冷冻大鼠肾上腺自体移植的超微结构观察

应用电子显微镜分别对经冷冻保存的和新鲜自体移植的大鼠肾上腺组织进行观察。结果表明,47只大鼠新鲜肾上腺组织自体移植成活40只(85.1%)冷冻保存的47只大鼠肾上腺自体移植成活27只(57.7%)。未行移植单纯冷冻保存的肾上腺组织,复温后多数细胞的形态无改变,部分细胞核内出现不规则腔隙,核质分布不均匀。未冷冻的肾上腺自体移植后,5d细胞开始再生,25～30d细胞成熟;冷冻的肾上腺移植后,与前者相比再生略缓慢,再生细胞数量少。47只未冷冻的肾上腺组织移植后,有5只髓质细胞成活。提示冷冻保存法对肾上腺组织有一定的保存作用。     

大鼠同种异体肾移植实验模型的研究现状

动物肾移植模型被广泛用于器官保存及免疫等相关研究。目前已有多种动物模型用于实验研究，如猪、狗、兔、大鼠及小鼠等。其中以大鼠模型最为常用。其优点有：1．动物来源多，经济；2．饲养简单；3．移植手术的无菌要求较大动物要小；4．应用近交系大鼠可在术后不用免疫抑制剂，自体移植器官能够长期存活；5．血管和输尿管的直径满足吻合的要求。     

同种异体甲状旁腺移植的进展

甲状旁腺(PTG)移植分自体移植、异种异体移植和同种异体移植三类。后者临床应用价值最大,是当今研究的重点。其适应症有:1.特发性PTG机能低下,2.医源性PTG机能低下,3.先天性PTG发育不全(Di George's综合症)。现在国外多做PTG组织片移植,国内主要采用胎儿带血管的PTG移植。 一、甲状旁腺组织移植 1.游离细胞移植:先把PTG制成细胞悬液,然后注入两面都有多个0.45μm微孔的扩散盒内,再把扩散盒埋入肱二头肌,盒内PTG细胞所产生的激素通过多孔滤膜排到盒外发挥治疗作用。Swan用狗作实验,结果不能对此作出评价。蔡全忠用此法移植2例,1例症状缓解2年,另1例缓解半年。随着时间的延长,膜表面的微孔易被吞噬细胞、白细胞和纤维素所堵塞,从而失去作用,现已很少有人采用。     

超速冷冻法冷冻保存大鼠卵巢组织的实验研究

目的探讨超速冷冻法对SD大鼠卵巢组织的冷冻保存功效。方法采用液氮直投法冷冻大鼠卵巢组织。计数冻融后卵巢组织内形态正常始基卵泡数量。将卵巢组织自体移植，术后35天处死动物，测定血清雌二醇（E2）水平。结果新鲜和冻融后的大鼠卵巢组织中形态正常始基卵泡数量无统计学差异（P〉0.05）。移植术后35天，两移植组血清E2水平无统计学差异（P〉0.05）。结论液氮直投超速冷冻法可以有效的保存卵巢组织，对大鼠卵巢组织学和功能的影响较小。     

犬自体单肺移植的实验研究

目的:通过同种自体犬单肺移植,掌握供肺的灌注,切取,保存,和修剪方法以及单肺移植的手术方法.方法:11例犬进行了一次犬自体单肺移植动物实验,其中8例自体左单肺移植.3例自体右单肺移植,吻合顺序先支气管后肺动脉,最后心房袖,移植后于不同时点暂阻断对侧肺动脉并检测植入肺的功能参数.结果:所有移植肺通气和换气功能均正常,移植后犬恢复自主呼吸.9例在24小时内死亡,2例在术中死亡.结论:冷缺血再灌注损伤是早期死亡的主要原因.     

同种异体移植子宫内膜异位症大鼠模型的建立

目的 建立Wistar大鼠同种异体子宫内膜异位症模型.方法 采用外科诱导法对24只Wistar雌性大鼠行异体子宫组织移植手术,30只行自体子宫组织移植手术.结果 Wistar大鼠造模12周后,异体移植成模率83.3%,与自体移植成模率76.7%比较差异有统计学意义(P<0.05);异体、自体模型异位内膜上皮细胞功能活跃,有类似正位子宫内膜的周期变化和生理特征;且造模后大鼠体液免疫反应敏感性与假手术对照组比较,C3、C4、IgG升高,IgM降低,差异有统计学意义,IgA没有明显变化;异体移植模型IgG比自体移植模型高,差异有统计学意义.结论 应用Wistar大鼠进行同种异体移植手术可以成功的建立子宫内膜异位症模型.     

供移植用狗肾保存72小时获得初步成功

临床上,移植用肾以简单低温法保存,其安全时间仅24～36小时。为了延长保存时间,国外研究狗肾保存48～72小时后有移植成功的报道,但不稳定。我院器官移植研究所自制一种新的细胞内液型保存液(WMO 1号液)作保存狗肾72小时后自体移植的研究。本年度已供24次。除因手术操作失误和保存温度不合规定标准者不计外,正式有效实验为18次。移植后肾     

自体脂肪干细胞治疗心肌梗死的实验研究

目的探讨自体脂肪干细胞治疗心肌梗死的作用机制。方法用9μmol/L的5-氮胞苷(5-aza)培养基诱导自体脂肪干细胞24h,2、3周后免疫细胞化学法染色鉴定心肌特异肌钙蛋白T(cTnT)和肌纤蛋白(desmin)的表达。冷冻法制作大鼠心肌梗死模型。1周后,经尾静脉移植自体脂肪干细胞。TTC染色测梗死体积,插管测左心室血流动力学变化。结果移植自体脂肪干细胞后,梗死体积减小(P<0.01),移植3w后,实验组左心室收缩压及左心室等容收缩期/舒张期内压最大上升或下降速率较对照组升高(P<0.05～0.01),左心室舒张末压较对照组减小(P<0.05)。结论经5-aza诱导的大鼠自体脂肪干细胞能减小心肌梗死体积及改善大鼠心肌梗死后的心脏功能。     

羊膜为基底的小片状自体结膜移植治疗碱球结膜烧伤

目的:探讨保存羊膜移植联合小片状自体结膜移植治疗碱烧伤的可行性。方法:大耳白兔3O只。右眼(实验组)和左眼(对照组)上方球结膜碱烧伤24h后分别行以羊膜为基底的小片状自体结膜移植和单纯小片状自体结膜移植。结果:实验组植片成活率显著高于对照组(p<0.05)。实验组术后第3周结膜外观恢复正常,而对照组则用了5周。结论:大面积结膜碱烧伤早期应用羊膜可改善自体结膜植片的基底,有利于提高植片成活率,达到结膜结构与功能重建的目的。     

移植自体嗅黏膜对脊髓损伤大鼠脊髓内MMP-2、9表达的影响

目的探讨自体嗅黏膜移植干预脊髓损伤(spinal cord injury,SCI)大鼠脊髓组织内基质金属蛋白酶-2、9(MMP-2、9)的表达与动物神经功能恢复的关系。方法制作SD大鼠半横断脊髓损伤模型,按随机数字表法分为模型对照组和自体嗅黏膜组,每组20只(同时取5只大鼠设为正常对照组)。自体嗅黏膜组在损伤处植入自体嗅黏膜,移植后1、3、7、14d(每时间点5只大鼠)采用BBB评分评价神经功能、免疫组织化学法检测脊髓内MMP-2、9表达。结果 SCI后自体嗅黏膜组BBB评分高于模型对照组(P<0.05)。各时间点SCI大鼠脊髓内MMP-2、9的表达显著升高(正常对照组大鼠未见MMP-2、9的表达)。伤后同一时间点自体嗅黏膜组脊髓内的阳性细胞数少于模型对照组(P<0.05)。结论自体嗅黏膜移植可明显促进神经运动功能恢复情况;此种作用可能与其能够抑制MMP-2、9过度表达有关。     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
