目的 克隆丹参碱性螺旋-环-螺旋(bHLH)类转录因子SmbHLH93,并初步探究该基因的功能.方法 采用PCR和RT-PCR技术分别从DNA和cDNA水平克隆得到了SmbHLH93基因,进一步通过BD Walking获得了该基因的5'启动子区.生物信息学分析SmbHLH93蛋白的理化性质、结构特点和系统进化关系.结合启动子区分析软件和qPCR,研究SmbHLH93在丹参不同部位和环境诱导下的表达模式.结果 获得SmbHLH93基因序列954 bp和5'启动子区1 583 bp,其中开放阅读框(ORF)657 bp,有3个内含子和4个外显子,编码218个氨基酸,含有bHLH和ACT_UUR-ACR-like结构域,无跨膜区域,可能定位于细胞核.表达模式分析结果显示该基因的表达量在根中最高,茎中最低,且随着花的形成逐渐降低.光和低温诱导SmbHLH93的高表达,而水杨酸(SA)抑制其表达.结论 克隆得到丹参bHLH类转录因子新成员SmbHLH93,初步预测其参与了丹参花的发育过程和丹参次生代谢途径的调控.  相似文献

目的 对川藏香茶菜Rabdosia pseudo-irrorata C. Y. Wu的化学成分进行研究。方法 采用硅胶柱色谱法进行分离、纯化,通过理化性质和现代波谱技术对化合物进行结构鉴定。结果 从川藏香茶菜全草95%乙醇提取物的醋酸乙酯萃取部分分离得到7个化合物,分别鉴定为川藏香茶菜壬素(1)、川藏香茶菜庚素(2)、川藏香茶菜己素(3)、川藏香茶菜乙素(4)、β-谷甾醇(5)、熊果酸(6)和β-胡萝卜苷(7)。结论 化合物1为新化合物,化合物57均为首次从该植物中分离得到。  相似文献

目的獉獉:探讨感觉寻求、负性情绪、风险认知三个因素对戒毒人员复吸倾向的影响。方法獉獉:应用复吸倾向问卷、风险认知状况问卷、正性负性情绪量表、感觉寻求量表简明版等对兰州强制戒毒所和美沙酮门诊的共293名戒毒人员进行测查。结果獉獉:风险认知、感觉寻求、负性情绪均对复吸倾向有显著影响,风险认知对感觉寻求、负性情绪具有一定的中介作用,通过风险认知的中介作用能够影响感觉寻求、负性情绪对复吸倾向的作用。  相似文献

目的从秦岭濒危药用桃儿七(Sinopodophyllum hexandrum)植株内分离出产鬼臼毒素(podophyllotoxin)的内生真菌,研究其发酵液的抑瘤活性。方法采用组织分离法分离、纯化桃儿七植株根、茎部位的内生真菌,逐一鉴定其代谢产物,筛选出能够产鬼臼毒素的内生真菌。采用动物体内抑瘤实验研究内生真菌发酵液对小鼠S180肉瘤的抑制作用,并对荷瘤小鼠眼眶取血进行白细胞及淋巴细胞计数。结果分离出一株产鬼臼毒素的内生真菌T8,鉴定为Cephalosporium sp.。T8发酵液实验动物组的肿瘤质量低于模型组,大于环磷酰胺组(P<0.05),且其抑制率低于环磷酰胺(P<0.05)。T8发酵液能够促进白细胞和淋巴细胞的增殖。结论 T8发酵液具有一定的抑制肿瘤生长的作用,经减毒处理后可以作为肿瘤治疗的辅助药物。  相似文献

Background: Since the outbreak of COVID-19 caused adults suffer from mental disorders, it would be an essential for psychologist to help individuals overcome mental disorders. Objective: This study aim to explore whether the intervention of mindfulness is an efficient method to enhance adolescents’ emotional intelligence (EI) and psychological capital (PC) to provide a theoretical basis and future directions for both targeted crisis intervention and psychological trauma recovery plans. Design: This research was designed as randomized controlled trial and total of 798 students were evaluated statistically. Methods: We used paired-sample t-tests and repeated measures ANOVA to compare every factor defined above by time and group. Then, we used Mplus to build LGCM to examine the trajectory of changes in EI and PC in the experimental group. Results and Conclusions: (1) The EI and PC scores had significantly different time points in the experimental group but not in the control group. (2) EI and PC tended to increase with intervention, and there were significant individual differences in the initial level and development speed. (3) Individuals’ EI could promote the growth of PC. This research indicated that mindfulness should be given increasing consideration to enhance mental health during the worldwide outbreak of COVID-19.  相似文献

Optical measuring technologies coupled with machine learning algorithms can be used to build a home-made sensor system. We built such a sensor system using a smartphone and a diffraction grating sheet. Diffraction images were captured under white light illumination and converted into a data matrix for data analysis. In this paper we present a systematic evaluation of this sensor system on the task of differentiating organic apples from conventional ones. We used the sensor system to measure 150 organic and conventional apples as rainbow images. We processed the rainbow images using computer vision techniques, built machine learning and chemometrics models, and used the resultant models to classify testing samples. Moreover, a comparative study was conducted where the same set of apples were scanned by a commercial spectrometer resulting in spectral data of the apple samples and classification was undertaken using partial least squares discriminant analysis (PLS-DA). Experimental results show that state of the art machine learning algorithms such as support vector machine (SVM) and locally weighted partial least squares classifier (LW-PLSC) are effective in handling low-quality image data with classification accuracies of 93 − 100%. These results suggest that the sensor system is convenient and low-cost, and provides a fast, effective, non-destructive and viable solution for in-line food authentication.  相似文献

Propiconazole (PCZ) is an ergosterol biosynthesis inhibiting fungicide. Carvacrol (CAR) is a monoterpenoid phenol that has various beneficial health effects. The current research was designed to study the impact of PCZ on the behavior of rats and its ability to induce DNA damage in neurons as well as to clarify the ameliorative effect of CAR against these toxic impacts. Sixty Sprague-Dawley rats were randomly and equally divided into 4 experimental groups and treated daily by oral gavage for 2 months as follows: Group 1 (control); group 2 treated with PCZ (75 mg/kg); group 3 treated with CAR (50 mg/kg) and group 4 treated with both PCZ and CAR. Behavioral tests demonstrated that exposure to PCZ had a deleterious effect on psychological, motor and cognitive neural functions. Additionally, antioxidant enzyme activities, SOD and GSH-Px, were declined in brain tissue following exposure to PCZ. Moreover, comet assay revealed a high percent of DNA damage in the brain of rats exposed to PCZ. On the other hand, CAR administration ameliorated the harmful effects induced by PCZ through a protective mechanism that involved the improvement of neural functions and attenuation of oxidative stress and DNA damage.  相似文献

??OBJECTIVE To observe the effects of lappaconitine sulfate on proliferation, cycle and apoptosis of human cerical neoplasm Hela cells and its possible mechanism.METHODS Cell Counting Kit-8 (CCK-8) was adopted to observe the inhibitory effects of lappaconitine sulfate on the proliferation of Hela cells and calculated its half maximal inhibitory concentration (IC50). The technology of flow cytometry, propidium iodide (PI) single stained method and Annexin-V-FITC/PI double stained method were employed to measure the cell cycle and apoptosis of Hela cells after lappaconitine sulfate treatment.RESULTS Lappaconitine sulfate could inhibit the proliferation of human cerical neoplasm Hela cells in vitro, and the inhibition presented obvious dose-effect relationship (r=0.990 5, P??0.01). The IC50 of lappaconitine sulfate on Hela cells was 0.421 mg??mL-1. After dealt with lappaconitine sulfate, Hela cells indicated remarkable cell cycle arrest at G0/G1 phase, and the apoptotic ratio was gradually increased with the concentration of lappaconitine sulfate.CONCLUSION Lappaconitine sulfate could inhibit the growth of human cerical neoplasm Hela cells, change the distribution of cell cycles and induce their apoptosis.  相似文献
