literature to ascertain the most appropriate treatment option in these patients.Case presentationA 11-year-old girl who developed progressive severe abdominal pain and dysmenorrhea was referred to our hospital. Uterine malformation and right hematosalpinx was confirmed with magnetic resonance imaging (MRI). Pre-operative treatment with a gonadotropin-releasing hormone agonist enabled improvement in the symptoms. Laparoscopic exploration was scheduled. The right fallopian tube was resected laparoscopically and a 3Fr tube was inserted into its cut end. Indigo carmine injected in the RH through the tube. No leakage of indigo carmine was found from the vagina, indicating the diagnosis of the uterine malformation is an UUNCRH and we performed the resection of the RH safely.Clinical discussionIn pediatric patients transvaginal detailed examination is not easy to perform. Therefore, diagnostic and operative laparoscopy is critically important for the safe treatment. In addition, laparoscopic removal of a RH can be used to decrease the incidence of adhesions.ConclusionWe found LIUICI technique before the resection of the RH is safe, technically feasible and minimally invasive approach for pediatric UUNCRH patients.  相似文献

目的研发内镜自动染色水汽装置,对内镜水汽瓶进行改造,使内镜水汽与染色剂整合在一个装置内,通过转换钮控制内镜水汽和染色剂都分别通过内镜水汽管道进入消化道。并与常规染色方法进行对比,以此来验证内镜自动染色水汽装置的临床效果。方法分为自动染色组与常规染色组两组,每组100例,选取靛胭脂及复方碘溶液作为化学染色剂,对比两组染色剂的使用量和操作时间。结果自动染色组碘液使用剂量和操作时间明显少于常规染色组[碘液使用剂量分别为(9.53±1.01)和(10.73±1.82)mL,操作时间分别为(16.10±2.46)和(25.45±2.43)s],两组比较,差异均有统计学意义(P 0.05);自动染色组靛胭脂使用剂量和操作时间明显少于常规染色组[靛胭脂使用剂量分别为(8.75±1.20)和(9.95±1.43)mL,操作时间分别为(8.60±1.35)和(10.45±2.01)s],两组比较,差异均有统计学意义(P 0.05)。结论内镜自动染色水汽装置可以用于内镜下化学染色,使内镜下染色既高效又便捷。  相似文献
