目的观察枸杞糖肽对心肌细胞缺氧性损伤的保护作用,建立心肌细胞缺氧损伤病理模型。方法从细胞形态学、细胞存活率、心肌细胞乳酸脱氢酶(LDH)外漏释放及细胞凋亡等方面,观察枸杞糖肽对缺氧损伤心肌细胞的保护作用。结果枸杞糖肽能明显抑制缺氧所致的心肌细胞死亡(P<0.05,P<0.01)、细胞内LDH的漏出(P<0.05,P<0.01),并能降低缺氧诱导的心肌细胞凋亡。结论枸杞糖肽对心肌细胞缺氧性损伤具有明显的保护作用。  相似文献

目的研究GMC0403注射液对氰化钠诱导原代培养心肌细胞损伤的保护作用。方法采用1μmol/ml氰化钠与低糖DMEM、共培养复制心肌细胞缺糖、缺氧损伤模型,通过细胞线粒体琥珀酸脱氢酶活力(MSDH)、乳酸脱氢酶(LDH)外漏率、台盼蓝染色率研究GMC0403注射液对原代培养心肌细胞氰化钠(NaCN)诱导缺糖、缺氧模型的保护作用。结果GMC0403注射液能显著降低缺糖、缺氧损伤心肌细胞线粒体琥珀酸脱氢酶活力,抑制LDH的外漏,降低台盼蓝染色率。结论GMC0403注射液对NaCN诱导原代培养心肌细胞损伤具有保护作用。  相似文献

目的探讨环维黄杨星D(CVB-D)新衍生物对酒精诱导的PC12细胞损伤模型的影响。方法建立酒精诱导PC12细胞损伤模型,通过MTT染色、Hoechst 33258染色和乳酸脱氢酶(LDH)活性的测定研究环维黄杨星D新衍生物对PC12细胞的保护作用。结果环维黄杨星D新衍生物抑制酒精对PC12细胞的增殖抑制作用,凋亡率下降,同时LDH的漏出减少。结论环维黄杨星D新衍生物对酒精诱导的PC12细胞损伤具有保护作用。  相似文献

目的研究色满卡林(Cromakalim,CRK)对缺氧复氧致大鼠乳鼠心肌细胞损伤的保护作用,并探讨其机制。方法用原代培养的Sprague—Dawley(SD)大鼠乳鼠心肌细胞建立缺氧2h复氧30min的损伤模型。采用乳酸脱氢酶(LDH)试剂盒测培养液中LDH的活性;采用黄嘌呤氧化酶法测定细胞内超氧化物岐化酶(SOD)活性;硫代巴比妥酸显色法测定细胞内丙二醛(MDA)含量;AV—PI双标记后应用流式细胞仪检测细胞的凋亡率。结果色满卡林对缺氧复氧造成的心肌细胞损伤有保护作用,表现为可以对抗缺氧复氧引起的LDH活性增加、MDA含量增加、SOD活性下降、凋亡率增加。结论色满卡林对缺氧复氧致培养大鼠乳鼠心肌细胞的损伤具有保护作用,此作用与抑制脂质过氧化、减少心肌细胞凋亡有关。  相似文献

目的 研究葛根素(Pue)处理后对培养大鼠乳鼠心肌细胞缺氧/复氧损伤的保护作用及其机制.方法 原代培养新生大鼠心肌细胞,建立缺氧/复氧损伤模型,分为缺氧/复氧对照组和缺氧/复氧 葛根素保护组(葛根素浓度分别为1、0.1、0.01g/L).观察各组心肌细胞形态结构及超微结构的变化,测定心肌细胞超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、乳酸脱氢酶(LDH)含量和一氧化氮(NO)分泌量,以及Pue对上述指标的影响.流式细胞仪计数各组细胞凋亡率.结果 Pue可明显提高SOD和LDH活性,降低MDA含量,减少NO分泌量,差异均有高度统计学意义(均P<0.01);电镜下治疗组心肌细胞结构接近正常;流式细胞仪计数治疗组细胞凋亡率明显低于对照组,差异有高度统计学意义(P<0.01).结论 葛根素处理对体外培养心肌细胞模拟缺氧/复氧损伤模型有明显的保护作用,其保护机制可能与葛根素稳定心肌细胞膜和减轻氧自由基损伤,并且抑制缺氧/复氧损伤中的心肌细胞凋亡有关.  相似文献

目的探讨金丝桃苷对原代培养的大鼠心肌细胞缺氧-再给氧损伤的保护作用。方法采用显微镜和M TT法观察缺氧-再给氧导致培养的大鼠原代心肌细胞的损伤和金丝桃苷的保护作用;流式细胞术观察金丝桃苷对心肌细胞凋亡的影响;并观察乳酸脱氢酶(LDH)水平的变化和细胞内C a2 浓度的变化。结果金丝桃苷对缺氧-再给氧导致的心肌细胞损伤有一定的保护作用,可以抑制心肌细胞凋亡的发生;0.5~50μm o l/L金丝桃苷可以剂量依赖性地抑制心肌细胞中LDH的形成和细胞内C a2 超载。结论金丝桃苷具有对抗缺氧-再给氧导致心肌细胞损伤的作用,其机制可能与抑制心肌细胞凋亡、钙超载和抗自由基形成有关。  相似文献

[目的]探讨甘草对双氧水(H_2O_2)诱导大鼠H9c2心肌细胞损伤的保护作用。[方法]建立H_2O_2诱导心肌细胞损伤模型(100μmol/L,4h),甘草(800、400μg/mL)作用4 h,检测心肌细胞活力、乳酸脱氢酶(LDH)释放量、超氧化物歧化酶(SOD)和丙二醛(MDA)含量、细胞内氧自由基(ROS)含量、钙离子含量、线粒体膜通透性转换孔(mPTP)的开放及心肌细胞凋亡率。[结果]甘草能够不同程度提高心肌细胞活力和SOD活力,减少LDH、MDA、细胞内ROS和钙离子含量,抑制mPTP开放、降低细胞凋亡率(P0.05或P0.01)。[结论]甘草对H_2O_2诱导的大鼠H9c2心肌细胞损伤具有保护作用。  相似文献

目的:探讨神经再生素(nerve regeneration factor,NRF)对缺氧/再复氧(hypoxia/reoxygenation,H/R)心肌细胞的保护作用。方法:取Winstar乳鼠心脏,制成心肌细胞悬液,贴壁法培养心肌细胞并进行缺氧/再复氧损伤,观察神经再生素对缺氧/再复氧心肌细胞形态及细胞内SOD、MDA及LDH水平的影响,采用流式细胞仪双标法检测心肌细胞凋亡百分率。结果:神经再生素能升高SOD活性,降低MDA含量和LDH活性,减少心肌细胞凋亡率。结论:神经再生素对缺氧/再复氧心肌细胞损伤具有一定的保护作用。  相似文献

目的 探讨小鼠乳鼠心肌细胞在缺氧和缺氧/复氧下心肌细胞损伤及凋亡情况及T3对心肌细胞缺氧/复氧损伤的影响。 方法 采用1~2天新生昆明小鼠,分离培养心室肌细胞,分别给予不同时间缺氧及缺氧/复氧处理,观察细胞状态及细胞搏动次数,流式细胞仪分析细胞凋亡;给予T3预处理后进行缺氧/复氧损伤,流式细胞仪检测细胞凋亡,JC-1染色检测细胞线粒体膜电位;应用Western blot法检测心肌细胞凋亡蛋白。结果 缺氧可造成心肌细胞凋亡,缺氧时间越长,心肌细胞损伤越重,缺氧/复氧可进一步加重心肌细胞损伤,增加细胞凋亡率;T3预处理可减少缺氧/复氧所致损伤,减少细胞凋亡率,降低凋亡蛋白caspase-3、Bax的表达,减少细胞线粒体膜电位损伤。结论 缺氧可诱导心肌细胞损伤,缺氧时间越长,心肌细胞损伤越重,且缺氧/复氧损伤对心肌细胞损害更大;T3预处理可以保护心肌细胞并减少细胞凋亡。  相似文献

目的:研究环维黄杨星D(CVB-D)对Na2S2O4诱导EVC304细胞损伤的保护作用.方法:采用体外培养人脐静脉内皮细胞EVC304,Na2S2O4作为耗氧剂建立细胞缺氧损伤模型.用MTT法测定细胞存活率与细胞培养液乳酸脱氢酶(LDH)活力反映细胞损伤的程度,培养液一氧化氮合酶(NOS)活力与内皮素(ET)含量分析EVC304细胞的功能.结果:CVB-D能显著抑制Na2S2O4诱导EVC304细胞OD<,570>的降低和细胞培养液中LDH活力的增加,提高培养液中NOS活性,降低ET的含量(与模型组比较,P<0.05或P<0.01).结论:CVB-D对Na2S2O4诱导EVC304细胞损伤具有保护作用,其作用机制可能与改善NOS和ET失调有关.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献

目的 探讨猪肺磷脂注射液联合经鼻持续气道正压通气(NCPAP)对呼吸衰竭早产儿的临床疗效及肌酸激酶同工酶活性(CK-MB)的影响.方法 选取呼吸衰竭早产儿80例,分为观察组和对照组各40例.对照组采用NCPAP给氧治疗,观察组给予NCPAP给氧联合猪肺磷脂气管内给药.观察两组患儿治疗前及治疗12h、24 h后PaO2、PaCO2、血氧饱和度(SaO2)、pH的变化情况,检测治疗前及治疗5d后血清CK-MB水平;评估两组患儿的临床治疗效果.结果 两组患儿PaO2、PaCO2、SaO2、pH比较,差异均有统计学意义(P<0.05),其中观察组治疗后的PaO2、SaO2、pH均高于对照组,PaCO2则低于对照组.两组的PaO2、SaO2、pH均随观察时间延长而升高(P<0.05),PaCO2均随观察时间的延长而降低(P<0.05).观察组治疗有效率为87.5%,显著高于对照组的70.0% (P <0.05).治疗5d后两组患儿血清CK-MB水平均较前降低(P<0.05),且观察组明显低于对照组(P<0.05).结论 猪肺磷脂注射液气管内给药联合NCPAP可以显著降低呼吸衰竭早产儿CK-MB的含量,提高治疗有效率,起到很好的呼吸循环支持作用.  相似文献

Evidence obtained from randomized controlled trials (RCTs) has been generally accepted as the gold standard in the evaluation of clinical effectiveness. Readers need to understand the trial design, implementation, results, analysis and interpretation, so as to fully Jnderstand the results of RCTs. Thus, the investigators of RCTs have to report these items in a complete, accurate and clear manner. Since 1998, we have conducted several evaluations on the reporting quality of RCTs published in Chinese journals on traditional Chinese medicine (TCM) and results have shown that there is an urgent need for higher quality RCTs on TCM.  相似文献

Ankylosing spondylitis is a chronic and progressive disorder with inflammation mainly involving the central axis joints. It mainly affects the cervical spine and the lumbosacral area, with the pathogenesis closely related to the kidney and the Governor Vessel (GV). TCM holds that the syndrome is deficiency in origin and excess in superficiality, which is due to insufficiency of the kidney, deficiency of GV, and blocking of the channels with the invasion of exogenous evil, leading to poor circulation of qi and blood and malnutrition of the bones, muscles and joints. The TCM method of tonifying the kidney and strengthening GV to regulate circulation of qi and blood and check the arthralgia pain should be adopted, with the Kidney-Tonifying and GV Strengthening Decoction (益肾强督汤) prescribed.  相似文献
