ObjectiveAnnexin A5 (Anxa5) is associated with the progression of some cancers, while its role and regulation mechanism in tumor lymphatic metastasis is rarely reported. This study aims to investigate the influence of Anxa5 knockdown on the malignant behaviours of murine hepatocarcinoma Hca-F cell line with high lymph node metastatic (LNM) potential and the underlying regulation mechanism.MethodsRNA interfering was performed to silence Anxa5 in Hca-F. Monoclonal shRNA-Anxa5- Hca-F cells were obtained via G418 screening by limited dilution method. Quantitative real-time RT-PCR (qRT-PCR) and Western blotting (WB) were applied to measure Anxa5 expression levels. CCK-8, Boyden transwell-chamber and in situ LN adhesion assays were performed to explore the effects of Anxa5 on the proliferation, migration, invasion and adhesion capacities of Hca-F. WB and qRT-PCR were used to detect the level changes of key molecules in corresponding signal pathways.ResultsWe obtained two monoclonal shRNA-Anxa5-transfected Hca-F cell lines with stable knockdowns of Anxa5. Anxa5 knockdown resulted in significantly reduced proliferation, migration, invasion and in situ LN adhesion potentials of Hca-F in proportion to its knockdown extent. Anxa5 downregulation enhanced E-cadherin levels in Hca-F. Moreover, Anxa5 affected Hca-F behaviours specifically via ERK2/p-ERK2/c-Jun/p-c-Jun(Ser73) instead of p38MAPK/c-Jun, Jnk/c-Jun and AKT/c-Jun pathways.ConclusionsAnxa5 mediates the in vitro malignant behaviours of murine hepatocarcinoma Hca-F cells via ERK2/c-Jun/p-c-Jun(Ser73) and ERK2/E-cadherin pathways. It is an important molecule in metastasis (especially LNM) and a potential therapeutic target for hepatocarcinoma.  相似文献

The aim of this study is to estimate whether the occupational exposure to low dose anesthetic gases could cause alterations of blood parameters in health care workers. 119 exposed subjects and 184 not exposed controls were included in the study. Each worker underwent the complete blood count test (CBC), proteinaemia, leukocyte count, serum lipids, liver and kidney blood markers.The liver blood markers show statistically significant differences in health care workers compared with controls (p < 0.05), a statistically significant decrease in neutrophils and an increase of lymphocytes in health care workers compared with controls (p < 0.05). The prevalence of values outside the range for GPT, GGT, total bilirubin, lymphocytes and neutrophils was statistically significant in health care workers compared with controls (p < 0.05). The results suggest that occupational exposure to low dose anesthetic gases could influence some haematochemical hepatic and hematopoietic parameters in exposed health care workers.  相似文献

Plant growth-promoting bacteria are a promising alternative to improve agricultural sustainability. Gluconacetobacter diazotrophicus is an osmotolerant bacterium able to colonize several plant species, including sugarcane, coffee, and rice. Despite its biotechnological potential, the mechanisms controlling such osmotolerance remain unclear. The present study investigated the key mechanisms of resistance to osmotic stress in G. diazotrophicus. The molecular pathways regulated by the stress were investigated by comparative proteomics, and proteins essential for resistance were identified by knock-out mutagenesis. Proteomics analysis led to identify regulatory pathways for osmotic adjustment, de novo saturated fatty acids biosynthesis, and uptake of nutrients. The mutagenesis analysis showed that the lack of AccC protein, an essential component of de novo fatty acid biosynthesis, severely affected G. diazotrophicus resistance to osmotic stress. Additionally, knock-out mutants for nutrients uptake (Δtbdr and ΔoprB) and compatible solutes synthesis (ΔmtlK and ΔotsA) became more sensitive to osmotic stress. Together, our results identified specific genes and mechanisms regulated by osmotic stress in an osmotolerant bacterium, shedding light on the essential role of cell envelope and extracytoplasmic proteins for osmotolerance.  相似文献

目的检测冠心病(CHD)患者血小板内皮细胞黏附分子1(CD31)及血浆纤维蛋白原(FIB)水平,分析其与CHD严重程度的相关性并探讨其临床意义。方法入选研究对象97例,分为稳定型心绞痛(SAP)组(21例),急性冠脉综合征(ACS)组(43例)和正常对照组(33例)。采用流式细胞术检测血小板CD31,凝固法测定血浆FIB的表达水平。结果 ACS组血小板表面活性标志物CD31[(93.85±14.84)%],血浆FIB水平[(3.66±0.91)g/L]均明显高于对照组[(78.69±27.09)%,(3.06±0.62)g/L]和SAP组[(82.80±28.00)%,(3.14±0.56)g/L(均为P<0.05)];而对照组与SAP组CD31、FIB表达水平差异无统计学意义(均为P>0.05)。相关分析显示CD31与FIB呈显著正相关(r=0.337,P<0.01)。结论检测CHD患者CD31、FIB水平,能较好地反映CHD严重程度,对临床有重要的应用价值。  相似文献

We evaluated the effects green and mate teas on oxidative and DNA damages in rats exposed to ultraviolet radiation. Were utilized 70 adult male Wistar rats that received daily oral or topic green or mate tea treatment during exposed to radiation by seven days. After, animals were killed by decapitation. Thiobarbituric acid-reactive species levels, protein oxidative damage were evaluated in skin and DNA damage in blood. Our results show that the rats exposed to ultraviolet radiation presented DNA damage in blood and increased protein carbonylation and lipid peroxidation in skin. Oral and topic treatment with green tea and mate tea prevented lipid peroxidation, both treatments with mate tea also prevented DNA damage. However, only topic treatment with green tea and mate tea prevented increases in protein carbonylation. Our findings contribute to elucidate the beneficial effects of green tea and mate tea, here in demonstrated by the antioxidant and antigenotoxic properties presented by these teas.  相似文献

Aspartame is an artificial sweetner added to many low-calorie foods. The safety of aspartame remains controversial even though there are many studies on its risks. In this study, to understand the physiological effects of trace amounts of artificial sweetners on cells, the effects of aspartame on apoptosis were investigated using a PC12 cell system. In addition, the mechanism of apoptosis induced by aspartame in PC12 cells and effects on apoptotic factors such as cytochrome c, apoptosis-inducing factor, and caspase family proteins were studied by Western blotting and RT-PCR.Aspartame-induced apoptosis in PC12 cells in a dose-dependent manner. In addition, aspartame exposure increased the expressions of caspases 8 and 9, and cytochrome c. These results indicate that aspartame induces apoptosis mainly via mitochondrial pathway involved in apoptosis due to oxigen toxicity.  相似文献

The present study was aimed to investigate the effect of the veterinary food additive copper sulphate (CuSO4) on the eco-toxicological responses of earthworms Eisenia fetida (E. fetida). The following biomarkers were measured: catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione-S-transferase (GST) activities. Gene expression analyses such as metallothionein (MT) and heat shock protein 70 (Hsp70) were also examined. A time-dependent increase of CAT activity was found at 400 mg/kg and SOD activity at 200 and 400 mg/kg. The highest expression of Hsp70 (4.4-fold) was observed at day 15 at 400 mg/kg. Our results indicated that the measured antioxidant enzymes (except GST) had the ability to provide antioxidant defenses against the stressor; and compared to expression of MT, expression of Hsp70 could be more reliable molecular tools with predictive possibilities to monitor the eco-toxicity of stressors such as feed additive CuSO4.  相似文献

目的 探讨iNOS在SAH后迟发性血管痉挛中的作用。方法 SAH模型采用大鼠枕大池二次注血法。HE染色测量SAH不同时期基底动脉腔的周长,比较痉挛程度;免疫组化和RT-PCR方法观察在痉挛最严重时基底动脉血管壁中iNOS的表达。结果 第7天时血管痉挛最为明显。免疫组化染色及RT—PCR分析均可见第7天时基底动脉血管壁中有iNOS的表达而在非SAH组为阴性。结论 iNOS在SAH后脑血管痉挛发生过程中起重要作用。  相似文献
