目的观察中药复方菊藤胶囊对慢性应激性高血压大鼠血压、血浆血管紧张素Ⅱ(AngⅡ)及血液流变学的影响。方法大鼠随机分为6组(每组7只):应激+蒸馏水组(model),应激+菊藤胶囊高剂量组(JT—H),应激+菊藤胶囊中剂量组(JT—M),应激+菊藤胶囊低剂量组(JT—L),应激+卡托普利组(captopril),正常对照组(control);除正常对照组外,其余5组均采用低频低压交流电间断电击法。电击大鼠足底28d,制作应激性高血压模型,同时各组均加入不同的干预。后应用经尾动脉测量血压和心率,酶联免疫吸附法(ELISA)检测大鼠血浆血管紧张素Ⅱ,血液流变学和细胞流变学的各项指标。观察比较菊藤胶囊不同剂量组、卡托普利组对应激致大鼠血压及相关指标的影响。结果与正常对照组相比,模型组大鼠血压和血浆血管紧张素Ⅱ含量明显升高(P〈0.01)。全血黏度、血浆黏度、纤维蛋白原及红细胞聚集指数均明显升高(P〈0.05),红细胞变形指数无变化。与模型组相比,菊藤胶囊高、中组均能抑制应激大鼠的血压及血浆血管紧张素Ⅱ含量的升高(P〈0.01),菊藤胶囊高剂量能明显改善应激大鼠的血液流变性(P〈0.05),低剂量组应激大鼠的血液流变学无明显改变(P〉0.05)。结论菊藤胶囊能够降低应激性高血压大鼠的血压,其机理可能是通过降低血浆中血管紧张素Ⅱ的含量和改善血液流变和细胞流变性实现的。  相似文献

Kappa opioid receptors stimulation with U50,488 is known to modulate behaviors during the early postnatal period, but the specific neuroanatomical locus of many of these effects is unexplored. In the present study, we infused U50,488 into the midbrain periaqueductal gray (PAG) and investigated the effects of this drug on behavior and heart rate of 1-, 2-, and 3-week-old rats. U50,488 increased activity most potently in 1- and 2-week-old subjects. Ultrasonic vocalization (USV) production was increased in 1-week-old subjects, but not in 2- or 3-week-old pups. Heart rate changes were similarly seen in younger aged subjects. At 1 week, U50,488 decreased heart rate, but at 2 weeks it increased heart rate. There was no effect of this drug on heart rate at 3 weeks. At 1 week, USVs were more potently elicited from dorsal than lateral PAG infusion sites. No other site-specific effects within the PAG were seen. The age-related decline in behavioral effects elicited by U50,488 is consistent with other published reports, and to the extent that kappa receptor activity mediates infant separation responses, implicates the PAG as a modulator of those responses.  相似文献

为了确定可吞服式电子胶囊在人体内的位置,基于矩形永磁体空间磁场解析式,设计了定位系统,提出了相应的定位算法,并进行了实验。系统中两个三轴磁传感器封装在胶囊内,由胶囊和永磁体的空间位姿关系模型,建立了与传感器输出和胶囊位姿变量相关的一组非线性方程。对方程进行了数值求解,并和期望值进行了比较,结果发现该方法是切实可行的。  相似文献

IntroductionHepatocellular carcinoma (HCC) is one of the most common cancers worldwide. Despite the therapeutic advances in HCC in the past few decades, the mortality rate of HCC is still high. Hepatitis C (HCV) infection is one of the major etiological risk factors of HCCs. However, the underlying mechanisms of HCV-induced hepatocarcinogenesis remain largely unclear.Material and methodsOur study represented the comprehensive analysis of differentially expressed lncRNAs in HCV-positive HCC for the first time by analyzing the public dataset GSE17856. Co-expression network and gene ontology (GO) analysis revealed the functions of those differentially expressed lncRNAs.ResultsWe identified 256 upregulated lncRNAs and 198 downregulated lncRNAs in HCV- positive HCC compared to the normal liver tissues. Co-expression network and GO analysis showed that these lncRNAs were involved in regulating metabolism, energy pathways, proliferation and the immune response. Seven lncRNAs (LOC341056, CCT6P1, PTTG3P, LOC643387, LOC100133920, C3P1 and C22orf45) were identified as key lncRNAs and co-expressed with more than 100 differentially expressed genes (DEGs) in HCV-related HCC. Kaplan-Meier analysis showed that higher expression levels of LOC643387, PTTG3P, LOC341056, CCT6P1 and lower expression levels of C3P1 and C22orf45 were associated with shorter survival time in the TCGA dataset.ConclusionsWe believe that this study can provide novel potential therapeutic and prognostic biomarkers for HCV-positive HCC.  相似文献

The aim of this study was to explore and compare the magnitude and time-course of the shift in the angle-force curves obtained from maximal voluntary contractions of the elbow flexors, both before and 4 consecutive days after eccentric and isometric exercise. The maximal isometric force of the elbow flexors of fourteen young male volunteers was measured at five different elbow angles between 50° and 160°. Subjects were then divided into two groups: the eccentric group (ECC, n=7) and the isometric group (ISO, n=7). Subjects in the ECC group performed 50 maximal voluntary eccentric contractions of the elbow flexors on an isokinetic dynamometer (30°.s–1), while subjects in the ISO group performed 50 maximal voluntary isometric muscle contractions with the elbow flexors at a lengthened position. Following the ECC and ISO exercise protocols, maximal isometric force at the five angles, muscle soreness, and the relaxed (RANG) and flexed (FANG) elbow angles were measured at 24 h intervals for 4 days. All results were presented as the mean and standard error, and a quadratic curve was used to model the maximal isometric force data obtained at the five elbow angles. This approach not only allowed us to mathematically describe the angle-force curves and estimate the peak force and optimum angle for peak force generation, but also enabled us to statistically compare the shift of the angle-force curves between and within groups. A large and persistent shift of the angle-force curve towards longer muscle lengths was observed 1 day after eccentric exercise (P<0.01). This resulted in a ~16° shift of the optimum angle for force generation, which remained unchanged for the whole observation period. A smaller but also persistent shift of the angle-force curve was seen after isometric exercise at long muscle length (P<0.05; shift in optimum angle ~5°). ECC exercise caused more muscle damage than ISO exercise, as indicated by the greater changes in RANG and ratings of muscle soreness (P<0.05). It was suggested that the shift in the angle-force curve was proportional to the degree of muscle damage and may be explained by the presence of overstretched sarcomeres that increased in series compliance of the muscle.  相似文献

Summary A long lasting inhibition (> 8 s) of the soleus Hoffmann reflex (H-reflex) was evoked by a preceding soleus H-reflex, by a brief voluntary ankle flexor or extensor muscle contraction or by a tap applied to the Achilles tendon. The time course of this long lasting inhibition was similar in all these cases, suggesting that the same spinal mechanism is involved. Furthermore, it was shown that the post-activation depression may interfere with the determination of inhibitory or facilitatory effects on the H-reflex. It is stressed that when the onset of inhibitory or facilitatory effects on the soleus H-reflex is to be determined in relation to start of an ankle movement, either very long stimulus intervals (> 8 s) must be used, or the onset must be determined in relation to a reference value of the soleus H-reflex, which may be influenced by the long lasting inhibitory effect, but not yet by the succeeding muscle contraction.  相似文献

目的:研究长链非编码RNA HOTAIR调控糖皮质激素受体的表达对急性淋巴细胞白血病细胞增殖及凋亡的作用。方法:采用RT-qPCR法检测人正常骨髓基质细胞系HS-5及急性淋巴细胞白血病细胞系MOLT-4、CCRF-CEM和CEM-C1中HOTAIR和糖皮质激素受体的表达。用si RNA沉默CEM-C1细胞中HOTAIR的表达,CCK-8法检测si-HOTAIR对CEM-C1细胞活力的影响; Brd U法检测si-HOTAIR对CEM-C1细胞增殖的影响以及对地塞米松抑制CEM-C1细胞增殖的增效作用; Hoechst 33342染色法和caspase 3/7活性检测法研究si-HOTAIR对CEM-C1细胞凋亡的影响;并采用Western blot法检测糖皮质激素受体的蛋白表达水平。结果:人急性淋巴细胞白血病细胞系MOLT-4、CCRF-CEM和CEM-C1中HOTAIR的表达显著高于人正常骨髓基质HS-5细胞(P 0. 01)。在CEM-C1细胞中干扰HOTAIR表达后,细胞活力降低,细胞增殖被抑制并发生凋亡,地塞米松抑制CEM-C1增殖的作用被增强,糖皮质激素受体表达上调(P 0. 01)。结论:长链非编码RNA HOTAIR增强急性淋巴细胞白血病的活力,促进其增殖并抑制其凋亡,该作用可能与其抑制糖皮质激素受体的表达有关。  相似文献
