目的 探讨免疫活性细胞和免疫调节因子对骨髓间充质干细胞(mesenchymal stem cells,MSCs)中血红素加氧酶-1(HO-1)的影响.方法 体外培养人骨髓间充质干细胞,在IFN-γ及PHA活化的T细胞刺激下,应用RT-PCR方法检测HO-1 mRNA的表达情况.应用流式细胞术分析细胞凋亡情况.结果 正常骨髓间充质干细胞中HO-1 mRNA在IFN-γ及活化的T细胞刺激后表达下调.流式细胞术结果显示,在IFN-γ锌原卟啉-Ⅸ(Znpp-Ⅸ)、IFN-γ+Znpp-Ⅸ的刺激下,间充质干细胞出现明显的凋亡,凋亡率分别为(56.50±0.16)%、(56.85±2.27)%、(82.53±2.65)%,较正常MSCs组显著增高[(7.56±1.43)%,P<0.05].结论 免疫活性细胞及免疫调节因子使骨髓间充质干细胞中HO-1 mRNA表达下凋,进而促进MSCs凋亡.  相似文献

小鼠肝癌细胞(H22)源外泌体的体内抗肝癌作用研究   总被引:2,自引:1,他引:1  
目的在荷瘤动物模型观察小鼠肝癌细胞(H22)来源的外泌体(exosomes)激发宿主抗肝癌免疫应答的效应。方法用自行分离纯化的小鼠肝癌细胞H22源外泌体免疫小鼠,然后给小鼠皮下注射H22肿瘤细胞,观察肿瘤的生长状况。用MTT法检测小鼠脾细胞增殖情况和细胞毒活性;用免疫组织化学法检测肿瘤组织中CD4^+、CD8^+T淋巴细胞浸润情况。结果该外泌体使肿瘤出现时间延迟,肿瘤生长缓慢,小鼠生存率提高。促进脾淋巴细胞增殖并增强其细胞毒活性,促进CD4^+和CD^8+T细胞活化。结论小鼠肝癌细胞(H22)源外泌体能诱导抗肿瘤应答,有效地诱导特异性的杀伤肿瘤细胞活性,有针对亲本细胞的免疫保护作用。  相似文献

目的:探讨骨髓间充质干细胞(MSCs)体外对异体外周血T淋巴细胞的抑制作用及机制.方法:从骨髓中分离培养MSCs,反复贴壁进行纯化;用尼龙棉柱法纯化异体外周血T淋巴细胞;观察MSCs对植物血凝素(PHA)刺激下T淋巴细胞增殖的影响,用ELISA方法检测IFN-γ,IL-4水平变化.结果:MSCs对PHA诱导的T淋巴细胞增殖有抑制作用,并呈剂量依赖性;同时MSCs亦可抑制T淋巴细胞的IFN-γ分泌,但可促进IL-4的分泌.结论:MSCs对异体外周血T淋巴细胞的增殖有抑制作用,其作用可能与其影响T淋巴细胞分泌IFN-γ和IL-4有关.  相似文献

目的研究确立全骨髓贴壁法培养小鼠骨髓间充质干细胞体外培养和优化组合最佳实验条件,建立一个简便易行的小鼠骨髓间充质干细胞体外扩增方法.方法观测不同周龄小鼠、不同浓度胎牛血清和不同种植密度条件下的细胞增殖活性.光镜下观察最佳实验条件培养骨髓间充质干细胞的形态.流式细胞检测细胞周期和表面抗原标志.结果 4~6周龄小鼠、5%~10%的胎牛血清和5~8×107/L的细胞种植密度最适宜MSCs的体外培养,细胞形态、表面抗原标志和细胞周期检测均具有骨髓间充质干细胞的特征.结论本实验室优化选择的培养条件可成功分离纯化、扩增小鼠骨髓间充质干细胞,为下一步深入研究奠定基础.  相似文献

目的探讨间充质干细胞株C3H10T1/2对小鼠骨髓来源树突状细胞(dendritic cells,DCs)体外分化、成熟的影响。方法采用密度梯度离心法分离小鼠骨髓细胞,在mGM-CSF和mIL-4刺激下获得大量未成熟DCs。将DCs和C3H10T1/2细胞共培养2d,同时加入LPS刺激,分别通过倒置显微镜、流式细胞仪,混合淋巴反应以及ELISA检测DCs的成熟。结果经C3H10T1/2细胞体外共培养的DCs,形态观察呈散在分布,细胞边缘圆滑,流式细胞仪检测显示其低表达CD11c、MHC-Ⅱ、CD80、CD86、CD40;共培养组的DCs刺激同种异型小鼠脾细胞增殖的能力在各个浓度均明显低于对照组(P〈0.05或0.01);ELISA检测培养上清中IFN-γ和IL-10的浓度也明显低于对照组(P〈0.05或0.01)。结论间充质干细胞株C3H10T1/2可以明显抑制小鼠骨髓来源DCs的体外成熟。  相似文献

目的:观察小鼠骨髓间充质干细胞(mesenehymal stem cells,MSCs)移植对H22原位肝癌移植小鼠生存期的影响,探讨小鼠骨髓MSCs在肝癌微环境中是否向肝细胞分化和可能的抗肿瘤机制.方法:体外利用贴壁培养法联合免疫磁珠阴性分选CD45-、CD11b-细胞法制备BALB/c小鼠骨髓MSCs,流式细胞仪行细胞表面标志鉴定,采用绿色荧光染料羧基荧光素二醋酸盐琥珀酰亚胺酯[5(6)-carboxyfluoresce indiacetate N-succinimidyl ester,CFSE]对其标记后备用.随机取12只8周龄BALB/c小鼠,肝脏原位注射法建立小鼠H22原位肝癌移植模型,建模1周后随机分为MSCs移植组和对照组,开腹亢视下分别注射入肝癌组织和/或同一肝叶的正常肝组织内.观察荷瘤小鼠生存期,利用免疫荧光法和激光共聚焦技术检测小鼠肝组织白蛋白表达,并行常规组织学检查.结果:MSCs移植组平均生存期为25 d(95%可信区间:22~28 d),对照组平均生存期为21 d(95%可信区间:20~23 d),但组间生存期差异无统计学意义(P=0.0713).激光共聚焦显微镜观察到在肿瘤边缘和瘤体内可见CFSE标记细胞有白蛋白表达;另与对照组相比,MSCs移植组肝癌组织内出现大片坏死.结论:小鼠骨髓MSCs可在原位肝癌移植小鼠模型的肝脏定植,并且能分化为具有肝细胞功能的肝细胞样细胞,同时可能诱发肿瘤细胞坏死,机制有待进一步研究.  相似文献

目的:探讨体外间充质干细胞(MSCs)对BALB/c鼠T,B细胞的免疫调节作用及可能机制.方法:从5~6周龄BALB/c鼠骨髓中分离培养MSCs并鉴定其纯度.用EZ-SepTM Mouse 1X分离异体BALB/c鼠脾脏淋巴细胞,分别在刀豆球蛋白A(ConA)、脂多糖(LPS)刺激下,用不同比例MSCs处理,MTT法检测淋巴细胞的增殖,流式细胞术检测T,B细胞CD69,CD28和CD86的表达和凋亡,实时定量PCR测定细胞因子IL-10和IFN-γ的mRNA水平.结果:MSCs明显抑制T,B细胞增殖和T细胞的凋亡,促进IFN-γ、抑制IL-10的mRNA表达(P<0.01),并且随着MSCs数量的增加而更加明显.MSCs对T,B细胞CD69,CD28和CD86的表达无显著影响.结论:MSCs对BALB/c鼠T,B细胞有免疫负调节作用,但机制复杂,可能与协同抑制细胞增殖和凋亡、干预相关细胞因子mRNA的表达有关.  相似文献

目的:在荷瘤 BALB/c 小鼠观察热休克小鼠肝癌细胞(H_(22))源 Exosomes(HS-Exo) 激发宿主抗肝癌免疫应答的效应.方法:用自行分离纯化的小鼠肝癌细胞 H_(22) 源 Exosomes 及 HS-Exo 免疫小鼠,然后给小鼠皮下接种 H_(22) 肿瘤细胞,观察肿瘤的生长状况.用MTT法检测小鼠脾细胞增殖情况和细胞毒活性;用免疫组织化学法检测肿瘤组织中CD_4~+、CD_8~+T淋巴细胞浸润情况.BALB/c 小鼠皮下接种 H_(22) 细胞,肿瘤长至可触及大小后,分别注射Exosomes及HS-Exo进行治疗,观察各组肿瘤大小及其小鼠存活期.结果:与对照组 Exosomes 相比,HS-Exo 能使肿瘤出现时间延迟,肿瘤生长缓慢(P<0.05);更能促进脾淋巴细胞增殖并增强其细胞毒活性(P<0.05),能更有效的诱导 CD_4~+ 和 CD_8~+T 细胞活化(P<0.05);更为显著的抑制了小鼠的肿瘤生长(P<0.05).结论:HS-Exo 具有更强的免疫活性,能更好的诱导小鼠抗 H_(22) 肝癌作用,有针对亲本细胞的免疫保护作用,并且具有更为显著的肿瘤治疗作用.  相似文献

目的:观察并比较常用不同品系H22肝癌小鼠中医证候的特征与差异。方法:将KM、BALB/c、ICR、C57共4种品系小鼠各30只,分别随机分成正常对照组10只,肝癌组20只。肝癌组小鼠右腋下接种H22细胞造成荷瘤小鼠模型,观察出瘤后各品系肝癌小鼠生存率差异,采用小鼠四诊工作站及其方法,检测小鼠证候的发生与演变。结果:4种品系小鼠腋下接种后均形成实体瘤。在生存率方面,早期各品系肝癌小鼠无明显差异;中晚期差异大,BALB/c肝癌小鼠最低,ICR与KM肝癌小鼠其次,C57肝癌小鼠最高。瘤体积比较,KM肝癌小鼠增长最快,ICR肝癌小鼠增长最慢。去瘤体质量方面,中晚期各荷瘤小鼠呈现下降趋势,ICR小鼠下降最明显,其次是BALB/c与KM小鼠,C57小鼠去瘤体质量变化小。另外,不同品系肝癌小鼠邪毒证与气虚证贯穿疾病始终,且程度逐渐加重,阴虚证与阳虚证在中晚期发生且逐渐加重,其中BALB/c与ICR肝癌小鼠气虚、阴虚、阳虚最严重。结论:KM、BALB/c、ICR、C57等不同品系H22肝癌小鼠均会自发形成证候,KM小鼠最适合于肝癌证候与辨证论治的研究。  相似文献

重组人白细胞介素-1α(rhI1-1α,IL-1)对C3H和BALB/c小鼠的辐射防护作用有明显的种属差异。在C3H小鼠产生防护作用的IL-1剂量显著高于BALB/c小鼠的剂量.IL-1能增加BALB/c和C3H小鼠早期多能造血干细胞(CFU—S_(12))的数目,但C3H小鼠的CFU—S_(12)对IL-1的反应性低于BALB/c小鼠。BALB/c小鼠的骨髓基质细胞层在无TL-1存在时能很好地维持骨髓细胞的生长,而C3H小鼠的骨髓基质细胞层需有IL-1的存在才能维持骨髓有核细胞的生长。上述事实提示,IL-1对C3H和BALB/c小鼠的辐射防护作用存在种属差异。  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献

目的 探讨猪肺磷脂注射液联合经鼻持续气道正压通气(NCPAP)对呼吸衰竭早产儿的临床疗效及肌酸激酶同工酶活性(CK-MB)的影响.方法 选取呼吸衰竭早产儿80例,分为观察组和对照组各40例.对照组采用NCPAP给氧治疗,观察组给予NCPAP给氧联合猪肺磷脂气管内给药.观察两组患儿治疗前及治疗12h、24 h后PaO2、PaCO2、血氧饱和度(SaO2)、pH的变化情况,检测治疗前及治疗5d后血清CK-MB水平;评估两组患儿的临床治疗效果.结果 两组患儿PaO2、PaCO2、SaO2、pH比较,差异均有统计学意义(P<0.05),其中观察组治疗后的PaO2、SaO2、pH均高于对照组,PaCO2则低于对照组.两组的PaO2、SaO2、pH均随观察时间延长而升高(P<0.05),PaCO2均随观察时间的延长而降低(P<0.05).观察组治疗有效率为87.5%,显著高于对照组的70.0% (P <0.05).治疗5d后两组患儿血清CK-MB水平均较前降低(P<0.05),且观察组明显低于对照组(P<0.05).结论 猪肺磷脂注射液气管内给药联合NCPAP可以显著降低呼吸衰竭早产儿CK-MB的含量,提高治疗有效率,起到很好的呼吸循环支持作用.  相似文献

Evidence obtained from randomized controlled trials (RCTs) has been generally accepted as the gold standard in the evaluation of clinical effectiveness. Readers need to understand the trial design, implementation, results, analysis and interpretation, so as to fully Jnderstand the results of RCTs. Thus, the investigators of RCTs have to report these items in a complete, accurate and clear manner. Since 1998, we have conducted several evaluations on the reporting quality of RCTs published in Chinese journals on traditional Chinese medicine (TCM) and results have shown that there is an urgent need for higher quality RCTs on TCM.  相似文献

Ankylosing spondylitis is a chronic and progressive disorder with inflammation mainly involving the central axis joints. It mainly affects the cervical spine and the lumbosacral area, with the pathogenesis closely related to the kidney and the Governor Vessel (GV). TCM holds that the syndrome is deficiency in origin and excess in superficiality, which is due to insufficiency of the kidney, deficiency of GV, and blocking of the channels with the invasion of exogenous evil, leading to poor circulation of qi and blood and malnutrition of the bones, muscles and joints. The TCM method of tonifying the kidney and strengthening GV to regulate circulation of qi and blood and check the arthralgia pain should be adopted, with the Kidney-Tonifying and GV Strengthening Decoction (益肾强督汤) prescribed.  相似文献
