The aqueous extract of Vernonia colorata (Willd.) Drake (Composeae) leaves is used by African traditional medicine practitioners as a remedy for the treatment of diabetes. Our previous studies have shown the hypoglycaemic activity of the aqueous extract of Vernonia colorata leaves (300 mg/kg, per os) in normoglycaemic rats. The aim of the present study was to investigate the hypoglycaemic and antidiabetic activity of acetonic and hexanic extracts of the leaves of Vernonia colorata in order to further discriminate the type of extract which provides a better antidiabetic activity. Experiments were performed in normoglycaemic and alloxan-induced diabetic rats. The acetonic extract of the leaves of Vernonia colorata (AELVC) (100 mg/kg, per os) induced a significant decrease of blood glucose in normoglycaemic rats. The glycaemia varied from 4.72+/-0.11 to 3.72+/-0.22 mmol/l (p<0.05, n=5) 3 h after AELVC administration per os. In contrast, the hexanic extract of the leaves of Vernonia colorata (HELVC) increased significantly the glycaemia in normoglycaemic rats. Like glibenclamide, AELVC has an antihyperglycaemic effect in oral glucose tolerance test (OGTT) and alloxan-induced diabetic rats. These results have shown that: (i) AELVC and HELVC have an opposite effect on basal blood glucose in normoglycaemic rats, suggesting that the mechanisms of action of both above-mentioned extracts are different; (ii) AELVC has also an antidiabetic activity in hyperglycaemic rat models.  相似文献

The leaves of Mangifera indica were assessed for antidiabetic properties using normoglycaemic, glucose-induced hyperglycaemia and streptozotocin (STZ) induced diabetic mice. The aqueous extract produced a reduction of blood glucose level in normoglycaemic and glucose-induced hyperglycaemia, but did not have any effect on streptozotocin-induced diabetic mice. The hypoglycaemic effect of the aqueous extract was compared with that of an oral dose of chlorpropamide under the same conditions. The results of this study indicate that the aqueous extract of the leaves of Mangifera indica possess hypoglycaemic activity.  相似文献

The hypoglycaemic effect of Anthocleista vogelii was studied in mice, rats and rabbits. Aqueous extract of the plant obtained by infusion from finely pulverized root was used. The extract (100, 400 and 800 mg/kg) induced significant hypoglycaemic activity in a dose-related fashion at 2 h after oral administration in mice and rats with ED25 of 250 mg/kg and 350 mg/kg respectively. The extract (800 mg/kg, orally) similarly induced statistically significant lowering of blood glucose levels at 8 h in normoglycaemic rabbits. The extract (400 mg/kg and 800 mg/kg, orally) also caused reduction of blood glucose levels in alloxan-induced diabetic animals. The results of this study indicate that the aqueous extract of the roots of Anthocleista vogelii possess favourable hypoglycaemic activity both in normo and hyperglycaemic animals compared to chlorpropamide as a standard.  相似文献

The leaves of Mangifera indica are used as an antidiabetic agent in Nigerian folk medicine. To determine whether or not there is a scientific basis for this use, the effect of the aqueous extract of the leaves on blood glucose level was assessed in normoglycaemic, glucose - induced hyperglycaemic and streptozotocin (STZ)-induced diabetic rats. The aqueous extract given orally (1 g/kg) did not alter the blood glucose levels in either normoglycaemic or STZ-induced diabetic rats. In glucose - induced hyperglycaemia, however, antidiabetic activity was seen when the extract and glucose were administered simultaneously and also when the extract was given to the rats 60 min before the glucose. The hypoglycaemic effect of the aqueous extract was compared with that of an oral dose of chlorpropamide (200 mg/kg) under the same conditions. The results of this study indicate that the aqueous extract of the leaves of Mangifera indica possess hypoglycaemic activity. This action may be due to an intestinal reduction of the absorption of glucose. However, other different mechanisms of action cannot be excluded.  相似文献

Screening of aqueous extract of Phyllantus niruri (PL), Zingiber zerumbet (ZG), Eurycoma longifolia (TA-a and TA-b) and Andrographis paniculata (AP) to determine their blood glucose lowering effect were conducted in normoglycaemic and Streptozotocin-induced hyperglycaemic rats. Significant reduction in blood glucose level at 52.90% was shown when hyperglycaemic rats were treated with 50 mg/kg body weight (BW) aqueous extract of AP. This effect is enhanced when freeze-dried material was used, where 6.25 mg/kg BW gave 61.81% reduction in blood glucose level. In the administration of TA-a and TA-b, positive results in hyperglyacaemic rats were only obtained when 150 mg/kg BW of the aqueous extract was used. No significant reduction in blood glucose level were shown in hyperglycaemic rats treated with PL and ZG at all concentrations used (50, 100 and 150 mg/kg BW). In normoglycaemic rats, no significant reduction was noted when all the same extracts were used.  相似文献

AIM OF THE STUDY: Hypoglycaemic and hypolipidemic properties of the ethanolic and aqueous extracts, respectively, from Chinese juniper (Juniperus chinensis L.) berries were investigated in alloxan-induced diabetic rats. MATERIALS AND METHODS: After oral administration of each extract singly or repeatedly to alloxan-induced diabetic rats, the blood glucose, glutamate-pyruvate transferase (GPT), glutamate-oxaloacetate transaminase (GOT), total cholesterol (TC) and triglyceride (TG) levels were assayed. RESULTS: The blood glucose levels after a single oral administration of the ethanolic extract significantly reduced in a time-dependent manner, which is much faster and more than that of glibenclamide. The blood glucose levels of alloxan-induced diabetic rats treated with the ethanolic extract were reduced to 94, 81%, 66%, 45% and 40% at 1, 3, 5, 7 and 9h, respectively (p<0.05), while the aqueous extract had no effect at all. Repeated oral administration of the ethanolic extract also effectively reduced the GPT value to 58% of the diabetic rats, but slightly reduced the GOT value to 87% of the diabetic rats (p<0.05). On the other hand, the repeated oral administration of aqueous extract effectively reduced the GOT value to 43% of the diabetic rats, without affecting the GPT level. Effects of both extracts on the TC and TG levels were different. There was no significant difference in the TC and TG levels between diabetic control and diabetic groups when repeatedly administered orally with ethanolic extract. On the other hand, the aqueous extract brought down the TC value to 57% and the TG value to 37% of the diabetic control rats (p<0.05). CONCLUSIONS: The results suggested that the ethanolic extract of Chinese juniper berries possesses a potential hypoglycaemic effect while the aqueous extract has a potential hypolipidemic effect.  相似文献

Diabetes mellitus is a chronic metabolic disorder characterized by hyperglycemia. Enicostemma littorale Blume is a small herb and recently we have reported its blood glucose lowering potential in alloxan induced diabetic rats. A single dose of aqueous extract of E. littorale (15 g dry plant equivalent extract per kg) had shown significant increase in the serum insulin levels in alloxan-induced diabetic rats at 8 h. The insulinotropic action of aqueous extract of E. littorale was further investigated using rat pancreatic islets. Extract has the potential to enhance glucose-induced insulin release at 11.1 mM glucose from isolated rat pancreatic islets and was partially able to reverse the effect of diazoxide (0.25 mM). Incubation with Ca(2+) chelator (EGTA) and Ca(2+) channel blocker (nimodipine) did not affect the glucose-induced insulin release augmented by the extract. Above results suggest the glucose lowering effect of aqueous extract of E. littorale to be associated with potentiation of glucose-induced insulin release through K(+)-ATP channel dependent pathway but did not require Ca(2+) influx.  相似文献

The aqueous root extract of Morinda lucida (Rubiaceae) Benth, exhibited potent hypoglycaemic effects in both normoglycemic and alloxan-induced diabetic mice by oral administration. This effect was dose-dependent and more potent than that observed with chlorpropamide (1-(p-chlorobenzene-sulphanyl)-3-propylurea). In the normoglycaemic mice, the extract (140 mg/kg body weight) produced a fall in blood sugar of 32% + 1% while at 280 mg/kg body weight there was a 52% + 2% fall over the same time interval. In alloxan-induced diabetic mice, the reaction time of the extract was longer with a single dose of the extract producing a significant hypoglycaemic effect 4 h after administration. In diabetic mice, a dose of 140 mg/kg body weight of the plant extract caused a fall in blood sugar of 51% + 1% while a dose of 280 mg/kg produced a fall of 60% + 2%.  相似文献

Tap roots of Potentilla fulgens L. traditionally chewed along with betel nut (Areca catechu) and betel leaves (Piper betel), are commonly used by local practitioners for various types of ailments. The crude methanolic extract of the roots was tested for its effects in normoglycemic and alloxan-induced diabetic mice. Hypoglycemic activity was observed to be dose- and time- dependent. The extracts reduced blood glucose level 2 h following administration in both normal and alloxan-induced diabetic mice. In alloxan-induced diabetic mice blood glucose was markedly reduced by 63%, while in normal mice a 31% reduction was observed 24 h after the effective dose of extract was administered. Further, in the diabetic mice a prolonged anti-hyperglycemic action was observed where glucose levels was, found to be significantly low (79%) when compared with control even on the third day. Glucose tolerance was also improved in both normal and diabetic mice. The results were compared against those of insulin, glibenclamide, metformin, and the probable mechanism of action is discussed.  相似文献

Anti-diabetic activity of Picrorrhiza kurroa extract   总被引:3,自引:0,他引:3  
An alcoholic extract of Picrorrhiza kurroa was found to lower blood glucose in basal conditions and after a heavy glucose load in normal rats. Maximum reduction in serum glucose was observed after 2 h at a dose level of 75 mg extract/kg of body weight. P. kurroa extract was also found to reduce the increase of blood sugar in alloxan-induced diabetic rats (43% at 75 mg/kg body weight and 60% at 150 mg/kg body weight). Chronic administration of the extract significantly reduced the blood sugar in alloxan-induced diabetic rats for several days (10 days). The extract was also found to reduce the increased blood urea nitrogen and serum lipid peroxides in alloxan-induced diabetic animals and to inhibit the body weight reduction and leukopenia induced by alloxan administration. These results indicate that P. kurroa extracts are able to ameliorate biochemical damages induced by alloxan in diabetic rats.  相似文献
