目的:通过测定中药制剂参芪扶正注射液中Cu、Zn、Mn、Se、Mo、Fe、Cr、Ni、V、Sn、Pb、Cd、Hg、As等14种微量元素的含量,探讨该中药注射液中元素的分布是否存在某种规律性.方法:微波消解法制备供试品,电感耦合等离子体-质谱(ICP-MS)法测定.结果:参芪扶正注射液中Cu、Fe、Zn、Cr、Se的相对含量较高,有毒元素Pb、Cd、Hg、As中As含量接近美国FAD药品与功能性食品要求标准,Pb、Cd、Hg含量均明显低于最高限量.结论:测定结果为探讨中药制剂中微量元素与抗肿瘤作用的关系提供有用的数据.  相似文献

Ginger (Zingiber officinale Roscoe, Zingiberacae) is a medicinal plant that has been widely used in Chinese, Ayurvedic and Tibb-Unani herbal medicines all over the world, since antiquity, for a wide array of unrelated ailments that include arthritis, rheumatism, sprains, muscular aches, pains, sore throats, cramps, constipation, indigestion, vomiting, hypertension, dementia, fever, infectious diseases and helminthiasis. Currently, there is a renewed interest in ginger, and several scientific investigations aimed at isolation and identification of active constituents of ginger, scientific verification of its pharmacological actions and of its constituents, and verification of the basis of the use of ginger in some of several diseases and conditions. This article aims at reviewing the most salient recent reports on these investigations. The main pharmacological actions of ginger and compounds isolated therefrom include immuno-modulatory, anti-tumorigenic, anti-inflammatory, anti-apoptotic, anti-hyperglycemic, anti-lipidemic and anti-emetic actions. Ginger is a strong anti-oxidant substance and may either mitigate or prevent generation of free radicals. It is considered a safe herbal medicine with only few and insignificant adverse/side effects. More studies are required in animals and humans on the kinetics of ginger and its constituents and on the effects of their consumption over a long period of time.  相似文献

The skeletal system, which contains bones, joints, tendons, ligaments and other elements, plays a wide variety of roles in body shaping, support and movement, protection of internal organs, production of blood cells and regulation of calcium and phosphate metabolism. The prevalence of skeletal diseases and disorders, such as osteoporosis and bone fracture, osteoarthritis, rheumatoid arthritis, and intervertebral disc degeneration, increases with age, causing pain and loss of mobility and creating a huge social and economic burden globally. Focal adhesions (FAs) are macromolecular assemblies that are composed of the extracellular matrix (ECM), integrins, intracellular cytoskeleton and other proteins, including kindlin, talin, vinculin, paxillin, pinch, Src, focal adhesion kinase (FAK) and integrin-linked protein kinase (ILK) and other proteins. FA acts as a mechanical linkage connecting the ECM and cytoskeleton and plays a key role in mediating cell–environment communications and modulates important processes, such as cell attachment, spreading, migration, differentiation and mechanotransduction, in different cells in skeletal system by impacting distinct outside-in and inside-out signaling pathways. This review aims to integrate the up-to-date knowledge of the roles of FA proteins in the health and disease of skeletal system and focuses on the specific molecular mechanisms and underlying therapeutic targets for skeletal diseases.  相似文献

本文利用real-time PCR方法对不同产地甘草的3-羟基-3-甲基戊二酰CoA还原酶 (3-hydroxy-3-methylglutaryl-CoA reductase, HMGR)、鲨稀合成酶1 (squalene synthetase 1, SQS1)、β-香树脂醇合成酶 (β-amyrin synthase, β-AS) 基因的拷贝数进行了研究, 发现不同产地的HMGR基因存在1~3个拷贝数变异, SQS1基因存在1~2个拷贝数变异, 未发现β-AS基因拷贝数变异。甘草HMGR、SQS1、β-AS基因拷贝数存在5种自然组合类型: A型 (2+1+1)、B型 (1+1+1)、C型 (3+2+1)、D型 (2+2+1) 和E型 (3+1+1), 其中内蒙古杭锦旗甘草存在A、B两种类型, A与B的比例为1∶1.3; 内蒙古赤峰甘草也存在A、B两种类型, 但A与B比例为3∶1; 宁夏盐池甘草存在A、B、C、D 4种类型, A/B/C/D比例为1∶5.1∶1∶2, A/B比例为1∶5.1; 甘肃民勤甘草存在A、B、E 3种类型, A/B/E比例为4.1∶2.1∶1, A/B比例为2∶1。本研究证明中药功能基因基因组拷贝数变异 (copy number variations, CNVs) 与产地具有相关性, 可能是道地药材形成的机制之一。  相似文献

In addition to metabolic differences, the anatomical, physiological, and biochemical differences in the gastrointestinal (G.I.) tract of the human and common laboratory animals can cause significant variation in drug absorption from the oral route. Among the physiological factors, pH, bile, pancreatic juice, and mucus and fluid volume and content can modify dissolution rates, solubility, transit times, and membrane transport of drug molecules. The microbial content of the G.I. tract can significantly affect the reductive metabolism and enterohepatic circulation of drugs and colonic delivery of formulations. The transit time of dosage forms can be significantly different between species due to different dimensions and propulsive activities of the G.I. tract. The lipid/protein composition of the enterocyte membrane along the G.I. tract can alter binding and passive, active, and carrier-mediated transport of drugs. The location and number of Peyer's patches can also be important in the absorption of large molecules and particulate matter. While small animals, rats, mice, guinea pigs, and rabbits, are most suitable for determining the mechanism of drug absorption and bioavailability values from powder or solution formulations, larger animals, dogs, pigs, and monkeys, are used to assess absorption from formulations. The understanding of physiological, anatomical, and biochemical differences between the G.I. tracts of different animal species can lead to the selection of the correct animal model to mimic the bioavailability of compounds in the human. This article reviews the anatomical, physiological, and biochemical differences between the G.I. tracts of humans and commonly used laboratory animals.  相似文献

氧化苦参碱可防治高糖、高脂引起的心血管并发症,脂肪肝并发症,糖尿病视网膜病变,糖尿病肾病以及肺、骨并发症。氧化苦参碱防治这些并发症的药理机制可能归因于:降血糖、调血脂及抗氧化作用,从源头上减少了氧化型低密度脂蛋白和糖基化终末产物的生成;抗氧化、抗炎和细胞保护作用可保护肝脏,促进肝糖原合成,在脂质转运、氧化和合成3个环节改善脂质代谢紊乱,产生降血糖、调血脂作用,也可保护受累的心血管系统、肾脏、肝脏、视网膜、肺和骨等其他器官,减轻胰岛素抵抗,提高胰岛素敏感性。因此氧化苦参碱有望开发为治疗代谢综合征的药物。  相似文献

摘要:阿维菌素是阿维链霉菌发酵产生的十六元大环内酯类抗生素,主结构与米尔贝霉素相似,用于农业害虫防治和动物 的杀虫杀螨,还具有抗病毒、抗肿瘤、抗菌、消炎和醒酒等功效。它通过抑制RNA复制酶,阻止病毒蛋白进入细胞核以及封闭 剌突蛋白等方式,阻止病毒的复制感染和传播。已经产生抗药性的分枝杆菌和金黄色葡球菌也能被阿维菌素抑制。阿维菌素世 界上年需求量为5000吨左右,基本由中国企业生产。本文回顾了阿维菌素在菌种技术、发酵技术、提取技术、制剂技术发展和 组合生物学手段在本产业中的应用,建议通过菌种筛选技术、胞壁通透性调整、途径工程指导的基因改造、发酵过程的代谢流 控制、气升式发酵罐应用及细胞融合育种来提高发酵水平,将阿维链霉菌与米贝霉素链霉菌细胞融合育种。  相似文献

The paper reviews the early toxicologic and pharmacologic studies carried out by the author and his associates from 1943 to 1947, which were largely responsible for launching DDT as an agent for the control of typhus, malaria, yellow fever, and related vector-borne diseases. After reviewing recent studies conducted at the University of Miami, which dealt with organochlorine pesticides in human tissues, the tumorigenicity of aldrin, dieldrin and endrin (rat), six-generation mouse and three-generation dog reproduction studies, synergism of DDT and aldrin (dog), and the fate of DDT and aldrin during a period of severe starvation (rat), it is pointed out that it is primarily the overuse and misuse of DDT in pest control that have caused the pollution in our ecology. It is emphasized that the requirements for pest control differ the world over and that it must therefore be left to the national regulatory agencies to legislate the safe use of DDT and related pesticides. It is recommended that future human and animal studies with DDT and its derivatives give consideration to: (a) the balance and metabolism of the various hormones, (b) reproduction (estrus, libido, mammary development, milk production, (c) hepatic microsomal enzyme activities, (d) cancer prevention and cancer production, (e) excessive body weight changes induced by disease, unbalanced diet or starvation, and (f) the effects of DDT and its derivatives when absorbed in combination with other related and even unrelated compounds.Presented at the joint meeting of the Scandinavian and German Pharmacological Societies, Copenhagen, Denmark, July 20–23, 1971.  相似文献

目的 探讨心肌损伤标志物[血清肌钙蛋白Ⅰ( cTnI)、肌红蛋白(Mb)、肌酸激酶同工酶(CKMB)]单项及联合检测在急性心肌梗死( AMI)患者早期诊断中的意义.方法 检测AMI患者(AMI组)发病后2、4、8h血液中Mb、CK-MB、cTnI浓度,与非AMI心胸疾病(UA)患者(UA组)进行比较,并分析Mb、CK-MB、cTnI单项及多项联合指标对诊断AMI的敏感性和特异性.结果 AMI组在2、4、8h各项指标与UA组比较,经统计学分析,差异具有显著性(P< 0.05).发病2h,Mb敏感性和特异性与其他单项指标和联合指标相比,经统计学分析,差异具有显著性(P<0.05).发病4h,Mb、CK-MB、cTnI的敏感性为87.5%、53.13%、43.75%,特异性为70.97%、74.19%、80.65%,经统计学分析,差异具有显著性(P<0.05);联合指标Mb+cTnI、Mb+CK-MB和cTnI+CK-MB敏感性和特异性较高.发病8h,Mb、cTnI和CK-MB敏感性均达到100%,特异性分别为87.09%、90.32%、96.77%;联合指标以Mb+cTnI+CK-MB敏感性及特异性最高.结论 对AMI患者早期进行cTnI、Mb和CK-MB的联合检测,既可尽早确诊,又可避免误诊,对提高AMI诊断的敏感性和特异性,提高确诊的速度和确诊率,具有重要的临床意义.  相似文献

1. The disposition of diflunisal (DF) was investigated in both bile-exteriorized and intact rats given 10 and 100 mg/kg doses intravenously (i.v.). 2. In addition to the phenolic glucuronide (DPG) and acyl glucuronide (DAG) conjugates, the sulfate conjugate (DS) was found to be a major metabolite. The glucuronides were excreted preferentially in bile, whereas DS was excreted almost exclusively in urine. 3. In bile-exteriorized animals, recoveries of DPG, DAG and DS in bile were 12.2%, 23.8%, 0.4%, respectively, and in urine, 10.3%, 5.6% and 15.2%, respectively, at the 10 mg/kg dose; and in bile, 11.3%, 41.6% and 1.0% respectively, and urine 2.9%, 1.1% and 17.0%, respectively, at the 100 mg/kg dose. 4. Total plasma clearance of DF and formation clearance of DF to DPG were reduced at the higher dose, suggesting saturation of this glucuronidation pathway. Formation clearances of DF to DAG and DS were little affected by the dose change. 5. Considerable enterohepatic recirculation of DF was apparent from the prolongation of DF and its conjugates in plasma of rats with an intact bile flow into the gut. The net metabolic effect of such cycling was enhancement of overall DS formation, from 15.6% and 18.0% of the 10 and 100 mg/kg doses, respectively, in bile-exteriorized rats to 28.5% and 42.1% of the doses respectively, in the intact animals.  相似文献
