为阐明骨髓基质细胞和内皮细胞联合种植在生物复合材料上构建组织工程化骨 ,在促进成骨过程、加速局部血管生成中的作用 ,将大鼠骨髓基质细胞和人脐静脉内皮细胞联合种植在左旋聚乳酸 /β-磷酸三钙 (PL L A/β- TCP)多孔复合支架材料上作为实验组 ,仅种植骨髓基质细胞在该材料上作为对照组。将此种有活细胞的复合材料种植于裸鼠大腿后侧肌袋内 ,分别于种植后 1、4、8、12、16周处死动物 ,取出复合材料 ,行组织学检测 ,并用图像处理技术计算实验组和对照组的成骨面积百分比和材料面积百分比 ,观察其变化规律。结果显示 :实验组成骨面积百分比增加比对照组明显加快 ,而材料面积百分比减少比对照组迅速 ;其内毛细血管网形成情况也要明显优于对照组。表明骨髓基质细胞和内皮细胞联合种植于支架材料构建组织工程化骨不仅有利于成骨作用 ,促进新生骨区内部血管网形成 ,还有利于复合材料的降解吸收 ,这在构造组织工程化骨过程中具有重要意义。  相似文献

Congenital diaphragmatic hernia is generally recognized as a sporadic malformation with little or no risk of recurrence. A family with three affected individuals in two generations is presented. In addition, new prenatal diagnostic techniques including ultrasonography and amniography are discussed. A comparison of associated physical characteristics in isolated versus twelve familial cases of diaphragmatic hernia is presented. In the familial group, there was a higher incidence of affected males (M:F ratio = 2.1 versus 0.67), a higher incidence of bilateral defects (20% versus 3%) and a lower incidence of additional life-threatening malforamtions 3.6% versus 47%). Analysis of available pedigree data favors multifactorial inheritance with a high male: female sex ratio as the most probable mode of transmission.  相似文献

Outbred NMRI mice were sensitized for high IgE production either by subcutaneous injections of low doses of untreated bovine milk or homogenized bovine milk in combination with intraperitoneal injections of Freund's Complete Adjuvant or by oral administration of untreated or homogenized bovine milk without adjuvant. When analysed in murine passive cutaneous anaphylaxis test both types of milk resulted in production of reaginic antibodies against bovine milk proteins when given subcutaneously. When given orally, homogenized milk resulted in reagin production in 10 out of 14 mice, whereas untreated milk resulted in reagin production in only one out of 12 mice. The sensitized mice, and control mice, were orally challenged with either untreated milk, homogenized milk or 0.9% NaCl. Examination of the intestines 40 min after oral administration revealed that homogenized milk, contrary to untreated milk or 0.9% NaCl, resulted in a large increase in the mass of the proximal gut segment of mice sensitized orally with homogenized milk compared with control mice orally challenged with saline (P less than 0.001), and only mice both sensitized and challenged orally with homogenized milk showed degranulation of mast cells in the intestinal wall. By contrast, subcutaneously sensitized mice or mice sensitized orally with untreated bovine milk showed no significant intestinal reaction upon oral challenge with either homogenized or untreated bovine milk. These observations may indicate that the route of sensitization is of great importance when intestinal reactions are to be studied, and that homogenization of bovine milk may render the milk more aggressive with respect to its ability to induce intestinal reactions. The study indicates that mice may be an attractive experimental animal model for mimicking the intestinal anaphylactic reactions of cow milk-allergic humans.  相似文献

A female child is described with multiple anomalies including epicanthus, frontal bossing, short sternum, polydactyly, cleft of the larynx, renal cysts, and unusual dermatoglyphics. She died aged 3 months and was found to have a unique de novo deletion of chromosome No. 4 (q22-q25). This case is compared with other long arm deletions of 4q and reference made to assignment of genetic markers to chromosome No. 4.  相似文献

目的数值模拟神经系统在发育过程中轴突的生长情况。方法根据神经发育原理,建立具有一定刚性的非线性混合抛物型偏微分方程组,运用ADI差分格式和改进的欧拉法作数值分析。结果 (1)在一个正方形平面区域内有10个轴突以相等的间距围成圆形,当中心有1个靶细胞时,轴突生长路径的数值模拟结果与前人所做的有关结果一致;(2)轴突的初始位置同(1),当中央有4个两两上下并排分布的靶细胞时,数值模拟结果很好地反映了轴突生长时的趋化性;(3)在区域的近上、下边界分别有一串靶细胞和一串轴突时,数值模拟结果较好地反映了轴突结集生长时的成束和解束现象。结论本文给出的数学模型和数值主法能够较好地模拟有关实验所观测到的主要现象。  相似文献

Medullary thyroid carcinoma occurs sporadically or as a part of the inherited cancer syndrome multiple endocrine neoplasia (MEN) type 2. The MEN 2 gene has been identified as the RET proto-oncogene on chromosome 10. In MEN 2A, RET mutations are detectable in one of five cysteine codons within exons 10 and 11 and in MEN 2B in codon 918 (exon 16). Direct DNA testing for RET proto-oncogene mutations is the method of first choice in presymptomatic screening of MEN 2 families. Gene carriers should be offered prophylactic thyroidectomy. The process of DNA analysis for RET proto-oncogene mutations is demonstrated in one family with hereditary medullary thyroid carcinoma. The flexibility of the scoliotic spine is an important biomechanical parameter to take into account in the planning of surgical instrumentation. The objective of the paper was to develop a method to characterisein vivo the mechanical properties of the scoliotic spine using a flexible multi-body model. Vertebrae were represented as rigid bodies, and intervertebral elements were defined at every level using a spherical joint and three torsion springs. The initial mechanical properties of motion segments were defined fromin vitro experimental data reported in the literature. They were adjusted using an optimisation algorithm to reduce the discrepancy between the simulated and the measured Ferguson angles in lateral bending of three spine segments (major or compensatory left thoracic, right thoracic and left lumbar scoliosis curves). The flexural rigidity of the spine segments was defined in three categories (flexible, nominal, rigid) according to the estimated mechanical factors (α). This approach was applied with ten scoliotic patients under-going spinal correction. Personalisation of the model resulted in an increase of the initial flexural rigidity for seven of the ten lumbar segments (1.38≤α≤10.0) and four of the ten right thoracic segments (1.74≤α≤5.18). The adjustment of the mechanical parameters based on the lateral bending tests improved the model's ability to predict the spine shape change described by the Ferguson angles by up to 50%. The largest differences after personalisation were for the left lumbar segments in left bending (40±30). Thein vivo identification of the mechanical properties of the scoliotic spine will improve the ability of biomechanical models adequately to predict the surgical correction, which should help clinicians in the planning of surgical instrumentation manoeuvres.  相似文献
