Objective:To study the effects of the Chinese medicine Jinmaitong Capsule(筋脉通胶囊,JMT) on the pathomorphology of sciatic nerves,ciliary neurotrophic factor(CNTF),and the mRNA expressions of CNTF in rats with streptozotocin-induced diabetes mellitus(STZ-DM).Methods:The animal model was established by one time intraperitoneal injection of streptozotocin.The rats were simply divided by random into 5 groups including model group,low-dose JMT group(JL),medium-dose JMT group(JM),high-dose JMT group(JH) and neurotropin group.For each of the above 5 groups,a group of 10 normal Wistar rats matched in body weight,age and gender were set as normal group.Intragastric administrations were started after the animal model established.The JL group were administered with five times the JMT dose recommended for a human adult;the JM group were administered with ten times the JMT dose recommended for a human adult;the JH group were administered with twenty times the JMT dose recommended for a human adult.The neurotropin group was administered with ten times the neurotropin dose recommended for a human adult.All rats were given intragastric administration for 16 weeks and then killed.In the 4th,8th,12th,16th week,body weight and blood glucose level were detected before and after the intervention.The morphologic changes of the sciatic nerves were observed by optical microscope and transmission electron microscope.The CNTFmRNA expressions were detected by real-time fluorescent quantitative polymerase chain protein,and the CNTF protein expressions were detected by immunohistochemical method.Results:The blood glucose levels of the STZ-DM rats were much higher than normal group(P<0.01),and there was no apparent difference between any treatment groups and the model group(P>0.05).Before and after the intervention in the 4th, 8th,12th,16th week,there were no significant differences in the body weight among all the groups(P>0.05). The sciatic nerves of STZ-DM rats might have pathomorphological changes in axons,myelin sheaths,and interstitium.The levels of CNTF and CNTF-mRNA expressions in the STZ-DM rats were both significantly decreased(P<0.01).The sciatic nerves of STZ-DM rats might have pathomorphological changes in axons, myelin sheaths,and interstitium.Conclusion:JMT could improve the pathomorphology of sciatic nerves by increasing CNTF’s and CNTF-mRNA expressions in sciatic nerve tissues,and promote the repair and regeneration of damaged nerve fibers.  相似文献

血浆TGFβ1水平与糖尿病视网膜病变关系的研究   总被引:2,自引:0,他引:2  
目的探讨血浆TGFβ1水平与糖尿病视网膜病变的关系.方法采用ELISA法测定24名对照组和78例2型糖尿病患者血浆TGFβ1水平,并分析血浆TGFβ1水平与不同糖尿病视网膜病变及不同病程的关系.结果①2型糖尿病患者血浆TGFβ1水平明显高于对照组(P<0.01);②背景型和增殖型视网膜病变组血浆TGFβ1水平均较对照组和糖尿病无眼底病变组显著升高(P<0.01);③病程5~10年和10年以上者血浆TGFβ1水平显著高于5年以下者(P<0.05).结论糖尿病视网膜病变(DR)患者血浆TGFβ1水平升高,并且血浆TGFβ1水平可能与糖尿病视网膜病变的发生和发展有关.  相似文献

目的:观察胰岛素样生长因子-1(IGF-1)与糖尿病周围神经病变(DPN)的关系。方法:测定97例2型糖尿病患者(其中DPN组32例,无DPN组65例)的血清IGF-1及其各项临床及生化指标。结果:DPN组血清IGF-1明显低于无DPN组和正常对照组(P<0.01),无DPN组和正常对照组间血清IGF-1无差异(P>0.05),在2型糖尿病患者中血清IGF-1与年龄、HbA1C、病程呈负相关。结论:DPN患者血清IGF-1显著下降,提示IGF下降可能与DPN的发生有关。  相似文献

目的:研究胰岛素样生长因子-1(IGF-1)和胰岛素抵抗(IR)与原发性高血压及左室肥厚的关联性。方法:78例分为3组,原发性高血压(EH)组29例,EH合并左室肥厚(LVH)组28例,正常对照组21例。采用二维超声心动图测定3组左心室重量指数(LVMI),采用放射免疫法测定3组血清IGF-1及胰岛素(INS)水平,并计算胰岛素敏感指数(ISI)。结果:EH+LVH组血清IGF-1和INS高于EH组,EH组高于正常对照组,差异均有统计学意义(P〈0.01);相关分析结果显示:IGF-1与INS呈正相关,与ISI呈负相关。结论:EH患者血清IGF-1和INS水平升高,合并LVH时升高更显著;IR促进LVH的作用可能与其升高IGF-1的水平有关。  相似文献

目的:测定血清胰岛素样生长因子-1(IGF-1)、胰岛素样生长因子结合蛋白-3(IGF-BP3)水平,探讨其与2型糖尿病肾病的关系。方法:选择2型糖尿病患者86例,根据其尿微量白蛋白排泄率(UAER)结果分为单纯糖尿病组(SDM组)33例、早期糖尿病肾病组(EDN组)28例和临床期肾病组(CDN组)25例。检测IGF-1、IGF-BP3、UAER等指标,分析IGF-1、IGF-BP3与糖尿病肾病的发病和病情轻重的关系。结果:CDN组血清IGF-1、IGF-BP3和UAER均明显高于SDM与EDN组(P <0.01),CDN组血清IGF-1、IGF-BP3和UAER亦均明显高于EDN组(P <0.01)。 IGF-1与肌酐、尿素氮、UAER和IGF-BP3均呈正相关关系(P <0.01),与年龄和糖化血红蛋白均无相关关系(P >0.05),而与空腹血糖呈负相关关系(P <0.01)。结论:血清IGF-1、IGF-BP3水平与2型糖尿病肾病的发生发展有相关性,可作为糖尿病肾病早期的诊断指标之一。  相似文献

胰岛素样生长因子-1 (insulin-like growth factor-1,IGF-1)是1种由70个氨基酸组成的单链多肽,几乎所有组织中都含IGF-1,现将IGF-1的研究现状作一综述.1 IGF-1的发现和命名Salmon和Daughaday首次发现IGF-1能促进小鼠软骨组织对硫酸盐35S的吸收,当时命名为"硫酸化因子".1963年,Froesch等将它描述为具有不可抑制的胰岛素样活性物质.1972年,又将它命名为"促生长因子".1976年Rinderknecht和Humbel从人血浆中分离出活性物质,由于它的结构与胰岛素原相似,重新将它命名为胰岛素样生长因子-1[1].  相似文献

目的 研究在下颌骨发育过度导致的骨性Ⅲ类错(牙合)畸形患者中,胰岛素样生长因子-1(IGF-1)基因的表达,探讨IGF-1基因表达与下颌骨生长的关系.方法 选取骨性Ⅲ类错(牙合)畸形患者27例,骨性Ⅰ类患者(包括个别正常(牙合))27例,应用荧光定量PCR技术和酶联免疫吸附剂测定(ELISA)技术研究下颌骨中IGF-1基因的表达变化.结果 RT-PCR检测结果显示,骨性Ⅲ类错(牙合)畸形组中IGF-1mRNA的表达量为(5.5410±2.044 7)μg/μl,骨性Ⅰ类组中IGF-1mRNA的表达量为(1.2821±0.273 1)μg/μl,2组比较具有统计学差异(P<0.05).根据1GF-1基因光密度值与浓度关系曲线,得回归系数0.998.ELISA检测结果显示,骨性Ⅲ类错(牙合)畸形组中IGF-1蛋白浓度为(84.1259±29.294 7) ng/L,骨性Ⅰ类组中IGF-1蛋白浓度为(22.406 4±4.9312) ng/L,2组相比具有显著差异(P<0.05).结论 下颌骨生长量越大,IGF-1基因mRNA和蛋白表达越高.  相似文献

目的:探讨瘦素促胎儿生长作用及其与胰岛素和胰岛素样生长因子1(IGF-1)的关系.方法:采用放射免疫法检测100例足月新生儿脐血血清瘦素、胰岛素和IGF-1水平,根据新生儿出生体重分为大于胎龄儿(LGA)组19例,适于胎龄儿(AGA)组65例,小于胎龄儿(SGA)组16例.结果:(1)脐血瘦素水平与新生儿出生体重呈正相关关系(r=0.57,P<0.01),其中LGA组脐血瘦素水平为(13.38±6.75)μg/L,显著高于AGA组(7.40±4.45)μg/L;SGA组(2.79±1.54)μg/L低于AGA组,差异有显著性(P<0.01);(2)脐血胰岛素和IGF-1水平分别与新生儿出生体重呈显著正相关关系,相关系数(r)分别为0.30和0.35;(3)脐血瘦素和胰岛素水平呈正相关关系(r=0.32,P<0.01),脐血瘦素与IGF-1水平之间元相关关系(r=018,P>0.05).结论:脐血瘦素水平与胎儿生长发育状态密切相关,瘦素和胰岛素对胎儿体重有双向调节作用,IGF-1和瘦素具有各自独立的调控胎儿生长发育体系.  相似文献

目的:探讨霉酚酸酯(MMF)对糖尿病(DM)大鼠肾组织中胰岛素样生长因子1(IGF-1)及基质金属蛋白酶2(MMP-2)表达的影响。方法:雄性SD大鼠随机分为对照组(n=12)、糖尿病组(DM组,n=11)、MMF组3组(n=10)。于8周末检测24h尿蛋白量、血糖、内生肌酐清除率(Ccr)、相对肾质量(左肾质量/体质量);PAS染色观察肾脏病理改变;免疫组化、Western印迹检测肾脏IGF-1、MMP-2的表达。结果:3组大鼠相对肾质量、血糖、24h尿蛋白和Ccr差异有统计学意义(F分别为51.097、150.925、161.315和103.307,P<0.001),MMF组相对肾质量、24h尿蛋白和Ccr均低于DM组,高于对照组(P<0.05)。与对照组比较,DM组肾小球体积增大,系膜区基质明显增生,肾小球毛细血管袢受压,基底膜增厚,而MMF组较DM组有所改善。3组大鼠肾组织IGF-1、MMP-2蛋白表达差异有统计学意义(F分别为15.715和20.449,P<0.001),DM组中IGF-1的表达较对照组增加,MMP-2的表达较对照组下降;MMF组IGF-1较DM组下降,MMP-2增加(P<0.05)。结论:MMF可能通过减少肾脏IGF-1的表达,同时上调MMP-2表达,起到保护肾脏的作用。  相似文献

目的:检测食管癌患者血清胰岛素样生长因子-1(IGF-1的水平.方法:采用ELISA法检测52例食管癌患者手术前后血清IGF-1含量,以22名正常健康人为对照.结果:手术前食管癌患者血清IGF-1水平为(292.57±100.53 μg/L),手术后为(280.50±81.46 μg/L,正常人血清IGF-1水平为(196.75±58.31 μg/L).食管癌患者手术前后血清IGF-1水平均高于正常人(P<0.05).手术前后食管癌患者血清IGF-1水平差异无统计学意义(P>0.05).结论:IGF-1参与食管癌的发病过程,是食管癌发生发展过程中重要的生长因子之一.  相似文献
