Previous studies in our laboratory have documented that in utero chlordane exposure caused a significant enhancement in the survival of the offspring to influenza A virus infection, and a depressed delayed type hypersensitivity (DTH) response to oxazolone. To correlate these 2 effects, we assayed influenza A virus-specific DTH response, and found that it was significantly decreased in chlordane-treated offspring. Virus-specific T-cell blastogenesis was also assayed in chlordane-treated animals. No significant differences due to the chlordane treatment were found in virus-specific T-cell blastogenesis, suggesting that the DTH depression did not result from a paucity of antigen-reactive T-cells. To determine whether enhanced survival was due, in part, to the effects of chlordane on virus replication, rather than on immunological alteration alone, the kinetics of influenza virus replication in the lungs of chlordane- and vehicle-treated animals were determined. In utero chlordane treatment caused no significant differences in in vivo virus replication. These data suggest that increased survival was due to a decrease in virus-specific DTH and its associated pathology.  相似文献

In a rat model of macrophage-dependent glomerular immune injury induced by administration of antibody against the glomerular basement membrane (anti-GBM), the authors assessed the anti-proteinuric effect of Heme Oxygenase-1 (HO-1) induction. Rats received anti-GBM antibody alone, anti-GBM antibody and treatment with the HO-1 inducer, hemin, or non-immune serum (controls). Urine protein, creatinine, and nitrite/nitrate excretion were measured on days 5, 7, and 14 after administration of the anti-GBM antibody. In hemin-treated animals with anti-GBM antibody-induced immune injury, HO-1 immunolocalized in macrophages infiltrating glomeruli and in tubular epithelial cells. In these animals, proteinuria was decreased. There was also a decrease in blood urea nitrogen (BUN) levels without a change in serum creatinine or systemic blood pressure. The observations establish the anti-proteinuric effect of hemin induction. This effect could be mechanistically linked to blunting of the ability of infiltrating macrophages to cause injury or to changes in tubular handling of filtered protein.  相似文献

Three saponins, oleanolic acid-28- O -ß-D-glucopyranoside (1), chikusetsusaponin V (2), and 3- O -ß-D-glucopyranosyl-oleanolic acid-28- O -ß-D-glucopyranoside (3) were isolated from the roots of Achyranthes bidentata Blume (Amaranthaceae). No activity was shown in the granulocyte phagocytosis test nor in the test of the potentiation of the cytotoxicity of cisplatin in human colon cancer cells. This is the first report of compounds 1, 2 and 3 isolated from Achyranthes species. Furthermore, the NMR data of 2 completed the partially published data.  相似文献

目的观察竹节参的有效成分竹节参总皂苷(TSPJ)对类风湿性关节炎大鼠(RA)抗炎效果,并探讨其抗炎机制。方法建立RA大鼠模型后,灌胃给TSPJ 10d,采用冰醋酸致大鼠腹腔毛细血管通透性增高实验检测TSPJ抗炎效果,测定大鼠巨噬细胞碳清除率、血清IgG、二硝基氟苯诱导的大鼠迟发性超敏反应(DTH),研究TSPJ对类风湿性关节炎模型大鼠抗炎机制。结果与模型组比较,TSPJ能抑制冰醋酸所致RA大鼠腹腔毛细血管通透性的增高,TSPJ对RA大鼠碳清除率无明显影响,能降低RA大鼠血清IgG水平1,00mg/kg TSPJ对DTH反应有抑制作用。结论 TSPJ对RA大鼠有抗炎作用,其机制可能是通过抑制RA大鼠血清IgG水平及DTH反应而实现的。  相似文献

The dissemination of Paracoccidioides brasiliensis cells to the heart, omentum/pancreas, spleen, liver and lungs, assessed by colony forming unit (CFU) counts, the levels of specific antibodies to this fungal agent (by ELISA), and the specific DTH reaction were studied in susceptible (B10.A) and resistant (A/Sn) mice. The animals were infected intraperitoneally with P. brasiliensis yeast cells and were evaluated 2, 4, 12 and 16 weeks later. The most remarkable differences between the two mouse strains were observed 16 weeks after infection, when B10.A mice displayed high numbers of CFU in all examined organs, except the heart, high antibody titres, and depressed DTH response. At this point, A/Sn mice presented low or absent CFU in all organs, low antibody titres and expressive DTH response. The CFU counts were shown to be a reliable parameter to discriminate susceptible from resistant animals. The fungal load in the most affected organs correlated with the antibody titres and was inversely correlated with the intensity of the DTH reaction. The patterns of immune response in this model mimic human paracoccidioidomycosis, in which high specific antibody levels and depressed DTH reactions are found in multifocal and severe forms of the disease.  相似文献

将picryl chloride(PC)所致的迟发型变态反应(PC-DTH)导入肝脏诱发肝损伤,方法是将PC涂于昆明种小鼠腹部致敏,6天后用PC在肝脏攻击,测量血清谷丙转氨酶(sGPT)的活性并进行肝组织病理学检查结果发现,sGPT活性明显上升,肝细胞坏死、门管区粒细胞和淋巴细胞浸润等。在PC致敏前用环磷酰胺处理明显地增强了这种肝损伤。但给正常小鼠肝脏注射PC则不能诱发肝损伤。这些结果表明,该肝损  相似文献

静脉注射BaIb/c小鼠(H-2 ̄d)脾细胞和骨髓细胞,48小时后,腹腔注射环磷酰胺和抗胸腺细胞血清,可诱导B6小鼠(H-2 ̄b)对BaIb/c小鼠皮肤移植的长期耐受(>80天).耐受B6小鼠可正常排斥无关品系CBA/J小鼠(H-2 ̄k)的皮肤,显示了耐受的特异性。上述诱导方案可导致受体小鼠脾细胞对ConA、LPS诱导的增植反应的抑制,但这种全身性免疫抑制可在短期内恢复,而供体特异性的同种异型反应,如MLR和DTH则不能恢复。  相似文献

用荷瘤BALB/c小鼠为模型,研究了冬虫夏草多糖的抑瘤作用,对荷瘤宿主MΦ吞噬功能、外周血淋巴细胞酸性非特异酯酶阳性(ANAE~+)细胞及迟发性变态反应(DTH)的影响。结果表明,冬虫夏草多糖能抑制小鼠S180肿瘤生长,增加外周血ANAE~+细胞百分率,增强DTH及MΦ吞噬活性。  相似文献
