目的探讨不同强度跑台训练对大鼠膝关节软骨胶原纤维形态学及基因表达的影响。 方法采用随机数字表法将48只雄性SD成年大鼠分为对照组、低强度运动组、中强度运动组及高强度运动组。3个运动组大鼠分别进行低、中、高强度跑台运动。于实验进行8周后处死所有实验动物,取膝关节软骨标本行天狼星红-饱和苦味酸染色观察胶原纤维排列情况,采用免疫组化技术检测Ⅱ型胶原(Col2)含量;另外本研究同时采用实时荧光定量逆转录聚合酶链反应检测软骨biglycan(BGN)、fibromodulin(FMOD)及Ⅱ型胶原(Col2)mRNA表达。 结果对照组膝关节软骨胶原纤维排列规则;低、中强度运动组与对照组间无明显差异;而高强度运动组胶原排列不规则、细纤维数量增多;高强度运动组Ⅱ型胶原含量显著低于对照组。与对照组比较,低强度运动组软骨细胞Col2 mRNA表达明显增高,高强度运动组BGN mRNA表达明显增高。 结论低、中强度运动有助于维持关节软骨正常结构及功能;高强度运动能促使关节软骨Ⅱ型胶原纤维减少、排列紊乱,但软骨自我修复可能同时存在。  相似文献

摘要 目的:观察负重游泳运动对膝关节骨性关节炎(OA)大鼠关节软骨及运动能力的影响,探讨负重游泳运动防治膝OA的机制,为临床采用负重游泳运动防治膝OA提供理论依据。 方法:将50只雄性Wistar大鼠随机分为空白组(20只)和模型组(30只),模型组大鼠采用木瓜酶注射法建立膝关节骨性关节炎模型,造模完成后在空白组和模型组大鼠中各抽取10只,进行以下测定:①肉眼及光镜下观察大鼠膝关节软骨形态及病理学改变并评分;②免疫组化法观察大鼠膝关节软骨中基质金属蛋白酶(MMP-13)阳性细胞的数量;③评价大鼠的运动能力。将剩余的模型组大鼠随机分为负重游泳组(10只)和对照组(10只)。剩余空白组及对照组大鼠不予以任何干预,负重游泳组大鼠接受6周的负重游泳运动后,对三组大鼠进行上述3项指标的测定。 结果:负重游泳运动后,负重游泳组大鼠关节软骨面大体观分值、镜下观Mankin分值、软骨中MMP-13阳性细胞数较对照组大鼠明显减少;运动能力较对照组明显增加(P<0.05)。 结论:负重游泳运动能够延缓膝OA模型大鼠膝关节软骨细胞的进一步破坏,并能够提高膝OA模型大鼠的运动能力。负重游泳运动可作为治疗膝OA的运动疗法。  相似文献

目的探讨高强度跑台运动对大鼠关节液及滑膜白细胞介素-1β(IL-1β)的影响.方法将24只雄性Sprague-Dawley大鼠随机分为安静对照组和高强度运动组各12只.高强度运动组进行6周高强度跑台运动.取各组大鼠膝关节股骨内侧髁软骨及滑膜组织,HE染色观察其组织变化,免疫组化法检测IL-1β在大鼠滑膜中的表达,酶联免疫吸附法(ELISA)检测大鼠关节液中IL-1β水平.结果高强度运动组大鼠膝关节出现关节软骨损伤和滑膜炎表现,关节液中IL-1β的表达高于安静对照组(P<0.05),滑膜组织中IL-1β的表达明显高于安静对照组(P<0.01).结论高强度运动导致的关节软骨损伤、滑膜炎症,以及关节液、滑膜中IL-1β的异常表达可能是创伤性关节炎发生的机制之一.  相似文献

背景:适量的运动是维持正常关节组织形态结构及生理功能的必要条件,过度运动或制动均可导致关节软骨退变。目的:观察不同强度跑台运动对大鼠膝关节软骨的影响。方法:将18只雄性成年Wistar大鼠随机分安静组、低强度运动组、高强度运动组。6周后ELISA法测定血清基质金属蛋白酶3水平,并行蕃红-O染色、基质金属蛋白酶3与Ⅱ型胶原免疫组织化学染色及Mankin评分,反转录聚合酶链反应检测软骨基质金属蛋白酶3的mRNA表达。结果与结论:高强度运动组Mankin评分、血清及软骨中基质金属蛋白酶3表达均显著高于安静组与低强度运动组(P<0.05),基质糖氨多糖及Ⅱ型胶原含量均显著低于安静组与低强度运动组(P<0.05);低强度运动组与安静组差异无显著性意义。说明高强度运动可造成大鼠膝关节软骨退行性变,且软骨运动性损伤可能与基质金属蛋白酶3表达增强有关。  相似文献

目的探讨高强度运动对大鼠关节液软骨寡聚基质蛋白(cartilage oligomeric matrix protein,COMP)及关节软骨的影响。方法 SD大鼠20只随机分为对照组和高强度运动组各10只,高强度运动组行8周高强度跑台运动,对照组不进行运动训练;8周后取大鼠股骨髁软骨行组织病理学检查观察其组织学变化,采用ELISA法检测2组大鼠关节液COMP水平。结果高强度运动组大鼠膝关节出现不同关节软骨损伤的表现,关节液COMP水平((14.1±2.3)μg/L)明显高于对照组((5.6±1.4)μg/L))(P〈0.05)。结论高强度运动可造成关节软骨损伤,关节液中COMP表达水平与创伤性关节炎发生有关。  相似文献

目的研究跑步运动对大鼠不稳定膝关节软骨的影响。 方法将20只切断膝关节前交叉韧带的8周龄SD大鼠按随机数字表法分为自由活动组(对照组)和跑步训练组(实验组),每组10只大鼠,2组再根据处死取材时间分为造模成功3周和6周各2个亚组,每个亚组5只大鼠。实验组按15m/min强度进行跑步训练,每天训练1h;对照组自由活动,不接受任何干预。于造模成功3周和6周后采用苏木精-伊红(HE)、甲苯胺蓝及免疫组化染色、透射电镜等方法分别观察和比较2组大鼠膝关节的软骨厚度、Mankin评分、蛋白多糖含量、软骨基质Ⅱ型胶原含量及软骨形态结构。 结果造模成功6周后,2组大鼠关节软骨的厚度和Mankin评分与组内造模成功3周后比较,差异均有统计学意义(P<0.05);造模成功3周和6周后,实验组软骨厚度分别为(154±13)μm和(131±15)μm,Mankin评分分别为(9.93±1.36)分和(11.23±1.57)分,分别与对照组同时间点比较,差异均有统计学意义(P<0.05)。造模成功6周后,2组大鼠膝关节软骨甲苯胺蓝染色阳性光密度与组内造模成功3周后比较,差异有统计学意义(P<0.05);造模成功3周和6周后,实验组甲苯胺蓝染色阳性光密度分别与对照组同时间点比较,差异均有统计学意义(P<0.05)。造模成功6周后,2组大鼠膝关节软骨Ⅱ型胶原纤维免疫组化染色阳性光密度与组内造模成功3周后比较,差异有统计学意义(P<0.05);造模成功3周和6周后,实验组Ⅱ型胶原纤维免疫组化染色阳性光密度分别与对照组同时间点比较,差异均有统计学意义(P<0.05)。造模成功3周后,实验组透射电镜显示软骨细胞减少,软骨表面有部分断裂;造模成功6周后,实验组透射电镜可见软骨表面多处有破损,软骨细胞坏死。 结论跑步运动对不稳定膝关节软骨具有破坏效应,可加重关节损伤和软骨基质的破坏,加速软骨细胞的退行性变。  相似文献

目的探讨不同强度跑台运动对大鼠胫骨软骨下骨三维结构的影响。 方法采用随机数字表法将24只雌性SD大鼠分为对照组、低强度跑步组、中强度跑步组及高强度跑步组。3个运动组大鼠根据各自运动方案进行跑步训练,对照组大鼠不给予特殊运动训练。于实验进行8周后取各组大鼠胫骨近端组织进行Micro-CT扫描分析。 结果与对照组比较,高强度运动组软骨下骨板变厚、骨密度增加、孔隙率降低;软骨下骨松质骨骨小梁数量增加并多呈现板状结构改变;而低强度组和中强度组胫骨软骨下骨未见明显改变。 结论跑台运动对大鼠软骨下骨显微结构的影响具有强度依赖性;高强度跑步能导致胫骨软骨下骨结构显著改变。 【关键词】跑台运动;大鼠;软骨下骨;显微CT;强度  相似文献

目的:研究关节内骨折错位时不同纵向错位程度及运动方式对关节运动功能的恢复及关节软骨损伤修复的影响。方法:建立兔膝关节股骨内侧髁全厚(0.5mm)及两倍厚(1.Omm)关节软骨的骨折错位模型,分别对这些实验动物膝关节采用外固定、间歇性主动活动、持续性被动活动3种方法处理。在12周时,利用活体观测,并用光学显微镜和透射电镜对关节骨折错位的治疗和重建进行研究。结果:患膝关节活动范围,持续被动运动组和间断主动运动组之间差别不明显,二者均好于制动组。光学显微镜下,制动组1.0mm错位的兔膝关节软骨退变最明显。0.5mm错位组修复组织为类透明软骨;而1.Omm错位组修复组织则为纤维软骨,透射电镜发现,0.5mm错位组较1.Omm错位组修复组织软骨细胞更为成熟,细胞器丰富,细胞间纤维组织含量少。结论:兔膝关节内骨折错位的修复组织性质与错位的程度有关。运动有利于关节软骨损伤的修复,对关节运动功能的恢复有积极作用。  相似文献

目的:研究并探讨不同程度纵向错位时关节运动功能的恢复及关节软骨损伤修复的形态学改变。方法:建立兔膝关节股骨内侧髁全厚(0.5mm)及两倍厚(1.0mm)关节软骨的骨折错位模型。在12周时,利用活体进行观测,并用光学显微镜和透射电镜对关节骨折错位的治疗和重建进行研究。结果:患膝关节运动功能,0.5mm错位较1.0mm错位恢复好。光学显微镜下,0.5mm错位组修复组织为类透明软骨;而1.0mm错位组修复组织则为纤维软骨。透射电镜发现,0.5mm错位较1.0mm错位修复组织软骨细胞成熟度高。结论:兔膝关节内骨折纵向错位不超过关节软骨厚度(≤0.5mm)时,软骨损伤自身修复效果满意。  相似文献

背景:研究发现,基质金属蛋白酶和胶原参与关节软骨组织机体生理重建及病理破坏.目的:观察膝关节骨软骨缺损及表面软骨缺损动物模型关节软骨组织中胶原及基质金属蛋白酶的表达变化.方法:雌性SD大鼠48只随机分为3组:骨软骨缺损组在双膝关节制作骨软骨缺损模型,表面缺损组在双膝关节制作表面软骨缺损,对照组双膝关节制作关节囊切开.分别于术后4、8、12周取股骨髁标本,行苏木精-伊红染色,免疫组化检测Ⅰ型胶原、Ⅱ型胶原、基质金属蛋白酶3的表达.结果与结论:骨软骨缺损组术后4周缺损中有少量新生组织生成,8及12周可见到纤维组织填充,修复组织细胞外基质Ⅰ型胶原免疫组化染色阳性,Ⅱ型胶原免疫组化染色阴性,关节软骨组织中基质金属蛋白酶3表达增高.表面缺损组表面软骨缺损4及8周未见修复迹象,12周可见微量纤维组织填充,细胞外基质Ⅰ型胶原免疫组化染色阳性,Ⅱ型胶原免疫组化染色阴性,术后表面缺损组关节软骨组织基质金属蛋白酶3表达增高.对照组关节软骨组织Ⅰ型胶原免疫组化染色阴性,Ⅱ型胶原免疫组化染色阳性,基质金属蛋白酶3低表达,无形态学异常改变.说明机械性损伤可以导致关节软骨细胞外基质成分发生改变,丧失其原有的生物学特性而退变,基质金属蛋白酶3在损伤后的软骨组织中表达增高,使细胞外基质的降解增加,是导致关节软骨退变的重要因素.  相似文献

目的:探讨单核细胞趋化因子-1(MCP-1)在等长收缩运动训练促进缺血心肌侧支动脉生成中的作用。方法:将造模成功的24只雄性SD大鼠,体重(200±20)g,随机分为4组:对照组(CG)、心肌缺血组(MI)、运动训练组(ET)、MCP-1抑制剂组(LG),每组均有6只大鼠。CG组连续2周皮下注射生理盐水;MI组连续2周皮下注射异丙肾上腺素(10mg/kg/d),造成心肌缺血;ET组连续2周皮下注射异丙肾上腺素并且进行等长收缩运动训练;LG组在ET组的基础上进行MCP-1抑制剂来氟米特灌胃。训练8周结束后,麻醉大鼠,取左心室心肌,采用微球法测定缺血区心肌相对侧支循环血流量(RCBF);采用免疫组化法测定缺血区心肌小动脉密度和单核细胞数量;以Western blot和荧光定量PCR法测定缺血心肌中MCP-1的蛋白及mRNA的表达。结果:ET组大鼠的相对侧支血流量(RCBF)、小动脉密度(AD)明显高于其余3组(P0.001),LG组的RCBF和AD与MI组相比无显著性差异;ET组大鼠缺血心肌中单核细胞数量、MCP-1 mRNA以及MCP-1蛋白表达均显著高于其余各组(P0.001),而LG组大鼠缺血心肌中单核细胞数量、MCP-1 mRNA以及MCP-1蛋白表达与MI组相比无显著性差异。结论:持续8周的等长收缩运动训练可以增加大鼠缺血心肌中MCP-1的表达,从而促进缺血心肌侧支动脉的生成。  相似文献

目的探讨腹膜透析液对大鼠腹膜结缔组织生长因子(CTGF)的表达及细胞外基质(ECM)合成的影响。方法将中南大学湘雅二医院动物实验中心提供的SD大鼠随机分为4组:正常对照组(Con)、生理盐水组(NS)、低糖透析液组(LG)、高糖透析液组(HG)。4周后,评价腹膜的功能。VG染色镜下测量腹膜胶原组织厚度。免疫组织化学方法检测CTGF、TGF-β1及纤连蛋白(FN)蛋白质的表达。采用RT-PCR检测大鼠腹膜组织中CTGF和TGF-β1mRNA的表达。结果LG、HG组与Con、NS组相比较,超滤量均显著下降(P〈0.05);HG组与LG组比较,其超滤量明显减少(P〈0.05)。D/Pcr的比较:HG组比Con、NS组升高(P〈0.05);D/DO的比较:HG组比Con、NS组明显下降(P〈0.05),LG组比Con组低(P〈0.05)。HG组腹膜胶原厚度比与其余各组均增厚(P〈0.05);LG组比Con组增厚(P〈0.05);HG组CTGF、TGF-β1、FN蛋白表达水平显著高于其他各组(P〈0.05);LG组显著高于Con、NS组(P〈0.05)。在Con及NS组中,TGF-β1、CTGFmRNA呈低水平表达,TGF-β1、CTGFmRNA在HG组中表达最高(P〈0.05)。结论腹膜透析液尤其是高糖腹膜透析液,能明显诱导大鼠腹膜CTGF的表达增强,并促使腹膜纤维化发生。  相似文献

Enhanced denaturation of type II collagen fibrils in femoral condylar cartilage in osteoarthritis (OA) has recently been quantitated immunochemically (Hollander, A.P., T.F. Heathfield, C. Webber, Y. Iwata, R. Bourne, C. Rorabeck, and A.R. Poole. 1994. J. Clin. Invest. 93:1722-1732). Using the same antibody that only reacts with denatured type II collagen, we investigated with immunoperoxidase histochemistry (results were graded for analysis) the sites of the denaturation (loss of triple helix) of this molecule in human aging (at autopsy, n= 11) and progressively degenerate (by Mankin grade [MG]) OA (at arthroplasty, n= 51) knee condylar cartilages. Up to 41 yr, most aging cartilages (3 of 4) (MG 0-4) showed very little denaturation. In most older cartilages, (4 of 7) (MG 2-4), staining was observed in the superficial and mid zones. This pattern of collagen II denaturation was also seen in all OA specimens with increased staining extending to the deep zone with increasing MG. Collagen II staining correlated directly both with MG and collagen II denaturation measured by immunoassay. Cartilage fibrillation occurred in OA cartilages with increased penetration of the staining for collagen II denaturation into the mid and deep zones and where denaturation was more pronounced by immunoassay. Thus in both aging and OA the first damage to type II collagen occurs in the superficial and upper mid zone (low MG) extending to the lower mid and deep zones with increasing degeneration (increasing MG). Initial damage is always seen around chondrocytes implicating them in the denaturation of type II collagen.  相似文献

[Purpose] We examined the impact of exercise on the expression pattern of nerve growth factor in the spinal cord of rats with induced osteoarthritis of the knee joint. [Subjects and Methods] To produce monosodium iodoacetate-induced arthritis, rats were administered 3 mg/50 µL monosodium iodoacetate through the interarticular space of the right knee. The animals were randomly divided into four groups: rats sacrificed 3 weeks after 0.9% saline solution injection (shame group, n = 10), rats sacrificed 3 weeks after monosodium iodoacetate injection (control group, n = 10), rats with 4 weeks rest from 3 weeks after monosodium iodoacetate injection (no exercise group, n = 10), and rats with 4 weeks treadmill training from 3 weeks after monosodium iodoacetate injection (exercise group, n = 10). Serial coronal sections of the lumbar spine were cut and processed for immunohistochemistry. [Results] The expression of nerve growth factor was significantly increased in the EG compared with the SG, CG, and NEG. [Conclusion] Increased nerve growth factor expression in the spinal cord due to exercise-induced stimulation can be effective in treating chronic pain. Such treatment will contribute not only to improving the joint function of patients with chronic pain but also their quality of life.Key words: Osteoarthritis, Treadmill exercise, Nerve growth factor  相似文献

[Purpose] The purpose of this study was to investigate the effects of treadmill training on functional recovery by analyzing the ankle joint as well as the knee and hip joints with 2D video analysis during gait by rats with sciatic nerve injury. [Subjects and Methods] Twenty-four male Sprague-Dawley rats were used in this study. The sham group (SG) received only a sham operation without any sciatic injury; the training group (TG) performed treadmill training for 4 weeks after sciatic injury; and the control group (CG) wasn’t provided with any therapeutic intervention after sciatic injury. [Results] The ankle, knee, and hip ROM of TG and CG during the initial, mid stance, and toe-off phases of gait at post-test were significantly different from SG. [Conclusion] Physical exercise, like treadmill training, is beneficial for the improvement of the ankle, knee and hip joints of rats with crushed sciatic nerve injury.Key words: Sciatic nerve injury, Physical exercise, 2D video analysis  相似文献

Changes in the structure of the proteoglycan aggrecan (PG) of articular cartilage were determined immunochemically by RIA and gel chromatography and related to cartilage degeneration documented histologically by the Mankin grading system. Monoclonal antibodies to glycosaminoglycan epitopes were used. In all cartilages, three chondroitin sulfate (CS)-rich populations of large size were observed in addition to a smaller keratan sulfate (KS)-rich population. In grades 7-13 OA cartilages (phase II), molecules were significantly larger than the equivalent molecules of grades 2-6 (phase I). CS chain lengths remained unchanged. In most OA cartilages, a CS epitope 846 was elevated in content, this being most marked in phase II (mean: fivefold). Loss of uronic acid, KS, and hyaluronic acid were only pronounced in phase II OA because of variations in normal contents. Aggregation of PG was unchanged (50-60%) or reduced in OA cartilages, but molecules bearing epitope 846 exhibited almost complete aggregation in normal cartilages. This study provides evidence for the capacity of OA cartilage to synthesize new aggrecan molecules to replace those damaged and lost by disease-related changes. It also defines two phases of PG change in OA: an early predominantly degenerate phase I followed by a net reparative phase II accompanied by net loss of these molecules.  相似文献

背景:采用传统方法冻存后,关节软骨细胞存活率低,且软骨表层与深层的软骨细胞存活率差别较大,移植物易发生退行性变,导致手术失败.目的:经打孔梯度降温冻存膝关节软骨后并行异体移植,观察打孔梯度降温冻存对兔关节软骨的影响.方法:自2月龄新西兰白兔膝关节股骨膑面取骨软骨移植物,随机分为3组:实验组在软骨面以3 mm×3 mm矩阵打孔,梯度降温冷冻保存.以非打孔经梯度降温冷冻保存组、非打孔超低温冷冻保存组为对照.复温后移植到成年新西兰白兔相应膝关节缺损部位,观察各组移植物大体形态学、组织化学、免疫组织化学染色效果的差别.结果与结论:实验组、非打孔经梯度降温冷冻保存组大体形态学、组织化学、免疫组织化学染色效果均明显优于非打孔超低温冷冻保存组.实验组与非打孔经梯度降温冷冻保存组效果差别不明显,但实验组明显加强了对中间层软骨组织的保护程度.提示关节软骨的梯度降温冷冻保存效果优于快速降温冷冻保存;关节软骨打孔冷冻保存对深层细胞有一定的保护作用,提高了软骨细胞存活率,延缓了移植软骨组织的退变过程.  相似文献

Headache in children with unknown aetiology is an increasing phenomenon in industrial countries, especially during growth spurts. During this growth phase, the Long Sitting Slump (LSS) can be a useful tool for measurement of neurodynamics and management. This study investigated the difference in cervical flexion and sensory responses (intensity and location) during the LSS tests in children (n=123) aged 6-12 years, between a migraine (primary headache group=PG), cervicogenic headache (secondary headache group=SG) and control group (CG). The results indicated that the intensities of the sensory response rate were highest in the PG and SG when compared to CG. The responses in the legs were predominantly found in the PG (81.9%) and responses in the spine in the SG (80%). The sacrum position varied significantly between both headache groups (PG and SG) and the CG (p<0.0001), but there was no significant difference between the CG and the PG (p>0.05). No significant difference in the neck flexion range was measured in LSS, nor in standardized knee flexion between the PG and CG (p>0.05). The cervical flexion ranges differed significantly (p<0.0001) between the SG on the one hand and the PG and CG on the other. The biggest difference in neck flexion during knee extension was between the SG and CG.  相似文献
