Binding of sulfaethidole to bovine serum albumin was studied by equilibrium dialysis, fluorescence probe technique, uv difference spectrophotometry and circular dichroism. Equilibrium dialysis method enabled us to estimate the total number of drug binding sites of albumin molecule. For sulfaethidole, albumin had 6 primary and 40 secondary binding sites. The primary and secondary binding constants were 0.9×105 M−1 and 0.2×106 M−1, respectively. 1-Anilino-8-naphthalenesulfonate (ANS) and 2-(4′-hydroxylbenzeneazo)-benzoic acid (HBAB) were used as the fluorescence probe and the uv spectrophotometric probe, respectively. In fluorescence probe technique, results indicated that the number of higher affinity drug binding site of albumin was 1 and the number of lower affinity drug binding sites of albumin was 3, and the primary and secondary drug binding constants for bovine serum albumin were 2.15×105 M−1 and 1.04×105 M−1, respectively. In uv difference spectrophotometry, binding sites were 3 and binding constant was 1.88×105 M−1. The above results suggest that several different methods should be used in ompensation for insufficient information about drug binding to albumin molecule given by only one method.  相似文献

本实验用荧光组化技术及记录显微自动曝光时间方法研究了马桑内酯所致的Wistar大鼠癫痫持续状态黑质-腹侧被盖区多巴胺含量的变化。结果显示癫痫发作高峰组(10只)和发作后组(8只)该区多巴胺含量明显低于对照组(10只)和发作前组(10只)。由于多巴胺是一种抑制性神经递质,它的减少可能表明抑制作用的下降对癫痫的发作有一定的调控作用。  相似文献

Microorganisms are very powerful tools for the supply of information about the toxic effects of lipophilic compounds, since an impairment of cell growth usually occurs as a result of perturbations related, in most cases, with the partition of toxicants in membranes. The thermophilic eubacterium Bacillus stearothermophilus has been used as a model system to identify α- and β-endosulfan interactions with the membrane possibly related with the insecticide toxicity. Two approaches have been pursued: (a) bacterial growth is followed and the effects of endosulfan isomers determined; (b) biophysical studies with the fluorescent fluidity probe 1,6-diphenyl-1,3,5-hexatriene (DPH) were performed to assess the effects of α- and β-endosulfan on the organization of the membrane lipid bilayer. The effects on growth were quantitatively evaluated by determination of growth parameters, namely the lag phase, the specific growth rate and the cell density reached by cultures in the stationary phase. Growth inhibition by α and β-endosulfan dependent on the concentration is diminished or removed by the addition of 2.5 m Ca2+ to bacterial cultures. Fluorescence DPH polarization consistently showed opposite effects of Ca2+ and α- and β-endosulfan on the physical state of bacterial polar lipid dispersions.  相似文献

Objective Pretargeting with radioactivity has significantly improved tumor to normal tissue radioactivity ratios over conventional antibody imaging in both animal studies and clinical trials. This laboratory is investigating DNA analogues such as phosphorodiamidate morpholinos (MORFs) for pretargeting using technetium-99m (99mTc) for detection. However, the unique properties of fluorescence activation and quenching combined with oligomers with their unique properties of hybridization may be particularly useful when used together for pretargeting with optical detection. The use of linear fluorophore-conjugated oligomer duplexes have been little used in animals, and to our knowledge, have not previously been considered for pretargeting applications. Methods A MORF/cDNA pair was selected such that when hybridized, the fluorescence of the Cy5.5-conjugated 25 mer MORF (Cy5.5–MORF25) is inhibited with a BHQ3-conjugated 18 mer complementary DNA (BHQ3–cDNA18). The short BHQ3–cDNA18 was selected to dissociate in the presence of a long cMORF25 in the pretargeted tumor, thus releasing the inhibitor from the Cy5.5 emitter. In this manner, the Cy5.5 fluorescence will be inhibited everywhere but in the target. The dissociation was first examined in vitro by adding the duplex to the cMORF25 both in solution and immobilized on polystyrene microspheres and by surface plasmon resonance (SPR). Thereafter, biotinylated cMORF25 immobilized on streptavidin polystyrene microspheres were administered intramuscularly in one thigh of hairless SKH-1 mice as target while an identical weight of the identical microspheres but without the cMORF25 was administered in the contralateral thigh as control. The animals then received IV the Cy5.5–MORF25/BHQ3–cDNA18 duplex or equal molar dosage of single-chain Cy5.5–MORF25 and were imaged. Results The SPR studies showed that the immobilized cDNA18 rapidly captured the flowing MORF25 to provide a duplex with a slow dissociation rate constant. Furthermore, when cMORF25 was next allowed to flow over the now immobilized duplex, the cDNA18 was unable to prevent dissociation of the heteroduplex and the formation and release of the cMORF25-MORF25 homoduplex. Images of animals obtained soon after receiving the Cy5.5–MORF25 singlet showed intense whole body fluorescence obscuring the target thigh. However, only 5 minutes after receiving the Cy5.5–MORF25/BHQ3–cDNA18 duplex, the target thigh was clearly visible along with only the kidneys. Conclusions This first study of optical pretargeting provides a proof of concept that oligomer pretargeting found to be useful with radioactivity detection is applicable with fluorescent detection as well. In addition, our results demonstrate that by using linear oligomers for optical pretargeting, chain lengths (and base sequences) may be manipulated to provide duplexes with stabilities and fluorescence inhibition optimized for pretargeting and other in vivo applications of optical imaging.  相似文献

目的 确定UF - 10 0型全自动尿液沉渣检测仪检测健康人尿沉渣检测的参考值范围。方法 用UF - 10 0型全自动尿液沉渣检测仪随机检测 5 2 0例健康人不离心尿液标本 ,对尿液沉渣进行分析。结果 尿液中红细胞 (RBC)、白细胞 (WBC)、上皮细胞 (EC)、管型 (CAST)各年龄组之间检测值经 F 检验 ,差异无显著意义 (P >0 .0 5 ) ,但男、女性别之间差异有显著意义 (P <0 .0 5 )。结论 尿液RBC、WBC、EC的多少与年龄无关 ,但与性别有关。健康人参考值的确立 ,为尿液常规检验定量标准化和临床应用提供了依据  相似文献

在酸性介质中对香英兰素与儿茶素的显色反应进行了研究,改进了反应条件,以506nm为测定波长,测定TP口含片中儿茶素的含量,线性范围7.58μg~53.05μg(r=0.9998),回收率97.97%,RSD1.85%,认为改良法简便,灵敏,口含片辅料无干扰,可用于原料的药和制剂的含量分析。  相似文献

Eight commercially available HPD-photosensitizers intended for photodynamic therapy were tested in a murine tumour model with regard to their therapeutic efficacy. The regrowth delay of the fibrosarcoma SSK-2 on the mouse C3H, Neuherberg-line, was determined 3, 24, 48 and 72 h after injection of the drugs (dose: 9 mg kg–1 body weight). The corresponding pharmacodynamics, as measured by regrowth delay, were approximated by an exponential function and the characterizing coefficients derived. These coefficients served to quantify the photodynamic properties of the drugs.The pharmacodynamics of five substances were compared with those obtained fluorometrically. The latter showed shorter decay constants than the therapy-correlated substances which indicates different metabolic behaviour of the therapeutic and diagnostically useful fluorescent components of haematoporphyrin-derived photosensitizers.  相似文献

本文探讨了用DPH作为荧光探剂,研究家蝇飞翔肌线粒体膜脂流动性的方法。实验表明:DPH确是用来研究家蝇生物膜流动性的有效探剂。同时,初步观察了不同杀虫剂对DPH与线粒体膜结合后荧光强度的影响。其中氯菊酯和倍硫磷、马拉硫磷、敌百虫、氧化乐果,可使DPH探剂与线粒体膜结合后的荧光相对强度增加;而氰戊菊酯、氯氰菊酯,溴氰菊酯及辛硫磷、乙酰甲胺磷,则可使荧光强度减弱。  相似文献
