用McAb对陕西疫区分离的31株HFRS病毒进行抗原性鉴定。结果表明该疫区患者中主要为野鼠型病毒株流行,但个别患者来源的毒株McAb反应谱类似家鼠型,并有野鼠家鼠混合型毒株出现。提示同一疫区有可能存在不同型別的HFRS病毒,这对本疫区HFRS流行病学分析和血清学诊断均有意义。  相似文献

肾综合征出血热病毒血凝素单克隆抗体的建立与鉴定   总被引:13,自引:0,他引:13  
用HFRSV血凝素免疫BALB/c小鼠,取脾细胞与SP2/0骨髓瘤细胞融合,共获得56株分泌抗HFRSV McAb的杂交瘤细胞系。对其中10株细胞系应用血凝抑制试验证明有9株为特异性分泌抗HFRSV血凝素McAb的杂交瘤细胞系。应用微量细胞培养中和试验证明6株具有中和活性。z株(3D_8和3G_1)具有高滴度的血凝抑制活性和中和活性。10株HFRSV血凝素McAb均能与从不同疫区,不同宿主分离的HFRSV反应,说明HFRSV血凝素具有群共同性。  相似文献

应用“Hirt上清法”直接从感染细胞中提取HSV-1(Sm44)标准株DNA,经BamHI酶切获得DNA片段混合物制备探针。实验结果表明,该探针可与HSV-1 (SM44)、HSV-2(SAV)标准株和HSV-1分离株以及重组质粒的DNA杂交,而不与ADV(8型)、CMV及人肺炎支原体和pBR322质粒的DNA提取物杂交,显示此DNA探针的特异性。为将HSV DNA探针用于临床标本检测建立了初步的实验基础。  相似文献

本文报道从16例上海流行性出血热患者血清中分离出12株流行性出血热病毒。血清标本来自1985年冬季流行性出血热流行高峰期间上海市第八人民医院住院患者。病毒分离阳性率高达75%。其中1份血清EHFIgG荧光抗体高达1:1280;1份血清在发病10天后采集,仍能分离出病毒。分离毒株经EHFV多克隆和单克隆抗体分析,发现病毒抗原性存在差异。其中7株毒株根据与McAb反应结果,暂分为三种类型:2株与76-118、A9株有相似的抗原决定簇;3株带有与R22相似的抗原决定簇;另2株同时具有这两种抗原决定簇。  相似文献

近 10 a来用于抗单纯疱疹病毒 (HSV- 1和 HSV- 2 )的无环鸟甙及其它核甙衍生物已出现抗药性。为寻找新的药物 ,对金线吊乌龟中 49种生物碱的抗 HSV活性进行了研究。材料采用猴肾脏细胞 ,方法采用噬斑法。结果 :金线吊乌龟的甲醇提取物在 10 0 μg/ m L 浓度时可完全抑制 HSV- 1,水提取物也有显著抗 HSV- 1作用。从甲醇提取物中分离得到的 49种生物碱中有 17种具有抗 HSV- 1活性 ,包括 13种二苄基异喹啉生物碱 ,1种原小檗碱型生物碱 ,2种吗啡烷型生物碱和 1种 proaporphine生物碱 ,而苄基异喹啉型和莲花烷型生物碱则无此活性。其中 ,…  相似文献

目的 探讨目前单纯疱疹病毒感染的临床特点及在疾病发生、发展过程中的作用.方法 用单克隆抗体(McAb)酶联免疫吸附试验(ELISA)双抗体夹心法进行HSV型抗原检测.结果 1009例妇女生殖器炎症标本,检出单纯疱疹病毒2型(HSV-2Ag)阳性标本268例,阳性检出率为26.58%;宫颈糜烂、淋病、滴虫性和霉菌性阴道炎HSV-2Ag阳性检出率分别为11.69%、5.45%、6.54%和2.88%.将宫颈糜烂程度分为3度进行检测,结果Ⅱ°和Ⅲ°的HSV-2Ag检出率分别是Ⅰ°的2.34倍和2.50倍.Ⅲ°宫颈糜烂所检出的21例HSV-2Ag阳性标本有1例为宫颈癌患者.结论 提示上述疾病均可感染或混合感染HSV,HSV又可能是疾病发生和发展的原因之一;宫颈病变程度与HSV-2Ag阳性检出率正相关;HSV-2的感染与宫颈癌的发生关系密切.  相似文献

运用杂交瘤技术,制备出7株分泌抗人轮状病毒短型S_2株单克隆抗体的杂交瘤,其中6株分泌IgG_(2a),1株分泌IgG_(2b)抗体。7株杂交瘤培养上清的免疫荧光抗体(IFA)滴度为1:32~256,腹水IFA滴度为1:8000~25600。双夹心ELISA腹水抗体滴度达10~(-4)~10~(-6)。7株McAb均与HSV、CMV、RSV、ADV等其它常见病毒无交叉反应。7株McAb均能与HRV长型Wa株反应,滴度为10~(-3)~10~(-6)。将其中二株或数株McAb混合后用ELISA检测其相应的OD值较每一单株为高,但相加指数(AI)值均在10%左右。结果表明,7株McAb均属RV群特异性抗体,但各自作用的抗原决定簇可能稍有差异。  相似文献

以HSV-1(F株)或HSV-2(G株)免疫血清结合SPA,然后用于交叉吸收HSV-2或HSV-1抗原,经吸收的抗原作ELISA,目的是检测HSV型特异性抗体。在未经吸收的抗原反应中,八份兔抗血清的同型反应与交叉反应差别不大,其抗体效价比值在2倍以内,而经交叉吸收的抗原反应中,抗体以同型反应为主,其抗体效价比值都在4倍以上,最高达16倍。经多次交叉吸收抗原后,抗体的交叉反应进一步降低,但其同型反应也有所降低。  相似文献

在Ⅲ型Polio病毒McAb的研究中,用ELISA法筛选出60株阳性杂交瘤细胞系,其中11株中和试验阳性。ELISA试验型特异的McAb有10个,中和试验型特异的McAb有5个。McAb C_(36)和C_(50)只能与疫苗株和类疫苗株起中和反应而不能与非类疫苗株病毒起反应。因而这二株McAb具有区分Ⅲ型Polio野毒株与疫苗株相关病毒株的能力。另外应用McAb初步分析了17株Polio Ⅲ型病毒的抗原性。  相似文献

将我国HFRS各疫区不同宿主(包括家兔、家猫)分离的病毒,苏联棕背(鼠平)分离株,北美草原田鼠分离株和日本HFRS病毒代表株,经HFRS病毒Hantaan-76-118株、SR-11株和A_9株、R_22株多种McAb分析,可分为五型:姬鼠型、大鼠型、棕背(鼠平)型、田鼠型和未定型,后者以从湖南疫区病人分离的H_79。株为代表。根据型特异性McAb的比较分析,姬鼠属和大鼠属来源的极少数毒株的抗原性与宿主来源不相一致。  相似文献

INTRODUCTION: This study was performed to compare nested-polymerase chain reaction (PCR) with viral culture as a diagnostic tool for genital herpes simplex virus (HSV) infection in a sexually transmitted infection (STI) clinic in Singapore. METHODS: 103 consecutive patients presenting with clinical features suggestive of genital herpes were enrolled in the study. Two swabs were obtained from each patient. On one swab, cell culture and typing was performed, and on the second swab, nested-PCR was performed and the infecting viral type was determined by using type-specific primers. RESULTS: 63 patients (61.2 percent) had a positive PCR test for HSV. Of these, 13 patients (20.6 percent) had HSV type 1 (HSV-1), 50 patients (79.4 percent) had HSV type 2 (HSV-2). HSV-1 and HSV-2 were detected by viral culture in only six patients and 24 patients, respectively. The sensitivity of cell culture compared to PCR was 46.1 percent for HSV-1 infection and 48 percent for HSV-2 infection. PCR further detected an additional 52.4 percent of HSV cases. The specificity of PCR was 100 percent. CONCLUSION: Nested-PCR has been shown in this study to be an effective diagnostic and typing method for HSV infection in a STI clinic in Singapore with its higher sensitivity and specificity to routine viral isolation in cell culture.  相似文献

Four McAbs (1A7E7, 2F9D4, 2G3H5, 1A7H12) against outer membrane antigens of L. interrogans serovar Lai strain 017 were produced by hybridoma technique. McAb 1A7E7 was identified to be IgG2a, the rest IgG1 by immunodiffusion. Outer membrane antigens of three pathogenic leptospiral strains (017, 601, 156) and two non-pathogenic leptospiral strains (Patoc I, 3055) were analysed by SDS- polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting with McAbs. It was found that McAb 1A7E7 recognized specifically the 42kd antigenic band of strain 017; McAb 2F9D4 the 42kd antigenic bands of strain 017, 601, 156, and McAbs 2G3H5 and 1A7H12 the 31 kd antigenic bands of 017, 601, 156. Further study of antigenic properties possessed by pathogenic leptospira may provide some new clues to look for specific diagnostic and protective antigens.  相似文献

目的分析由各脏器分离出的柯萨奇B3病毒(CVB3)抗原决定簇的异同,并探讨其与致病性的关系. 方法利用自制的26种柯萨奇B3病毒mAb(CVB3- McAb)与心脏、肝脏、肾脏及肺脏分离出的CVB3株进行免疫荧光分析,观察各分离株抗原决定簇的变化,并将各分离株再次注入动物体内,观察心、肝、肾及肺的病理改变. 结果 CVB3在不同器官的细胞中繁殖后,其抗原决定簇发生改变, CVB3-McAb 2A4, 2B2, 2D3, 2D4与肝、肺、心、肾分离株均发生反应;26种CVB3-McAb中有4种不与心脏分离株反应;有15种不与肝脏分离株反应;有21种不与肺脏分离株反应;有11种不与肾脏分离株反应. 且各分离株对器官的致病程度亦有差异. 结论 CVB3各分离株抗原决定簇有差异;病毒抗原决定簇的不同是影响其致病性的重要因素之一.  相似文献

We reported the production of monoclonal antibodies (McAbs) against chorionic gonadotropin hormone (CG) receptor by fusing spleen cells of BALB/c mice which had been immunized by purified bacteria (Pseudomonas maltophilia) CG receptor with mouse myeloma line SP2/0. Four hybridoma cell lines secreting CG receptor McAbs were obtained (ED490 DG390, AB890 and GE590). The titers of specific antibodies of both mice ascites and culture supernatant were 10(-2)-10(-6) and 1-10(-2) respectively, by solid phase ELISA. Double-Immunodiffusion test showed that the McAb GE590 was IgG1, and the McAbs ED490, DG390 and AB890 were IgG2b Immunoprecipitation indicated that 125I-HCG could bind the HCG receptor which had reacted with McAbs ED490, AB890 and DG390, suggesting that they may recognize the receptor with different antigenic determinant. Interaction of McAb GE590 with the receptor showed that the increased concentration of GE590 was in inverse proportion to the amount of 125I-HCG binding receptor, indicating that both the McAb and 125I-HCG could recognize a common site of receptor and that increased concentration of McAb GE590 may induce some change in conformation and structure of the receptor. Our study suggested that these McAbs may be used for studying structure of CG receptor.  相似文献

Thirty-five strains of HFRS virus isolated from the patients andseveral host animals in various areas in China were analyzed by IFATwith 10 McAbs specific for HFRS virus.It has been proved that thereare antigenic differences among the strains.The HFRS virus strainsrevealed nine different reactions for the McAb,showing that there are atleast nine different antigenic determinants,including group-,type-andstrain-specific.The analytical result shows that antigenic differencesamong the HFRS virus strains are mainly related to difference in the hostanimals.  相似文献
