Improved Orange and Red Ca2+ Indicators and Photophysical Considerations for Optogenetic Applications

HMGA1在葡萄膜黑色素瘤中的表达及对细胞增殖和侵袭的影响

目的：探讨高迁移率族蛋白A1(HMGA1)蛋白在葡萄膜黑色素瘤(UM)组织中的表达,以及抑制该基因表达对细胞增殖和侵袭的影响。

方法：选取2014-02/2019-08在我院接受手术治疗的UM患者53例53眼,同期留取因外伤摘除眼球的正常葡萄膜组织34例34眼。采用免疫组织化学法检测组织中HMGA1蛋白表达。将体外培养的人UM细胞系M23分为HMGA1下调组、阴性对照组和空白组,分别转染HMGA1干扰序列、阴性对照序列和不作任何处理,采用实时荧光定量PCR术检测HMGA1 mRNA表达,CCK-8法检测细胞增殖能力,Transwell法检测细胞迁移和侵袭能力。

结果：UM组织中HMGA1蛋白阳性表达率为77%,高于正常葡萄膜组织中的29%(P<0.001); 与未发生巩膜浸润、未累及睫状体和未发生眼外生长相比,HMGA1蛋白在发生巩膜浸润、累及睫状体和发生眼外生长的组织中阳性表达率升高(均P<0.05)。与阴性对照组和空白组相比,HMGA1 mRNA在HMGA1下调组细胞中相对表达量降低,且HMGA1下调组细胞培养24、48、72、96h时吸光度OD值降低,迁移细胞数和侵袭细胞数均明显减少(均P<0.05)。

结论：UM组织中HMGA1蛋白阳性表达率升高,下调M23细胞中HMGA1表达可减少细胞增殖,抑制细胞迁移和侵袭。     

关于应用健康相关生活质量作为肝血管瘤手术指征的初步探讨

肝血管瘤是临床上最常见的肝脏良性肿瘤,对于肝血管瘤的治疗目前临床上尚存在争议,特别是手术治疗,对于手术指征的把握,不同学者之间意见未能统一,本文对其手术指征作一综述,提出健康相关生活质量作为肝血管瘤手术指征,探讨其在肝血管瘤临床治疗中的应用.     

Evidence-based practice: A quality indicator analysis of peer-tutoring in adapted physical education

The purpose of the research was to conduct a quality indicator analysis of studies investigating peer-tutoring for students with a disability in adapted physical education. An electronic search was conducted among English journals published from 1960 to November 2012. Databases included ERIC, PsycINFO, and SPORTDiscus. Fifteen research studies employing group-experimental (Gersten et al., 2005) or single-subject designs (Horner et al., 2005) met inclusion criteria. Each study was assessed for the presence and clarity of quality indicators. Group designs met an average of 62.5% essential and 69% desirable indicators. An average of 80% of indicators was present for single-subject designs. Results suggest claims of peer-tutoring being an evidence-based practice are premature. Recommendations for clarifying and applying the quality indicators are offered.     

乳腺癌患者PICC护理质量敏感性指标的建立与实施

目的探讨乳腺癌患者PICC护理质量敏感性指标的建立与实施效果。方法将实施PICC护理质量敏感性指标前(2014年)行PICC置管的203例患者作为对照组,实施后(2015年)231例患者作为研究组。对照组实施常规护理,研究组采用自行研制的PICC护理质量敏感性指标监测管理,比较实施前后PICC相关并发症发生率、PICC专科护理质量及患者满意率。结果实施前后患者PICC相关并发症发生率、PICC维护护理质量评分比较,差异有统计学意义(P0.05,P0.01);实施后患者对护理工作满意率明显提高。结论建立并实施PICC专科护理质量敏感性指标监测与控制,能有效降低PICC相关并发症发生率,提高PICC专科护理质量,提高患者满意率。     

Using leading and lagging indicators to select safe contractors at the prequalification stage of construction projects

The contractor-selection decision at the prequalification stage is critical to the project success. An insufficient prediction of contractors’ safety capacities using only lagging indicators may hinder the continuous improvement of safety performance in the construction industry. This research enhanced construction management and practices by proposing a comprehensive safe contractor selection model which integrated both leading and lagging indicators. First, a set of leading and lagging safety indicators were identified based on literature review and expert opinions. Then, the grey correlation analysis (GCA) was utilized to assign weights to individual indicators. We found that management commitment, safety training and education, safety risk management, and safety rules and procedures were four most influential factors to the safety performance of contractors. In addition, the fuzzy technique of ordering preference by similarity to ideal solution (Fuzzy TOPSIS) was used to condense individual indicators and create a composite safety performance indicator (c-SPI). Finally, the feasibility of the decision support tool for safe contractor selection was verified using a real-case railway construction project.
