我们建立的K562/ADM耐药细胞株,在ADM浓度为2.4μg/m1(4.46μM)中已稳定培养3.5个月,传了30—35代,K562/ADM亦具有多药耐受件(Multidrug Resistance,MDR)的特点,对ADM、VCR、AT—1258和DDP的耐受性分别为K562的114.7、94.0、13.3和7.4倍,但对5—FU不产生交叉耐药。K562和K562/ADM的倍增时间分别为19.2h和52.8h,集落生成率分别为37.5%和11.1%,K562染色体数为34—68,中位数为56;K562/MDM染色体数为32—90,中位数为50,K562/ADM可做为耐药机理和克服耐药措施研究的极好模型。  相似文献

Flecainide. Flecainide is a Class IC antiarrhythmic agent whose primary electrophysiologic effect is a slowing of conduction in a wide range of cardiac tissues. It is well absorbed and effective in suppressing isolated premature ventricular contractions (PVCs) or nonsustained ventricular arrhythmia but has only a modest efficacy when electrophysiologic testing is used as an endpoint. Us adverse effect on mortality in the CAST trial suggested a propensity to proarrhythmia-a phenomenon to which the Class IC agents appear particularly prone. Despite the applicability of the CAST study only to patients with a prior myocardial infarction, there has been a shift away from flecainide in ventricular arrhythmia, but the low noncardiac side effect profile of the agent allows for its continued use in a wide variety of supraventricular arrhythmias. We report the findings of spinal magnetic resonance imaging (MRI) in 2 patients who had undergone intrathecal chemotherapy and presented with the subacute onset of ascending numbness and weakness. MRI revealed a symmetric hyperintensity at the posterior columns of the spinal cord from the lower cervical region down to the conus medullaris level on T2-weighted images, and no abnormal enhancement. The imaging findings are similar to those seen in subacute combined degeneration (SCD), but the serum vitamin B(12) levels were normal in these 2 cases.  相似文献

Objective: In order to document the stereoselective distribution in joints of a chiral nonsteroidal anti-inflammatory drug, the relative affinities of the enantiomers of tiaprofenic acid in synovium and for cartilage were compared. Methods: The distribution of tiaprofenic acid in synovium and in cartilage was studied 25 h after administering the racemic drug for 2 days (600 mg of a sustained-release preparation, once daily), in 12 inpatients with osteoarthritis of the hip requiring arthroplasty. Enantiomers were quantified in plasma and freeze-ground tissues by a chiral HPLC assay. Results: Plasma concentrations of the dextrorotatory (R) enantiomer (0.40 μg/ml) were higher than those of its antipode. The concentration of racemate in synovium (in dried and fresh tissues, 150% and 40%, respectively, of the concentration in plasma) was much higher than that in cartilage (in dried tissues 32% of the plasma concentration). The ratio of the active, dextrorotatory (R) enantiomer to its antipode was higher in synovial tissue than in plasma. Conclusion: Tiaprofenic acid is distributed stereoselectively in plasma and synovium, which contain a higher concentration of the active, dextrorotatory (R) enantiomer. Summary An electron microscopic analysis of the synapses in Clarke's column of the cat, using fresh short term degeneration, long term degeneration for detection of the persisting elements, and a newly devised two-step degeneration technique using fresh short term degeneration for further analysis of the persisting synaptic elements in chronic degeneration cases. — Three kinds of synaptic terminals can be identified in the lower part (L3 segmental level) of Clarke's column: (1) very large (so-called giant) axon terminals with spheric vesicles of primary muscle afferents establishing multiple (climbing-type) contacts mainly with the large dorsal spinocerebellar tract neurons; (2) small bouton-type terminals having spheric synaptic vesicles and originating mainly from spinal interneurons, localized below L4 in the lumbar enlargement and contacting small distal dendrites and cell bodies of small cells; (3) two types of nerve terminals, originating from local neurons, and characterized by flattened vesicles. One type of (3) contacts mainly the large Clarke neurons directly, whereas the other establishes axo-axonic synapses with the giant terminals (1) of primary muscle affer ents. The possible functional significance of these findings is discussed.  相似文献

Class I molecules of the major histocompatibility complex bind peptides derived from cytosolic proteins and display them on the cell surface. This function alerts cytotoxic T cells to the presence of intracellular pathogens. Class I molecule assembly requires the association of the heavy chain with β2-microglobulin, accompanied by peptide loading via specific transporters. This study localizes where these assembly steps take place, using monoclonal antibodies recognizing class I molecules in different assembly states to analyze subcellular fractions of the early secretory pathway. The distribution of peptide-loaded class I molecules was more localized than the distribution of the total pool of class I molecules in the early secretory pathway. Loaded molecules colocalized with the peptide transporter, free heavy chains, and the chaperone calnexin in high density rough endoplasmic reticulum (RER) membranes. These data suggest that subunit assembly and peptide acquisition occur at the same intracellular site. Class I molecules also localized to less dense subfractions of the early secretory pathway, which contained comparatively less peptide-loaded molecules than the high density RER fractions, at steady state. Following a 15 °C temperature block, class I molecules accumulated in these less dense membrane fractions, indicating that these fractions represent the intermediate compartment where empty class I molecules are trapped in mutant cells. In the presence of cycloheximide, a pool of class I molecules recycling to the RER was detected, suggesting empty molecules recycle to acquire peptide.  相似文献

The classification of some of the extractable birch pollen antigens as allergens was established by crossed radioimmunoelectrophoresis (CRIE). In CRIE the major allergen (antigen 23) exhibited the strongest “radiostaining” and only a few other components of birch pollen extract were visibly radiostained. The major allergen and a preparation containing mainly the minor allergens, antigens 25 and 19, were isolated from a crude aqueous birch pollen extract by a combination of anion-exchange, size-exclusion, and chelate chromatography. Antigen 23 was purified to near homogeneity. The molecular weights and the pIs of antigens 23, 25, and 19 were determined to be 17,000 daltons, pI 5.25 (5.5, 5.0); 25,000 daltons, pI 5.0 (4.9, 5.4); and 29,000 daltons, pI 6.2 (5.4), respectively. The classification of antigen 23 as the major allergen in birch pollen was supported by results of RAST inhibition experiments, RAST screening, and skin prick testing.  相似文献

本文应用荧光素双标记法研究了后索核、孤束核、三叉神经脊束核向下丘和丘脑投射神经元的分布及其分支投射。作者将Propidium Iodide注入大白鼠右侧丘脑,Bisbenzimide注入右侧下丘,结果如下:楔束核内被标记的神经元中有88.2%向丘脑投射,它们位于核的主部;11.2%向下丘投射,它们在尾侧主要位于核的边缘部,向吻侧(至闩平面)主要位于核的背外侧端。薄束核内被标记的神经元中向下丘投射者(10.9%)与向丘脑投射者(86.7%)混杂在一起。在此两核内仅有极少量分支投射的双标神经元。孤束核和三叉神经脊束核内被标记的神经元中约有2/3投向丘脑,1/3投向下丘,未见分支投射的双标神经元。  相似文献
