人血浆中盐酸氟桂嗪HPLC测定及其药代动力学

建立了盐酸氟桂嗪的ＨＰＬＣ测定法。血浆样品碱化后用正已烷提取，以ＭｉｃｒｏＰａｃＭＣＨ－５为固定相，６５％甲醇，３５％水（含０．０２ｍｏｌ／Ｌ四丁基溴化铵，ｐＨ３．０）为流动相，紫外检测波长２５４ｎｍ。最低检出浓度为５．０ｎｇ／ｍｌ，线性范围１０．０～２００．０ｎｇ／ｍｌ（ｒ＝０．９９９８，ｎ＝６），平均回收率为（９７．５５±２．６８）％（ＲＳＤ２．１３％。用本法测定了８名健康男性志愿者分别服用杨森产盐酸氟桂嗪胶囊和新昌产盐酸氟桂嗪片剂的血药浓度。结果表明，其血药浓度－时间曲线均符合一房室模型。新昌产盐酸氟桂嗪片的主要药代动力学参数Ｔ１／２Ｋａ＝（０．７７±０．１１）ｈ，Ｔ１／２Ｋｅ＝（５．０７±０．５２）ｈ，Ｔｍａｘ＝（２．７９±０．２４）ｈ，Ｃｍａｘ＝（９５．００±４．５２）ｎｇ／ｍｌ，ＡＵＣ＝（９９７．６±６２．５）ｎｇ·ｈ／ｍｌ，相对生物利用度为９６．４％。     

RP—HPLC测定人血浆中洛伐他汀浓度及药动学研究

用ＲＰ－ＨＰＬＣ测定人血浆中洛伐他汀浓度。以甲醇－水（８３∶１７，Ｖ／Ｖ）为流动相，色谱柱采用ＨＹＰＥＲＳＩＬＢＤＳ－Ｃ１８（５μｍ）不锈钢柱，紫外２３７ｎｍ波长检测。血浆样品经环己烷－异丙醇（９５∶５）萃取浓缩后进样测定。洛伐他汀浓度在２．５～８０ｎｇ／ｍｌ范围内线性良好，ｒ＝０．９９６８。测定含洛伐他汀２０．０ｎｇ／ｍｌ的血浆样品，其日内（ｎ＝７）及日间（ｎ＝７）的ＲＳＤ分别为９．８％和８．５％。回收率平均为（１０１．３±５．５）％。测定了１０名健康志愿者单次ｐｏ洛伐他汀片剂８０ｍｇ后不同时间的血药浓度，计算了相应的药动学参数。     

同位素示踪沉淀法测定神经生长因子血药浓度

目的 建立测定小鼠血浆神经生长因子（ＮＧＦ）浓度的同位素示踪沉淀法。方法 采用放射性同位素（＾１２５Ｉ）标记示踪法结合三氯醋酸（ＴＣＡ）沉淀法测定ＮＧＦ的浓度。结果 本方法的最低检测浓度为２．０ｎｇ／ｍｌ,日内、日间变异系数（ＣＶ）均小于１０％,血浆中ＮＧＦ在１２．５－４００ｎｇ／ｍｌ浓度范围内均呈良好的相关性,相关系数ｒ＝０．９９９０,血浆中ＮＧＦ的平均回收率为９５．２１％,。结论 同位素标记示     

反相高效液相色谱法测定血浆中普萘洛尔的浓度

以硫酸奎宁为内标，反相ＨＰＬＣ－ＵＶ法定量测定血浆中普萘洛尔的含量。固定相为Ｓｐｈｅｒｉｓｏｒｂ５ＯＤＳ（２），流动相为甲醇－水－０．２ｍｏｌ／Ｌ磷酸二氢钾（１００：１５：１）。紫外检测波长为２３２ｎｍ。检测浓度可低达１０ｎｇ／ｍｌ。线性范围为１０～２００ｎｇ／ｍｌ（ｒ＝０．９９９７）。本法灵敏，准确，简便，可用于血浆普萘洛尔测定及其药代动力学研究。     

高效液相色谱法测定奥美拉唑血浆药物浓度

用反相高效液相色谱法测定人体血浆中奥美拉唑的浓度。采用国产填料（ＹＷＧ－Ｃ１８，１０ｕｍ）反相色谱柱，１．０％三乙胺甲醇：水（６０：４０，Ｖ：Ｖ）为流动相，心律平为内标。紫外检测波长为３０２ｎｍ。方法回收率分别为９８．０３％（５０ｎｇ／ｍｌ）、１０１．５５％（４００ｎｇ／ｍｌ）和１０３．１３％（１０００ｎｇ／ｍｌ），相应变异系数（ＣＶ）分别为２．９６％（ｎ＝３）、１．９２％（ｎ＝３）和１．０４％（ｎ＝３）。本方法最小检测量为３ｎｇ，最低检测血药浓度为１０ｎｇ／ｍｌ。至少在５０ｎｇ／ｍｌ至１０００ｎｇ／ｍｌ范围内呈良好线性关系。本方法专一性强、灵敏度高、准确、简便、快速。     

HPLC法测定经皮渗透接受液中双氯灭痛的浓度

采用ＲＰ－ＨＰＬＣ以４＇－羟基－５－氯双氯灭痛为内标，测定了经皮渗透接受液中双氯灭痛的含量。考察了流动相组成、ｐＨ值等对双氯灭痛色谱行为的影响情况。方法简便准确，回收率为９８．４４％～１０２．３％，ＲＳＤ＜２．４％，双氯灭痛浓度在０．５～１６μｇ／ｍｌ范围内，线性关系良好（ｒ＝０．９９９９），最低检测限５０ｎｇ／ｍｌ。色谱条件为：Ｃ（１８）Ｈｙｐｅｒｓｉｌ（５μｍ，１００ｍｍ×４．６ｍｍ）不锈钢柱，甲醇ｐＨ７．０磷酸盐缓冲液（０．０５ｍｏｌ／Ｌ）（５６：４４）作流动用，检测波长２８０ｎｍ，进样量２０μｌ。     

HPLC法测定人血浆中维拉帕米浓度

建立了高效液相色谱测定维拉帕米血药浓度的新方法。血浆样品用正己烷－乙醚（１：１，Ｖ／Ｖ）提取，以Ｚｏｒｂａｘ Ｓｉｌｉｃａ为固定相，０．０５ｍｏｌ／Ｌ磷酸二氢铵－乙腈－甲醇－三乙胺（５０：２５：２５：０．２５，Ｖ／Ｖ）为流动相，紫外２３２ｎｍ处测定。采用外标法峰高定量。血药浓度在２．５￣４００．０ｎｇ／ｍｌ线性关系良好，ｒ＝０．９９９６。最低检测浓度为２．０ｎｇ／ｍｌ。回收率达９５．１１％￣１０１     

大鼠纹状体灌流谷氨酸对局部羟自由基生成的影响

采用脑区微灌流系统和水杨酸为捕捉剂的ＨＰＬＣ羟自由基检测法，观察到大鼠纹状体灌流０．１ｍｍｏｌ／Ｌ谷氨酸１５～３０ｍｉｎ内，灌流液中羟自由基和水杨酸反应生成的衍生物２，５－ＤＨＢＡ的浓度为３１０．４±１７１．８ｎｇ／ｍｌ（ｘ±ｓ，ｎ＝５）较谷氨酸灌流前的８４．８±３７．０ｎｇ／ｍｌ（ｘ±ｓ，ｎ＝５）含量显著增高（Ｐ＜０．０１）．而给予氯胺酮０．６ｍｇ／ｍｌ加灌组，谷氨酸灌流１５～３０ｍｉｎ内的２，５－ＤＨＢＡ浓度为１６５．４±８８．１ｎｇ／ｍｌ（ｘ±ｓ，ｎ＝５）。结果提示纹状体细胞外谷氨酸含量增加能使局部羟自由基生成增加且能被氯胺酮部分抑制。     

HPLC测定人血浆中罗痛定浓度及药代动力学研究

建立了测定人血浆中罗痛定药物浓度的高效液相色谱法。采用ＹＷＧＣ１８色谱柱（２５０ｍｍ×４．６ｍｍ），以甲醇－水－三乙胺（７０∶３０∶０．３，ｖ／ｖ／ｖ，ｐＨ６．３）为流动相，维拉帕米为内标，用紫外检测器在２８１ｎｍ处检测。血浆样品用正己烷－正丁醇混合液（１２∶１）在碱性条件下提取。血药浓度在２．５～１０００ｎｇ／ｍｌ范围内呈线性关系，ｒ＝０．９９９１，最小检测浓度１．５ｎｇ／ｍｌ。日内、日间ＲＳＤ分别＜６．９％，＜８．７％。方法回收率＞９２．１％。应用该法研究了五名健康志愿者口服罗痛定片后的药代动力学。     

HPLC法同时测定人血浆中四种氟喹诺酮药物的血药浓度

研究了用ＨＰＬＣ同时测定人血浆中氧氟沙星、诺氟沙星、洛美沙星和环丙沙星血药浓度的方法。采用ＳｐｈｅｒｉｓｏｒｂＣ（１８）色谱柱（２５０ｍｍ×４．６ｍｍ，５μｍ），以甲醇－０．００８ｍｏｌ／Ｌ磷酸盐缓冲液－０．５ｍｏｌ／Ｌ四丁基溴化铵（１６：７５：１．４，ｐＨ２．６）为流动相，检测波长２８０ｎｍ，血浆样品用乙腈沉淀蛋白后进样，简便快速，回收率稳定，对氧氟沙星，诺氟沙星，洛美沙星，环丙沙星四种氟喹诺酮药物检测的线性范围分别为：０．１０～５．００μｇ／ｍｌ；０．１１～５．４０μｇ／ｍｌ；０．１３～５．００μｇ／ｍｌ；０．１０～１０．００μｇ／ｍｌ。其方法检测限为：２０ｎｇ／ｍｌ；２０ｎｇ／ｍｌ；２５ｎｇ／ｍｌ及４０ｎｇ／ｍｌ，回收率均高于８６％。应用本法观察了健康人口服氧氟沙星和环丙沙星后的血药浓度变化。     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
