抗AIB1-C单克隆抗体的制备及初步应用

目的:建立分泌抗AIB1单克隆抗体的杂交瘤细胞株,并对分泌的单克隆抗体进行特异性鉴定。方法:以纯化GST-AIB1-C蛋白为免疫原,免疫BAL b/c小鼠,取其脾细胞与小鼠骨髓瘤细胞SP2/0融合,杂交瘤细胞采用间接ELISA筛选。结果:成功筛选出4株稳定分泌抗AIB1-C单克隆抗体的杂交瘤细胞株,经Western-blot和免疫细胞化学法鉴定,这些McAb特异性高,亲和力强。结论:成功研制出抗AIB1-C单克隆抗体,为双抗体夹心法ELISA试剂盒的研制奠定基础。     

抗人ζ珠蛋白链不同表位单克隆抗体的制备和鉴定

目的制备和鉴定抗ζ珠蛋白链单克隆抗体。方法用纯化的重组ζ珠蛋白链免疫Balb/c小鼠,取其脾细胞与NS-1细胞融合制备杂交瘤,经重组的ζ珠蛋白链筛选和3次克隆化,从制备的腹水中纯化单克隆抗体。结果和结论获得3株杂交瘤细胞1A12、3H9及4D11,其分泌单抗的亚型分别为IgG2a、IgG1和IgG1;双抗体夹心ELISA结果证明3株单抗均可结合重组的及东南亚(--SEA)缺失型地中海贫血基因携带者血中的ζ珠蛋白链,且其中1A12和3H9识别的位点为不同表位。这三株抗体将为筛选地中海贫血基因携带者的免疫检测提供有力工具。

鼠抗人TCR/CD3单克隆抗体的制备及其初步应用

目的应用B淋巴细胞杂交瘤技术,制备鼠抗人CD3单克隆抗体,并用于T淋巴细胞亚类检测。方法以本室纯化的CD3蛋白免疫Balb/c小鼠,取免疫小鼠脾细胞与SP2/0骨髓瘤细胞融合,成功地获得1株抗人CD3单克隆抗体。经间接ELISA测定,杂交瘤细胞培养腹水的抗体效价。结合SDS-PAGE实验考察单克隆抗体对CD3促T淋巴细胞增殖活性的作用。结果杂交瘤细胞培养腹水的抗体含量为20 mg/mL,效价为10-7,并对T淋巴细胞有促增殖活性,检测正常人外周血阳性T细胞百分率为（73±7）%。结论制备的抗人CD3单克隆抗体具备较高的纯度和活性,可用于T细胞亚类的检测。     

抗蛋白S单克隆抗体的制备

目的 制备抗蛋白S(protein S)单克隆抗体.方法 用微量蛋白S抗原对Balb/c小鼠脾内包埋免疫,取小鼠脾细胞与SP2/O细胞融合,融合后对杂交瘤细胞进行筛选克隆化培养,建立了10株稳定分泌抗蛋白s单克隆抗体的杂交瘤细胞株.结景对其中的一株2E3进行研究,其分泌的抗体为IgG1亚类.腹水效价1:106阳性,采用饱和硫酸铵盐析法纯化腹水,Western blot测定显示单克隆抗体2E3可特异性地识别分子量为69,000的抗原分子.此杂交瘤细胞株体外传代培养6个月以上,ELISA检测抗体滴度无明显降低.结论脾内包埋法成功制备抗蛋白S单克隆抗体,使蛋白S的临床深入研究和应用成为可能.     

抗炭疽PA15单克隆抗体的制备及免疫学检测方法的建立

目的:制备抗炭疽保护性抗原15(protective antigen,PA15)单克隆抗体,初步建立双抗体夹心ELISA检测炭疽感染者血清中的保护性抗原?方法:以纯化的PA63蛋白为免疫原免疫小鼠,利用杂交瘤技术制备单克隆抗体,SDS-PAGE电泳检测抗体纯度,间接ELISA?Western blot?免疫沉淀(IP)和蛋白质谱分析单抗特异性,并建立双抗体夹心ELISA检测方法?结果:制备了2株抗PA15单克隆抗体,命名为3D7和8E9?SDS-PAGE电泳可见抗体的重链和轻链,间接ELISA?Western blot发现单抗3D7和8E9可与PA15?PA63特异性结合,IP和蛋白质谱分析发现单抗3D7可与PA83特异性结合,双抗体夹心ELISA方法检测炭疽感染血清中保护性抗原的最低检出浓度为16 ng/ml?结论:成功制备了抗PA15单克隆抗体,并建立了双抗体夹心ELISA方法检测炭疽感染血清中的保护性抗原?     

抗粉尘螨主要变应原Der f1单克隆抗体的制备与鉴定

目的制备和鉴定鼠抗粉尘螨主要变应原Derf1的单克隆抗体（monoclonal antibody,McAb）。方法利用重组Derf1蛋白为免疫原,免疫BALB／c小鼠,取免疫小鼠脾细胞与小鼠骨髓瘤NS-1细胞融合。通过间接ELISA法筛选特异性分泌的杂交瘤细胞。用杂交瘤细胞株诱生小鼠腹水,应用蛋白A亲和层析法进行抗体纯化。采用Ig类与亚类鉴定试剂盒鉴定该单克隆抗体的Ig亚型;通过间接ELISA、Western Blotting鉴定该单克隆抗体的特性和交叉性。结果获得6株可稳定分泌鼠抗粉尘螨主要变应原Derf1的单克隆抗体,其Ig亚型均为IgG1,且6株单抗均具有良好的效价。ELISA和Western—blotting分析表明,该6株单抗均能识别重组Derf1蛋白和天然的粉尘螨提取物。其中,除了484不识别屋尘螨提取物,其他均能识别。结论成功制备了6株鼠抗粉尘螨主要变应原Derf1的单克隆抗体,为建立粉尘螨主要变应原Derf1检测及纯化方法奠定了基础。     

全人源抗狂犬病病毒单克隆抗体的制备与鉴定

目的:利用人源IgM转基因小鼠制备全人源抗狂犬病病毒单克隆抗体,并对其进行初步鉴定?方法:以灭活的狂犬病病毒CTN株作为抗原免疫人IgM转基因小鼠?采用杂交瘤技术结合酶联免疫吸附试验(ELISA)高通量交叉筛选技术(HTS)制备全人源抗狂犬病病毒单克隆抗体?通过双抗体夹心ELISA鉴定单抗的人源性和抗体型,间接ELISA?斑点杂交(Dot Blot)实验及Western blot检测单抗的特异性,BiaCore X-100测定单抗结合抗原的亲和力?结果:建立了2株稳定分泌抗狂犬病病毒人源性单抗的杂交瘤细胞株,分别命名为9E3和9F2?双抗体夹心ELISA结果显示2株单抗均为人源性免疫球蛋白IgM型?间接ELISA?斑点杂交实验结果表明2株单抗均能特异性识别灭活的狂犬病病毒CTN株?Western blot结果显示2株单抗均能与狂犬病病毒糖蛋白特异性结合?2株单抗与狂犬病病毒CTN株抗原结合的亲和力分别为 2.62 × 10-10 mol/L?4.06 × 10-11 mol/L?结论:筛选制备了2株特异性?高亲和力的全人源抗狂犬病病毒单克隆抗体?本研究为进一步筛选人源抗狂犬病病毒免疫预防性中和抗体及其免疫治疗的应用奠定了基础?     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
