背景 对蓝光照射所致视网膜色素上皮(RPE)细胞损伤与细胞内钙离子含量变化关系的深入研究可能对探讨视网膜变性类疾病的发生机制及防治具有重要意义. 目的 建立人RPE细胞蓝光损伤模型,探讨RPE细胞损伤与细胞内钙离子含量变化的关系.方法 将人RPE细胞系培养传代,锥虫蓝染色评估细胞活力.第4代人RPE细胞分别用(2000±500) lx蓝光照射3、6、9、12h,终止细胞培养24 h后,采用TUNEL法检测不同时间点细胞的凋亡情况,确定最适宜的光照度.然后将RPE细胞分为无光照组、光照组、硝苯地平组、光照+硝苯地平组、(-)BayK8644组、光照+(-)BayK8644组.除无光照组外,其他各组细胞均用蓝光照射6h,终止细胞培养24h,采用激光扫描共焦显微镜检测细胞内游离钙离子含量,激发光波长为488 nm,发射光波长为505 nm,每组随机选取30个细胞,扫描细胞图像,采用分析软件分析各组细胞内游离钙离子的荧光强度.结果 锥虫蓝染色证实RPE细胞的活力均在90%以上,TUNEL染色和免疫组织化学染色证实光照3h组未见细胞凋亡,而光照后6、9、12h发现不同数量的凋亡细胞.硝苯地平组RPE细胞内游离钙离子荧光强度比无光照组和光照+硝苯地平组低,差异均有统计学意义(均P=0.000);光照组、(-)BayK8644组、光照+(-)BayK8644组RPE细胞内钙离子荧光强度高于无光照组,差异均有统计学意义(均P=0.000);光照+硝苯地平组与无光照组相比较,(-)BayK8644组与光照+(-)BayK8644组比较,差异均无统计学意义(P=0.339、0.410). 结论 光照度(2000±500)1x、光照6h、终止培养24 h为建立蓝光照射致人RPE细胞损伤模型的最适合条件.蓝光照射所致的人RPE细胞损伤与细胞内游离钙离子含量增加有关.  相似文献

目的:观察外源性超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(caralase,CAT)对培养的人视网膜色素上皮(retinal pigment epithelium,RPE)细胞光化学损伤的保护作用. 方法:将铺满期原代培养的人RPE细胞分为三组.A组直接作光照处理,B组在培养液中加入SOD(90U/mL)和CAT(900U/ml)后30分钟进行光照,c组为对照组.光照强度2 400 Lx,时间6小时.处理结束后将三组细胞以2.4X105/ml的密度分别接种于24孔板,每孔0.4m1l.于接种后1、3、5和7天观察细胞形志,井进行细胞计数. 结果:光照后3天时,A、B组细胞生长缓慢,C组细胞生长活跃,3天后B组细胞生长接近C组,比A组明显活跃(P<0.05)相似文献

目的:探讨生长激素释放多肽(Ghrelin)对高糖环境下人视网膜色素上皮(retinal pigment epithelium,RPE)细胞氧化应激的影响。

方法:将体外培养的RPE细胞分为阴性对照组、高糖组、Ghrelin低浓度组、Ghrelin高浓度组。通过CCK-8法检测细胞存活率,氧敏感荧光探针H2DCFDA染色法观察细胞氧化损伤程度,流式细胞技术检测细胞内活性氧(reactive oxygen species,ROS)的变化,分光光度计比色法检测细胞内超氧化物歧化酶(superoxide dismutase,SOD)活力和丙二醛(malondialdehyde,MDA)含量。

结果:CCK-8结果显示,分别用10-9mol/L、10-6mol/L Ghrelin预处理后,RPE细胞存活率分别为54.79%±3.43%和79.16%±3.29%,与高糖组(41.65%±3.42%)比较,差异均有统计学意义(P<0.05)。H2DCFDA荧光探针染色结果显示,Ghrelin预处理后RPE细胞内ROS生成量下降,氧化损伤细胞减少。分光光度计比色法结果显示,与高糖组相比,Ghrelin组细胞SOD活力增加,MDA含量下降。

结论:Ghrelin可以抑制高糖诱导的人RPE细胞氧化损伤,其在糖尿病视网膜病变的发生发展过程中可能具有一定的细胞保护作用。  相似文献

蓝光诱导体外培养的人视网膜色素上皮细胞凋亡   总被引:5,自引:0,他引:5  
目的 观察蓝光光照对体外培养的人视网膜色素上皮(RPE)细胞凋亡的影响。 方法 用蓝光照射体外培养的人RPE细胞,分为3组,A组:不同光照强度;B组:同一光照强度,不同光照时间;C组:同一光照强度和光照时间,不同细胞培养终止时间。利用末端脱氧核酸转移酶介导的原位缺口末端标记(TUNEL)、荧光素标记的连接素V/碘化吡啶(Annexin V-FITC/PI)双染流式细胞检测、透射电子显微镜等手段观察细胞凋亡。 结果 TUNEL染色阳性细胞的体积缩小变圆,细胞核浓缩,边聚成新月形或帽状,细胞核碎裂成数个,细胞膜出胞。透射电子显微镜观察发现细胞内线粒体肿胀,线粒体内膜嵴消失;粗面内质网扩张;溶酶体增加,内含代谢产物。A组低于一定阈值[(500±100)lx]的蓝光光照对RPE细胞损伤较轻,细胞凋亡及坏死随光照强度的增强而增加;B组随着光照时间延长,RPE细胞凋亡数量没有增加,而细胞坏死逐渐增加;C组光照后6、12h,损伤以细胞凋亡为主,但随着光照时间延长,凋亡继发坏死细胞和坏死细胞明显增加。 结论 蓝光照射可引起体外培养的人RPE细胞损伤,损伤形式有凋亡、凋亡继发坏死及坏死;损伤程度呈光照强度和光照时间依赖性。 (中华眼底病杂志, 2005, 21: 384-387)  相似文献

目的 观察艾塞那肽(exendin-4,EX4)对4-羟基壬烯醛(4-hydroxynonenal,4-HNE)诱导人视网膜色素上皮(retinal pigment epithelium,RPE)细胞氧化应激损伤的抑制作用。方法 体外培养人RPE细胞(ARPE-19),采用CCK-8法检测不同浓度EX-4处理后细胞增殖情况;利用倒置显微镜观察对照组、EX4处理组、4-HNE组和4-HNE+EX4处理组细胞形态变化;利用荧光显微镜观察各组细胞的活性氧自由基(reactive oxygen species,ROS)生成;运用流式细胞仪检测各组RPE细胞内ROS含量变化;化学比色法检测细胞中超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)的含量变化。结果 100.0 nmol·L-1 EX4对RPE细胞具有较强的保护作用,在此浓度下对10 μmol·L-1 4-HNE引起的氧化应激损伤具有较强的抑制作用(P<0.05)。荧光显微镜观察发现,与对照组相比,4-HNE组亮绿色高荧光信号明显增强;与4-HNE组相比,4-HNE+EX4处理组荧光信号强度明显减弱,细胞内ROS含量明显减少。流式细胞仪检测也发现,与对照组相比,4-HNE组细胞内ROS含量升高,差异有统计学意义(P<0.01);与4-HNE组比较,4-HNE+EX4处理组细胞内ROS含量明显降低(P<0.05)。4-HNE组SOD含量明显低于对照组,差异有统计学意义(P<0.01);与4-HNE组比较,4-HNE+EX4处理组SOD含量明显升高,差异有统计学意义(P<0.05)。与对照组相比,4-HNE组MDA含量明显升高(P<0.01);4-HNE+EX4处理组MDA含量较4-HNE组明显下降,差异有统计学意义(P<0.05)。结论 EX4对4-HNE诱导人RPE细胞的氧化应激损伤具有保护作用,对老年性黄斑变性的治疗具有一定的潜在价值。  相似文献

目的 探讨雷公藤甲素对人视网膜色素上皮(retinal pigment epithelium,RPE)细胞氧化损伤的抑制作用及其机制。方法 RPE细胞传代培养,分为对照组、氧化损伤组(100 μmol·L-1 H2O2)和雷公藤甲素(100 μmol·L-1、200 μmol·L-1)干预组。应用MTT比色法检测细胞增殖率,流式细胞技术测定细胞内活性氧(reactive oxygen species,ROS)含量,Hoechst33258染色观察细胞凋亡,Western-blot检测RPE细胞内超氧化物歧化酶(superoxide dismutase,SOD)及丙二醛(malondialdehyde,MDA)蛋白的表达水平。结果 雷公藤甲素能明显抑制H2O2诱导的RPE细胞活性的下降,用不同浓度雷公藤甲素处理后,RPE细胞存活率分别提高到(56.00±2.76)%和(70.33±3.85)%,与氧化损伤组[(32.67±3.08)%]相比差异均有统计学意义(均为P<0.05)。雷公藤甲素能减少细胞内ROS生成,抑制细胞凋亡,不同浓度雷公藤甲素处理后,RPE细胞凋亡率分别下降到(50.33±4.61)%和(46.67±4.73)%,与氧化损伤组(67.00%±3.42%)相比差异均有统计学意义(均为P<0.05)。雷公藤甲素组SOD蛋白表达显著高于氧化损伤组,MDA蛋白表达显著低于氧化损伤组(P<0.05)。结论 雷公藤甲素对RPE细胞氧化损伤具有抑制作用,其作用机制与上调SOD的表达及下调MDA的表达有关;雷公藤甲素有望成为治疗视网膜病变的有效药物。  相似文献

背景 视网膜色素上皮(RPE)细胞的光损伤模型是多种视网膜变性类疾病的研究工具,光诱导RPE细胞损伤的主要病理基础是凋亡及炎症反应,但是内质网应激(ERS)反应是否参与其病理机制的研究国内外少有报道. 目的 探讨ERS对光损伤诱导的人RPE细胞凋亡的作用及其机制.方法 体外培养人RPE细胞株(ARPE-19),将培养的细胞分为正常对照组及光照3、6、12和24 h组,各光照组在培养箱内以(2 000± 500) lx的白色荧光灯光照细胞建立光损伤模型,正常对照组细胞在暗环境中培养且不给予光照射,筛选实验最适光照时间.将细胞分为正常对照组、光照组(光照12h)和苯基丁酸(4-PBA)预处理+光照组,4-PBA预处理+光照组先用ERS抑制剂4-PBA培养细胞30 min,然后光照细胞12h.采用流式细胞仪检测各组人RPE细胞的凋亡率和细胞内活性氧(ROS)的荧光强度;采用ELISA法检测各组细胞上清液中炎性因子白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)质量浓度;分别采用实时荧光定量PCR和Western blot法检测人RPE细胞中ERS标志物活化转录因子-6(ATF-6)、C/增强结合蛋白同源蛋白(CHOP)和细胞凋亡标志物半胱氨酸天冬氨酸蛋白酶-12(caspase-12) mRNA及其蛋白的表达. 结果 光照后细胞形态呈长梭形改变,边界不清,细胞质脱颗粒,细胞碎片增多,且随光照时间的延长而加重.流式细胞仪检测发现,随光照时间的延长,人RPE细胞内ROS含量逐渐增加,细胞凋亡率逐渐升高,差异均有统计学意义(F=763.00、119.30,均P<0.01).ELISA法检测发现,与正常对照组比较,光照后6h细胞上清液中IL-1β和TNF-α质量浓度均明显升高,12h达峰.实时荧光定量PCR和Western blot检测显示,与正常对照组比较,光照后人RPE细胞中ATF-6、CHOP和caspase-12 mRNA及其蛋白的相对表达量均明显升高,均于光照后12h达峰或升高,故选择光照12 h为最适光照时间作为后续研究.4-PBA预处理+光照组细胞中ATF-6、CHOP和caspase-12 mRNA的相对表达量均明显低于光照组,差异均有统计学意义(F=281.69、473.88、308.45,均P<0.01);ATF-6、CHOP和caspase-12蛋白的相对表达量均明显低于光照组,差异均有统计学意义(F=47.86、57.93、106.59,均P<0.01);细胞凋亡率、细胞上清液中IL-1β和TNF-α质量浓度均明显低于光照组,差异均有统计学意义(F=88.64、245.47、101.01,均P<0.01). 结论 (2 000±500)lx的光照可诱导人RPE细胞内ROS增加,并激活细胞的ERS反应,导致RPE细胞凋亡及炎症反应.ERS抑制剂4-PBA可抑制光损伤导致的ERS反应,进而降低RPE细胞凋亡率并抑制炎症反应过程.  相似文献

目的观察维生素C(Vit C)对光照后兔视网膜色素上皮(retinal pigment epithelium,RPE)细胞内B细胞淋巴瘤(Bcl-2)表达的影响。方法使用酶消化法体外分离培养色素兔的RPE细胞,用可见光照射造成其凋亡。观察不同剂量的维生素C(Vit C)在不同的给药时间对兔RPE细胞Bcl-2表达的影响。结果色素兔RPE细胞经可见光照射后细胞内Bcl-2均有不同程度的表达,其表达数量与Vit C剂量呈正相关。结论Vit C可以增加光照后兔RPE细胞Bcl-2的表达,它可能起到抑制光损伤、减少凋亡的作用。  相似文献

目的:观察手机光照刺激体外培养人视网膜色素上皮细胞(retinal pigment epithelium; RPE)后形态及B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl2-Associated X(Bax)、半胱天冬酶-3(Caspase-3)的mRNA表达变化。

方法:将体外培养的人RPE细胞随机分成4组,根据RPE细胞光照时间长短分为照射3h光照组、6h光照组、12h光照组和0h光照组。在特制的培养箱内,将智能手机屏幕开到最大亮度(200±20Lx)做为光源,持续静音情况下循环播放彩色图片。然后应用苏木精-伊红(HE)染色法,末端脱氧核糖核酸转移酶介导的原位缺口末端标记(TUNEL)染色,四甲基偶氮唑盐微量酶反应比色(MTT)法观察各实验组RPE细胞形态学改变,并运用聚合酶链反应(PCR)技术检测各组细胞中Bcl-2、Bax、Caspase-3的mRNA表达变化。

结果:用HE染色、TUNEL染色、MTT法观察发现各光照组RPE细胞形态学无明显改变,差异均无统计学意义(P>0.05)。应用PCR技术检测发现RPE细胞在光照刺激3h光照组和6h光照组,细胞内的Bcl-2、Bax、Caspase-3的mRNA表达与0h光照组的差异均无统计学意义(P>0.05)。但随着光照时间持续延长(12h),细胞内的Bcl-2表达下调,Bax、Caspase-3表达水平均上升,差异均有统计学意义(P<0.05)。

结论:手机屏幕等作为人造光源在长期持续高亮度照射下会引起体外培养人视网膜色素上皮细胞的损伤。  相似文献

目的探讨过度光照诱导大鼠视网膜色素上皮(retinal pigment epithelium.RPE)细胞表达诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)的作用及其病理学意义。方法应用免疫组织化学的方法分别检测处于自然光照环境中的正常大鼠及接受强光过度照射后大鼠的视网膜RPE细胞中iNOS的表达情况。结果正常大鼠RPE细胞中未见iNOS的表达,而过度光照后大鼠RPE细胞的胞浆中表达高水平的iNOS,同时视网膜外核层感光细胞发生结构损伤。结论过度光照可诱导RPE细胞表达iNOS.这一异常改变可能是视网膜感光细胞结构损伤的重要原因。  相似文献

目的:探讨用心理物理的方法检测糖尿病患者黄斑部功能在糖尿病视网膜病变(diabeticretinopathy,DR)诊断中的意义。 方法:对15名正常人的30只眼和41例糖尿病患者的82只眼(无视网膜病变27只眼,单纯型55只眼)黄斑部光敏感度进行检测并与图形视觉诱发电位(pattern visual evoked potential,P-VEP)检测结果进行对比. 结果:糖尿病患者黄斑部光敏感度明显低于正常组(P<0.01);在糖尿病组,黄斑部光敏感度异常检出率为62.19%,P-VEP异常率为69.51%. 结论:黄斑部光敏感度检测对糖尿病患者视网膜病变和早期视功能损害的检测有一定临床价值。 (中华眼底病杂志,1996,12:223-224)  相似文献

PURPOSE: To study whether the difference in the demographic characteristics of participants and non participants could result in biased prevalence estimates and associations. AIM: To compare the non-participant and participant characteristics, and to ascertain if non-response bias is present in the rural population of the Chennai Glaucoma Study (CGS). METHODS: Rural participants and non-participants were compared with regard to socio-demographic variables (age, gender, religion, mother tongue, literacy and employment). RESULTS: 4800 subjects aged 40 years or over were enumerated, 82% (3934: 45% male and 55% female) responded. Gender did not influence participation (adjusted OR-1.11, CI: .91-1.36). Subjects in the 70-79 year age group were more likely to respond (OR-1.76; CI-1.31-2.38). Hindus had a higher participation rate than Christians or Muslims (adjusted OR-2.63, CI: 1.80-3.84). The other predictors of participation were illiteracy (adjusted OR-1.44, CI: 1.22-1.70), unemployment (OR-1.28, CI: 1.04-1.58), place of residence (main villages) (OR-6.66, 95% CI: 4.6-9.64). CONCLUSION: Based on our study findings, it does not seem likely that participation bias will affect the study results.  相似文献

PURPOSE: To correlate functional damage over time detected by standard automated perimetry (SAP) and frequency doubling technology (FDT) with central corneal thickness (CCT) in patients with ocular hypertension (OHT). METHODS: Seventy-eight OHT patients underwent CCT measurements, SAP, and FDT (the latter two also after 12 and 18 months). Patients were divided into three equally sized groups of 26 patients each: thin (< 540 microm), normal (540-580 microm), and thick cornea (> 580 microm). The frequency of abnormal FDT and SAP results was analyzed over time (Pearson chi2 test). RESULTS: Six of 26 patients with thin corneas (23.1%) presented an abnormal FDT test at baseline, compared to 1 of 26 (3.8%) in the normal thickness cornea group and 1 of 26 (3.8%) in the thick cornea group. After 12 months, the abnormal FDT tests were as follows, respectively: 9 of 26 (34.6%), 2 of 26 (7.7%), and 2 of 26 (7.7%). For SAP the abnormal results were as follows, respectively: 8 (30.1%), 5 (19.2%), and 2 (7.7%). After 18 months, the abnormal FDT tests were as follows, respectively: 16 (61.5%), 5 (19.2%), and 5 (19.2%). For SAP, the abnormal results were as follows, respectively: 10 (38.5%), 5 (19.2%), and 2 (7.7%). CONCLUSIONS: OHT patients with thinner corneas have a greater risk of developing functional damage over time.  相似文献

目的:探讨血浆内皮素(endothelin,ET)和血清血管紧张素转换酶(angiotensin converting enzyme,ACE)水平与糖尿病视网膜病变(diabetic retinopathy,DR)的关系。 方法:用直接放射免疫法和紫外分光光度法测定62例糖尿病(diabetes mellitus,DM)患者和30名正常人血浆ET和血清ACE水平。 结果:DR患者血浆ET和血清ACE水平显著高于无视网膜病变者(P<0.01)。随着DR程度增加,血浆ET水平显著升高,血清ACE水平逐渐升高。 结论:DR患者血浆ET和血清ACE水平增高可能与DR的发生与进展有关。 (中华眼底病杂志,1996,12:177-179)  相似文献

PURPOSE: To determine wether the cataract extraction method and intraocular lens material affect bacterial contamination of the eye during surgery. METHODS: This retrospective study evaluated microbial contamination of the anterior chamber fluid in three group of patients. Group 1: extracapsular extraction and PMMA lens (354 cases). Group 2: phacoemulsification and PMMA lens (474 cases). Group 3: phacoemulsification and silicone lens with PMMA haptic (264 cases). RESULTS: Microbial contamination rate was in group 1: 5.65%, in group 2: 4.75%, and in group 3: 4.54%. The difference was not statistically significant (chi: 0.321). All contaminants were Gram positive. CONCLUSION: Bacterial contamination rate of the anterior chamber is similar during extracapsular extraction and phacoemulsification, and is not modified when using PMMA or silicone lens.  相似文献

AIM: To report the incidence, risk factors and visual outcomes for postoperative endophthalmitis (POE) based on 7-year data from the Malaysian Ministry of Health Cataract Surgery Registry (MOH CSR). METHODS: Data was collected from the web-based MOH CSR. All consecutive cataract surgery patients from 1st June 2008 to 31st December 2014 were identified. Exclusion criteria were traumatic cataract or previous ocular surgery. Demographic data, ocular co-morbidities, intraoperative details and postoperative visual acuity (VA) at final ophthalmological follow-up were noted. All eyes were taken for analysis. Subjects with POE were compared against subjects with no POE for risk factor assessment using multiple logistic regressions. RESULTS: A total of 163 503 subjects were screened. The incidence of POE was 0.08% (131/163 503). Demographic POE risk factors included male gender (OR: 2.121, 95%CI: 1.464-3.015) and renal disease (OR: 2.867, 95%CI: 1.503-5.467). POE risk increased with secondary causes of cataract (OR: 3.562, 95%CI: 1.740-7.288), uveitis (OR: 11.663, 95%CI: 4.292-31.693) and diabetic retinopathy (OR: 1.720, 95%CI: 1.078-2.744). Intraoperative factors reducing POE were shorter surgical time (OR: 2.114, 95%CI: 1.473-3.032), topical or intracameral anaesthesia (OR: 1.823, 95%CI: 1.278-2.602), posterior chamber intraocular lens (PCIOL; OR: 4.992, 95%CI: 2.689-9.266) and foldable IOL (OR: 2.276, 95%CI: 1.498-3.457). POE risk increased with posterior capsule rupture (OR: 3.773, 95%CI: 1.915-7.432) and vitreous loss (OR: 3.907, 95%CI: 1.720-8.873). Postoperative VA of 6/12 or better was achieved in 15.27% (20/131) subjects with POE. CONCLUSION: This study concurs with other studies regarding POE risk factors. Further strengthening of MOH CSR data collection process will enable deeper analysis and optimization of POE treatment.  相似文献

AIM: To evaluate the six months follow-up outcome of combined intravitreal triamcinolone acetonide (IVTA) and photodynamic therapy (PDT) for subfoveal choroidal neovascularization compared to PDT alone. STUDY DESIGN: Prospective interventional pilot study. MATERIALS AND METHODS: Patients with six months follow-up of IVTA following PDT (Group I, eight eyes) and PDT alone (Group II, eight eyes) were included. Four mg/ 0.1 ml of IVTA was injected 7-10 days following PDT. The patients were reevaluated every month for the first two months and every three months thereafter in both the groups. RESULTS: Group I: The mean age was 65.8+/-11.8 years (range: 47-79 years). Five patients were male. The total treatment sessions in six months were 11 (mean: 1.36). At six months, one eye had >or= 10 letters gain and three eyes had > 10 letters loss. Four eyes had stable vision. Two eyes (25%) developed increased intraocular pressure (>40 mmHg) during follow-up. Group II: The mean age was 58.7+/-11.7 years (range: 46-76 years). Five patients were male. The total treatment sessions in six months were 17 (mean: 2.13). At six months, six eyes had >or= 10 letters gain and none had > 10 letters loss. Two eyes had stable vision. CONCLUSION: The mean number of treatment sessions following combination therapy of IVTA (4 mg) and PDT appears relatively less (1.36 at six months) compared to PDT alone (mean: 2.13). PURPOSE: To evaluate posterior capsule opacification (PCO) in fellow eyes, 1 receiving a silicone intraocular lens (IOL) and the other, an acrylate IOL. SETTING: Department of Ophthalmology, Isala Clinics, Zwolle, The Netherlands. METHODS: This retrospective study comprised 107 patients (214 eyes). In each patient, 1 eye was randomly selected to have implantation of an acrylate IOL (AcrySof MA30BA or MA60BM, Alcon) and the other eye, a plate-haptic silicone IOL (AA4203VF, Staar). Outcome measures were the total PCO index, percentage of neodymium:YAG (Nd:YAG) capsulotomies performed, and logMAR best corrected visual acuity (BCVA). The follow-up was 3 years. RESULTS: The total PCO index was significantly lower in the AcrySof group than in the plate-haptic silicone group (P<.0001). There was no significant difference in logMAR BCVA between groups (P>.05). The percentage of Nd:YAG laser treatments was significantly lower in the AcrySof group (2.8%) than in the plate-haptic silicone group (23.1%) (P<.05). CONCLUSIONS: There was significantly less PCO and a lower Nd:YAG laser capsulotomy rate after AcrySof IOL implantation than after plate-haptic silicone IOL implantation. These results did not seem to affect the logMAR BCVA as there were no significant differences between groups in this parameter.  相似文献

BACKGROUND: We report our experience with stereotactic radiosurgery using the Gamma-knife in large uveal melanoma unsuitable for brachytherapy (Ru106). PATIENTS AND METHODS: We treated 35 patients (16 male, 19 female; age: median 59 years (95% Confidence interval (CI): 31-84 years; 18 right eyes, 17 left eyes). 7 tumors were located juxtapapillary, 16 tumors were located in the mid-periphery and 12 tumors were located in the ciliary body, The localization and/or the dimension of the tumors did not allow for radiation brachytherapy (Ru106). All patients underwent regular clinical, echographical and neuroradiological follow-up examinations. RESULTS: The follow-up time was median 12 months (95% CI: 4-20 months). A local tumor control defined as either continuous tumor regression or stopping of tumor progression was achieved in 34 (97%) of the 35 treated patients within the observation period. The eye of one patient was enucleated due to tumor regrowth. Maximum apical tumor height according to standardized A-scan before treatment was median 9.1 mm (95% CI: 3.2-13.9 mm) and after treatment median 6.4 mm (95% CI: 2.1 bis 11.9 mm). The difference was statistically highly significant (p < 0.001, one-tailed paired t-test). CONCLUSIONS: Our results in 35 patients indicate that radiosurgery using the Gamma-knife is beneficial in retaining the eyes of patients with large uveal melanomas that are not suitable for brachytherapy (Ru106).  相似文献

PURPOSE: We reviewed the surgical results of vitrectomy for macular holes (MHs) associated with myopic foveoschisis (MF). DESIGN: Noncomparative, interventional case series. METHODS: setting: Institutional. participants: Eight eyes of eight patients with MHs associated with MF. intervention procedure: All patients underwent vitrectomy, internal limiting membrane peeling, and gas tamponade. main outcome measures: Slit-lamp-based biomicroscopy, optical coherence tomography examinations, and best-corrected visual acuity (BCVA) measurement. RESULTS: The MHs closed in two eyes (25%), and the BCVA improved more than 2 lines in three (37.5%) eyes, remained unchanged in three (37.5%), and worsened in two (25%). The MH size increased significantly in a case with an unresolved MH (P < .05). The postoperative BCVA was significantly correlated with the preoperative BCVA (P < .05). CONCLUSIONS: Although significant visual improvement occurs in less than 50% of cases, vitrectomy can be beneficial for some cases.  相似文献
