CTLA4Ig基因转染对大鼠胰岛移植的影响

目的 研究ＣＴＬＡ４Ｉｇ基因在糖尿病大鼠体内表达及其产物延长胰岛移植物存活的作用。方法 利用Ｌｉｐｏｆｅｃｔｉｎ载体包裹ＣＴＬＡ４Ｉｇ ｃＤＮＡ质粒后转染鼠胰岛和肌肉细胞，检测移植后ＣＴＬＡ４ｉＧ基因表达水平，观察ＣＴＬＡ４Ｉｇ基因表达在延长糖尿病鼠胰岛移植物和大鼠存活时间的作用。结果 Ａ组胰岛移植第７天２只大鼠血清ＣＴＬＡ４Ｉｇ呈阳性（阳性率２０％），分别为１４ｎｇ＝ｍｌ、３１ｎｇ／ｍｌ。Ｂ组胰

目的：克隆人淋巴毒素ｃＤＮＡ并测序。方法：采用ＲＴ－ＰＣＲ技术，从ＰＨＡ／ＰＭＡ活化的人Ｔ细胞系Ｊｕｒｋａｔ细胞总ＲＮＡ中扩增人淋巴毒素ｃＤＮＡ，并定向克隆于ｐＵＣ１８、ｐＵＣ１９质粒载体，Ｓａｎｇｅｒ双脱氧链终止法测序。结果：ＲＴ－ＰＣＲ扩增出一个５４１ｂｐ的ＤＮＡ片段，限制性内切酶图谱分析和测序结果显示，该５４１ｂｐ片段为编码人淋巴毒素成熟肽的ｃＤＮＡ，与公布的人淋巴毒素ｃＤＮＡ序列完全一致。结论：本实验成功地克隆了人淋巴毒素ｃＤＮＡ，为在大肠杆菌表达人淋巴毒素并进一步研究其功能，以及淋巴毒素的开发与临床应用奠定了基础。     

人单核细胞趋化蛋白1基因的克隆及在大肠杆菌中的表达

目的：克隆表达具有重要生物功能的人单核细胞核细胞趋化蛋白１（ｈｕＭＣＰ－１）。方法：从人外周血单核细胞（ＰＢＭＣ）中提取ｐｏｌｙ（Ａ）＾＋ＲＮＡ,用ＲＴ－ＰＣＲ法扩增出ｈｕＭＣＰ－１ｃＤＮＡ。将ｈｕＭＣＰ－１ｃＤＮＡ插入融合表达载体ｐＧＥＸＩＮ,１ｍｍｏｌ／Ｌ异丙基－β－Ｄ－硫代半乳糖苷（ＩＰＴＧ）诱导后获得高效表达。结果：Ｗｅｓｔｅｒｎ ｂｌｏｔ证明与ｈｕＭＣＰ－１单克隆抗体有明显交叉反应。结论

目的;在大肠杆菌宿主系统中表达Ｔｒｏｐｉｃ１８０８基因编码的蛋白。方法：用ＰＣＲ方法从质粒中扩增出Ｔｒｏｐｉｃ１８０８ｃＤＮＡ开放阅读框架片段,并重组人表达载体ｐＥＴ－２１ａ中,转化大肠杆菌ＢＬ２１（ＤＥ３）,用ＩＰＴＧ诱导Ｔｒｏｐｉｃ１８０８融合蛋白的表达,经ＳＤＳ－ＰＡＧＥ,表达蛋白条带转至ＰＶＤＦ膜进行氨基酸序列分析,结果：构建了ｐＥＴ－２１ａ－１８０８重组质粒,外源性Ｔｒｏｐｉｃ１８０８基     

饰胶蛋白（Decorin）基因的cDNA克隆与表达

目的 通过基因克隆技术获得饰胶蛋白基因ｃＤＮＡ克隆并表达。为研究饰胶蛋白的生物学功能及应用打基础。方法 应用ＰＣＲ技术从Ｔ细胞ｃＤＮＡ文库获得饰胶蛋白全长基因ｃＤＮＡ片段。并克隆到ｐＵＣ１９载体中,采用Ｓａｎｇｅｒ双脱氧末端终止法测定全部ｃＤＮＡ序列,将测序正确的饰胶蛋白ｃＤＮＡ序列插入ｐＧＥＸ－４Ｔ－１表达载体中,经ＢａｍＨＩ和ＥｃｏＲＩ双酶切后１．２％凝胶电泳鉴定,ＩＰＴＧ诱导表达产物经ＳＤＳ－ＰＡＧＥ分析。结果 克隆的饰胶蛋白ｃＤＮＡ序列与ＧｅｎＢａｎｋ中ＡＦ１３８３００记载的序列完全一致,所表达的融合蛋白质分子量与理论计算值（６５．６ＫＤ）也一致并为可溶性蛋白。结论 本研究成功地克隆了饰胶蛋白基因和表达了ＧＳＴ－Ｄｅｃｏｒｉｎ融合蛋白。     

利用PCR技术构建GM—CSF／Fcγ2融合蛋白载体

目的：构建 Ｇ Ｍ Ｃ Ｓ Ｆ／ Ｆｃγ２ 融合蛋白载体。方法：利用 Ｐ Ｃ Ｒ技术,将人 Ｇ Ｍ Ｃ Ｓ Ｆ基因与人免疫球蛋白 Ｉｇ Ｇ２ 的 Ｆｃ 段基因联接,构建融合基因ｈ Ｇ Ｍ Ｃ Ｓ Ｆ Ｈ Ｖ２;并将此片段定向克隆到真核表达质粒ｐ Ｒｃ／ Ｃ Ｍ Ｖ２ 载体。结果：琼脂糖凝胶电泳结果显示 Ｐ Ｃ Ｒ扩增出了 １．３ ｋｂ 的融合基因片段;限制性内切酶图谱分析,成功地完成了 Ｇ Ｍ  Ｃ Ｓ Ｆ／ Ｆｃγ２ 融合蛋白载体的构建。结论：（１）经 Ｐ Ｃ Ｒ 扩增技术由质粒 Ｐ Ｃ Ｄｈ Ｇ Ｍ Ｃ Ｓ Ｆ和 Ｐ Ｓ Ｖ Ｖ Ｎ Ｐ Ｈ Ｖ２ 分别扩增到了所需的ｈ Ｇ Ｍ Ｃ Ｓ Ｆ ｃ Ｄ Ｎ Ａ 片段和 Ｉｇ Ｇ２ 从绞链区到 Ｃ Ｈ３ 的基因组 Ｄ Ｎ Ａ 片段。（２）利用 Ｐ Ｃ Ｒ介导,扩增出了融合基因片段ｈ Ｇ Ｍ Ｃ Ｓ Ｆ Ｈ Ｖ２。（３）构建了真核细胞表达载体ｐ Ｒｃ／ Ｃ Ｍ Ｖ２／ｈ Ｇ Ｍ Ｃ Ｓ Ｆ Ｈ Ｖ２ 。     

人源乙型肝炎病毒表面抗原抗体Fab片段与IFN-α融合蛋白表达载体的筛选

目的探讨如何在无药物抗性改变;无转化细胞生物指示系统改变的情况下准确快速地筛选出人源抗乙型肝炎病毒表面抗原（ＨＢｓＡｇ）抗体Ｆａｂ片段／ＩＦＮ－α融合蛋白原核表达载体。方法在 ＩＦＮ－α基因两端加上 ＸｂａⅠ酶切位点后用 ＰＣＲ法扩增,再重组入以抗体库技术筛选得到的人源抗ＨＢｓＡｇ－Ｆａｂ片段表达载体的相应位点上,构建 ａｎｔｉ－ＨＢｓＡｇ－Ｆａｂ／ＩＦＮ－α融合蛋白表达载体。取 ＰＣＲ扩增得到的 ＩＦＮ－α ＤＮＡ,用地高辛标记系统进行化学标记,制备 ＩＦＮ－α ＤＮＡ探针。取构建的融合蛋白表达载体转化受体菌．增菌后,以溶菌酶加煮沸法批量制备载体 ＤＮＡ,点样于硝酸纤维膜上, ８０℃烤２ｈ,预杂交液 ４２℃,２ｈ膜预杂交,标记探针 ４２℃, ４ｈ杂交,洗膜后用酶标抗地高辛抗体显色。同时设单纯人源抗 ＨＢｓＡｇ－Ｆａｂ片段表达载体为阴性对照, ＩＦＮ－α质粒为阳性对照。阳性克隆再以 ＳａｃＩ和 ＸｂａⅠ酶切,琼脂糖电泳鉴定。结果４０株转化细胞中筛选出７个阳性克隆,酶切鉴定显示两者符合率达１００％,成功地筛选出了ａｎｔｉ－ＨＢｓＡｇ－Ｆａｂ／ＩＦＮ－α融合蛋白表达载体。结论人源ＨＢｓＡｇ抗体Ｆａｂ片段／ＩＦＮ－α融合蛋白表达载     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
