生物发光法测定ATP制剂含量

本文采用虫荧光素－虫荧光素酶生物发光法测定ＡＴＰ制剂含量，在１－２０μｇ／ｍｌ范围内ＡＴＰ浓度和发光强度呈线性。ＣＶ％４．５，平均回收率１０２％，方法简便，结果准确。     

顺铂诱导肺腺癌H1650细胞线粒体氧化应激损伤的实验研究

目的:研究顺铂诱导肺腺癌H1650细胞氧化应激致线粒体功能损伤。方法:用不同剂量的顺铂(0、0.5、1.0、2.0μg/mL)处理肺腺癌H1650细胞后,分别采用荧光探针DCFH-DA和JC-1检测细胞内活性氧(reactive oxygen species,ROS)及线粒体膜电位(mitochondrial membrane potential,MMP)水平,荧光素酶测定细胞内三磷酸腺苷(adenosine triphosphate,ATP)浓度。结果:顺铂处理后,H1650细胞内的ROS含量在5 min时达到最高水平,之后逐渐下降,对照组(培养液不加顺铂)内的ROS含量没有变化。不同浓度顺铂处理细胞24 h后,随着顺铂浓度的增加,细胞的绿色荧光强度逐步增加,红色荧光强度逐步减弱,表明细胞内MMP水平随顺铂浓度的增加而降低。ATP含量测定显示,随着顺铂浓度增加,ATP含量逐步减少。结论:顺铂通过诱导肺腺癌H1650细胞产生ROS导致氧化应激,使MMP降低,ATP含量减少,线粒体功能受损,细胞能量供应受限而发挥抗肿瘤作用。     

肿瘤体外药敏试验三磷酸腺苷生物发光法（ATP-TCA）检测技术在化疗中的应用（简报）

肿瘤体外药敏试验三磷酸腺苷生物发光法(ATP-TCA)，是近年发展起来的针对恶性实体肿瘤有较高可评估率的新方法。ATP-TCA法的检测原理：由于ATP是活细胞最基本的能量来源，其水平与活细胞数呈正相关线性关系。用荧光素-荧光素酶试验定量测定细胞内源性ATP的含量，荧光酶在有氧条件下，可以和底物结合后催化ATP转变成ADP，并且释放出荧光(波长562mm)。通过测定内源性ATP的含量所产生的荧光强度，直接反映出细胞的数量。     

氯丙烯对鸡胚脑神经细胞钙稳态与能量合成影响的研究

进一步研究氯丙烯的中毒机理,探讨氯丙烯对神经细胞内钙稳态与能量合成的影响。方法用Fura-2／AM、电子探针X线微区分析、ATP生物发光分析等方法测定氯丙烯染毒后鸡胚脑神经细胞内游离Ca2+、钙百分含量及ATP、ADP、AMP含量的改变。 结果随着氯丙烯浓度的增加,细胞内游离Ca2+浓度、钙百分含量明显增加(P     

心衰患儿红细胞内外钙离子浓度的变化及临床意义

为了探讨心肌细胞内钙离子浓度的变化与心力衰竭发生的关系。方法；原子吸收分光技术测定红细胞内外Ｃａ＾２＋含量。我们对住院的５６例心力衰竭患儿进行了红细胞内及血浆钙离子浓度的测定，并与３０例正常小儿对照。结果：心衰患儿红细胞内Ｃａ６２＋浓度明显升高，血浆Ｃａ６２＋浓度降低，与对照组相比有非常显著性差异。

目的：为了深讨心肌细胞内钙离子浓度的变化与心力衰竭发生的关系。方法：原子吸收分光技术测定红细胞内外Ca2+含量。我们对住院的56例心力衰竭患儿进行了红细胞内及血浆钙离子浓度的测定，并与30例正常小儿对照。结果：心衰患儿红细胞内Ca2+浓度明显升高，血浆Ca2+浓度降低，与对照组相比有非常显著性差异（P＜0．001）。其升高程度与心衰呈正相关（r=0．63，P＜0．01），即心衰越重，心功能越差，红细胞内Ca2+浓度升高亦越明显。而当心衰纠正，心功能恢复后，红细胞内Ca2+浓度则明显下降，血浆Ca2+浓度也恢复正常。而不同病因所致的心衰，其间红细胞内Ca2+浓度升高程度无明显差异。结论：提示钙离子参与了心力衰竭的发生与发展。

生物发光法短时内判断温缺血断肢再植时限的实验研究

目的 :在大量筛选实验的基础上 ,选择一种能短时间内判断温缺血断肢再植时限的方法。方法 :应用沸重蒸水沸法提取、生物发光技术连续测定兔离断肢体骨骼肌 ATP的含量 ,同时对每一标本作病理检查。结果 :温缺血条件下兔离断肢体骨骼肌 ATP含量下降程度与骨骼肌的变性程度呈对应关系 ,当 ATP含量下降 95%以后 ,骨骼肌发生不可逆变性。结论 :应用生物发光法可在短时间内 (约 0 .5h)判断温缺血断肢的再植时限 ,是一种有前途的检测方法。     

老化和被氧化的红细胞内ATP含量的变化

本文用荧光素－荧光素酶生物发光法测定了老化和被氧化红细胞内ＡＴＰ含量的变化，结果表明老化的红细胞和被外源性Ｈ２Ｏ２氧化的红细胞，其内ＡＴＰ含量均明显降低，而麦芽酚对红细胞氧化具有保护作用，能减少氧化红细胞内ＡＴＰ含量的变化。     

生物荧光法检测结核分枝杆菌ATP含量

目的：建立生物荧光技术检测结核分枝杆菌ATP含量的方法并观察几种杭结核药物异烟肼和利福平的作用。方法：结核分枝杆菌标准株H37Rv的菌悬液,经高温裂解,提取ATP,用生物荧光技术检测ATP含量。条件稳定后,观察菌数和ATP浓度的关系,以及加入不同浓度的抗结核药物后1周之内ATP的动态变化。结果：菌液加入裂解液后100℃煮沸8分钟,迅速降至室温,加入荧光素酶,3—5分钟时测荧光值。按上述步骤提取和测定时菌数和ATP浓度成正比关系。含药液体培养情况：随药物浓度增加,ATP量下降,培养至第7天时,药物的敏感性可以初步进行判断。结论：所建立的生物荧光法检测结核分枝杆菌ATP含量,具有简便、快速、敏感、可重复性强等特点,有望成为结核菌体外药敏试验的一种有用的方法．     

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.     

A preliminary screening study on the associated proteins in human psoriasis vulgaris by serum proteomics technologies

Objective：To investigate the optimum screening conditions of associated proteins in human psoriasis vulgaris by serum proteomics technique, and to screen the different expression proteins related with psoriasis vulgaris. Methods：Serum samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 matched healthy persons. Serum albumin IgG was removed by filtering with ProteoExtract Albumin/IgG. After comparative proteomics analysis the different protein spots were identified using 2-DE and MS. Results ：Electrophoresis figures with high resolution and reproducibility were obtained. Three different expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other different proteins expressing from healthy volunteers. Conclusion：The protein expression was different in the serum between the psoriasis vulgaris patients and healthy volunteers. It was hoped that we could find the biomarkers related to psoriasis vulgaris by using proteomics.

Acupuncture Treatment of Chest Bi Syndrome

CASE HISTORY A male,a retired officer of 61 years old,paid his first visit on May 12,2006.The patient stated that 2 weeks before he suddenly got paroxysmal precordial choking pain,with irregular attacks.Each attack would last several seconds to several minutes.The attack was accompanied with heavy oppressed sensation in the precordial region,palpitation,lassitude,and in severe cases,sweating,which was once diagnosed by a hospital as'coronary heart disease'.Recalling the case history,the patient said that several days before he got angry with others because of some affairs in work,and he was not so used to the life pattern since he had just retired from his busy working post.At usual times,he was quick tempered and peevish.In recent days,he had poor appetite,and loose stool.
