从恙虫病立克次体(Rt)Karp株的群特异性抗原基因序列设计两对引物建立套式PCR检测现场鼠体采集的恙螨幼虫体内及恙虫病现症病人外周血单核细胞内的RtDNA。除一份已用过药病人标本外,其余9份均可检见88bpDNA扩增产物;检测结果表明:现场鼠体采集的地里纤恙螨(Leptotrombidiumdeliense)幼虫的Rt携带率是13.3%,证实套式PCR可用于急性期恙虫病的诊断和媒介恙螨流行病学的调查。 相似文献

Queensland tick typhus (QTT), caused by Rickettsia australis, is usually a relatively mild illness but can occasionally be severe. We describe three cases of probable QTT with unusual clinical features, namely splenic infarction, fulminant myopericarditis and severe leukocytoclastic vasculitis. QTT may present with uncommon clinical features in addition to the more common manifestations. A high index of suspicion enables specific antibiotic therapy that may hasten recovery. 相似文献

We assessed the prevalence of antibodies to spotted fever group rickettsiae in human beings and dogs by indirect immunofluorescence in the region of 'Vallés Occidental', Barcelona (Spain). In the group of 150 serum samples from patients without former history of Mediterranean spotted fever, 12 had antibodies to Rickettsia conori. The overall seroprevalence was 8% (95% confidence interval, 4.6% to 13.5%). There were no statistically significant differences between the mean ages of patients with positive and negative antibodies to R. conorii. However, seropositivity was significantly more common among patients living in semi-rural areas. In the group of 138 dog serum samples, 36 (26.1%) sera had antibodies to R. conorii. When the present results were compared with those obtained in a previous seroepidemiological survey carried out in the same geographical region in 1987, no significant differences were found. Therefore, although the epidemiological markers have dropped, this does not absolutely confirm the decrease of the presence of R. conorii in this area. 相似文献

Rickettsia typhi and R. felis, etiologic agents of murine typhus and fleaborne spotted fever, respectively, were detected in Oriental rat fleas (Xenopsylla cheopis) collected from rodents and shrews in Java, Indonesia. 目的评估全国9省、市CDC实验室(排名不分先后)立克次体检测技术质量。方法国家实验室统一向参加单位发放3种血清及5种PCR质控品及必要相关试剂,各单位在本实验室现有仪器设备、常规使用试剂及各种量器等条件下按质控要求采用间接免疫荧光实验和PCR进行检测,规定时间内反馈检测结果。我们对IgG抗体滴度值和PCR的特异性、灵敏度进行评估。结果9个参加单位,除一个单位对s3血清质控品不合格外,其他均合格。除一个单位对P4 PCR质控品检测不合格外,其他检测均达到要求。结论参加本次质控的9个实验室,7个通过此次考核。2个实验室须进一步实验室优化调整。 相似文献

目的克隆与表达普氏立克次体(Rickettesia prowazekii)主要蛋白抗原基因,分析所表达重组蛋白的抗原特异性。方法采用PCR法从普氏立克次体全基因组中扩增其主要抗原基因,将扩得的基因片段插入原核表达质粒;将基因重组表达质粒转化大肠杆菌,用IPTG诱导转化菌表达重组蛋白。将普氏立克次体、莫氏立克次体、立氏立克次体、恙虫病东方体、黑龙江立克次体、贝氏柯克斯体等实验感染小鼠血清与重组蛋白做免疫印迹。结果经过PCR扩增后获得9个目的基因片段,用其构建出9个原核表达质粒;9个原核表达质粒转化菌均高效表达出相应的重组蛋白;结果显示FtsZ、GroEI。Rp828、EF—Ts等4个蛋白特异性最好,仅与普氏立克次体感染小鼠血清反应;其次是Omp,它除与普氏立克次体感染小鼠血清反应外,仅与莫氏立克次体感染血清反应。结论FtsZ等4个重组蛋白为普氏立克次体的特异性抗原,可以作为研制流行性斑疹伤寒血清学诊断试剂的候选抗原。 相似文献

AIM: To investigate the chemical constituents of the roots of Polygala sibirica L. (Polygalaceae) METHOD: The isolation was performed by solvent extraction and various chromatographic techniques, including silica gel, Sephadex LH-20, ODS, semi-preparative HPLC, and preparative TLC. The chemical structures were elucidated based on extensive spectroscopic analysis, including HR-ESI-MS and 1 D- and 2D-NMR spectroscopic data. RESULTS: A total of sixteen compounds, including five xanthones (5, 7-10), five saccharide esters (1, 3, 4, 12, 13), two flavon- oids (14, 16), two triterpenoids (11, 15), one phenylpropanoid (6), and one benzophenone glycoside (2) were isolated. Their struc- tures were determined as sibiricose A7 (1), sibiriphenone A (2), polygalatenoside A (3), polygalatenoside C (4), lancerin (5), 3, 4, 5-trimethoxycinnamic acid (6), 6-hydroxy-1, 2, 3, 7-tetramethoxyxanthone (7), 1, 3, 7-trihydroxy-2-methoxyxanthone (8), onjix- anthone II (9), 1, 2, 3, 6, 7-pentamethoxyxanthone (10), presenegenin (11), 3'-O-3, 4, 5-trimethoxycinnamoyl-6-O-4-methoxy ben- zoyl sucrose (12), tenuifoliside C (13), 5, 3'-dihydroxy-7, 4'-dimethoxyflavonol-3-O-β-D-glucopyranoside (14), tenuifolin (15), and rhamnetin 3-O-β-D-glucopyranoside (16).
