Objective: To observe the effect of moxibustion on intestinal propulsion rate, content of serum D-xylose, content of adenosine triphosphate (ATP) and activities of adenosine triphosphatases (ATPase) in jejunum tissues of spleen-deficiency rats, and to explore the effect of moxibustion on the production of ATP and activities of membrane proteins, for analyzing the mechanism of moxibustion in reinforcing the spleen-stomach. Methods: Forty healthy Sprague Dawley rats were randomly divided into four groups, namely group A (blank control group), group B (model control group), group C (moxibustion group) and group D (herbs group). The animal model of spleen-deficiency was established by intragastric administration with Da Huang (Radix et Rhizoma Rhei) infusion. The rats in group C received moxibustion at Zusanli (ST 36), Zhongwan (CV 124 Guanyuan (CV 4), Pishu (BL 20) and Weishu (BL 21). The rats in group D received intragastric administration with 5i Jun Zi decoction. The intestinal propulsion was measured by toner method, and the content of D-xylose in serum was detected by phloroglucinol method. Colorimetry method was used to detect the content of ATP and activities of Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATPase in jejunum tissues. Results: Compared with group A, the intestinal propulsion rate, content of D-xylose in serum, content of ATP, activities of Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATP in jejunum tissues decreased significantly in group B (P〈0.05 or P〈O.O1). Compared with group B, the intestinal propulsion rate, content of D-xylose in serum, content of ATP, activities of Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATPase in jejunum tissues increased significantly in group C and D (P〈0.05 or P〈0.01), while there were no significant differences between group C and group D. Conclusion: Moxibustion can increase the intestinal propulsion rate, content of D-xylose in serum and ATP in jejunum tissues, as well as the activities of Na~-K+-ATPase and Ca^2+-Mg^2+-ATPase of spleen-deficiency rats, which suggests that moxibustion can enhance the motility and absorption functions of small intestine by promoting the production of ATP in intestinal epithelial cells, increasing the content of membrane proteins, and improving transmembrane transport.  相似文献

Objective: To observe the effect of electroacupuncture (EA) on cytochrome c oxidase (COX)activity of hippocampal mitochondria in senescence-accelerated mouse prone 8 (SAMP8) mice, and to explore the EA mechanism on Alzheimer disease (AD) in improving energy metabolic disorder.
Methods: Twelve SAMP8 mice were randomly divided into a model group and an EA group, with six in each group. Six senescence-accelerated mouse resistance 1 (SAMR1) mice were prepared as blank group. Mice in the EA group received EA on Baihui (GV 20) and Yongquan (KI 1), once a day for 7 d as a course, altogether 3 courses with one day intervalbetween two courses. Mice in the model group and the blank group were manipulated and fixed as those in the EA group. After interventions, Morris water maze was employed to test spatial learning and memory ability to evaluate EA effect; spectrophotometry was used to detect the activity of hippocampal mitochondria COX.
Results: Compared with the blank group, mean escape latenciesof the EA group and model group were prolonged significantly in Morris water maze tests (P〈0.01), the residue duration in the former platform quadrant significantly decreased (P〈0.01). Compared with the model group, mean escape latencies on 1 d, 2 d and 3 d of the EA group were significantly reduced (P〈0.05), and those on 4 d and 5 d continued the decreasing tendency (P〈0.01), the residue duration on the former platform quadrant was significantly prolonged (P〈0.05). The COX activity tests showed that, compared with the blank group, COX activities of the model group and the EA group were significantly decreased (P〈0.01); compared with the model group, COX activity of the EA group was significantly elevated (P〈0.01).
Conclusion: It's plausible that EA improves AD learning and memory ability by increasing mitochondria COX activity, protecting the structure and function, and improving energy metabolism.  相似文献

OBJECTIVE: To study the effects of a decoction of Fuzhengzengxiao formula on lung adenocarcinoma regarding the inflammatory protein S100A9 known to enhance cancer cell sensitivity.METHODS: A nude mouse model of human lung adenocarcinoma was established. The mice were randomly divided into four groups using the random number table method: Group Ⅰ, control;Group Ⅱ, treatment with a decoction of the Fuzhengzengxiao formula alone; Group Ⅲ, treatment with radiotherapy alone; and Group Ⅳ, treatment with radiotherapy plus a decoction of Fuzhengzengxiao formula. When the tumor body was 1 cm~3 in diameter, the tumor bearing mice in GroupsⅢ and Ⅳ were irradiated at a single dose of 10 Gy and the tumor inhibition rate was evaluated.The expression of S100A9 was determined using Western blotting and q-PCR(Real-time Quantitative PCR Detecting System). The sensitivity of cells containing RNAi S100A9 to radiotherapy was evaluated using the Click multiple target model,and the cell cycle was analyzed using flow cytometry.RESULTS: Relative to the control group,the expression of S100A9 in the tumors in each treatment group was decreased,especially in Group Ⅳ. The sensitizing enhancement ratio(SER) Dq was 1 after RNAi S100A9; it decreased the surviving fraction after a 2 Gy dose exposure,and also the D_0 and Dq of the tumor cells; in addition, the radiosensitivity of G_2/M cells was significantly increased.CONCLUSION: The decoction of the Fuzhengzengxiao formula downregulated the expression of S100A9 in lung adenocarcinoma cells.  相似文献

Objective To explore the mechanism of Huagan Jiedu decoction in treatment of chronic hepatitis B by testing the influence of the decoction on Serum IL-2, IL-4 and IFN-γ of rat models of chronic hepatic injury. Methods The model of Chronic hepatic injury was established by subcutaneously injecting 40% CCl4-olive oil solution in rats according to the ratio of 3ml per kg. of body weight. 60 healthy rats were divided randomly into a normal group, a model group, a low dose of Huagan Jiedu decoction group, a large dose of Huagan Jiedu decoction group, and a control group (Qingre Jiedu tablet group), 15 cases in each group. The Serum IL-2, IL-4 and IFN-γ were detected in each group separately. Results Compared with the normal group, the Serum IL-2 and IFN-γ were decreased (t=2.401, 2.337, P=0.0349, 0.0378) and the Serum IL-4 was increased (t=2.896, P=0.00861 ) in the model group; Compared with the model group, the Serum IL-2 and IFN-γ were increased (t=2.417, 2.344, P=0.0341, 0.0372, P<0.05; t=3.114, 2.988, P=0.0052, 0.0068) in the large Huagan Jiedu decoction group and the control group, while the Serum IFN-γ was increased (t=3.049, P=0.0062) in the low dose of Huagan Jiedu decoction group and the Serum IL-4 was decreased (t=3.102, 3.017, 2.979, P=0.0061, 0.0065, 0.0069)in both Huagan Jiedu decoction groups and the control group. Conclusion Large dose of Huagan Jiedu decoction has sound therapeutic effects on rats with chronic hepatic injury by decreaseing the content of the Serum IL-4 and increasing IL-2 and IFN-γ, and adjusting th1/th2 balance.  相似文献

OBJECTIVE: To investigate the effect of Danggui Buxue Tang(DBT), a decoction from Traditional Chinese Medicine, on bleomycin-induced pulmonary fibrosis in rats, and to propose the possible underlying mechanism.METHODS: Forty male Sprague-Dawley rats were randomly divided into sham group, model group,prednisone group and DBT group. Pulmonary fibrosis rat model was established by intratracheal injection with bleomycin. Body weight and lung index were monitored. Histopathologic examination and collagen deposition were determined using Hematoxylin and eosin(HE) and Masson's trichrome staining. Immunohistochemistry staining was applied to observe the expression of alpha-smooth muscle actin(α-SMA). m RNA expression of α-SMA,collagen Ⅰ and collagen Ⅲ were measured by realtime fluorescence quantitative PCR(RT-q PCR). Inflammatory cytokines, including tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6) and IL-1β in serum were detected by Enzyme-linked immunosorbent assay. Alkali hydrolysis method was conducted to investigate the content of hydroxyproline(HYP). Transforming growth factor-β1(TGF-β1),Smad3 and plasminogen activator inhibitor-1(PAI-1) protein level were examined by Western blot assay.RESULTS: DBT significantly reduced the severity of bleomycin-induced pulmonary fibrosis and inflammation as indicated by minimizing the lost of weight, and by lowering the levels of lung index, inflammation score, Ashcroft score, collagen volume fraction(%), HYP, α-SMA, collagen Ⅰ, collagen Ⅲ,TNF-α, IL-6, IL-1β, TGF-β1, Smad3 and PAI-1, consistent with the effect of prednisone.CONCLUSION: Our findings suggest that DBT is able to ameliorate the pulmonary fibrosis, the possible mechanism may involve inhibition of pulmonary inflammation and collagen deposition, possibly via suppressing TGF-β1/Smad3/PAI-1 signaling pathway.  相似文献

Background: Huangqi Guizhi Wuwu decoction (HQGZWW) exhibits good effects when administered to treat multiple sclerosis (MS) and its animal model, experimental autoimmune encephalomyelitis (EAE). Understanding the precise mechanism of this decoction is thus important. Based on the findings of our previous study, the aim of the present study was to understand the role of antigen?specific CD8+ T-cells on the pathogenesis of MS/EAE when HQGZWW is administered as treatment. Methods: Myelin oligodendrocyte glycoprotein (MOG) 35-55-induced mice were administered distilled water, prednisone, and high dose or low dose HQGZWW.After purified CD4+ and CD8+ T-cells were stimulated with the MOG35-55 peptide, proliferation and cytokine secretion assays were performed. To establish the adoptive transfer EAE model, naïve mice were injected with MOG35-55 -CD8+ or CD4+ T-cells. Results: Significant improvements in EAE score and pathology were observed in the high dose HQGZWW and prednisone groups. Compared to the low dose HQGZWW and distilled water groups, lower antigen?specific responses, lower levels of interferon-gamma, and higher levels of interleukin (IL)-4 and IL-10 from CD8+ and CD4+ T cells were observed in the high dose HQGZWW and prednisone groups. Finally, the EAE score was observed to be similar between the high dose HQGZWW group and prednisone group; however, this finding was not observed in the low dose HQGZWW group. Conclusion: Our findings suggest that high dose HQGZWW has similar effects on cell proliferation, cytokine secretion, and EAE score to prednisone, while low dose HQGZWW does not have such effect. The protective role of HQGZWW against EAE might thus depend on the Th2 cytokine secretion profile induced by either MOG35-55 specific CD8+ or CD4+ T-cells.  相似文献

OBJECTIVE: To investigate the effect of catgut implantation at acupoints on the expressions of γ-amino butyric acid B receptor(GABAB ) and metabotropic glutamate receptor 1(mGluR1) in the brain stem of rats with spasticity after stroke.METHODS: In total, 60 male Sprague-Dawley rats were randomly divided into three groups: a sham group(n=10), a model group(n=25) and a treatment group(n=25). The rats in both the model group and the treatment group were subjected to middle cerebral artery occlusion to establish a model of focal cerebral ischemia. Rats with limb-spasm met the inclusion criteria. Only the left carotid artery was isolated in sham group rats. Three days after modeling, the treatment group was subjected to catgut implantation at Dazhui(GV 14), Guanyuan(CV 4), and Zhongwan(CV 12). Neurological deficit symptoms were assessed with the Zea-Longa neurological deficit score. The Modified Ashworth Scale(MAS), and isolated muscle tone were used to evaluate spasticity before and after treatment. Immunohistochemistry was applied to determine the expression of GABAB and mGluR1 in the rat brain stem after treatment.RESULTS: After treatment, neural impairment symptoms had significantly improved in the treatment group when compared to the model group(P〈0.05). Both MAS and isolated muscle tone in the treatment group were significantly decreased when compared with the model group(P〈0.05),and were also lower than before treatment. GABAB expression was significantly higher and mGluR1 was lower in the treatment group when compared with the model group(P〈0.01 and P〈0.05, respectively).CONCLUSION: Catgut implantation at Dazhui(GV 14), Guanyuan(CV 4), and Zhongwan(CV 12), can relieve limb spasticity by increasing the expression of GABAB and reducing the expression of mGluR1 in the brain stem of rats after stroke.  相似文献

To compare the acupuncture plus oral administration of Chinese herbal decoction with simple oral administration of Chinese herbal decoction in the treatment of vascular headache. Methods: Sixty two patients were randomly divided into a treatment group (32 cases) and a control group (30 cases). Acupuncture at Baihui (GV 20), Fengchi (GB 20), Shuaigu (GB 8), Xingjian (LR 2), Neiguan (PC 6), Sanyinjiao (SP 6) and Ashi points combined with oral administration of Chinese herbal decoction, was applied in the treatment group, and simple oral administration of Chinese herbal decoction was applied in the control group. Results: The total therapeutic effect in the treatment group was better than that in the control group (P〈0.05). After treatment, the frequency, and duration of the attacks were reduced and shortened, and headache greatly alleviated in both groups (P〈0.01). The alleviation in the treatment group was more obvious than that in the control group (P〈0.05). Conclusion: Acupuncture combined with oral administration of Chinese herbal decoction provided remarkable therapeutic effects in treating vascular headache.  相似文献

Objective

To investigate the protective role of Sijunzi decoction in neuromuscular junction (NMJ) and muscle cell mitochondria ultrastructure; as well as its effects on the amount of adenosine triphosphate (ATP) and the activities of mitochondrial respiratory chain complexes I, II, III, and IV in autoimmune myasthenia gravis rats.

Methods

An experimental autoimmune myasthenia gravis (EAMG) rat model was established by inoculating rats with acetylcholine receptors extracted from Torpedo. Rats were divided into three groups: model, prednisone, and Sijunzi decoction, and were fed physiological saline, prednisone, or Sijunzi decoction, respectively. NMJ and muscle cell mitochondria ultrastructure were observed by transmission electron microscope. The amount of ATP was assessed by high performance liquid chromatography. The activities of mitochondrial respiratory chain complexes I, II, III, and IV was determined using the Clark oxygen electrode method.

Results

In the model group, there were sparse muscle fibers, with decreased mitochondria, and sparse, diffluent, or absent NMJ folds. After intervention with Sijunzi decoction, the above pathology changes were improved: muscle fiber structure was clear and complete; the mitochondria count was higher; and the NMJ structure was close to normal. Gastrocnemius muscle mitochondria in the model group produced significantly less ATP than those in the prednisone group (P<0.01). Conversely, the ATP of Sijunzi decoction group was significantly higher than prednisone group (P<0.01). The activities of gastrocnemius muscle mitochondrial respiratory chain complexes I, II, III, and IV in both the prednisone and Sijunzi decoction groups was dramatically higher compared with the model group (P<0.05). The activities of complexes I and III in the Sijunzi decoction group were significantly higher than those in the prednisone group (P< 0.05), but there was no obvious difference in complex II or IV activities between the two groups (P>0.05).

Conclusion

Sijunzi decoction improved pathological changes in muscle mitochondria and NMJ, enhanced the amount of ATP in gastrocnemius muscle mitochondria, and improved the activities of respiratory chain complexes I, II, III, and IV (especially I and III) of the EAMG rats.  相似文献

[目的]观察四君子汤加味治疗高脂血症眩晕的临床疗效。[方法]36例患者应用四君子汤加味治疗,1剂/d,水煎分2次内服。[结果]治疗1~3个疗程。治愈29例,显效6例,无效1例,总有效率97.22%。[结论]四君子汤加味治疗可明显改善血脂代谢,减轻临床症状,疗效显著。  相似文献

目的:观察四君子汤合痛泻药方在临床上用于治疗腹泻型肠易激综合征的疗效。方法:将66例患者分成两组,其中治疗组36例,服用四君子汤合痛泻药方治疗;对照组30例,服用思密达和思连康治疗。两组均治疗6周后观察各组的疗效。结果:治疗组的总有效率为94.40%,对照组的总有效率为80.80%,治疗组的临床疗效要明显优于对照组(X^2=4.3914,P〈0.05)。结论:四君子汤合痛泻药方用于治疗腹泻型肠易激综合征的临床疗效显著,复发率低,提高了患者的生活质量,值得临床推广应用。  相似文献

目的:探讨加味四君子汤含药血清对肝癌Hep-G2细胞增殖、凋亡、周期的影响及其作用机制。方法:不同浓度加味四君子汤含药血清处理Hep-G2细胞后,采用细胞活性检测试剂盒(CCK-8)检测细胞增殖;Annexin V/碘化丙啶(PI)流式细胞术检测细胞凋亡率和周期;hoechst33342荧光染色观察细胞凋亡形态;免疫印迹法(Western blot)检测细胞中磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)/雷帕霉素靶蛋白(m TOR)信号通路相关蛋白表达水平。结果:加味四君子汤含药血清呈浓度依赖性抑制肝癌Hep-G2细胞的增殖,诱导Hep-G2细胞凋亡,阻滞细胞于G0/G1期。hoechst33342染色后,随着含药血清浓度的增加,Hep-G2细胞核显著呈碎块状致密浓染。同时,加味四君子汤含药血清能够抑制Hep-G2细胞Akt,m TOR,核糖体S6蛋白激酶(S6),真核翻译起始因子4E结合蛋白1(4EBP1)的磷酸化,从而上调Bcl-2相关X蛋白(Bax)和下调细胞周期蛋白(Cyclin D1),B淋巴细胞瘤-2(Bcl-2)的表达。加味四君子汤含药血清与300 nmol·L-1的PI3K/m TOR双重抑制剂VS-5584联合使用具有协同作用,含药血清能增强PI3K/m TOR双重抑制剂VS-5584对Hep-G2细胞中PI3K/Akt/m TOR信号通路靶点Akt和m TOR磷酸化的抑制作用。结论:加味四君子汤含药血清能抑制Hep-G2细胞的增殖,诱导其凋亡,阻滞其于G0/G1期,其机制可能通过阻断PI3K/Akt/m TOR信号通路而实现。  相似文献

目的:观察加味四君子汤联合甲硝唑、叶酸三联疗法对萎缩性胃炎及癌前病变,中医辨证属脾虚血瘀证患者增殖细胞核抗原(PCNA)表达的影响及胃黏膜病理改变与临床疗效。方法:68例脾虚血瘀型胃脘痛,经胃镜检查,病理证实为萎缩性胃炎和(或)肠上皮化生及异型增生患者,随机分为三联治疗组(n=34),给予加味四君子汤100ml 2次/日,联合甲硝唑200 mg 2次/日、叶酸10 mg 3次/日,对照组(n=34)仅给予甲硝唑200mg 2次/日、叶酸10 mg3次/日。以上药物除甲硝唑服用1周外,其他服用至第24周。治疗前后观察临床症状、黏膜病理、PCNA表达。结果:三联治疗组治疗后PCNA表达较对照组减弱(P<0.01),三联治疗组临床症状缓解、病理改变逆转总有效率(79.4%)均优于对照组(39.4%)(P<0.05或P<0.01)。结论:加味四君子汤三联疗法能有效改善症状,一定程度逆转胃黏膜萎缩、肠化和异型增生,并对胃黏膜异常活跃的增殖状态有明显抑制作用,脾虚血瘀与萎缩性胃炎及癌前病变关系密切。  相似文献

该研究首先筛选了四君子汤(红参)等药液组体外干预H9c2心肌细胞的给药浓度,优选其中高、中、低3个剂量组进行后续实验;建立了体外H2O2诱导H9c2心肌细胞凋亡模型以考察四君子汤(生晒参/红参)对其保护作用,从而为优选用于临床治疗缺血性心脏病的四君子汤中的人参炮制品提供参考,以更好地体现其疗效;并考察其对SOD,MAD和LDH等指标影响以初步阐明作用机制。结果表明,四君子汤中用红参较生晒参对H2O2诱导心肌细胞损伤保护作用为好,二者均可提高SOD活性,减少MDA产生和LDH释放,从而明显减少心肌细胞凋亡数量,起到一定的保护作用。  相似文献

目的探讨四君子汤治疗中医内科疾病脾胃气虚证的临床疗效及应用价值。方法 78例经中医临床诊断为脾胃气虚证患者,随机分为研究组和对照组,2组均给予常规治疗,研究组在此基础上加用四君子汤辅助调理,评价2组临床疗效及预后情况。结果研究组治疗总有效率95.0%,高于对照组的84.2%(P0.05,χ~2=-13.61)。随访对照组复发5例,研究组1例,研究组复发率明显低于对照组(P0.05,χ~2=-15.32)。结论四君子汤加减治疗内科脾胃气虚证有对症效果,复发率低,价格低廉,可作为中医治疗脾胃气虚证的常用方。  相似文献

目的研究四君子汤总多糖(SJZPS)对化疗荷瘤小鼠脾脏、派氏结(PPs)、肠系膜淋巴结(MLNs)淋巴细胞的影响。方法将BALB/c小鼠随机分为对照组、肿瘤模型组,环磷酰胺(CY)化疗组和四君子汤总多糖治疗(CY+SJZPS)组。CY组和CY+SJZPS组每天腹腔注射CY20 mg.kg-1,CY+SJZPS组同时灌胃SJZPS 1g.kg-1,连续给药9 d。应用流式细胞术检测脾脏、小肠PPs和MLNs中T、B淋巴细胞比例、活化水平以及T细胞亚群的变化。结果 SJZPS能提高肠道PPs中B淋巴细胞的比例和活化水平,降低MLNs中CD4+/CD8+的比值,改善化疗所致的肠道黏膜免疫损伤;SJZPS能提高化疗荷瘤小鼠脾脏淋巴细胞中B淋巴细胞的比例,及T、B淋巴细胞的活化水平,同时降低CD4+/CD8+的比值以改善化疗荷瘤小鼠系统免疫功能,其中SJZPS对B细胞的作用更加明显。结论 SJZPS能有效拮抗化疗药物所致的小鼠肠黏膜免疫功能损伤,同时对整体免疫功能具有改善作用。  相似文献

目的:探讨四君子汤对妊娠期糖尿病(GDM)患者临床疗效、内皮功能、围产期及新生儿情况的影响。方法:124例GDM患者随机分为观察组和对照组,每组62例。对照组患者采用饮食、运动疗法及胰岛素治疗,观察组患者在对照组基础上给予口服四君子汤。观察比较两组患者治疗前后血糖代谢指标、临床治疗效果,治疗前后内皮功能相关指标,围产期情况、妊娠结局及新生儿结局。结果:观察组与对照组相比,治疗后空腹血糖(FBG)、餐后2 h血糖(2hPG)和糖化血红蛋白(HbA1C)水平分别为(5.20±0.78)mmol/L、(6.87±1.16)mmol/L和(6.14±1.23)%均较低。与观察组比较临床治疗总有效率为91.94%较高,血浆一氧化氮(NO)、一氧化氮合酶(NOS)水平分别为(92.26±15.47)μmol/L、(26.71±5.45)U/ml较高,非对称性二甲基精氨酸(ADMA)水平为(0.68±0.24)μmol/L较低,羊水过多及早产发生率较低,新生儿体重较高。结论:四君子汤治疗GDM患者不仅能有效改善患者血糖代谢,提高临床治疗疗效,降低患者围产期不良情况发生率,提高新生儿体重指数,还能有效改善患者血管内皮功能。  相似文献
