A case of small round and spindle cell sarcoma with neuronal differentiation and oncocyte-like features is presented. The tumor was encountered in a 32 year old Japanese woman with an initial presentation of palpable tumor in the left lateral region of the thorax. The resected tumor was a partially well encapsulated whitish medullary one and consisted of small round and spindle tumor cells, together with so-called rhabdoid cells in the small round cell area. Although pseudorosettes were often observed, true rosette formation could not be detected anywhere. Ultrastructurally, despite a histologic variety of tumor cells, most tumor cells possessed numerous mitochondria, some of which occasionally contained abnormal filamentous or crystalloid structures. Various amounts of microfilaments were present in most tumor cells and microtubules were present in a few. A minority of small round cells possessed a small number of neurosecretory granules, especially in short cytoplasmic processes. A positive immunoreaction for neuron specific enolase was found by immunohistochemical examination in several small round tumor cells and for neurofilaments in lesser numbers. Despite the lack of S-100 protein, MB2 was detected in both small round and spindle cells. On the basis of these findings, the tumor of the present case corresponds to malignant peripheral nerve sheath tumor with neuronal differentiation and oncocytic features.  相似文献

To study the role of the noradrenergic innervation in the production of peptide neurohormones by cells of the supraoptic nucleus (SON) the substance 6-hydroxydopamine (6-HDA), which causes selective destruction of noradrenergic terminals, was used. The intensity of fluorescence of these terminals in the region of SON 7 days after intraventricular injection of 6-HDA in a dose of 250 g was sharply reduced and morphological pictures reflecting activation of hormone production by SON cells were observed. According to the results the noradrenergic innervation evidently inhibits synthesis of peptide neurohormones by SON cells and regulates the transport and secretion of peptide neurohormones from the posterior lobe of the pituitary into the general circulation.Laboratory of Neuroendocrinology, I. The Merkel cell is a distinctive nondendritic, nonkeratinocytic, epithelial clear cell believed to migrate from the neural crest to the epidermis and dermis, which is usually located in or near the basal layer of the epidermis and associated with nerve terminations. Merkel first described these cells in 1875 as "Tastzellen" occurring in the snout of a mole. They are believed to function as slowly adapting mechanoreceptors that mediate the sense of touch. Tumors arising from Merkel cells have been reported to occur on the head and neck area, the trunk, arms, and legs, and resemble a primary cutaneous lymphoma or cutaneous metastasis of a lymphoma or a carcinoma. Electron microscopy, to locate the characteristic membrane-bound, dense core neurosecretory granules, is needed for accurate diagnosis. These tumors must be treated aggressively to minimize the chance of local recurrence and nodal or visceral metastases. The authors present a case of Merkel cell tumor occurring on the eyelid. The clinical history, light and electron microscopic findings are shown.  相似文献

This study has analyzed variations in the number of nucleoli and nuclear bodies, as well as in their ultrastructural and cytochemical organization, after the osmotically induced activation of supraoptic nucleus (SON) neurons of the rat. The number of nucleoli and nuclear bodies and also the nucleolar size were determined on smear preparations of previously block-impregnated SON. The mean number of nucleoli per cell was 1.35 +/- 0.6 (mean +/- SDM) in control rats. No significant variations in this value were registered either in dehydrated or rehydrated rats. The mean nucleolar volume and the total nucleolar volume per cell showed a significant increase in dehydrated rats with respect to the controls, whereas these two parameters tended to return to control values in rats rehydrated after dehydration. The mean number of nuclear bodies per cell increased significantly from 0.56 +/- 0.50 (mean +/- SDM) in control rats to 1.54 +/- 1.1 after 6 days of dehydration. By electron microscopy, SON neurons displayed a reticulated nucleolar configuration. After the osmotically induced neuronal activation, there was an increase in the proportion of the total nucleolar area occupied by the granular component, and also a reduction in the mean fibrillar-center area. The most characteristic nucleolar features in rehydrated rats were the tendency for the granular component to be segregated and the occurrence of intranucleolar vacuoles. Ultrastructural cytochemistry with a specific silver method revealed a selective silver reaction on the coiled threads of the nuclear bodies--identified as "coiled bodies"--and on the nucleolar fibrillar components in all animal groups studied. Since nucleoli play a major role in ribosome biogenesis, a relationship between these nucleolar changes and the level of cellular activity of SON neurons is proposed. Furthermore, the response of nuclear "coiled bodies" to neuronal activation suggests their participation in the processing and transport of rRNA precursors.  相似文献

目的:探讨支气管扩张症中神经内分泌免疫网络的异常和肥大细胞(MC)在该病发病中的作用及关系。方法:应用组织化学、免疫组化、组织化学与免疫组化结合的方法和形态计量学方法进行观测。结果:支气管扩张症中,支气管上皮蛙皮素(Bombesin)阳性细胞、固有膜S-100蛋白和神经特异性烯醇化酶(NSE)阳性神经纤维、IgE阳性细胞、MC和IgE阳性MC均显著增多,且在支气管相关淋巴组织(BALT)增生的区域上述肺内分泌细胞、神经纤维和IgE阳性细胞增多尤为显著,S-100蛋白和NSE阳性神经纤维分布于弥散淋巴组织和BALT中,MC与S-100蛋白阳性神经纤维紧密接触.MC表面有IgE阳性环状带,MC和IgE阳性细胞出现在支气管上皮间和肺泡壁。结论:支气管扩张症的发病与局部神经内分泌免疫网络异常有关;MC可能作为感受器、分泌细胞或靶细胞参与神经内分泌免疫网络,在支气管扩张症的发病中起重要作用。  相似文献

The present study aims to give detailed histomorphological features of the hippocampus of adult male New Zealand rabbits. Both histological and histochemical specimens were prepared to be examined microscopically by using a light microscope. The hippocampus appeared as C-shaped hippocampal proper, dentate gyrus, and subiculum. The hippocampal proper subdivided along its length according to the density and size of its major constituent pyramidal cells into four distinct regions named Cornu Ammonis (CA1, CA2, CA3, and CA4). With the histochemical preparations, each of these regions consisted of five layers, stratum alveolus, stratum oriens, stratum pyramidale, stratum radiatum, and stratum lacunosum-moleculare. The stratum pyramidale constituted the middle dark zone and contained the principal excitatory neurons and a few interneurons. Histochemically, the pyramidal neurons along all regions of the CA reacted positively to Grimelius silver impregnation, lead hematoxylin, Gomori's aldehyde fuchsin, aldehyde thionine, Gomori's chrome alum hematoxylin, and performic acid alcian blue stains. Immunohistochemically, the pyramidal neurons reacted positively to anti-NSE antibodies. The dentate gyrus was formed of three distinct layers. The subiculum was formed of proper subiculum, presubiculum, and parasubiculum.  相似文献

The tumor cells in the pleural effusions of a case of small cell undifferentiated carcinoma have been studied by light microscopy and by thin section and freeze-fracture electron microscopy. Evidence of exocytosis of neurosecretory granules from the tumor cells is presented. The tumor cells form clumps, with the cells joined by focal tight junctions and small desmosomes. These findings are discussed with reference to the neuroendocrine origin of these tumors and the role of the junctional complexes in the formation of tumor cell aggregates.  相似文献

Brain stem projections to the neurons in the caudal neurosecretory complex (CNC) of Poecilia sphenops (molly) have been studied with HRP retrograde tracing. Using light microscopic procedures, HRP-labelled neurons were located in the reticular nucleus of the medulla (RMN) and in the vicinity of the nucleus of the medial longitudinal fascicle (NMLF). The present study was undertaken to examine the cytology of the brain stem neurons that project to the caudal neurosecretory complex using combined HRP-electron microscopic methods. Cells in the midbrain NMLF containing HRP-filled profiles were located bilaterally close to the midline and just beneath the ependyma. HRP reaction product was found additionally in the neurosecretory cells of the midbrain dorsal tegmental magnocellular nucleus (DTMN) located dorsal to the NMLF. Cells containing HRP-labelled profiles were also seen in the RMN and in neurons dorsal-lateral to the RMN. This latter group of neurons contained small dense core vesicles in addition to HRP labelled dense bodies.  相似文献

The hypothalamic paraventricular nucleus (PVN) and angiotensin II (AngII) play critical roles in cardiovascular and neurohumoral regulation ascribed in part to vasopressin (VP) release. The AngII actions in the PVN are mediated largely through angiotensin II type 1 (AT1) receptors. However, there is indirect evidence that the functionally elusive central angiotensin II type 2 (AT2) receptors are also mediators of AngII signaling in the PVN. We used electron microscopic dual immunolabeling of antisera recognizing the AT2 receptor and VP to test the hypothesis that mouse PVN neurons expressing VP are among the cellular sites where this receptor has a subcellular distribution conducive to local activation. Immunoreactivity for the AT2 receptor was detected in somatodendritic profiles, of which 60% of the somata and 28% of the dendrites also contained VP. In comparison with somata and dendrites, axons, axon terminals, and glia less frequently contained the AT2 receptor. Somatic labeling for the AT2 receptor was often seen in the cytoplasm near the Golgi lamellae and other endomembrane structures implicated in receptor trafficking. AT2 receptor immunoreactivity in dendrites was commonly localized to cytoplasmic endomembranes, but was occasionally observed on extra- or peri-synaptic portions of the plasma membrane apposed by astrocytic processes or by unlabeled axon terminals. The labeled dendritic plasmalemmal segments containing AT2 receptors received asymmetric excitatory-type or more rarely symmetric inhibitory-type contacts from unlabeled axon terminals containing dense core vesicles, many of which are known to store neuropeptides. These results provide the first ultrastructural evidence that AT2 receptors in PVN neurons expressing VP and other neuromodulators are strategically positioned for surface activation by AngII and/or intracellular trafficking.  相似文献
