目的 探讨鼻咽癌铂类化疗敏感度与X射线交错互补修复基因1 codon194和codon399单核苷酸多态性的相关性。方法 收集广西医科大学第四附属医院2012年9月1日至2013年12月31日鼻咽部肿物活检确诊为鼻咽癌患者资料,采用限制性片段长度多态性聚合酶链反应技术检测鼻咽癌患者外周血DNA XRCC1 codon194和codon399单核苷酸多态性。顺铂+氟尿嘧啶方案诱导化疗2周期后复查MRI,按照RECIST 1.1标准评价其化疗敏感度,分析单核苷酸多态性(Single nucleotide polymorphism,SNP)与化疗敏感度的关系。结果 XRCC1 codon399 Gln/Gln基因型携带者化疗敏感度为Arg/Arg基因型携带者的3.500倍(P<0.05)。XRCC1 codon399不含Arg基因型(即Gln/Gln)携带者化疗敏感度为含Arg基因型(Arg/Arg 和 Arg/Gln)携带者的3.274倍,(P<0.05)。携带XRCC1 codon194各基因型患者化疗敏感度之间差异无明显统计学意义(P>0.05)。结论 XRCC1 codon399 单核苷酸多态性有可能成为鼻咽癌铂类化疗敏感度的预测因子。 相似文献

RNA interference (RNAi) is an evolutionary conserved mechanism for specific gene silencing. There are currently numerous cancer therapy clinical trials based on RNAi technology. Using an adenoviral system as a delivery mediator of RNAi, we investigated the therapeutic effects of targeting three genes simultaneously in vitro and in vivo. In this study, we constructed an recombinant adenoviral shRNA expression system as Adv-pEGFP-shVEGF-shTERT-shBcl-xl for multi-genes silencing. Our results showed that the adenoviral vector can achieve above 90% of transfection efficiency and induced obvious apoptosis in CNE-2 cell both in vitro and in vivo compared with targeting the TERT alone or controlled group. 相似文献

We have analyzed at high resolution the neuroanatomical connections of the juxtaparaventricular region of the lateral hypothalamic area (LHAjp); as a control and in comparison to this, we also performed a preliminary analysis of a nearby LHA region that is dorsal to the fornix, namely the LHA suprafornical region (LHAs). The connections of these LHA regions were revealed with a coinjection tract-tracing technique involving a retrograde (cholera toxin B subunit) and anterograde (Phaseolus vulgaris leucoagglutinin) tracer. The LHAjp and LHAs together connect with almost every major division of the cerebrum and cerebrospinal trunk, but their connection profiles are markedly different and distinct. In simple terms, the connections of the LHAjp indicate a possible primary role in the modulation of defensive behavior; for the LHAs, a role in the modulation of ingestive behavior is suggested. However, the relation of the LHAjp and LHAs to potential modulation of these behaviors, as indicated by their neuroanatomical connections, appears to be highly integrative as it includes each of the major functional divisions of the nervous system that together determine behavior, i.e., cognitive, state, sensory, and motor. Furthermore, although a primary role is indicated for each region with respect to a particular mode of behavior, intermode modulation of behavior is also indicated. In summary, the extrinsic connections of the LHAjp and LHAs (so far as we have described them) suggest that these regions have a profoundly integrative role in which they may participate in the orchestrated modulation of elaborate behavioral repertoires. 相似文献

This study identified LTBP-2 as a pleiotropic tumor suppressor in nasopharyngeal carcinoma, which safeguards against critical malignant behaviors of tumor cells. LTBP-2 expression was significantly decreased or lost in up to 100% of NPC cell lines (7/7) and 80% of biopsies (24/30). Promoter hypermethylation was found to be involved in LTBP-2 silencing. Using a tetracycline-regulated inducible expression system, we unveiled functional roles of LTBP-2 in suppressing colony formation, anchorage-independent growth, cell migration, angiogenesis, VEGF secretion, and tumorigenicity. Three-dimensional culture studies suggested the involvement of LTBP-2 in maintenance of tumor cell dormancy in a growth factor favorable microenvironment. 相似文献

To provide an anatomical basis for clinical implant esthetics,we evaluated the morphology of the nasopalatine canal(NPC) and analyzed labial and interproximal bone anatomy at the maxillary anterior region.We sought to investigate the effect of maxillary protrusion and tooth labiolingual inclination on labial bone anatomy in Chinese adults.Three dimensional(3D) images were reconstructed using cone-beam computed tomography(CBCT) images from 80 Chinese subjects and by SimPlant 11.04.The dimensions of the NPC,the thickness and profile of the labial bone,the width and height of the interproximal bone,angle sella-nasion-subspinale(SNA) and angle upper central incisor-nasion,subspinale(U1-NA) were measured.The incisive foramen of the NPC was markedly wider than its nasal foramen.The dimension of its labial bone wall demonstrated an increasing width from the crestal to apical measurements.The labial bone at the maxillary anterior region was rather thin,especially at 3 mm below the cemento-enamel junction(CEJ) and the mid-root level;the profile of the labial bone was more curved at the central incisor,and the interproximal bone became wider and shorter posteriorly.There were significant relationships between maxillary protrusion and labial bone profile,tooth labiolingual inclination and labial bone thickness(P 〈 0.02).To achieve optimal esthetic outcome of implant,bone augmentation is necessary at the maxillary anterior region.For immediate or early placement at the maxillary anterior region,the implant should be located palatally to reduce labial bone resorption and marginal recession;its apex should be angulated palatally to avoid labial perforation at the apical region.To protect the NPC,implants at the central incisor region should be placed away from NPC. 相似文献
