目的:检测人肺癌细胞中是否存在与Na^ /H^ 交换蛋白-1(NHE-1)基因转录相关的重要转录因子--C/EBP和HMG样蛋白。方法:采用电泳迁移率改变分析法(EMSA)检测人肺癌细胞株核蛋白中C/EBP和HMG样蛋白的表达及其水平。结果:EMSA检测到A549细胞和转染NHE-1反义载体的A549细胞均有转录因子C/EBP和HMG样蛋白的表达,且后者表达水平高于前者;采用50倍未标记的片段能完全抑制相应转录因子与^32P标记的相同片段的结合。结论:A549细胞核蛋白中存在能与C/EBP结合序列和Poly(dA:dT)区结合的转录因子,即C/EBP和HMG样蛋白,后者的表达水平高于前者。特异性竞争抑制试验表明外源性特异DNA片段能抑制相同片段与转录因子的结合。 相似文献

The most common graft in anterior cruciate ligament (ACL) surgery involves using the central one-third of the patellar tendon.
Knowledge concerning the postoperative disability after harvesting the patellar tendon is, however, limited. The aim of this
study was to evaluate the impact patellar tendon suture and bone grafting of the patellar bone defect might have in terms
of functional outcome and patellofemoral pain after harvesting the bone-tendon-bone graft, compared with leaving the harvested
site non-sutured and non-grafted. Sixty patients, scheduled for arthroscopically assisted ACL reconstruction, were randomly
allocated to two groups. In group I, suture of the patellar tendon and bone grafting of the patellar defect were performed.
In group II, the tendon gap and the patellar defect were left open. Preoperatively, there was no significant difference between
the groups when comparing objective knee stability, as measured with a KT-1000 laxity meter, Lysholm score, Tegner activity
level, IKDC score, or patellofemoral pain score. Both groups had a significantly improved Lysholm score at the 2-year follow-up,
without any difference between them. Tegner's activity level was significantly lower at follow-up, compared with the pre-injury
level in both groups. The patellofemoral pain score improved significantly after the reconstruction, without any difference
between the groups. Ultrasonography did not reveal any difference between the groups in terms of healing of the tendon gap.
This study revealed no differences in donor site morbidity, functional outcome, patellofemoral pain score or knee joint stability
between the two treatment groups. The conclusion is that suture of the patellar tendon and bone grafting of the patellar defect
do not improve the functional results or reduce donor site morbidity after arthroscopically assisted ACL.

目的:探讨人前病毒整合位点1(PIM1)诱导细胞衰老的分子机制。方法:构建过表达PIM1的2BS细胞系,通过Western印迹法和衰老相关β-半乳糖苷酶染色实验测定过表达PIM1是否诱导细胞衰老。免疫沉淀联合质谱分析测定PIM1是否能有效免疫沉淀核异质核糖核蛋白U(hnRNPU)蛋白。运用Real-timePCR和Western印迹法测定PIM1是否影响hnRNPUmRNA和蛋白表达水平。构建同时过表达PIM1和hnRNPU的2BS细胞系,运用Western印迹法和衰老相关β-半乳糖苷酶染色实验检测hnNRPU过表达对PIM1诱导的细胞衰老的影响。结果:与空载对照组相比,过表达PIM1组中衰老信号通路关键基因p53、p21和p16的表达显著增加(p53,t=4.36,P<0.05;p21,t=3.814,P<0.05;p16,t=4.72,P<0.01),衰老细胞的比例增加(t=6.831,P<0.01)。免疫沉淀联合质谱分析结果显示PIM1能有效免疫沉淀hnRNPU蛋白。PIM1过表达对hnRNPU的mRNA表达水平没有影响(t=0.295,P=0.783),但是能抑制hnRNPU蛋白的表达(t=33.85,P<0.001)。与只过表达PIM1细胞相比,同时过表达PIM1和hnRNPU细胞,衰老信号通路关键基因p53、p21和p16的表达下降(p53,t=15.317,P<0.001;p21,t=8.012,P<0.01;p16,t=14.08,P<0.001),衰老细胞的比例降低(t=10.38,P<0.01)。结论:hnRNPU过表达抑制PIM1诱导的细胞衰老。 相似文献

The interactions of acidic and basic drugs with 1-acid glycoprotein (1-AGP) were investigated using circular dichroism (CD) measurements. Extrinsic Cotton effects were generated by the binding of drugs to 1-AGP. The CD data suggested the presence of a single binding site on the 1-AGP molecule. The induced ellipticities of the acidic drug–1-AGP system decreased with increasing pH, while the ellipticities for the basic drugs increased with pH. The ellipticities for all drugs were reduced by the addition of fatty acids. Furthermore, the induced ellipticities decreased in the presence of cesium chloride for basic drugs bound to 1-AGP. The extrinsic Cotton effects therefore appear to result from hydrophobic interaction with 1-AGP for the acidic drugs and from hydrophobic and electrostatic interactions for the basic drugs. 相似文献

Hepatitis C virus (HCV) establishes persistent infection in the majority of infected individuals. The currently accepted hypothesis of immune evasion by antigenic variation in hypervariable region 1 (HVR1) of glycoprotein E2 does not however, explain the lack of subsequent immune recognition. Here, we show that the N-terminal region of E2 is antigenically and structurally similar to human immunoglobulin (Ig) variable domains. E2 is recognized by anti-human IgG antibodies and also possesses common amino acid (aa) sequence features of the conserved v-gene framework regions of human Ig light chains in particular but also heavy chains and T cell receptors. Using a position specific scoring system, the degree of similarity of HVR1 to Ig types correlated with immune escape and persistence in humans and experimentally infected chimpanzees. We propose a unique role for threshold levels of Ig molecular mimicry in HCV biology that not only advances our concept of viral immune escape and persistent infection but also provides insight into host-dependent disease patterns. 相似文献
