Objective To explore the relationship between power density of He-Ne laser with the growth of fibroblasts in hypertrophic scar(HS).Methods The cultured fibroblasts in HS were irradiated with He-Ne laser(Wavelenth 632.8 nm),various power densities such as 50mW/cm2,100mW/cm2 and 150mW/cm2 were respectively adopted once a day for 10 minutes.After 1,3 and 5 times of He-Ne laser for dose rate irradiation separatedly,the cell count and cell circle analysis were counted and examined by trypan blue staining and flow cytometry .Results The amount of cell after 1 times of 50mW/cm2 laser irradiation was markedly more than the control(P<0.01).The amount of cells after 5 times of 100mW/cm2,3 and 5 times of 150 mW/cm2 laser irradiation was less than the controls(P< 0.05).The result of cell circle analysis was corresponded with that of the cell count.Conclusion Both stimualtion and inhibition of He-Ne laser on the growth of scar fibroblasts can be obtained with various power densities.Power density of He-Ne laser associates with the effects on the growth of scar fibroblasts.  相似文献

目的探讨氦-氖激光能量密度与培养瘢痕成纤维细胞生长的关系。方法以30、60、90、180和270J/cm2能量密度氦-氖激光(功率密度100mW/cm2),分别照射培养的人增生性瘢痕成纤维细胞1、3和5次,1次/d,然后采用台盼蓝染色法进行细胞计数。结果30J/cm2照射1次后细胞总数大于非照射组(P<0.001);90和180J/cm2照射3次,60、90和180J/cm2照射5次后细胞总数均少于同期非照射组(P<0.05),270J/cm2照射3、5次后细胞总数明显少于非照射组(P<0.001)。结论氦-氖激光对培养瘢痕成纤维细胞生长的抑制效应与能量密度、照射次数有关。  相似文献

目的 探讨氦氖激光体外照射对培养的瘢痕成纤维细胞生长的抑制作用。方法 以6 32 .8nm波长氦氖激光 (功率密度 5 0mW /cm2 ) ,3J/cm2 ,30J/cm2 ,90J/cm2 ,180J/cm2 剂量 ,分别照射培养的人瘢痕成纤维细胞 1次 ,3次和 5次 ,每日 1次 ,然后进行细胞计数和细胞周期分析。结果180J/cm2 重复照射 3次和 5次 ,细胞总数都明显少于同期对照组 ,差异均有显著性 (P <0 .0 5 )。细胞周期分析表明 ,180J/cm2 照射细胞 3次和 5次后 ,S期细胞数从 5 1%降低到 2 0 %和 14% ,G0 /G1期细胞从 2 8%分别上升到 5 5 %和 6 0 % ,Sub -G2 期细胞百分比分别为 6 .7%和 9.8%。结论 一定剂量 (180J/cm2 )氦氖激光重复照射能抑制培养的瘢痕成纤维细胞生长 ,原因可能是由于氦氖激光引起G0 /G1期细胞停滞和细胞凋亡所致  相似文献

Objective To explore He- Ne laser inducing apoptosis of cultured fibroblasts in hypertrophic scars(HS).Methods Cultured fibroblasts in HS were irradiated with He- Ne laser. Apoptosis was examined by TUNEL technique and cell necrosis was counted by trypan blue staining. Results There was no cell apoptosis after repeated irradiation with 10mW/cm2 and 50mW/cm2 for 10 and 30 minutes. The percent of apoptosis was 7.20% and 12.73% after repeated irradiation with 100mW/cm2 for 10 and 30 minutes. The percent of apoptosis was 14.74% and 16.27% after laser irradiation with 150mW/cm2 for 10 and 30 minutes.No necrotic cell was presented after repeated irradiation for 10 and 30 minutes at various power densities. Conclusion Repeated He- Ne irradiation of HS- derived fibroblast cultures at power densities of 100mW/cm2 and 150 mW/cm2 can induce cell apoptosis. Power density is more important than energy density so far as laser- inducing apoptosis of scar fibroblasts concerned.  相似文献

目的探讨氦氖激光诱导瘢痕成纤维细胞凋亡的分子机制。方法将培养瘢痕成纤维细胞随机分为转染组、转染对照组和非转染组,转染组细胞与绿色荧光蛋白(GFP)质粒cDNA、含目的基因片段的表达载体质粒pcDNA3-FADD-DNcDNA共转染,转染对照组细胞与GFP质粒cD-NA、空载体质粒pcDNA3共转染,非转染组细胞没有进行转染,转染后24h分别对各组细胞进行氦氖激光照射(100mW/cm2照射30min),1次/d,连续照射3d,然后采用TUNEL技术检测细胞凋亡。结果转染组、转染对照组与非转染组细胞凋亡率分别为9.27%,15.73%和12.87%,转染组细胞凋亡率明显小于转染对照组(q=4.55,P<0.01)和非转染组(q=4.14,P<0.05)。结论FADD-DN基因转染能减少氦氖激光诱导的瘢痕成纤维细胞凋亡,FADD可能是氦氖激光激活死亡信号途径的基本元件之一。  相似文献
