目的 了解甘肃省严重急性呼吸综合征(SARS)患者、密切接触者和正常人群血清SARS冠状病毒特异性IgG抗体水平。方法 利用酶联免疫吸附试验(ELISA)法检测SARS病毒IgG抗体水平。检测对象包括甘肃省9例SARS患者的急性期和(或)恢复期系列血清,l109例直接护理SARS患者的医生、护士、实验室检测人员、疾控人员和曾与患者有接触的人员以及978例正常人血清。结果 9例临床诊断病例SARS冠状病毒特异性IgG抗体中6例为阳性,疾病恢复后12个月血清抗体仍为阳性;密切接触者1例特异性IgG抗体阳性;正常人3例特异性IgG抗体阳性。结论 病例抗体阳性符合临床诊断,密切接触者和正常人的抗体阳性数较低,提示可能不存在隐性感染。  相似文献

流感病毒血凝素基因工程抗体的抗病毒实验效果观察   总被引:1,自引:0,他引:1  
目的:对昆虫细胞系统表达纯化的中和性流感病毒血凝素基因工程全抗体IgG-IV-2,IgG-IV-6进行体外和体内抗病毒效果的研究。方法:对比抗体应用前后病毒在MDCK细胞中的病毒滴度和动物模型中粘膜用药前后鼠肺内病毒滴度的改变,验证抗体的粘膜抗感染效果。结果:两株基因工程抗体使4.5log10TCID50滴度的病毒下降1/2的剂量分别为0.8μg和0.5μg;动物模型的粘膜给药表明使4.0log10TCID50的病毒下降1/2所需的抗体剂量IgG-IV-2为0.25mg/kg体重,IgG-IV-6为0.1mg/kg体重,联合应用时为0.08mg/kg体重。结论:获得的基因工程抗体具有体内体外的抗病毒效果,能够中和病毒毒力。  相似文献

The potential of vaccine-elicited anti-HIV envelope antibodies to control HIV-infection was evaluated by immunizing macaques with the HIV envelope protein and transiently depleting them of their CD8+ cells before intravenous challenge with the pathogenic CCR5-tropic SIV/HIV chimeric virus, SHIV(SF162P4). Although sterilizing immunity was not achieved, all vaccinated animals effectively controlled infection and remained free of disease for the duration of observation (over 3 years). In contrast, during the same period, the control animals progressed to disease. Both the vaccinees and the controls developed robust cell-mediated antiviral and neutralizing antibody responses following infection. A comparative analysis of these responses suggests that the more effective long-term control of infection by the vaccinated animals is due to the more rapid development of anti-HIV envelope antibodies. These studies suggest that priming by vaccination of B cell anti-HIV envelope responses maybe crucial for the long-term control of HIV infection.  相似文献

PROBLEM: Conventional immunization using whole sperm containing multiple antigens as the immunogen followed by hybridoma technology usually gives antibodies to antigens invariably of testicular origin, probably because of the strong immunogenic nature of these antigens. Therefore, an alternate approach of neonatal tolerization or subtractive immunization has been utilized to raise antibodies specific to epididymis by suppressing immune response to testicular antigens. METHOD OF STUDY: Neonatal mice were tolerized with testicular sperm proteins on days 0 and 5. These animals were then immunized with epididymal sperm proteins on day 21, followed by two boosters at biweekly intervals. Sera from these mice were used to localize epididymis-specific antigens. RESULTS: Sera from mice that were tolerized to testicular sperm proteins and later immunized with epididymal sperm proteins reacted only with epididymal proteins. CONCLUSION: The results of this study demonstrate that neonatal tolerization with testicular sperm proteins, followed by immunization with epididymal sperm proteins, enhances the production of antibodies to proteins exclusively of epididymal origin.  相似文献

目的 研究SARS冠状病毒棘突蛋白受体结合部位S1的免疫原性,为SARS的实验诊断和新型疫苗的研究提供依据。方法 用克隆有哺乳动物细胞密码子优化的SARS-CoV S1基因的质粒pcDNA3.1/S1或P-S1Ig转染293T细胞,用细胞的上清液纯化S1蛋白。以pcDNA3.1/S1质粒对BALB/c小鼠进行2次基因免疫,以纯化的S1蛋白进行加强免疫。用ELISA法检测小鼠抗SARS-CoV的特异性IgG抗体,并在Vero E6细胞上做体外中和实验,检测中和抗体。结果 S1蛋白诱导小鼠产生抗SARS-CoV的特异性抗体;1:1499.68稀释的S1蛋白免疫的小鼠血清可保护50%的细胞对1000TCID50的病毒攻击,而阴性对照血清不能保护细胞对病毒的感染。结论 SAPS冠状病毒棘突蛋白受体结合部位S1能有效诱导机体产生具有高效保护作用的中和抗体免疫反应,可望发展成为理想的SARS棘突蛋白亚单位疫苗。  相似文献

In the event of smallpox bioterrorism, widespread vaccination may be required. Vaccinia immune globulin (VIG) has been used to treat complications from the smallpox vaccine. While the potency of VIG was defined by its ability to neutralize intracellular mature virus, a second form of vaccinia called the extracellular enveloped virus (EEV) is critical for virus spread in the host. The B5R-protein is one of many EEV-specific proteins. Immunoprecipitation and ELISA revealed that VIG recognizes the B5R-protein. An EEV plaque-reduction assay using a recombinant vaccinia that lacks the majority of the extracellular domain of B5R showed that the ability of VIG to neutralize EEV is principally directed at B5R. In addition, absorbing out the anti-B5R antibody present in VIG through the addition of recombinant B5R protein abrogated VIG's ability to significantly neutralize wild-type EEV. This work demonstrates the prominent role of B5R as a target of EEV-neutralizing activity of human antibodies.  相似文献

目的:研究CD20scFv嵌合T淋巴细胞靶向杀伤Daudi细胞时杀伤的效果和T细胞活化情况。方法:将两种质粒转染至PA317细胞中,用转染成功的PA317上清液感染外周血T淋巴细胞后经800 mg/L的G418筛选1周后去杀伤Daudi、K562细胞,分别在不同时点用流式细胞仪检测Daudi细胞AnnexinⅤ的阳性率 ,用ELISA检测细胞因子IL-2、IFNγ。结果:Daudi细胞AnnexinⅤ的阳性率在24 h内两实验组与K562组相比明显增高,而实验组之间没有显著差异。72 h时CD20scFv-IgGFc-CD28-ζ组比CD20scFv-IgGFc组IL-2(1 509.00 ng/L比220.54 ng/L)和IFNγ(912.16 ng/L比251.42 ng/L)的分泌量有显著增高。结论:①两种CD20scFv特异的T细胞在引起Daudi细胞早期凋亡无显著差异,说明在CD20scFv的靶向杀伤过程中CD28-ζ基因可能不主导Daudi细胞的早期凋亡。②CD20scFv-IgGFc-CD28-ζ组IL-2、IFNγ增幅更加明显,说明CD3ζ和CD28嵌合的T细胞可以自身活化而不受MHC的限制,从而增强了T细胞的活化和杀伤等功能。  相似文献

目的 探讨新型抗蝰蛇毒血清对国产圆斑蝰蛇毒及缅甸蝰蛇毒的作用 .方法 采用免疫电泳、SDS -聚丙烯酰胺电泳及动物实验等方法 .结果 国产圆斑蝰蛇毒的LD50 (腹腔注射 )为 0 .3 0 6± 0 .0 0 3mg/kg ,缅甸蝰蛇毒的LD50 (腹腔注射 )为 0 .2 90± 0 .0 0 3mg/kg .免疫电泳结果表明国产圆斑蝰蛇毒和缅甸蝰蛇毒均和新型抗蝰蛇毒血清产生明显的免疫沉淀线 .体外中和实验表明抗蝰蛇毒血清对国产圆斑蝰蛇毒的中和抗体效价为 2 75 0 μg/ml(约45 0LD50 ) ,对缅甸蝰蛇毒的中和抗体效价为 2 490 μg/ml(约 43 0LD50 ) .体内保护实验表明给予 0 .0 5ml抗蝰蛇毒血清可抵御 2 5 4μg(约41 5LD50 )国产圆斑蝰蛇毒或 116μg(约 2 0LD50 )缅甸蝰蛇毒的攻击 ,使小鼠全部存活 .结论 新型抗蝰蛇毒血清对国产圆斑蝰蛇毒及缅甸蝰蛇毒的中和抗体效价均较高 ,有较大的应用价值 .  相似文献

Summary Samples of 96 polyvalent and virus-specific immunoglobulin batches commercially available in West-Germany were tested by enzyme-linked immunoassay and immunoblot for the presence of anti-HTLV-III antibodies. 37% of the polyvalent and 87% of the virus-specific batches were positive. We studied the sensitivity of sensorimotor cortex neurons in rabbits to microiontophoretic administration of antibodies against acid fibroblast growth factor (anti-aFGF). Spontaneous activity and reaction of neurons to electrical stimulation of ‘feeding centers’ in the lateral hypothalamic area (LHA) were recorded. We analyzed impulse activity of 52 neurons in the sensorimotor cortex. It was shown that 14 neurons (25%) reacted to microiontophoresis of anti-aFGF (excitation and inhibition in 9 and 5 neurons, respectively). Microiontophoretic administration of anti-aFGF did not change the reaction (excitation or inhibition) of 27 neurons to electrical stimulation of LHA. Initially, 14 neurons did not response to LHA stimulation. After microiontophoretic administration of anti-aFGF, 6 of 14 neurons displayed pronounced reactions to electrical stimulation of LHA (excitation and inhibition in 2 and 4 neurons, respectively). These data suggest that aFGF plays an important physiological role in feeding motivations.  相似文献
