目的:构建丙型肝炎病毒(HCV)NS5A蛋白原核表达载体,获得高产量的纯化目的蛋白,并制备其多克隆抗体。方法:应用聚合酶链反应技术,PCR扩增获得NS5A基因,将NS5A基因插入至原核表达载体pET-32a(+)中,转化大肠埃希菌DH5α提取质粒,验证正确后,转化大肠埃希菌BL21中,以IPTG诱导,获得NS5A融合蛋白的可诱导性表达.通过SDS—PAGE电泳、Western blot免疫印迹分析证实融合蛋白表达的特异性。利用Ni^+亲和柱对表达蛋白进行纯化及柱上复性。纯化蛋白免疫新西兰兔,利用Western blot和ELISA法对多克隆抗体进行特异性和效价检测。结果:NS5A融合蛋白表达成功,成功获得了融合蛋白及兔抗NS5A多克隆抗体。结论:成功表达NS5A基因融合蛋白.获得高特异性、兔抗NS5A多克隆抗体,为研究NS5A基因的生物学功能提供了重要制剂。 相似文献

We studied 4 patients with progressive myoclonus epilepsy (Unverricht-Lundborg disease; ULD). Somatosensory evoked fields (SEFs), auditory evoked fields (AEFs), and spontaneous activity over the somatomotor cortex were recorded with a 24-channel SQUID gradiometer. All patients had “giant” 20–45 msec median nerve SEFs at the first somatomotor cortex, with 2–6 times larger amplitudes than the healthy control subjects. Later deflections were not similarly enhanced. The dependence of SEF amplitudes on interstimulus interval (0.2–4 sec) and on successive ulnar-median nerve stimulation (stimulus interval 40 msec) was comparable to that in controls. Cortical AEFs were attenuated and delayed. In 3 patients, the spontaneous activity consisted of 6–8 Hz mu rhythm, which originated within 2 cm from the sources of SEFs and was abolished by clenching of the contralateral fist. Control subjects had major spectral peaks around 10 and 20 Hz. The SEF amplitudes and the strength of the 6–10 Hz mu correlated strongly, suggesting that some components of evoked and spontaneous activity obtain contributions from overlapping neuronal populations. The results imply that ULD is associated with thalamo-cortical hyperreactivity in the sensorimotor but not in the auditory system. 相似文献

- 1.1. Prolongation of action potentials upon the addition of isoproterenol, forskolin, isobutylmethyl-xanthine (IBMX) and dibutyril cAMP (dbcAMP) under Ca-free EGTA condition was examined in isolated myocardial preparations from neonatal and adult rats, whose action potential configuration greatly differ.
- 2.2. The prolongation of the action potential was previously suggested to be produced by persistent sodium influx through calcium channel due to the lack of calcium-mediated inactivation of calcium channels under such experimental condition.
- 3.3. Preparations used were papillary muscles and free walls of the right and left ventricles from neonatal and adult rats.
- 4.4. In adult preparations, the prolongation produced by isoproterenol, forskolin and IBMX in the right free wall was smaller than those in the other three regions, while no regional difference was observed with dbcAMP.
- 5.5. The degree of prolongation by all of the four drugs were smaller in the neonate than in the adult. No regional difference was observed with any of the drugs in the neonate.
- 6.6. Our present results suggest that contribution of calcium-mediated inactivation of calcium channels to the repolarization of rat myocardium may increase postnatally to produce the developmental shortening of its action potential. Also, regional difference in the cAMP related mechanisms may appear postnatally.

ObjectivePart C Early Intervention (EI) services have been shown to reduce autism symptoms and promote healthy development among young children. However, EI participation remains low, particularly among children from structurally marginalized communities. We investigated whether family navigation (FN) improved EI initiation following positive primary care screening for autism compared to conventional care management (CCM).MethodsWe conducted a randomized clinical trial among 339 families of children (ages 15–27 months) who screened as having an increased likelihood for autism at 11 urban primary care sites in 3 cities. Families were randomized to FN or CCM. Families in the FN arm received community-based outreach from a navigator trained to support families to overcome structural barriers to autism evaluation and services. EI service records were obtained from state or local agencies. The primary outcome of this study, EI service participation, was measured as the number of days from randomization to the first EI appointment.ResultsEI service records were available for 271 children; 156 (57.6%) children were not engaged with EI at study enrollment. Children were followed for 100 days after diagnostic ascertainment or until age 3, when Part C EI eligibility ends; 65 (89%, 21 censored) children in the FN arm and 50 (79%, 13 censored) children in the CCM arm were newly engaged in EI. In Cox proportional hazards regression, families receiving FN were approximately 54% more likely to engage EI than those receiving CCM (1.54 (95% confidence interval: 1.09–2.19), P = .02).ConclusionsFN improved the likelihood of EI participation among urban families from marginalized communities. 相似文献

目的观察实验性大鼠脑出血后白细胞介素2和白细胞介素8的表达及其与脑出血后脑含水量变化的关系。方法通过采用未抗凝新鲜自体股动脉血注入大鼠尾状核建立脑出血动物模型,用放射免疫法测定脑出血后6 h1、2 h、24 h、3天6、天和10天等时间点白细胞介素2和白细胞介素8的表达,用干重—湿重法测定脑出血后脑含水量的变化,并与白细胞介素2和白细胞介素8进行相关分析。结果与对照组比较,血肿周围组织中白细胞介素2水平在脑出血后6 h和1天显著降低(P<0.01),3天时轻度降低(P>0.05),6天时达高峰(P<0.01),10天时接近正常(P>0.05)。白细胞介素8水平在脑出血后1天和3天时显著升高(P<0.05),6天时降至正常(P>0.05)。血肿周围组织含水量在脑出血后6 h轻度增加(P>0.05),1天时增加显著(P<0.01),3天时达高峰(P<0.01),10天时基本恢复正常。白细胞介素8的变化与脑含水量的变化趋势一致(R2=0.861)。结论白细胞介素2有利于抑制脑水肿的形成;白细胞介素8可能参与实验性脑出血后血肿周围水肿的发生、发展。 相似文献
