目的:观察血小板反应蛋白1(TSP-1)在转化生长因子β1(TGF-β1)诱导大鼠心肌成纤维细胞中的作用.方法:差速贴壁法分离新生Sprague-Dawley(SD)大鼠心肌成纤维细胞(CFs).MTT法测CFs生长,羟脯氨酸法测CFs胶原含量,RT-PCR法测TSP-1mRNA表达.结果:TGF-β1诱导CFs TSP-l的表达呈浓度一时间依赖性增加(P<0.01).TGF-β1(20ug/L)作用下24 h可诱导CFs增殖和胶原合成(P<0.01).TSP-l反义寡核苷酸作用后的CFs的MTTA值、胶原含量减少(P<0.01).结论:TGF-β1可诱导新生大鼠CFs TSP-1表达,TSP-1反义寡核苷酸对TGF-β1诱导大鼠CFs增殖与胶原合成具有明显的抑制作用,TSP-1在TGF-β1诱导大鼠心肌纤维化中可能具有重要的作用.  相似文献

目的探讨p27、p16、PCNA的蛋白表达与垂体腺瘤侵袭性的关系,为根治垂体腺瘤及减少肿瘤术后复发提供实验依据。方法应用免疫组化SP法检测43例侵袭性垂体腺瘤和37例非侵袭性腺瘤的p16、p27和PCNA蛋白表达水平,分析p16、p27蛋白表达水平与PCNA蛋白表达之间的相关性。结果垂体侵袭腺瘤p16、p27蛋白表达较非侵袭组明显减低;复发组p16蛋白表达的阳性率与非复发组之间无统计学意义。p27蛋白表达的阳性率较非复发组减低有显著差异。结论p16、p27蛋白表达异常与垂体腺瘤的发生及侵袭性有关,它们的表达情况能作为垂体腺瘤侵袭性的参考指标。  相似文献

目的探讨人垂体腺瘤中垂体肿瘤转化基因(PTTG)蛋白的表达与肿瘤侵袭性和增殖程度的关系。方法采用免疫组织化学染色方法检测手术切除石蜡包埋的63例垂体腺瘤(侵袭组45例,非侵袭组18例)组织中PTTG蛋白的表达,染色增殖细胞核抗原(PCNA),同时计数组织内微血管数量(MVD)。结果 PTTG在侵袭性垂体腺瘤中的表达水平显著高于非侵袭性垂体腺瘤。侵袭性垂体腺瘤中PCNA标记指数和微血管密度也显著高于非侵袭性垂体腺瘤。相关分析显示PTTG表达与垂体腺瘤内PCNA标记指数和微血管密度呈正相关(P<0.05)。结论 PTTG在垂体腺瘤形成过程中起重要作用,并与垂体腺瘤的侵袭性和增殖程度密切相关。  相似文献

目的 探讨P16蛋白表达与垂体腺瘤侵袭及复发的关系。方法 应用免疫组化染色技术检测57例垂体腺瘤中P16蛋白的表达水平,并对患者进行了5抻随访观察。结果 垂体腺瘤细胞中P16蛋白表达的阳性率为31.6%;侵袭性组中该蛋白表达的阳性率显著低于非侵袭性组(P<0.01)。复发组11例未见该蛋白表达。结论 P16蛋白的表达阳性率与垂体腺瘤激素分泌功能无明显的相关性,但其表达水平可作为临床评价垂体腺瘤的侵袭性和预测患者复发的指标。  相似文献

目的 观察血小板反应蛋白1(TSP-1)反义寡核苷酸在转化生长因子β1(TGF-β1)诱导大鼠心肌成纤维细胞中的作用。方法 差速贴壁法分离新生大鼠心肌成纤维细胞(CFs)。反义、正义、TSP-1寡核苷酸与TGF-β1(20?g/L)共培养24h ,采用MTT法测CFs生长数目,羟脯氨酸法测胶原含量,RT-PCR 法测TSP-1和VEGF的mRNA表达。结果TGF-β1可以明显促进CFsTSP-1mRNA的表达且呈浓度-时间依赖性(P<0.01)。VEGFmRNA的表达呈浓度-时间依赖性降低(P<0.01)。TSP-1反义寡核苷酸作用后的MTT A 值、胶原蛋白含量减少,VEGF的表达明显增高(P<0.01)。结论TSP-1反义寡核苷酸可抑制TGF-β1诱导的大鼠心肌成纤维细胞增殖、胶原合成和升高VEGF的表达。TSP-1反义寡核苷酸可能具有抗心肌纤维化的作用。  相似文献

目的探讨福辛普利(Fos)和缬沙坦(Val)对血管紧张素Ⅱ(AngⅡ)抑制NRK-52E细胞klotho基因表达的干预作用及klotho与转化生长因子-β1(TGF-β1)的关系。方法将大鼠肾小管上皮细胞(NRK-52E)分为对照组、AngⅡ(10-7mol/L)组、AngⅡ分别再加Fos(10-5mol/L)组、Val(10-5mol/L)组和Fos+Val组,培养24 h后,用反转录-多聚酶链反应(RT-PCR)检测klotho和TGF-β1mRNA表达;用免疫组织化学法和Western blot检测klotho和TGF-β1蛋白表达。对klotho和TGF-β1蛋白表达进行直线相关分析。结果 (1)AngⅡ诱导NRK-52E中TGF-β1mRNA表达上调,同时使klothomRNA表达下调,经Fos或Val或Fos+Val干预后,TGF-β1mRNA表达明显受抑制,而klothomRNA显著上调表达(P<0.05)。(2)Fos、Val和Fos+Val均明显上调AngⅡ诱导的klotho蛋白低表达(P<0.05),同时抑制TGF-β1蛋白高表达(P<0.05)。(3)klotho和TGF-β1蛋白表达呈...  相似文献

目的:研究血管内皮生长因子(VEGF)、血管生成素-1(ANG-1)、血管生成素-2(ANG-2)、血小板反应蛋白-1(TSP-1)的表达与胆管细胞性肝癌(CCC)血管生成和侵润转移的关系。方法: 对33例手术切除的CCC标本进行CD34、VEGF、 ANG-1、 ANG-2 和TSP-1的免疫组化染色,研究VEGF、ANG-1、ANG-2、TSP-1的表达与胆管细胞性肝癌血管生成和肿瘤门静脉侵犯、肝内转移、淋巴结转移以及肿瘤分化水平之间的关系。 结果: 本组CCC的微血管密度(MVD)为(87.2±52.6)/mm2,VEGF、ANG-1、ANG-2 和TSP-1的阳性率分别为75.6%、36.0%、57.6%和45.5%。VEGF和ANG-2的阳性表达与高MVD相关,TSP-1则与MVD负相关(P<0.01,P<0.05,P<0.01)。阳性TSP-1与肝内转移正相关(46.7% vs 5.6%,P<0.05)。结论: CCC瘤内的血管新生活跃,VEGF和ANG-2的阳性表达与CCC血管生成正相关,TSP-1则与其负相关,TSP-1的阳性表达还与肝内转移相关,VEGF、ANG-1、ANG-2的表达与肿瘤的侵润转移未见显著相关。  相似文献

目的探讨转化生长因子β1(TGF-β1)和低氧(CoCl2)对肝癌细胞血管内皮生长因子(VEGF)表达的影响。方法体外培养肝癌细胞系HepG2细胞,实验分为对照组、不同浓度TGF-β1和CoCl2的分别处理组、TGF-β1(100 pmol/L) CoCl2(200 μmol/L)组;用免疫细胞化学法检测VEGF蛋白表达;半定量RT-PCR技术检测细胞中VEGFmRNA相对表达量。结果各处理组VEGF蛋白表达均为阳性;TGF-β1或CoCl2组中VEGF mRNA相对表达量明显高于对照组(P<0.01),并且呈浓度依赖性;TGF-β1 CoCl2组中VEGFmRNA表达高于单因素刺激组(P<0.05)。结论TGF-β1能够刺激HepG2细胞表达VEGF,并且可以协同CoCl2增加VEGF的表达。  相似文献

目的 探讨MMP 9及其抑制因子TIMP 1在垂体腺瘤中表达与肿瘤生物学行为的关系。方法 应用免疫组化S P法检测上述基因蛋白在 2 3例侵袭性和 2 4例非侵袭性垂体腺瘤组织中的表达。结果 侵袭性垂体腺瘤组中MMP 9的表达和MMP 9表达超过TIMP 1的比例高于非侵袭性腺瘤组 (P <0 0 5 ) ;TIMP 1在侵袭性垂体腺瘤组的表达有降低的趋势 ,但无统计学意义 (P >0 0 5 ) ;MMP 9与TIMP 1表达呈正相关 (P <0 0 5 )。结论 MMP 9表达上调导致其和TIMP 1的表达失衡与垂体腺瘤侵袭性有关 ,MMP 9可作为评估垂体腺瘤侵袭性的分子生物学指标  相似文献

目的:观察长期吸烟对大鼠肺血管重塑及转化生长因子-β1(TGF-β1)的影响,探讨其发生的可能机制。方法:将36只健康SD雄性大鼠随机分为对照组、烟雾暴露2周(S-2W)和烟雾暴露12周(S-12W)组。苏木精-伊红染色和α-平滑肌肌动蛋白染色切片观察肺血管重塑的程度,免疫组化法检测增殖细胞核抗原(PCNA)和TGF-β1在肺动脉的相对表达;RT-qPCR检测肺动脉TGF-β1的mRNA表达。结果:随烟雾暴露时间延长,模型组血管壁厚度和血管直径比值(WT%)和完全肌化血管比例均较对照组明显增大,差异具有统计学显著性(P0.01)。模型组的PCNA及TGF-β1蛋白表达水平均较对照组增大,且两模型组之间差异有统计学显著性(P0.01)。与对照组比较,模型组TGF-β1的mRNA表达均显著增加(P0.05),其中S-2W组和S-12W组之间的差异亦有统计学显著性(P0.05)。TGF-β1的mRNA表达与WT%和完全肌化血管比例呈显著正相关(P0.01);TGF-β1的mRNA表达与PCNA蛋白表达呈显著正相关(P0.01)。结论:长期烟雾暴露可导致大鼠肺血管重塑的形成,其机制可能与吸烟上调大鼠肺血管TGF-β1的mRNA表达,诱导肺血管平滑肌细胞增殖有关。  相似文献

A further analysis of already published data supports the position that retardates of low ability level less frequently have retarded siblings, retarded parents, and parents low in occupational level than do retardates higher in ability level. The analysis supports the position that there are two types of retarded individuals, persons retarded as a result of gene or chromosomal anomalies, brain injury, etc., who more frequently occur in the lower-level retardate group, and persons whose retardation represents polygenic segregation, who more frequently occur in the higher-level group.  相似文献

Eighteen families in which both parents had refractions within the range of +4·0 D to −4·0 D and axial lengths seen in emmetropia (22·3-26·0 mm) showed coefficients of correlation of the order 0·5 indicative of polygenic inheritance. Such coefficients were seen for axial length (0·407) and for the cornea (0·487), but not for the lens (which is known to be yoked to the axial length). No such coefficients were seen in 19 families in which one of the parents had axial length outside the emmetropic range (nine families with long axes and 10 with short axes).

The pattern of polygenic inheritance for emmetropia (completely correlated optical components) and errors of refraction up to 4·0 D (inadequately correlated components: correlation ametropia) follows that seen in stature and other measurable characters. In contrast the high refractive errors with their abnormal axial lengths (component ametropia) are—like the extremes in stature—pathological anomalies with monofactorial inheritance.

1. Recovery of responsiveness of single cells in lateral geniculate nucleus of rat has been determined in both P and I cells. There are three types of recovery curve among P cells; (a) early recovery, (b) early partial recovery followed by depression and then complete recovery, (c) prolonged depression followed by cyclic recovery. Type (c) is by far the commonest recovery curve. In contrast to the spike in a P cell, the synaptic potential recovers to its full amplitude in about 20 msec. All I cells exhibit similar rapid recovery curves after a prolonged depression.2. Conditioning stimuli applied to visual cortex also produce a prolonged depression in most P cells but I cells can be re-excited at short intervals from cortex. Decortication does not prevent the prolonged depression of the multineuronal response produced by optic nerve stimulation.3. A neuronal model is proposed to explain these observations. It is supposed that I cells (interneurones) are innervated by axon collaterals of the P cells (principal cells, projecting to visual cortex) and that the I cells exert an inhibitory influence on the P cells.  相似文献

It was established, in experiments on isolated spinal ganglia of adult rats in concluons of intracellular recording, that dopamine (1 M/liter) elicits depolarized responses in 61% of neurons, hyperpolarized in 20% of neurons, and depolarized-hyperpolarized in 19% of neurons. The depolarized responses are associated with the activation of D1 dopamine receptors, and are governed by the shift of cAMP-dependent cation (sodium) channels to the conducting state. The hyperpolarized responses are triggered by the activation of D2 dopamine receptors, which by means of HTP-binding protein convert the potassium channels to the conducting state.
