Objective To investigate the renal protective activity of Hsian-tsao Mesona procumbens Hemsl. water extracts in diabetic rats. Methods Thirty Sprague-dawley female rats were randomly divided into three groups (n=10 each), "control group" with intraperitoneal saline injection, "diabetic group" with 60 mg of intraperitoneal streptozotocin injection per kg of body weight and "Hsian-tsao group" with intragastric administration of Hsian-tsao extraction everyday for 4 weeks after intraperitoneal streptozotocin injection. The body weight and blood sugar were measured before and after model induction in the three groups. Thrombospondin-1 (TSP-1) expressions in the kidney were monitored by immunohistochemistry. Kidney ultrastructural changes were also analyzed by using transmission electron microscopy. Results Before diabetic model induction, there were no significant differences among the three groups in body weight and blood sugar. Four weeks after the induction of diabetes, the differences became statistically significant. Electron microscopy also revealed disruption of the foot processes of the podocytes and other damages in diabetic group. These damages were significantly less severe in Hsian-tsao group when compared with the diabetic group. TSP-1 expressions in the kidney were significantly increased in both the diabetic group and Hsian-tsao group, but it was relatively lower in Hsian-tsao group than in diabetic group. Conclusion Our results showed that Hsian-tsao treatment in the diabetic rats effectively prevented the pathological alterations in the kidney and decreased the TSP- 1 expression. It was suggested that Hsian-tsao had protective effect on the kidneys of the diabetic rats.  相似文献

Objective To investigate the renoprotection effect of low molecular weight heparin(LMWH)in diabetic rats.Methods Diabetic rats were induced by injection of steptozotocin(STZ).Rats were randomly divided into three groups:control group,diabetic model group,and LMWH treatment group.The levels of total cholesterol(TC),triglyceride(TG),prothrombin time(PT),kaolin partial thromboplastin time(KPTT),24-hour urinary albumin excretion rate(UAER),and serum soluble P-selectin were determined after 8 weeks.Glomerular morphology was observed by light microscopy.Results The levels of TG,UAER,and P-selectin in LWMH treatment group were lower than those in model group(all P〈0.01).The levels of PT and KPTT in LWMH treatment group were shorter than those in model group.And LWMH improved the histological changes of diabetic rats.Conclusion LMWH has some renal protective effects in diabetic rats,partly through down-regulating the expression of P-selectin.  相似文献

Objective:To evaluate the prevention effect of diabetic retinopathy of Jiangtang Xiaozhi Recipe(降糖消脂方,JXR) in streptozotocin(STZ)-induced diabetic rats.Methods:Sprague-Dawley rats were randomly divided into normal control group and diabetic group.Rats in the diabetic group were induced by intraperitoneal administration of STZ(50 mg/kg),and subdivided into 5 groups.Rats in the diabetic control group were given saline;four treatment groups were given metformin(300 mg/kg),JXR(2,4 and 8 g/kg) respectively for 8 weeks,while rats in the normal control group were injected with citrate buffer and given the same volume of vehicle.Body weight and food intake were measured every week.The hypoglycaemic effects were determined by testing fasting blood glucose(FBG) every other week,and hemoglobin A1c(Hb A1c),insulin,and glucagon at the end of the treatment.The preventive effects of JXR on STZ-induced diabetic rats were determined by histopathological examination with hematoxylin and eosin staining,and periodic acid-schiff staining.The effects were further evaluated by serum superoxide dismutase(SOD) activity and malondialdehyde(MDA).Results:High-dose JXR significantly reduced FBG and Hb A1 c level at the 8th week of administration(P0.01,P0.05).JXR significantly increased insulin level(P0.05),and decreased glucagon level(P0.05).JXR showed the antioxidant defense with increased SOD activity and decreased MDA contents in diabetic rats.Histopathological studies revealed that there were no basement membrane thickening and mild destruction in the treated groups.Morphometric measurements of retina microvascular showed that acellular capillary and capillary density decreased in treated rats while pericyte and endothelial cell increasing after the treatment.Conclusion:JXR have protective effect of diabetic retinopathy and its mechanism may be associated with the obvious hypoglycemic and antioxidant effect.  相似文献

Summary: To study the efficacy and the mechanism of Colquhoumia root ( Tripterygium hypoglaucure (Le,vL) Hutch) in the treatment of mesangial proliferation glomerulonephritis (MsPGN), SD rats were injected with anti-thymoeyte serum (ATS) to make MsPGN model (anti-Thyl model). The rats were then divided into 3 groups: normal control group, anti-Thyl model group and treatment group. Histopathologieal (HE, PAS), immunohistoehemieal, RT-PCR technique and computer imaging analysis system were used to evaluate mesangial matrix production, the expression of TGF-β protein and mRNA in the tissues of kidney. Our result showed that proteinuria and the ratio of extraeellular matrix/glomerular capillaries area (ECM/CA) were increased significantly in model group. The expression of both TGF-β protein and mRNA in glomeruli was much higher in model group than in control group (P〈0.01). After the treatment with Colquhoumia root, proteinuria, ECM/CA and the expression of both TGF-β1 protein and mRNA in glomeruli were significantly decreased in treatment group as compared with those in model group. It is concluded that Colquhoumia root is effective in reducing proteinuria and mesangial matrix proliferation in MsPGN and it may achieve these effects by inhibiting the expressions of TGF-β1 protein and mRNA of mesangial cells.  相似文献

OBJECTIVE:To study the effect of Wuziyanzong treatment on the sperm quality and content of calcium ions(Ca 2+) in oligoasthenospermia rats.METHODS:A model of oligoasthenospermia was induced in 50 Sprague Dawley rats by treatment with tripterygium glycosides at 30 mg/kg per day for 8 weeks.They were divided randomly into a model group,a positive group(Huangjingzanyu capsule,3.01 g/kg),and low,medium and high dose Wuziyanzong treatment groups(2.30,4.60,9.20 g/kg crude drug respectively) with 10 in each group.Another 10 rats were used as a control group.The rats in the control and model groups were administered distilled water,while the rats in the remaining groups were administered Wuziyanzong for 30 d.The epididymides were removed,spermatozoa recovered and the sperm density and viability were measured.The spermatozoa were purified and the contents of Ca 2+ in the cytoplasm and mitochondria were detected by flow cytometry and atomic absorption spectrometry,respectively.RESULTS:After 8 weeks of treatment with tripterygium glycosides,the sperm density,sperm activity and the Ca 2 + content of spermatozoa in the model rats were all significantly decreased compared with the control group(all P<0.05).After 30 d treatment,the sperm density and activity improved and the Ca 2 + content of sperm were increased significantly in the medium and high dose Wuziyanzong treatment groups in comparison with the model group(all P<0.05).CONCLUSION:The Wuziyanzong treatment increased sperm density,improved sperm viability and enhanced the content of Ca 2+ in the sperm cytoplasm and mitochondria in this rat model of oligoasthenospermia.  相似文献

Objective:To observe the pharmaceutical effect of Chinese drugs for activating blood circulation (Xiongshao Capsule,XSC,芎芍胶囊)and for activating blood circulation and detoxification(Xiongshao Capsule and Huanglian Capsule,XSHLC,黄连胶囊)in terms of the indices of thrombosis,inflammatory reaction and tissue damage related factors in experimental carotid artery thrombosis rats.Methods:Fifty Wistar rats were randomly divided into the sham operation group,the model group,the Simvastatin group(SG),the activating blood circulation(ABC)group,and the activating blood circulation and detoxifying(ABCD)group,with 10 rats in each group.Simvastatin(1.8 mg/kg),XSC(0.135 g/kg)and XSHLC(0.135 g/kg)were administered to Simvastatin,ABC and ABCD group by gastrogavage,and an equal volume of normal saline was given to the sham operation group and the model group.After 2 weeks of successive medication,the rats in the model and all drug therapy groups were made into experimental carotid artery thrombosis model.The serum levels of matrix metalloproteinases(MMP-9),tissue inhibitors to metalloproteinase(TIMP-1),granule membrane protein-140(GMP-140),tissue-type plasminogen activator(t-PA),high-sensitivity C-reactive protein(hs-CRP) and interleukin-6(IL-6)were detected with enzyme-linked immunoassay 24 h later.Results:Compared with the model group,the levels of serum GMP-140,hs-CRP,IL-6 and MMP-9 were significantly decreased,and the level of t-PA was significantly increased in the ABC and ABCD group(P〈0.05),while the level of serum hs-CRP in ABCD group decreased significantly compared with that in the ABC group(P〈0.05).Conclusions:Chinese drugs both for activating blood circulation and for activating blood circulation and detoxifying have good effects on regulating indices of thrombosis,inflammatory reaction and tissue damage in experimental carotid artery thrombosis rats.The effect of activating blood circulation and detoxifying drugs on regulating the level of serum hs-CRP is superior to that of activating blood circulation drug alone.  相似文献

Objective To investigate the correlation of interleukin(IL)-1,IL-6 and IL-10 concentrations to ovarian hyperstimulation syndrome(OHSS) and whether intravenous immunoglobulin(IVIG) has the effects on ovarian hyperstimulated rats.
Methods Immature female Wistar rats were divided into control group, OHSS group (n=13) and IVIG group(n=13). For the latter two groups, pregnancy mare serum gonadotropin(PMSG)and human chorionic gonadotropin(hCG) were given to induce OHSS, and rats in IVIG group were treated with immunoglobulin. Forty-eight hours after administration of hCG, capillary permeability was evaluated from the Evans blue dye(EB) concentration in the ovaries and the EB concentration in peritoneal irrigated fluid at 30 min after the intravenous injection of EB. Rats' blood samples and ovaries were obtained to be measured for IL-1, IL-6 and IL-10 by ELISA.
Results In OHSS group, total weights of bilateral ovaries and the ovarian EB concentration were significantly higher than those in others(P〈0.05). Both serum and ovarian concentrations of IL-1 were significantly higher in OHSS and IVIG groups than those in control group (P〈0.05). The ovarian concentrations of IL-6 and IL-10 in IVIG group were significantly lower than those in control group(P〈0.05). Furthermore, the ovarian IL-10 concentration in IVIG group was significantly lower than that in OHSS group(P〈0. 05).
Conclusion Inflammation involved IL-1 in OHSS rats plays an important role. Vascular permeability was mostly increased in ovaries of hyperstimulated rats. It appears that ovaries of OHSS rats may be the primary places of inflammation. IVIG treatment resulted in statistically significant reductions in ovaries' weights and ovarian vascular permeability of OHSS rats, with a decreased level of ovarian IL-10. It implys that IVIG have a beneficial effect in reducing the severity of OHSS in the experimental model maybe by restrainning IL-10.  相似文献

Objective To investigate the influence of Zhenqing Recipe (ZQR) and Ligustri Lucidi Fructus (LLF) on diabetic rats and its possible mechanism. Methods The model of type 2 diabetic rats was established by feeding a high-sucrose-high-fat diet and injecting a low dose of Streptozotocin in Wistar rats. The model rats were randomly divided into three groups: diabetic model, ZQR-treated, and LLF-treated groups for 8-weeks treatment. The normal Wistar rats were as a normal control group. Results The level of fasting blood glucose in ZQR and LLF groups was decreased compared with model group (P < 0.01, 0.05, respectively). Both ZQR and LLF markedly reduced serum triglycerides (P < 0.01, 0.05, respectively), and increased the insulin sensitivity index (P < 0.05). Histopathology revealed that ZQR and LLF reduced pancreatic damage. Immunohistochemistry evaluation showed that the percentage of insulin positive cells in pancreatic island was higher than model group (P < 0.01, 0.05, respectively).The mRNA and protein expression of SREBP-1c in pancreas were significantly decreased in ZQR and FLL group (P < 0.01). Conclusion ZQR has therapeutic effect on type 2 diabetes, it ameliorates the histopathological changes of pancreas, protects β cells, improves insulin resistance, and attenuates the expression of SREBP-1c. This study also provides the anti-diabetic evidence of FLL even its effects are weaker than ZQR.  相似文献

The preventive effects of nitroglycerine (NG) on glucocorticoid-induced osteoporosis in growing rats were studied. Three-month-old female Wistar rats were randomly divided into control group (CON), dexamethasone group (DXM), DXM plus a low dose NG group (NG-L), DXM plus a middle dose NG group (NG-M) and DXM plus a high dose NG group (NG-H), 8 rats in each group. The rat model of osteoporosis was developed by intramuscular injection of dexamethasone twice a week. NG 0.2, 0.4 and 1.0 mg/kg was administered by oral gavages to the treatment groups every day for 12 weeks. Rats in CON group and DXM group were treated with normal saline of the same amount. After the treatment, the bone mineral density (BMD) and bone metabolism-associated biochemical markers were determined. Compared with CON group, BMD of lumbar spine and femur in DXM group was decreased significantly (P〈0.05 and P〈0.01 respectively), blood BGP levels and NO levels reduced (both P〈0.01), and TRAP level increased (P〈0.05). As compared with DXM group, BMD, serum BGP and NO were increased, and TRAP decreased in NG-L group and NG-M group, but had no significant difference in comparison to CON group. All the markers other than serum NO and TRAP levels had no significant difference between NG-H group and DXM group. It was concluded that low or middle doses of NG could prevent glucocorticoid-induced bone loss in growing rats, but high dose of NG could not. Supplement with NO donor could be considered as a preventive treatment for glucocorticoid-induced osteoporosis in a developing skeleton.  相似文献

Objective: To explore the effects and mechanisms of poly (ADP-ribose) polymerase (PARP) inhibitor 3-aminobenzamide on nerve lesions in streptozotocin-diabetic rats. Methods: Experimental rats were divided into normal control group(NC group), diabetic control group (DC group )and diabetic group treated with 3-aminobenzamide (DT group ).Nerve conduction velocity (NCV), serum superoxide dismutase (SOD) activity and serum malondialdehyde (MDA) concentration,phosphocreatine (Pcr) ,creatine (Cr) concentration in sciatic nerves were evaluated after 4 weeks. Results: SOD,Pcr activity,and NCV were higher (P 〈 0.05) and MDA concentration were significantly lower in DT group, compared with DC group (P 〈 0.01 ). Meanwhile, ATP and Cr in sciatic nerves were similar in DT group, compared with DC group (P 〉 0.05). Conclusion: 3-aminobenzamide could alleviate the established functional and metabolic abnormalities of early DPN in the streptozotocin-induced diabetic rat models,which pro- vided a novel approach for prevention and treatment of diabetic neuropathy.  相似文献

目的:观察甘草酸对糖尿病大鼠肾脏氧化应激及醛糖还原酶活性、基因表达水平的影响,探讨其保护肾脏、预防糖尿病肾病的可能机制。方法:雄性Wistar大鼠30只,随机分为正常对照组、模型对照组和甘草酸治疗组,腹腔注射链脲菌素(60mg/kg)诱导糖尿病模型,治疗组给予甘草酸(30mg/kg/d)治疗16周。16周后测定各组大鼠24h尿白蛋白量(UP24)、肾脏肥大指数(肾重/体重)、丙二醛(MDA)、超氧化物歧化酶(SOD)、醛糖还原酶(AR)活性及mRNA水平。结果:与正常对照组相比,模型组大鼠UP24、肾指数显著升高(P<0.01),血清SOD活性降低,MDA含量增加(P<0.01),肾脏AR活性及基因表达显著升高(P<0.01)。与模型组相比,甘草酸治疗组UP24、肾指数显著降低(P<0.01),血清SOD活性显著增强(P<0.05),同时MDA含量显著减少(P<0.01),肾脏AR活性显著降低、ARmRNA水平显著下降(均P<0.01)。结论:高血糖可诱导机体氧化应激增强、多元醇代谢途径激活及两者的协同效应,促进糖尿病肾病的发生发展;甘草酸可能通过抗氧化应激、减少肾脏AR活性及基因表达以抑制多元醇通路的激活,发挥肾脏保护作用。  相似文献

目的:复制大鼠糖尿病肾病模型,研究复方丹参联合阿司匹林对大鼠糖尿病肾病的治疗作用及其机制。方法:选择体重180~200g的健康雄性SD大鼠64只,随机选取54只复制大鼠糖尿病肾病(DN)模型,其余大鼠作为正常对照(NC)组。实验第8周后,随机将模型组大鼠分为糖尿病肾病(DN)对照组、阿司匹林(DN+A)治疗组、复方丹参(DN+S)治疗组、复方丹参联合阿司匹林(DN+A+S)治疗组各10只。自模型成功之日起分别按相应方案治疗。检测空腹血糖(FBG)、血尿素氮(BUN)、血肌酐(Scr)、尿24h微量白蛋白(mALb)、肾组织丙二醛(MDA)、血栓素B2(TXB2),光镜观察大鼠肾组织形态学变化。比较治疗后各组之间上述指标的差异。结果:与DN对照组相比,DN+A+S组FBG、BUN、Scr、24hmAlb、MDA、TXB2均明显降低(P<0.05);DN+A组BUN、Scr、MDA、TXB2也明显降低(P<0.05),但FBG、24hmAlb无显著降低(P>0.05);DN+S组也见FBG、BUN、Scr、24hmAlb、MDA、TXB2明显降低(P<0.05)。DN+A+S组同DN+A组相比,上述指标变化更显著(P<0.05);同DN+S组相比,上述指标的改善同样更明显,P<0.05(FBG除外)。结论:复方丹参联合阿司匹林对大鼠DN的改善作用与其降低血糖、抗氧化应激损伤和改善肾脏微循环等有关。  相似文献

目的:通过观察黄连素对2型糖尿病大鼠血管内皮功能的影响,探讨黄连素对2型糖尿病血管并发症的防治意义及其机制。方法:采用小剂量链脲佐菌腹腔注射加高脂饲料喂养的方法建立2型糖尿病大鼠模型,将模型动物随机分为模型组及黄连素治疗组,另设正常对照组以及正常黄连素组。药物干预治疗8 周后,处理动物,检测各组动物血清中空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)及空腹血清胰岛素(FINS)的水平;通过离体实验方法观察离体胸主动脉条的张力反应;HE染色观察胸主动脉形态学改变。结果:与模型组比较,黄连素治疗组大鼠FBG、TC及TG 水平明显下降,胰岛素敏感指数(ISI)升高 ( P<0.05);黄连素治疗组大鼠乙酰胆碱(ACh)引起的内皮依赖性血管舒张反应明显改善,而硝普钠(SNP)诱导的内皮非依赖性舒张反应各组间比较差异无显著性(P>0.05);黄连素治疗组大鼠糖尿病引起的血管内皮病变明显减轻;与正常对照组比较,正常大鼠给予黄连素灌胃,上述指标未出现明显改变。结论:黄连素对2 型糖尿病早期出现的血管内皮损伤有良好的保护作用,机制可能与其降血糖、调节血脂、改善胰岛素抵抗及内皮依赖性的血管舒张反应等作用有关。  相似文献

目的: 探讨姜黄素对糖尿病肾病(DN)大鼠肾脏结缔组织生长因子( CTGF ) 表达和肾脏排泄功能的影响,阐明姜黄素对DN大鼠肾脏的保护作用及其机制。方法:40只雄性SD大鼠分为正常对照组(n=12)和模型组(n=28),采用腹腔注射链脲佐菌素(STZ)的方法建立DN大鼠模型,24只造模成功的大鼠随机分为模型组(n=12)和姜黄素组(n=12)。于用药8周末检测大鼠血糖水平、体质量、肾质量、肾质量/体质量、24 h尿微量蛋白清除率(UAER)和血尿素氮(BUN)、血肌酐(Scr)水平;HE染色观察大鼠肾组织病理学变化;免疫组织化学法检测肾皮质CTGF蛋白表达强度。结果: 与模型组比较,姜黄素组大鼠一般情况明显改善,空腹血糖降低(P<0.01),体质量明显增加(P<0.01);与模型组比较,姜黄素组大鼠肾质量、肾质量/体质量、UAER、BUN和Scr明显降低(P<0.01);与模型组比较,姜黄素组大鼠肾组织着色程度明显减轻,CTGF表达显著下降(P<0.01);与模型组比较,姜黄素组大鼠肾小球肥大、系膜细胞增生、肾小球变形病变明显减轻。结论:姜黄素能明显改善大鼠肾脏功能,其机制可能与抑制肾脏CTGF表达有关。  相似文献

目的探讨芪莲汤颗粒剂(QiLian soup-granula)对糖尿病肾病(diabetic dephropathy,DN)大鼠肾脏病变的保护作用及机制。方法采用高脂饲料联合链脲佐菌素(streptozotocin,STZ45 mg/kg)腹腔注射法建立DN大鼠模型,模型成功后分组给药。设正常对照组、模型对照组、消渴丸组、芪莲汤颗粒剂高剂量组、芪莲汤颗粒剂低剂量组。按试验设计剂量灌胃给药,1次/d,每周6d。连续给药4个月后检测空腹血糖、血肌酐、做糖耐量试验、尿素氮、尿肌酐、尿蛋白。取肾脏组织作病理、免疫组织化学法检测肾小球Ⅳ型胶原蛋白的表达。结果芪莲汤颗粒剂可以显著降低大鼠空腹血糖、尿素氮、血肌酐和尿蛋白。并能抑制DN大鼠肾组织Ⅳ型胶原蛋白表达。结论芪莲汤颗粒剂对DN大鼠具有很好的治疗作用,可以延缓DN病理进程。该作用可能与降血糖及降低胰岛素抵抗和抑制肾组织Ⅳ型胶原表达有关。  相似文献

目的:探讨天合保肾合剂对糖尿病肾病(DN)大鼠的肾脏保护作用。方法:单肾切除及链尿佐菌素(STZ)尾静脉注射诱导DN模型;设正常组,模型组,卡托普利组,天合保肾合剂高、中、低剂量组;8 w后测定DN大鼠空腹血糖(GLU)、糖化血红蛋白(GHb)、尿素氮(BUN)、血肌酐(Scr),光镜观察肾脏病理组织学变化。结果:天合保肾合剂能够降低DN大鼠血糖、糖化血红蛋白(GHb)、尿素氮(BUN)、血肌酐(Scr)含量,与模型对照组比较有显著性差异(P〈0.01),肾脏病理改变较模型组有明显减轻。结论:天合保肾合剂可以降低糖尿病肾病大鼠血糖,抑制糖化血红蛋白生成,改善肾功能,减轻肾脏病理变化,对DN大鼠具有肾脏保护作用。  相似文献

目的 探讨茴香提取液对糖尿病大鼠肾脏损害的保护作用.方法 按血糖分层随机将40只大鼠分为正常对照组(n=8)和造模组(n=32),给予造模组大鼠腹腔注射链脲佐菌素制备糖尿病模型,同时给予正常对照组腹腔注射等体积柠檬酸缓冲液.成模后,将造模组按随机数表法进一步分为茴香高剂量组、茴香低剂量组、二甲双胍组、模型组,每组各8只.每组给予相应处理30 d后,比较各组大鼠FBG、血清尿素氮(BUN)、肌酐(SCr),肾脏组织谷胱甘肽(GSH)、丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性,并分析苏木精-伊红染色下各组大鼠肾脏组织形态学改变.结果 与正常对照组比较,模型组FBG、BUN、SCr及MDA水平均较高,而SOD和GSH水平较低(均P<0.05);与模型组比较,二甲双胍组、茴香组的FBG水平均较低;茴香高剂量组SCr、BUN水平均较低、肾组织SOD和GSH水平均较高,而茴香低剂量组仅SCr水平较低(均P<0.05).茴香组肾小管的变性、坏死较模型组轻且管型少,茴香高剂量组比茴香低剂量组病变轻且范围小.结论 一定浓度范围内的茴香提取液具有减轻糖尿病大鼠肾脏损伤、改善肾功能、提高肾组织抗氧化的作用.  相似文献

目的::探讨小檗碱对糖尿病肾病(diabetic nephrpathy,DN)大鼠肾脏损伤的保护作用。方法:健康成年雄性 SD 大鼠40只,随机分为正常对照组、模型组(DN 组)和小檗碱组。DN 组及小檗碱组大鼠行左侧肾摘除术,术后2周腹腔一次性注射链脲佐菌素(streptozotocin,STZ)60 mg/kg 诱导 DN 大鼠模型,72 h 后测非空腹血糖>16.8 mmol/L 为造模成功;小檗碱组大鼠给予小檗碱0.1 g/(kg·d)灌胃。定时检测各组大鼠血糖、尿微量白蛋白和肾小球滤过率的变化。结果:与 DN 组大鼠相比,小檗碱组大鼠血糖和24 h 尿微量白蛋白明显降低,肾小球滤过率增加(P <0.05),肾功能明显改善。结论:小檗碱可明显降低 DN 大鼠血糖,改善 DN 大鼠肾功能紊乱,对 DN 大鼠肾脏具有较强的保护作用。  相似文献

目的:观察温肾健脾活血汤治疗糖尿病肾病(DN)IV期的效果。方法:80例患者随机分为对照组和治疗组,两组采用相同基础治疗,治疗组予温肾健脾活血汤口服,对照组予洛汀新片治疗,疗程均为8周。观察两组治疗前后临床症状积分、空腹血糖(FBG)、餐后血糖(PBG)、糖化血红蛋白(HBA1c)、尿素氮(BUN)、血清肌酐(Scr)、尿白蛋白排泄率(UAER)、24h尿蛋白定量(UP)、C反应蛋白(CRP)等指标变化。结果:治疗组在改善临床症状积分、UAER、UP方面优于对照组;两组血糖、HBA1c比较无显著差异(P>0.05);治疗组治疗后CRP水平显著降低(P<0.05),与对照组比较亦有显著性差异(P<0.05)。结论:温肾健脾活血汤能改善DNⅣ期患者临床症状,通过降低尿蛋白、抑制肾脏内炎症反应实现对肾脏的保护作用。  相似文献

Background Podocyte has inflammatory role in the development of diabetic nephropathy (DN). Mycophenolate mofetil (MMF), an anti-inflammatory agent, can suppress macrophage infiltration and reduce renal injury in streptozotocin-induced diabetic rats. Angiotensin II receptor blocker (ARB), another renal protecting agent, can decrease podocyte loss in DN. In this study, we detected the expression levels of monocyte chemoattractant protein-1 (MCP-1) and nephrin to evaluate podocyte’s role in inflammatory reaction in DN, observe and compare the effect of MMF alone and in combination with valsartan, on preventing podocyte loss in streptozotocin (STZ) induced diabetic rats. Methods Diabetic model was constructed in uninephrectomized male Wistar rats by single peritoneal injection of STZ (65 mg/kg). The successfully induced diabetic rats were randomly divided into four groups: diabetes without treatment group (DM), valsartan treated group (DMV), MMF treated group (DMM), and combined therapy group (DMVM). Normal rats of the same sibling were chosen as control (NC). At the end of the 8th week, serum biochemistry, 24-hour urinary protein (UP) and the ratio of kidney weight/body weight (RWK/B) were measured. The rats were sacrificed for the observation of renal histomorphology through light and electron microscope. Nephrin, desmin and MCP-1 levels were detected by semi-quantitative immunohistochemical assays. Real-time quantitative PCR was used to detect the mRNA levels of nephrin and MCP-1.Results Compared with group NC, serum glucose level, 24-hour UP and RWK/B in group DM were significantly higher (P&lt;0.01), and the nephrin mRNA level in DM group was significantly lower (P&lt;0.05). The nephrin mRNA expression levels in group DMV, DMM and DMVM were all higher than that of DM group (P&lt;0.05) and no significant differences were found among the three treatment groups (P&gt;0.05). Treatment with MMF, valsartan or their combination could significantly decrease the 24-hour UP and RWK/B, and suppress glomerulosclerosis and interstitial fibrotic lesions in diabetic rats. In diabetic rats, the high expressions of desmin and MCP-1 in kidney were suppressed by valsartan, MMF or their combination.Conclusions Podocytes are involved in the inflammatory reaction of diabetic rats. MMF could suppress MCP-1 and desmin expression, enhance nephrin expression, and attenuate proteinuria in diabetic rats. The combined therapy of valsartan and MMF did not show any superiority over monotherapies on renal protection. MMF may have renoprotective effect in early stages of diabetic nephropathy through preventing podocytes loss and anti-inflammatory activity.  相似文献
