道诺霉素抑制视网膜色素上皮细胞炎前因子的表达   总被引:1,自引:1,他引:0  
目的 观察道诺霉素对视网膜色素上皮细胞细胞(RPE)炎前因子表达的影响 .方法 培养的人 RPE经 IL- 1β(10μg· L- 1 )刺激以及不同浓度的道诺霉素作用后 ,用EL ISA、免疫组化和原位杂交等方法 ,检测培养的人 RPE炎前因子 m RNA和蛋白质的表达 .结果  EL ISA显示对照组培养 RPE在 IL- 1β刺激 8h后 ,上清中 IL- 6与 IL- 8分别为2 0 0 0 pg· m L- 1· 10 - 6 cells和 5 0 0 0 pg· m L- 1· 10 - 6 cells.使用 10 0 μg·L- 1 道诺霉素作用于培养的 RPE后 IL- 6与 IL-8分别为 180 pg· m L- 1 · 10 - 6 cells (P <0 .0 1)和 32 0pg· m L- 1· 10 - 6 cells(P<0 .0 1) .免疫组化和原位杂交亦显示不同浓度道诺霉素可以不同程度地抑制 RPE内炎前因子蛋白质与 m RNA的表达 .结论 道诺霉素能有效地抑制 IL -1β诱导的培养的 RPE IL - 6和 IL - 8的表达  相似文献

目的 研究苏拉明 (suramin)对培养人视网膜色素上皮细胞 (retinal pigm ent epithelium,RPE)增殖的影响 .方法 将不同质量浓度的苏拉明 (1.5 ,15和 15 0 mg· L- 1 )加入RPE细胞培养液 ,采用四甲基偶氮唑盐 (tetrazolium,MTT)比色法 ,细胞分裂指数计数和核仁组成区嗜银染色(Ag NORs)检测苏拉明对 RPE增殖活力的影响 .结果 含有10 0 m L· L- 1小牛血清的培养液可以显著刺激 RPE的增殖(P<0 .0 1) ,无血清组 A值为 0 .19± 0 .0 1、含血清组 A值为0 .30± 0 .0 1;苏拉明抑制了 10 0 m L· L- 1血清条件下 RPE的增殖 ,呈剂量依赖性 ,最大抑制率达 5 1% ,3种浓度组 A值分别为 0 .2 9± 0 .0 1,0 .2 4± 0 .0 1和 0 .14± 0 .0 1,对细胞的形态无明显影响 .结论 苏拉明对血清刺激 RPE增殖有显著非毒性抑制作用  相似文献

目的:增生性玻璃体视网膜病变(PVR)是多种细胞和细胞因子参与的一种眼内创伤修复反应。单核细胞趋化蛋白-1(MCP-1)对巨噬细胞和淋巴细胞有特异趋化作用。本研究目的在于了解视网膜脱离后不同时期的组织中MCP-1的表达与巨噬细胞和淋巴细胞的关系。方法:选用视网膜手术取材标本,包括孔源性视网膜脱离的视网膜下液中的细胞成份,B级PVR玻璃体手术集取物以及PVR C/D的视网膜增殖膜,免疫组化法检测其中MCP-1,CD68和CD3表达状态。结果:视网膜下液中细胞成份MCP-1的表达均为阴性或弱阳性,B级PVR手术集取物以及增殖膜中,MCP-1表达阳性程度随PVR等级的升高而逐渐增高。CD68阳性细胞(巨噬细胞)与CD3阳性细胞(T-淋巴细胞)的比例随PVR病变程度的增加而降低。结论:MCP-1对视网膜脱离后的愈合过程和PVR增殖膜的形成有调节作用。  相似文献

目的 探讨缺氧和高氧环境对兔主动脉平滑肌细胞增殖的抑制作用 ,间接判断缺氧与冠脉再狭窄的关系和研究氧疗是否能用于 PTCA支架后冠脉再狭窄的防治 .方法 用含 10 0 m L· L- 1小牛血清的 RPMI16 40培养液体外培养新西兰白兔的主动脉平滑肌细胞至 3~ 7代 ,将含 5 0 m L· L- 1小牛血清的 RPMI16 40培养液与动脉平滑肌细胞的混悬液10 0 μL 加入 96孔培养板中培养 ,按照实验分组分别在 10 0 ,2 10 ,2 5 0 ,5 0 0和 75 0 m L· L- 1 O2 及纯氧环境中培养主动脉平滑肌细胞 48h.用 MTT法和 3H- Td R掺入法检测动脉平滑肌细胞增殖的数量 .结果 以 2 10 m L· L- 1 O2 环境下动脉平滑肌细胞增殖的数量为基准对照组 ,其他各组为实验组 .5 0 0 m L· L- 1 O2 至纯氧环境下动脉平滑肌细胞的增殖显著低于对照组 (P<0 .0 5 ) ,2 5 0 m L· L- 1 O2 与 2 10 m L·L- 1 O2环境下主动脉平滑肌细胞的增殖无显著差异 (P>0 .0 5 ) ,10 0m L· L- 1 O2 环境下动脉平滑肌细胞的增殖显著高于对照组(P<0 .0 5 ) .结论 动脉平滑肌细胞的增殖随着培养环境中氧浓度升高而降低 ,缺氧促进动脉平滑肌细胞的增殖 ,高氧对动脉平滑肌细胞增殖具有显著的抑制作用 ;氧疗可能有助于降低冠脉再狭窄率 ,并为高压氧治疗其他以动脉血管  相似文献

目的 观察视网膜色素上皮细胞 ( RPE)在炎性介质刺激下炎前因子 m RNA的表达 .方法 培养的人 RPE经IL - 1β ( 10μg· L- 1 )和脂多糖 ( 1m g· L- 1 )的刺激后 ,用生物素标记的寡核苷酸探针进行原位杂交 ,检测 RPE炎前因子IL - 1β,IL - 6,IL - 8和 TNF-α m RNA表达 .杂交结果使用图像分析法进行比较 .结果 原位杂交显示 RPE细胞质中均呈现浓淡不一的淡蓝色着色 ,胞核不着色 .其中 IL - 6m RNA在脂多糖刺激下染色较浅 ,增加脂多糖的剂量不能提高其表达 .图像分析显示在 IL - 1β ( 10μg· L- 1 )和脂多糖 ( 1m g· L- 1 )刺激下 ,RPE表达 IL - 6m RNA的吸光度分别为 10 8± 18和3 5± 14 ,二者之间有显著性差异 ( P<0 .0 1) .结论 在 IL - 1β和脂多糖刺激下 ,人 RPE可以表达炎前因子 IL - 1β,IL - 6,IL - 8和 TNF-α的 m RNA .  相似文献

金雀异黄素诱导培养人视网膜色素上皮细胞凋亡   总被引:3,自引:0,他引:3  
目的 :探讨金雀异黄素对培养人视网膜色素上皮细胞 (hRPE)的促凋亡作用 .方法 :将不同浓度的金雀异黄素作用于培养人视网膜色素上皮细胞 ,通过TUNEL染色和光、电镜的形态学观察 ,观察金雀异黄素对培养hRPE凋亡的诱导作用 .结果 :不同浓度的金雀异黄素可以诱导RPE凋亡 ,5 0mg·L-1 金雀异黄素作用 2 4h即有TUNEL阳性的凋亡细胞出现 ,作用时间在 2 4 ,4 8和 72h ,其凋亡细胞百分数的中位数分别为 7.6 % ,9.8%和 1 3.7% ;当金雀异黄素浓度为 75和1 0 0mg·L-1 时 ,以上 3个时间点凋亡细胞百分数的中位数分别为 1 0 .3% ,1 6 .4 %和 2 3.4 % ;1 5 .4 % ,2 1 .2 %和 35 .8% .透射电镜观察 ,5 0mg·L-1 浓度作用于细胞 ,胞核内染色体出现边集 ,表现凋亡的早期征象 ;75mg·L-1 作用呈现典型的凋亡特征 ;1 0 0mg·L-1 作用于细胞 ,除凋亡细胞外 ,部分细胞出现胞质的进行性溶解 ,有胞膜破坏现象 ,坏死细胞比例增多 .结论 :金雀异黄素可诱导RPE凋亡的发生 ,并有剂量和时间效应关系 .1 0 0mg·L-1 金雀异黄素使RPE坏死  相似文献

以兔腹腔巨噬细胞条件培养基作为趋化因子的来源,用微孔滤膜法检测其对培养的兔视网膜神经胶质细胞的趋化活性。结果表明。该条件培养基对培养的神经胶质细胞具有趋化活性。从而说明单核巨噬细胞系统在增殖性玻璃体视网膜病变(PVR)发病中的作用。  相似文献

目的 观察血管钠肽 (Vasonatrin peptide,VNP)对低氧状态下大鼠心肌成纤维细胞 (CFs)增殖及胶原合成的影响 ,探讨其防治低氧所致心肌间质结构重建的作用机制 .方法 培养新生大鼠 CFs,应用流式细胞仪分析细胞周期 ,以 3H- proline掺入率反映胶原合成速率 .结果 单纯低氧不改变 CFs的细胞周期 ,但增加低浓度血清 (2 5 m L· L- 1 )作用基础上的细胞增殖指数 PI(S+G2 / M) / (S+G2 / M+G0 / G1 )(P<0 .0 1) ,10 - 6 m ol· L- 1的 VNP抑制低氧诱导的 PI增加(P<0 .0 1) .低氧可以增加基础胶原合成 (增加 19.6 % ,P<0 .0 5 ) ,也增加低浓度血清 (2 5 m L· L- 1 )诱导的胶原合成速率 (增加 37.2 % ,P<0 .0 1) . 10 - 8~ 10 - 6 mol· L- 1 的 VNP浓度依赖性抑制低氧诱导的胶原合成速率 (P<0 .0 1) .结论 低氧增加胶原合成以及血清刺激的 CFs的增殖反应 ,VNP抑制上述作用 ,从而抑制低氧引起的心肌纤维化  相似文献

目的 :研究人参二醇组皂甙 ( panaxadiol saponins,PDS)对体外培养视网膜色素上皮( RPE)细胞增生的直接抑制作用及可能机制。方法 :通过活细胞计数法与 MTT比色法分别检测不同浓度的 PDS和 2 0 0 mg· L-1PDS在不同作用时间 ( 6~ 1 2 0 h)对 RPE细胞增生及代谢的影响。结果 :PDS组细胞增殖明显受抑并出现细胞脱落 ,40 0 mg· L-1PDS具有最强的抑制效应 ,其明显抑制效应在 6h出现 ,96h达高峰。PDS组 A值明显低于对照组 ,RPE细胞生存率下降。结论 :PDS可能通过阻滞钙通道降低细胞内钙浓度、干扰 RPE细胞代谢 ,对 RPE细胞增殖具有剂量依赖和时间依赖方式的抑制作用  相似文献

目的研究单核细胞源性趋化因子对培养的牛视网膜Müller细胞的趋化作用.方法以刀豆蛋白A激活小牛的单核细胞,以此单核细胞条件培养基作为趋化因子的来源,用改良的Boyden小室微孔滤膜法检测其对培养的牛视网膜 Müller细胞的趋化活性,并检测经明胶结合处理掉纤维连接蛋白(FN)后条件培养基的活性.结果该条件培养基对培养的 Müller细胞具有趋化活性,且该条件培养基在经明胶处理后其趋化活性降低,但仍然存在.结论该条件培养基对Müller细胞趋化作用的物质成份不是单一的,是多种因素作用的结果,单核巨噬细胞系统在增殖性玻璃体视网膜病变发病中具有重要的作用.  相似文献

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.  相似文献

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.  相似文献

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P<0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.  相似文献

Objective:To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods:Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results :Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion:The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.  相似文献

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.  相似文献
