Gastrodia elata is an achlorophyllous and fully mycoheterotrophic orchid which obtains carbon and other nutrients from Armillaria species in its life cycle. Many researchers suggested that plant hormones, as signing molecules, play a central role in the plant–fungi interaction. In the process of Armillaria gallica 012 m cultivation, both exogenous indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) distinctly stimulated the growth of mycelia in solid media. The differential expression genes (DEGs) of A. gallica 012 m with IAA versus blank control (BK) and IBA versus BK were investigated. The results showed that more than 80% of DEGs of the IAA group were coincident with the DEGs of the IBA group, and more than half of upregulated DEGs and most of the downregulated DEGs of the IAA group coincided with those DEGs of the IBA group. Above research implied that A. gallica 012 m could perceive IAA and IBA, and possess similar responses and signaling pathways to IAA and IBA. The overlapping differential genes of the IAA group and IBA group were analyzed by GO term, and the results showed that several DEGs identified were related to biological processes including positive regulation of the biological process and biological process. The downregulated NmrA-like and FKBP_C genes might be benefit to the growth of mycelia. Those results can explain that exiguous IAA and IBA improved the growth of A. gallica to some extent. We speculate that IAA and IBA are signaling molecules, and regulate the expression of growth-related genes of A. gallica 012 m by the same signaling pathway. 相似文献

运用原位DNA末端标记法研究细胞凋亡对乳腺癌预后的影响 总被引:11,自引:0,他引:11
研究临床乳腺癌标本中细胞凋亡的发生情况,评价其在预测后在的意义。方法搜集91例浸润性乳腺癌的石蜡组织切片,运用原位DNA断裂位点的末端标记法,检测细胞凋亡,计算凋亡细胞占肿瘤细胞的百分比,求得凋亡指数。结果本组病例细胞凋亡发生率为91.2%,凋亡指数分为两组:0~0.21和0.28~0.62,凋亡指数高低与腋淋巴结转称相关(P<0.01)。生存率单因素分析中,凋亡指数高的病例,无病生存率(P=0.0095)及总生存率(P=0.0348)均优于凋亡指数低的病例。但Cox模型多因素分析末能指示凋亡指数是一个独立的预后指标。结论在乳腺癌组织中,细胞凋亡是一种自发现象,其发生情况各有不同,研究初步提示了细胞凋亡在乳腺癌预后中的作用,了解了凋亡和腋淋巴结转移的相关性。 相似文献

探讨紫杉醇诱发人乳腺癌细胞凋亡的发生机制。方法应用细胞形态观察、琼脂糖凝胶电泳、流式细胞仪、活细胞视频观察和蛋白印迹免疫法进行检测和观察。结果癌细胞在紫杉醇作用下,细胞分裂阻滞在分裂期的中期,并诱导细胞发生凋亡。凋亡细胞表现为细胞固缩,核染色质凝聚或者断裂。细胞DNA裂解片段呈现典型的“阶梯状”排列的条带。凋亡抑制基因Bcl-2在细胞凋亡过程中呈低表达并发生修饰反应,而凋亡诱导基因Bax先呈高表达然后表达降低。结论紫杉醇诱发的人乳癌细胞的凋亡与细胞分裂期阻滞密切相关,Bcl-2和Bax在紫杉醇诱发的人乳癌细胞凋亡中可能起着重要的调控作用 相似文献

目的以mdm2基因为指标,研究与肝细胞癌侵袭性有关的分子机理。方法应用RT-PCR方法,研究mdm2基因在34例癌组织和19例癌周肝组织中的表达及其与肝细胞癌侵袭性的关系。结果mdm2基因表达值(x±s)癌组织高于癌周肝组织(50.18%±5.24%对27.70%±7.43%,P<0.05),侵袭性肝癌高于非侵袭性肝癌(59.09%±7.28%对37.87%±6.37%,P<0.05),但不同大小肝癌及不同包膜状况肝癌,其mdm2基因表达值差异无显著意义。结论mdm2基因表达与肝细胞癌的侵袭性有关。 相似文献

Recent studies have demonstrated that following estrogen ablation, estrogen responsive breast cancer cells undergo apoptosis. In addition, estrogen receptor (ER) expression has been strongly correlated with the expression of the bcl-2 gene product, p26Bcl-2 protein, which is known to inhibit apoptosis. In the present studies, we investigated whether estrogen affects the intracellular levels of p26Bcl-2 and thereby modulates taxol-induced apoptosis of estrogen responsive human breast cancer MCF-7 cells. Transfer of MCF-7 cells to a culture-medium without estrogens reduced their intracellular p26Bcl-2 levels by 50%. Inclusion of 0.1 M estradiol in the medium produced approximately a four-fold increase in p26Bcl-2, but not p29Bcl-xL or p21Bax levels; the expression of the c-myc and mdr-1 genes remained unchanged. Estradiol-induced four-fold increase in the ratio of the p26Bcl-2 to p21Bax levels caused a significant decline in the lethal, kilobase size DNA fragments of apoptosis, which had resulted when MCF-7 cells were cultured in a medium without estrogen. In addition, in MCF-7 cells, estradiol-induced increase in the intracellular p26Bcl-2 to p21Bax ratios was associated with a significant reduction in the large-sized DNA fragmentation induced by treatment with taxol. The increased ratios also protected MCF-7 cells against taxol-mediated cytotoxicity as assessed by the MTT assay. These results suggest that by modulating p26Bcl-2 levels, estrogens may affect the antitumor activity of taxol and potentially of other anti-breast cancer drugs against estrogen responsive human breast cancer cells. 相似文献

A novel ER-like mRNA containing a 69 nucleotideinsertion precisely between exon 5 and 6 sequenceswas previously identified in human breast cancer biopsysamples. Data are presented which suggest that the69 nucleotide sequence is normally present in intron5 of the human estrogen receptor gene. Theregion corresponding to and surrounding this 69 nucleotidesequence was cloned and the nucleotide sequence determined.Cloning and sequencing of the corresponding region ingenomic DNA isolated from a breast tumor expressingthe 69 nucleotide inserted ER mRNA, revealed anAG point mutation immediately 3 to the69 nucleotide sequence. This point mutation resulted inthe generation of a consensus splice donor site.A consensus splice acceptor site sequence is normallypresent immediately 5 to the 69 nucleotide sequence.These data are consistent with the 69 nucleotidesequence being recognized as an exon by thesplicing machinery, and resulting in processing of amature ER mRNA containing the 69 nucleotide insert. 相似文献
