Screening microarrays of novel monoclonal antibodies for binding to T-, B- and myeloid leukaemia cells 总被引:3,自引:0,他引:3

We have developed a microarray (DotScan) that enables rapid immunophenotyping and classification of leukaemias and lymphomas by measuring the capture of cells by immobilized dots of 82 CD antibodies [Belov, L., de la Vega, O., dos Remedios, C.G., Mulligan, S.P., 2001. Immunophenotyping of leukemia using a cluster of differentiation antibody microarray. Cancer Res. 61, 4483; Belov, L., Huang, P., Barber, N., Mulligan, S.P., Christopherson, R.I., 2003. Identification of repertoires of surface antigens on leukemias using an antibody microarray. Proteomics 3, 2147]. The DotScan technology has been used to investigate the properties of 498 new antibodies submitted to the HLDA8 Workshop. These antibodies have been applied as 10 nl dots to a film of nitrocellulose on a microscope slide to make an HLDA8 microarray. After blocking the remaining nitrocellulose surface, individual arrays were incubated with each of 7 cell types from a human leukaemia cell panel consisting of three cell lines, CCRF-CEM (a T-cell acute lymphocytic leukaemia), MEC-1 (derived from B-cell chronic lymphocytic leukaemia) and HL-60 (a promyelocytic leukaemia), and four leukaemias from patients: a T-cell prolymphocytic leukaemia, a B-cell chronic lymphocytic leukaemia, and two acute myeloid leukaemias. Leukaemia cells were captured by those immobilized antibodies for which they expressed the corresponding surface molecule. Unbound cells were gently washed off, bound cells were fixed to the arrays and dot patterns were recorded using a DotScan array reader and quantified using DotScan data analysis software. The data obtained show the unique expression profiles of the 7 cell types in the leukaemia cell panel obtained with the DotScan microarray, and the differential capture patterns for these 7 cell types screened against the 498 antibodies in the HLDA8 microarray constructed for this study. 相似文献

Summary Age-related cognitive impairments were studied in rats kept in semi-enriched conditions during their whole life, and tested during ontogeny and adult life in various classical spatial tasks. In addition, the effect of intrahippocampal grafts of fetal septal-diagonal band tissue, rich in cholinergic neurons, was studied in some of these subjects. The rats received bilateral cell suspensions when aged 23–24 months. Starting 4 weeks after grafting, they were trained during 5 weeks in an 8-arm maze made of connected plexiglass tunnels. No age-related impairment was detected during the first eight trials, when the maze shape was that of a classical radial maze in which the rats had already been trained when young. The older rats were impaired when the task was made more difficult by rendering two arms parallel to each other. They developed an important neglect of one of the parallel tunnels resulting in a high amount of errors before completion of the task. In addition, the old rats developed a systematic response pattern of visits to adjacent arms in a sequence, which was not observed in the younger subjects. None of these behaviours were observed in the old rats with a septal transplant. Sixteen weeks after grafting, another experiment was conducted in a homing hole board task. Rats were allowed to escape from a large circular arena through one hole out of many, and to reach home via a flexible tube under the table. The escape hole was at a fixed position according to distant room cues, and olfactory cues were made irrelevant by rotating the table between the trials. An additional cue was placed on the escape position. No age-related difference in escape was observed during training. During a probe trial with no hole connected and no proximal cue present, the old untreated rats were less clearly focussed on the training sector than were either the younger or the grafted old subjects. Taken together, these experiments indicate that enriched housing conditions and spatial training during adult life do not protect against all age-related deterioration in spatial ability. However, it might be that the considerable improvement observed in the grafted subjects results from an interaction between the graft treatment and the housing conditions. 相似文献

目的:研究葛根素(Pur)对脂多糖(LPS)诱导的小鼠急性感染性脑损伤的保护作用及机制。方法:36只清洁级雄性ICR小鼠随机分为正常对照组(NS)、LPS感染组(LPS)和葛根素组(Pur+LPS)。LPS腹腔注射建立急性脑损伤模型,立即腹腔注射给予葛根素注射液治疗,6和24h后旷场。暗场实验检测各组小鼠的自发活动,WesternBlot检测脑组织白细胞分化抗原14(CD14)和环氧化酶2(C0X-2)蛋白表达量,Nisl染色观察神经元存活情况。结果:与NS组相比,LPS组6和24 h旷场实验自主活动距离和穿格次数显著减少,海马神经元的存活数目显著减少,脑组织CD14和C0X-2蛋白表达量显著升高(P<0.05);与LPS组比较,Pur+LPS组6和24 h时海马神经元的存活率明显提高,且CD14和C0X-2蛋白表达量明显下降(P<0.05)。结论:葛根素注射液对LPS所致小鼠急性感染性脑损伤具有一定保护作用,其作用机制可能与抑制脑组织CD14和C0X-2表达有关。 相似文献

A method which optimizes on global properties of sample recordings is proposed for the definition of and the discrimination between electroencephalogram (EEG) classes. The sample was drawn from students at the University of Heidelberg from 1974 to 1978 and consists of 15 healthy index cases clinically ascertained as belonging to the low voltage EEG group. In addition, the three clinically defined groups: diffuse β (18 index cases), borderline α (12 index cases) and monomorphous α (18 index cases) have been included in the study, as well as the first degree relatives of the index cases, thus providing a clinical classification into four groups. The proposed method provides an automatic and reliable classification algorithm using discriminant and cluster analysis. The relation between such an automatized classification and clinical classification schemes is investigated. In particular, the inheritance of the low voltage, EEG, the question on sex differences and the question of a simple Mendelian mechanism had been examined. The method of random splittings had been applied for discriminant and cluster analysis. Our findings can be summarized as follows: (1) except for the monomorphous α EEG group, the clinical classification shows rather marginal separation (discriminating performance 60% to 75%), while a new and more reliable grouping scheme improves the discriminating performance up to 87% to 91%. The latter scheme leads to the concept of personal channel pattern (PCP) and was compared to the clinical classification scheme by means of contingency tables; (2) only a weak correlation between the clinically and PCP-based groups could be found (Cramér Index: 0.27). Accordingly, we continued to investigate the extent to which the proposed EEG classification scheme can nevertheless explain the genetic mechanisms apparently involved in the low voltage EEG. We thus considered the role of sex differences manifest in our proposed new grouping scheme; (3) males occurred more frequently in the new group 3 and females more frequently in the new group 1. In this regard, a much better correlation of the new groups between mothers and children than between fathers and children was observed; and (4) with help of our new PCP scheme, we have been able to reproduce a simple two gene Mendelian scheme to explain inheritance of the clinical low voltage EEG group. The aim is to describe a general approach to determining important electrode positions when measured electro-encephalogram
signals are used for classification. The approach is exemplified in the frame of the brain-computer interface, which crucially
depends on the classification of different brain states. To classify two brain states, e.g. planning of movement of right
and left index fingers, three different approaches are compared: classification using a physiologically motivated set of four
electrodes, a set determined by principal component analysis and electrodes determined by spatial pattern analysis. Spatial
pattern analysis enhances the classification rate significantly from 61.3±1.8% (with four electrodes) to 71.8±1.4%, whereas
the classification rate using principal component analysis is significantly lower (65.2±1.4%). Most of the 61 electrodes used
have no influence on the classification rate, so that, in future experiments, the setup can be simplified drastically to six

Male Sprague-Dawley rats with combined lesions to the dorsal and lateral aspects of the entorhinal cortex in one hemisphere and of the contralateral dorsolateral hippocampal formation were compared with both operated and unoperated controls on three different behaviors, monitored across a 53 day postoperative period. The rats with the combined entorhinal cortex-hippocampal lesions (EH) showed transient hyperactivity in the open field, transient reduction in spontaneous alternation levels in an unbaited T-maze and persistent deficits in learning spatial maze problems. The results of the present experiment are discussed in comparison with those from experiments on rats with bilateral hippocampal lesions and those from experiments on rats with bilateral entorhinal cortex lesions. Although some similarities among these findings suggest that these two brain regions probably function in a coordinated fashion with respect to these behaviors, differences in the various syndromes are also discussed. 相似文献

Research on visual attention has demonstrated that covert attention can be focused on particular locations within one hemifield,
but that a specific "meridian" cost may also be found for shifting attention between hemifields. These issues have received
less consideration for audition, even though reliable behavioral measures for the effects of spatial attention on hearing
are now available. We examined the spatial distribution of covert attention in an auditory task following spatially non-predictive
peripheral auditory cues (which should induce exogenous attention shifts), or following symbolic central cues that predicted
the likely location for the auditory target (to induce endogenous attention shifts). In both cases, we found that attention
can be focused not only on one hemifield versus another, but also within one hemifield in an auditory task. However, there
was no unequivocal evidence for a meridian effect in audition.

Cluster analysis of the MMPI has been utilized widely in the chronic low back pain literature to try to identify reliable patient subtypes predictive of treatment outcome. We extended this methodology to patients with heterogeneous chronic medical conditions by replicating prototypic MMPI cluster group profiles and by relating cluster groups to clinical baseline and outcome data. Subjects were two independent samples (n=254 and n=263) of chronically ill patients admitted to an inpatient medicine/psychiatry unit. Using a four-cluster solution, similar cluster profile groups were replicated in both samples. Consistent differences emerged between cluster groups on functional impairment, psychiatric diagnoses, depression, and psychosomatic symptoms. Cluster group membership also predicted changes in functional impairment and depression six months after treatment. Results are discussed in terms of similarities between chronic low back pain and chronic illness and tailoring treatment to different patient types.This research was supported in part by a grant from the Henry J. Summary The effects of massive destruction of granule cells of the fascia dentata on the spatial and temporal firing characteristics of pyramidal cells in the CA1 and CA3 subfields of the hippocampus were examined in freely moving rats. Microinjections of the neurotoxin colchicine were made at a number of levels along the septo-temporal axis of the dentate gyri of both hemispheres, resulting in destruction of over 75% of the granule cells. By contrast there was relatively little damage to the pyramidal cell fields. As assessed by three different behavioral tests, the colchicine treatment resulted in severe spatial learning deficits. Single units were recorded from the CA1 and CA3 subfields using the stereotrode recording method while the animals performed a forced choice behavioral task on the radial 8-arm maze. Considering the extent of damage to the dentate gyrus, which has hitherto been considered to be the main source of afferent information to the CA fields, there was remarkably little effect on the spatial selectivity of place cell discharge on the maze, as compared to recordings from control animals. There was, however, a change in the temporal firing characteristics of these cells, which was manifested primarily as an increase in the likelihood of burst discharge. The main conclusion derived from these findings is that most of the spatial information exhibited by hippocampal pyramidal cells is likely to be transmitted from the cortex by routes other than the traditional trisynaptic circuit. These routes may include the direct projections from entorhinal layers II and III to CA3 and CA1, respectively. 相似文献

Spatial filtering of surface electromyography (EMG) signal can be used to enhance single motor unit action potentials (MUAPs).
Traditional spatial filters for surface EMG do not take into consideration that some electrodes could have poor skin contact.
In contrast to the traditional a priori defined filters, this study introduces an adaptive spatial filtering method that adapts
to the signal characteristics. The adaptive filter, the maximum kurtosis filter (MKF), was obtained by using the linear combination
of surrounding channels that maximises kurtosis. The MKF and conventional filters were applied to simulated EMG signals and
to real EMG signals recorded with an electrode grid to evaluate their performance in detecting single motor units. The MKF
was compared with conventional spatial filtering methods. Simulated signals, with different levels of spatially correlated
noise, were used for comparison. The influence of one electrode with poor skin contact was also investigated. The MKF was
found to be considerably better at enhancing a single MUAP than conventional methods for all levels of spatial correlation
of the noise. For a spatial correlation of 0.97 of the noise, the improvement in the signal-to-noise ratio, where a MUAP could
be detected, was at least 6 dB. With a simulated poor skin contact for one electrode, the improvement over the other methods
was at least 19 dB. 相似文献
