目的:构建脂氧素A4受体样蛋白(LRLP)基因的真核表达载体,克隆后转染Hela细胞,观察LRLP基因在Hela细胞中的表达。方法:应用PCR方法.以小鼠睾丸cDNA为模板,扩增出LRLP基因片段,插入质粒pGEM—T中,转化人大肠杆菌JM109。扩增阳性重组质粒,经酶切后纯化并测序鉴定目的片段。构建含绿色荧光蛋白基因的真核表达载体pEGFP/LRLP,转化人大肠杆菌JM109扩增.酶切后再测序鉴定目的片段,抽提质粒后瞬时转染Hela细胞,置激光共聚焦显微镜下照像。结果:测序鉴定表明.克隆的LRLP序列与GenBank中LRLP原序列100%.符合,激光共聚焦显微镜下见Hela细胞中LRLP基因表达。结论:成功地构建了真核表达载体pEGFP/LRLP,并转染了Hela细胞。  相似文献

目的:构建一种具有生物学活性的血管活性肠肽(VIP)的真核表达质粒.方法:利用RT-PCR从小鼠胸腺淋巴细胞中克隆有信号肽序列的VIP cDNA,插入真核表达载体pcDNA3.1,构建含VIP的重组质粒pcDNA3.1-VIP,转染COS-7细胞,用ELISA和Western blot鉴定表达的蛋白,细胞因子释放抑制法测定生物学活性.结果:经酶切和基因测序证实,克隆的基因片段为带有信号肽序列的VIP基因;经转染的COS-7细胞表达VIP蛋白,并能有效地抑制巨噬细胞的TNF-α释放.结论:成功构建了能够表达具有生物学活性的真核表达质粒pcDNA3.1-VIP.  相似文献

目的:构建TPX2的真核表达载体pcDNA3.1-TPX2,并观察其在食管癌EC9706细胞中的表达。方法:在pQE-70-TPX2原核表达载体基础上,根据GenBank上提供的TPX2基因序列及测序结果,采用Primer Premier 5.0软件设计扩增TPX2基因编码区的引物序列,经HindⅢ和BamHⅠ双酶切,与真核表达载体pcDNA3.1连接后转化感受态大肠杆菌JM109,并进行酶切分析和序列测定。将构建的质粒pcDNA3.1-TPX2体外转染EC9706细胞,进行稳定筛选。用Western blot鉴定基因的表达。结果:DNA测序和BLAST比对表明克隆的人TPX2基因序列正确,进一步酶切分析显示,成功构建了真核表达载体pcDNA3.1-TPX2,并在EC9706细胞中获得稳定表达。结论:TPX2基因真核表达载体的成功构建为后续研究TPX2基因在肿瘤细胞中的功能奠定了基础。  相似文献

目的构建小鼠cyclin A2的正义和反义真核表达载体。方法采用小鼠胚胎组织取材,提取总RNA。设计引物,采用RT-PCR方法扩增小鼠cyclin A2 mRNA得到cyclin A2的DNA,将此DNA插入pGEM-T载体,转化入JM109感受态细菌。将鉴定得到的重组质粒进行扩增,EcoRⅠ酶切后,回收cyclin A2目的片段亚克隆入pcDNA3.1真核表达载体。酶切筛选鉴定重组的真核表达载体pcDNA3.1-cyclin A2。结果将小鼠基因的开放阅读框(ORF)重组到真核表达载体pcDNA3.1内,用EcoRⅠ酶切重组质粒后可得到一接近1660 bp的DNA条带,核酸测序证实和GenBank所公布序列相符。HindⅢ酶切鉴定重组质粒后,证明分别为正向和反向插入载体的质粒。DNA测序证明构建的真核表达载体pcDNA3.1-cyclin A2的序列是正确的。结论成功构建了小鼠cyclin A2的正义和反义真核表达载体。  相似文献

目的 构建小鼠CD160胞外段的真核表达载体,并稳定转染CHO细胞进行真核表达.方法 从C57BL/6小鼠脾脏中提取总RNA,经RT-PCR扩增CD160胞外段,然后将其克隆到真核表达载体pcDNA3.1和pEGFP-N1两种质粒中,构建重组质粒psCD160和pEGFP-sCD160.重组质粒经双酶切鉴定及测序正确后,用脂质体转染CHO细胞,并用RT-PCR和Western blot检测CD160胞外段的表达,流式细胞术检测重组psCD160与其配体的结合能力.结果 获得长度约为520 bp的小鼠CD160胞外段基因,经测序证实其序列正确.用脂质体将含有CD160胞外段基因的重组质粒载体转染CHO细胞后.RT-PCR和Western blot分析证实转染的CHO细胞可表达重组的psCD160基因,并且表达的可溶性CD160可与其配体结合.结论 成功构建小鼠CD160胞外段基因的真核表达载体,并转染CHO细胞后表达出相应蛋白,为进一步研究CD160胞外段的功能效应奠定实验基础.  相似文献

目的:构建lefty1基因与pcDNA3.1(+)相融合的真核表达载体,并在lefty1基因的下游加上Flag标签,转染人肾小管上皮细胞株验证重组质粒活性。方法:设计并合成lefty1基因上下游引物,同时加上Flag标签序列,PCR扩增基因,并将其连接到真核表达载体pcDNA3.1(+)上,双酶切图谱分析和扩增产物测序鉴定所构建的真核表达载体。脂质体转染法将重组质粒pcDNA3.1(+)-lefty1-Flag和pcDNA3.1(+)分别转染至人肾小管上皮细胞株HK-2,qPCR和Western blot检测lefty1在mRNA和蛋白水平的表达。结果:以lefty1基因质粒模板扩增得到的片段约1 150bp,双酶切得到目的基因片段,测序的结果显示与lefty1基因序列相同。转染肾小管上皮细胞后,pcDNA3.1(+)-lefty1-Flag能表达lefty1蛋白。结论:成功构建lefty1基因真核表达载体,为一步研究lefty1基因功能奠定了基础。  相似文献

目的:构建分泌型核心蛋白聚糖(DCN)真核表达载体,为研究DCN的生物学功能奠定基础。方法:利用特异性引物,应用PCR技术扩增DCN全长基因cDNA片段,与 pcDNA3.1载体进行连接,并转化到大肠杆菌JM109中扩增以获得重组载体,应用双酶切、PCR以及测序鉴定此重组载体。结果:获得了约1080 bp大小的特异性DNA片段;PCR产物与真核表达载体进行连接,经过双酶切、PCR以及测序鉴定证实分泌型DCN cDNA片段正确插入真核表达载体pcDNA3.1中。结论:成功克隆了分泌型DCN cDNA,并且构建了真核表达载体pcDNA3.1-DCN。  相似文献

目的 利用TA克隆法构建凋亡诱导因子(AIF)的真核表达载体AIF-pcDNA3.1(+),并研究其在人肺腺癌A549细胞中的表达.方法 根据GenBank上AIF的mRNA序列设计特异性引物,以A549细胞总RNA为模板,RT-PCR扩增AIF基因.并用TA克隆法,将AIF目的基因连接到pUC-T载体,行双酶切和DNA测序鉴定;回收目的片段并将其插入真核表达载体pcDNA3.1(+)构建重组质粒AIF-pcDNA3.1(+).最后,将A IF-pcDNA3.1(+)转染A549细胞,RT-PCR和Western blot检测转染细胞AIF基因的表达.结果 成功扩增617 bp的AIF目的基因片段并连接pUC-T载体,测序后回收目的片段并与真核表达载体pcDNA3.1(+)连接,DNA序列分析显示,连接在真核表达载体上的基因片段与GenBank中的AIF序列完全吻合.RT-PCR和Western blot检测结果显示,转染了AIF-pcDNA3.1(+)的A549细胞AIF mRNA和蛋白表达水平均高于未转染细胞(P<0.05).结论 成功构建了真核表达载体AIF-pcDNA3.1(+),转染后能够在A549细胞中表达.  相似文献

目的 构建人胰十二指肠同源盒(PDX-1)基因的真核表达载体,观察其在NIH3T3细胞中的表达.方法 以人胰岛细胞瘤cDNA为模板,采用聚合酶链式反应(PCR)扩增PDX-1基因编码区的全部序列,克隆入真核细胞表达载体pcDNA3.1中,经限制性内切酶双酶切及DNA序列分析鉴定目的基因.序列正确的重组质粒用脂质体转染NIH3T3细胞,Western blot观察基因表达情况.结果 PCR扩增的特异性片段长度为852 bp,以此构建的重组质粒pcDNA3.1-PDX-1, 经BamHⅠ和EcoRⅠ双酶切后显示5.5 kb和 850 bp左右的两条片段,测序结果与GenBank中的人PDX-1基因cDNA(GenBank 序列号NM_000209)序列一致.证明PDX-1基因已成功克隆到了真核细胞表达载体pcDNA3.1 中.Western blot证实pcDNA3.1-PDX-1转染NIH3T3细胞24 h后有PDX-1的表达.结论 成功构建了pcDNA3.1-PDX-1重组真核表达载体,并在NIH3T3细胞中表达.  相似文献

目的:克隆人类TSLC1基因并构建其真核表达载体pcDNA3.1( )/TSLC1.方法:从正常皮肤组织中提取总RNA,采用RT-PCR方法扩增TSLC1基因全长cDNA,克隆入pMD18-T载体并转化大肠杆菌JM109,经PCR、酶切鉴定均为阳性的克隆,进行核苷酸测序分析,再将TSLC1基因定向克隆入pcDNA3.1( )载体中构建表达载体.结果:RT-PCR扩增后的产物在约1413 bp处出现明显的特异性条带,TSLC1基因的cDNA片段被成功插入真核表达载体pcDNA3.1( )质粒的多克隆位点,经鉴定与GenBank收录的TSLC1 cDNA 序列一致.结论:TSLC1基因的cDNA片段被成功克隆,为以后的研究奠定了基础.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献

目的 探讨猪肺磷脂注射液联合经鼻持续气道正压通气(NCPAP)对呼吸衰竭早产儿的临床疗效及肌酸激酶同工酶活性(CK-MB)的影响.方法 选取呼吸衰竭早产儿80例,分为观察组和对照组各40例.对照组采用NCPAP给氧治疗,观察组给予NCPAP给氧联合猪肺磷脂气管内给药.观察两组患儿治疗前及治疗12h、24 h后PaO2、PaCO2、血氧饱和度(SaO2)、pH的变化情况,检测治疗前及治疗5d后血清CK-MB水平;评估两组患儿的临床治疗效果.结果 两组患儿PaO2、PaCO2、SaO2、pH比较,差异均有统计学意义(P<0.05),其中观察组治疗后的PaO2、SaO2、pH均高于对照组,PaCO2则低于对照组.两组的PaO2、SaO2、pH均随观察时间延长而升高(P<0.05),PaCO2均随观察时间的延长而降低(P<0.05).观察组治疗有效率为87.5%,显著高于对照组的70.0% (P <0.05).治疗5d后两组患儿血清CK-MB水平均较前降低(P<0.05),且观察组明显低于对照组(P<0.05).结论 猪肺磷脂注射液气管内给药联合NCPAP可以显著降低呼吸衰竭早产儿CK-MB的含量,提高治疗有效率,起到很好的呼吸循环支持作用.  相似文献

Evidence obtained from randomized controlled trials (RCTs) has been generally accepted as the gold standard in the evaluation of clinical effectiveness. Readers need to understand the trial design, implementation, results, analysis and interpretation, so as to fully Jnderstand the results of RCTs. Thus, the investigators of RCTs have to report these items in a complete, accurate and clear manner. Since 1998, we have conducted several evaluations on the reporting quality of RCTs published in Chinese journals on traditional Chinese medicine (TCM) and results have shown that there is an urgent need for higher quality RCTs on TCM.  相似文献

Ankylosing spondylitis is a chronic and progressive disorder with inflammation mainly involving the central axis joints. It mainly affects the cervical spine and the lumbosacral area, with the pathogenesis closely related to the kidney and the Governor Vessel (GV). TCM holds that the syndrome is deficiency in origin and excess in superficiality, which is due to insufficiency of the kidney, deficiency of GV, and blocking of the channels with the invasion of exogenous evil, leading to poor circulation of qi and blood and malnutrition of the bones, muscles and joints. The TCM method of tonifying the kidney and strengthening GV to regulate circulation of qi and blood and check the arthralgia pain should be adopted, with the Kidney-Tonifying and GV Strengthening Decoction (益肾强督汤) prescribed.  相似文献
