口服炎痛静溶液体内药动学研究

本文用荧光分光光度法测定了6名健康受试者单剂量口服炎痛静溶液50mg后血药浓度。用NONLIN程序计算机对数据进行药物动力学模型嵌合,求得炎痛静体内动力学参数为:吸收速率常数Ka1.3331h~(-1),消除速率常数K0.0706h~(-1),表观分布容积V74.18L。     

曲尼司特在人体内的药代动力学研究

用RP-HPLC法测定曲尼司特血药浓度,对12名健康志愿者口服曲尼司特200和400mg(每个剂量组6人)的药代动力学进行了研究.结果表明,志愿者口服曲尼司特,药-时曲线符合一级吸收-房室模型,各药代动力学参数在二个剂量组无显著性差异,但Cmax和AUC与剂量成比例,提示该药在400mg剂量以下时在人体内呈线性消除动力学.     

小剂量阿司匹林缓释微丸药代动力学及生物等效性研究

用荧光分光光度法测定了口服小剂量阿司匹林缓释微丸在人体内的血药浓度,10名健康志愿受试者体内药代动力学结果显示,缓释微丸的相对生物利用度为Ka=0.4869 h~(-1),K=0.1097 h~(-1),t_(1/2)=6.69 h,C_(mx)=4.47μg/ml,t_(max)=4.52 h。方差分析表明阿司匹林缓释微丸与普通片具有生物等效性。     

布洛芬缓释微丸的制备及其药物动力学研究

采用膜控法制备布洛芬缓释微丸,应用转篮法(50r/min,37±1℃)比较了缓释微丸和普通片剂的体外药物溶出速率,采用HPLC法测定了制剂的血药浓度,并研究了制剂在人体内的药物动力学。缓释微丸的体外药物溶出符合一级动力学过程(K_r=0.4505h~(-1));体内动力学过程符合一级吸收和一级消除的单室模型,k_a=0.27h~(-1),K=0.27h~(-1),V_d=7.54L,t_(max)=3.97h,C_(max)=19.52μg/ml。多剂量给药血药浓度测定及计算表明:缓释微丸具有维持有效血药浓度(8.5μg/ml)时间长,血药浓度波动范围小的特点。此外,缓释微丸的体内外数据具有显著的相关性(p<0.01)。     

曲尼司特在4种动物的药物动力学参数和体重的相关性

曲尼司特(tranilast,利喘贝),化学名为N-3′,4′-二甲氧基肉桂酰基)-邻氨基苯甲酸,是一种预防支气管哮喘的药物,作用机理类似于色甘酸钠。作者给小鼠、大鼠、兔和犬分别ⅳ曲尼司特5mg/kg,血药浓度作二房室模型曲线拟合,t_(1/2α)均短于10min,t_(1/2β)分     

高效液相色谱法测定曲尼司特滴眼液中曲尼司特及其有关物质的含量

目的:建立高效液相色谱法测定曲尼司特滴眼液中曲尼司特及其有关物质的含量。方法:以汉邦Lichrospher C18(200 mm×4.6 mm,5μm)为色谱柱,以甲醇-0.02 mol.L-1磷酸二氢钾溶液(用磷酸调节pH值至4.2)(60∶40)为流动相,UV检测波长333 nm,流速1.0 ml.min-1。结果:曲尼司特在25~200 mg.L-l范围内具有良好线性关系(r=0.999 9),平均加样回收率为98.86%,RSD为1.10%;3批样品的标示百分含量(%)分别为99.57、100.17和99.87,有关物质的百分含量分别为0.52、0.56和0.55。结论:本方法准确、灵敏、重现性好,可用于曲尼司特滴眼液及其有关物质的含量测定。     

健康志愿者单剂量毛冬青甲素的药代动力学研究及胶囊制剂的生物利用度

用高效液相色谱法测定了6名健康志愿者(4男2女)静注(20mg)和口服胶囊(120mg)毛冬青甲素后的血药浓度,用二房室药代动力学模型,分析了毛冬青甲素在体内的动力学过程.6名志愿者口服和静注的平均消除相常数和半衰期分别为0.0397 h~(-1),0.0460h~(-1)和17.7h,22.5h,体内总廓清率为4.6L/h。也用统计距分析法,求得口服毛冬青甲素的两个动力学参数MRT(体内平均滞留时间)和VRT(体内平均滞留时间的方差)分别为17.3h和1000 h~2,求得毛冬青甲素胶囊的绝对生物利用度为0.39。     

头孢拉定的儿童临床药动学及给药方案

本文报道6例呼吸道感染的儿童口服头孢拉定的临床药动学。用双波长分光光度法测定尿药浓度,计算药动学参数为t_1/21.18±0.25h,K_e0.61±0.16h~(-1),K_a1.99±1.12h~(-1)。观察和计算结果表明,头孢拉定对儿童口服给药,以每次9-15mg/kg,1次/6-8h的给药方案较好。     

曲尼司特对阿霉素肾病大鼠肾脏保护作用的实验研究

目的研究曲尼司特对阿霉素肾病肾小球硬化大鼠的肾脏保护作用,并探讨其可能的机制。方法雄性SD大鼠24只,随机分为正常对照组、模型组、曲尼司特组和安博维组,采用单侧肾切除加尾静脉注射阿霉素5mg/kg的方法建立阿霉素肾病模型。观察各组大鼠24h尿蛋白定量、血尿素氮(BUN)、血肌酐(Scr)及肾脏病理的变化。应用免疫组化方法测定肾组织转化生长因子β1(TGF-β1)和金属蛋白酶组织抑制因子-1(TIMP-1)的表达,应用原位杂交方法测定肾组织α-平滑肌肌动蛋白(α-SMA)mRNA的表达。结果曲尼司特能减少阿霉素肾病大鼠的尿蛋白,延缓Scr上升;减轻基质增生和肾小球硬化;与模型组比较,曲尼司特下调阿霉素肾病大鼠肾组织中TGF-β1〔(0.6927±0.169)vs(0.4540±0.168)〕、TIMP-1〔(0.1615±0.077)vs(0.0869±0.041)〕、α-SMA mRNA〔(0.2693±0.082)vs(0.1407±0.045)〕的表达。结论曲尼司特可能通过下调阿霉素肾病大鼠肾组织中TGF-β1、TIMP-1的表达以及抑制肾脏细胞表型转分化,调节细胞外基质的生成与降解,从而减轻肾脏病理损害,发挥其肾脏保护作用。     

乙酰水杨酸肠溶微丸的研制

采用薄膜包衣法制备乙酰水杨酸肠溶微丸,以转篮法(100 r/min,37 0.5℃)测定了体外溶出速率。实验结果表明:本制剂2 h在人工胃液中的释放量不超过15%,转入人工肠液后在40min内完全释放(Kr=2.975 h~(-1))。体内试验选择了六名健康受试者(平行法单剂量给药900 mg)服用乙酰水杨酸普通片及肠溶微丸,测定了二种制剂在体内的经时过程。药动学参数的求算表明:普通片:Ka=0.309 h~(-1),K=0.309 h~(-1),Cls=1.598 L/h,AUC=563.30 h·μg/ml;肠溶微丸:Ka=0.415h~(-1),K=0.134 h~(-1),Cls=1.235 L/h,AUC=728.66 h·μg/ml。     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
