背景:骨碎补促进骨损伤愈合疗效确切,但具体作用机制尚不清楚。目的:观察骨碎补总黄酮对兔骨髓间充质干细胞增殖、分化的影响。方法:冲洗兔股骨和胫骨骨髓腔获取骨髓,采用密度梯度离心法联合贴壁培养法体外纯化扩增兔骨髓间充质干细胞;以四甲基偶氮唑蓝法测定不同浓度骨碎总黄酮对兔骨髓间充质干细胞增殖的影响;经骨碎补总黄酮体外诱导后,行扫描电镜形态学观察,碱性磷酸酶染色、茜素红染色和VonKossa银染色了解细胞钙化情况。结果与结论:兔骨髓间充质干细胞可以通过密度梯度离心法联合贴壁培养法扩增和纯化;含骨碎补总黄酮血清培养后,四甲基偶氮唑蓝测定结果显示,骨碎补总黄酮浓度为10-6mmol/L时可明显促进兔骨髓间充质干细胞增殖。经骨碎补总黄酮诱导液诱导,扫描电镜下可见成骨细胞样形态和钙结节形成;细胞碱性磷酸酶染色、钙结节茜素红染色和VonKossa银染色均呈阳性,提示骨碎补总黄酮可促进兔骨髓间充质干细胞增殖和分化。  相似文献

PurposeThis study aimed to investigate the resting-state brain network related to visuospatial working memory (VSWM) in patients with right temporal lobe epilepsy (rTLE). The functional mechanism underlying the cognitive impairment in VSWM was also determined.MethodFifteen patients with rTLE and 16 healthy controls matched for age, gender, and handedness underwent a 6-min resting-state functional MRI session and a neuropsychological test using VSWM_Nback. The VSWM-related brain network at rest was extracted using multiple independent component analysis; the spatial distribution and the functional connectivity (FC) parameters of the cerebral network were compared between groups. Behavioral data were subsequently correlated with the mean Z-value in voxels showing significant FC difference during intergroup comparison.ResultsThe distribution of the VSWM-related resting-state network (RSN) in the group with rTLE was virtually consistent with that in the healthy controls. The distribution involved the dorsolateral prefrontal lobe and parietal lobe in the right hemisphere and the partial inferior parietal lobe and posterior lobe of the cerebellum in the left hemisphere (p < 0.05, AlphaSim corrected). Between-group differences suggest that the group with rTLE had a decreased FC within the right superior frontal lobe (BA8), right middle frontal lobe, and right ventromedial prefrontal lobe compared with the controls (p < 0.05, AlphaSim corrected). The regions of increased FC in rTLE were localized within the right superior frontal lobe (BA11), right superior parietal lobe, and left posterior lobe of the cerebellum (p < 0.05, AlphaSim corrected). Moreover, patients with rTLE performed worse than controls in the VSWM_Nback test, and there were negative correlations between ACCmeanRT (2-back) and the mean Z-value in the voxels showing decreased or increased FC in rTLE (p < 0.05).ConclusionThe results suggest that the alteration of the VSWM-related RSN might underpin the VSWM impairment in patients with rTLE and possibly implies a functional compensation by enlarging the FC within the ipsilateral cerebral network.  相似文献

目的探讨应用 Ilizarov 技术胫骨横向骨搬移术治疗合并全身性炎症反应综合征(systemic inflammatory response syndrome,SIRS)的重度糖尿病足(Wagner 3~5 级)的临床疗效。方法2014 年 8 月—2017 年 12 月,采用 Ilizarov 技术胫骨横向骨搬移术治疗合并 SIRS 的重度糖尿病足患者 33 例。其中男 27 例,女 6 例;年龄 34~79 岁,平均 60.6 岁。均患 2 型糖尿病,糖尿病病程 1~28 年,平均 10 年。糖尿病足病程 1~12 个月,平均 2.7 个月。根据 Wagner 分级,3 级 8 例,4 级 23 例,5 级 2 例。术后观察患足创面愈合情况, 并计算保肢率。评估体温、心率、呼吸频率、白细胞计数、红细胞沉降率和 C 反应蛋白浓度变化。测量足背皮肤温度;行疼痛视觉模拟评分(VAS)评价患足疼痛改善情况。结果术后 33 例患者均获随访,随访时间 3~30 个月,平均 14.1 个月。足部溃疡全部愈合,愈合时间 3~12 个月,平均 5.3 个月;保肢率 100%。术后体温、心率、呼吸频率、白细胞计数、红细胞沉降率和 C 反应蛋白浓度均较术前显著下降(P<0.05)。术后 1 个月患足皮温为(32.64±2.17)℃,较术前的(31.28±1.99)℃ 显著改善(t=0.05,P=0.00);但与健侧皮温(32.46±2.10)℃ 比较差异无统计学意义(t=2.04,P=0.41)。术后 1 个月 VAS 评分为(2.4±0.7)分,较术前的(4.3±0.8)分显著改善(t=3.10,P=0.00)。 结论Ilizarov 技术胫骨横向骨搬移术能有效治疗合并 SIRS 的重度糖尿病足,可促进患足溃疡愈合,避免截肢。  相似文献

Introduction/objectives

To seek significant features of systemic lupus erythematosus (SLE) by utilizing bioinformatics analysis.

Method

Liquid chromatography-tandem mass spectrometry (LC–MS/MS) was used to quantify lysine crotonylation (Kcr) and lysine 2-hydroxyisobutyrylation (Khib) in peripheral blood mononuclear cells (PBMCs) of systemic lupus erythematosus (SLE) patients and normal controls.

Results

Seventy-six differentially modified proteins (DMPs) dually modified by Kcr and Khib were identified between SLE patients and healthy people. GO enrichment analysis prompted significant enrichment of seventy-six DMPs in MHC class II protein complex binding and leukocyte migration. KEGG pathways were enriched in antigen processing and presentation pathway and leukocyte transendothelial migration pathway. Six DMPs (CLTC, HSPA1B, HSPA8, HSP90AB1, HSPD1, and PDIA3) were identified in antigen processing and presentation pathway, of which HSPA8 was the core protein. Significant changes of Kcr and Khib in HSPA8 may increase ATP hydrolysis and promote antigen binding to MHC II molecule. In leukocyte transendothelial migration pathway, 7 DMPs (ACTN1, ACTN4, EZR, MSN, RAC1, RHOA, and VCL) were identified. MSN was the protein with the most modification sites in this pathway. In amino terminal ferm region of MSN, Kcr and Khib expression change may lead to the adhesion between leukocytes and endothelial cells, which was an important step of leukocyte migration.

Conclusion

Kcr and Khib may promote the antigen presentation and jointly regulate the tissue damage mediated by leukocyte migration in SLE patients, which may play key roles in the pathogenesis of SLE probably.
