Objective The present studies aimed at studying the expression and significance of HIF-1α and VEGF-C in gastric carcinoma. Methods SP method was used for the immunohistochemical detection of HIF-1α and VEGF-C proteins in paraffin embedded archival samples from 44 patients with gastric carcinoma and 20 normal gastric tissues. Results In 44 cases, the positive rate of HIF-1αwas 59.09%(26/44)and 23 cases 52. 27%(23/44)showed VEGF-C positive in cytoplasm and they were significantly higher than those in gastric normal tissue. The positive rate of HIF-1α in gastric cancer tissue expression with lymph node metastasis was higher than that without lymph node metastasis(72. 41% 21/29 vs 33.33 % 5/15;x2 = 6.25, P < 0. 05).The positive rate of VEGF-C expression with lymph node metastasis and that without lymph node metastasis was 65. 52%(19/29)and 26. 67%(4/15)respectively(x2 = 4. 53, P < 0. 05). The expression of HIF-1α and VEGF-C was closely correlated with lymph node metastasis and the depth of tumor invasion(x2 = 5. 14,P < 0. 05, and x2 = 4. 38, P < 0. 05), but no relationship with tumor position, sex and age. The positive expression of HIF-1α was consistent with the positive expression of VEGF-C. Conclusion The up regulated expression of HIF-1α and VEGF-C were observed in gastric cancer. The expression of HIF-1α and VEGF-C were correlated with lymph node metastasis and they were of some value for predicting prognosis of gastric carcinoma.  相似文献

Objective : To investigate the mechanism of phenotypic knockout of CXCR4 on T-cell leukemia cell line Molt-4 via SDF-1α/54/KDEL intrakine technology, which the mutant SDF-1α/54, human stromal cell-derived Faceor-1 (SDF-1α) was deleted its C- terminal α-helix and attached with a endoplasimc reticulum retention signal 4-peptide- KDEL encoding gene, so that retain the newly synthesized receptor CXCR4 within the Molt-4 cells endoplasmic reticulum. Methods: The recombinant vector pEGFP-C3/SDF- 1α/54/KDEL were transfected into Cos-7 cells by liposome, SDF-1α/54/KDEL fusion protein was confirmed with western blot. The recombinant plasmids were transfected transiently into Molt-4 by electroporation. Results:Western blot confirmed SDF-1α/54/KDEL expression in Cos-7. A dramatic downregulation of CXCR4 expression on Molt-4 was demonstrated by flow cytometric (FCM) analysis. Conelusion:SDF-1α/54/KDEL and SDF- 1αKDEL have no significant deviation for phenotypic knockout of CXCR4. These suggest that the phenotypic knockout effects of SDF-1α/54 against CXCR4 are not influenced by deleting of SDF-1α helix in the C-terminal.  相似文献

This study was designed to determine the levels of early endothelial progenitor cells(EPCs),apelin,vascu-lar endothelial growth factor(VEGF) and stromal cell-derived growth factor-1(SDF-1) after acute myocardial infarction(AMI),and to investigate the relationships between these cytokines and early EPCs.Early EPCs,de-fined as CD133+,KDR+,and CD34+ cells,were quantified by flow cytometry.The levels of early EPCs and those cytokines in AMI patients were significantly different from those with coronary artery disease or controls(P < 0.05).Plasma apelin levels were inversely correlated with Gensini score and early EPCs(both P < 0.01).Early EPCs,VEGF and SDF-1 showed different patterns of changes in AMI patients during the first 24 h.The trend in the change of early EPCs was proportionally correlated with that of VEGF(P < 0.05).AMI patients exhibited in-creased early EPCs with remarkably decreased apelin levels and enhanced VEGF levels.  相似文献
