1 临床资料患者女,19岁.主因"左耳水肿红斑3 d"于2007年10月9日来空军总医院门诊就诊.患者3 d前无明显诱因自觉左耳烧灼、痒痛,次日晨耳前上部位突然高度水肿、胀痛难忍,自服"扑尔敏"、外用"肤轻松",症状不能缓解,为进一步治疗来我院就诊.患者病程中无发热,精神饮食睡眠均正常.既往及家族史无特殊.体格检查;系统检查未见异常.皮肤科检查:左耳廓前部红肿,触之坚硬,压痛明显,表面无破溃、渗出糜烂,耳垂正常.  相似文献

自我控制与理性和谐的生活秩序息息相关。近十年来关于自我控制的研究表明.自制力类似于肌肉的力量,是有限而易消耗的,休息后能自动恢复;为了克服有限的资源与无限的需要的矛盾.个体倾向于有选择地使用宝贵的自制力资源;通过预期、启动等途径能在一定程度上降低自制力消耗的效应.通过锻炼能够有效地提升自制力的强度。未来的研究最重要的是找到直接测量自制力资源的途径.确立有限自制力理论的说服力。  相似文献

目的 探讨大学生的实际消费心理及消费行为并进行分析.方法 采用问卷调查的方法,并结合一定量的访谈.结果 绝大多数大学生具有健康的消费心理,只有5%左右的大学生具有病态的消费心理,但却有约20%的大学生对消费意义的认知不清.结论 大学生的消费心理及消费行为应受到关注,学校应加强这方面的教育.  相似文献

目的 通过研究骨科创伤患者引发的血栓栓塞及肺动脉栓塞,以探讨对其发病机制、临床特征、高危因素及病死率的早期治疗及预防.方法 统计自2000年2月~2006年1月的骨科创伤患者,合并血栓栓塞及肺动脉栓塞共18例(960人).进行临床资料回顾性研究分析.结果 血栓栓塞占骨科创伤病人并发症的2.76%.肺动脉栓塞占血栓栓塞的10.02%(3人).且全部死亡.其中年龄大于50岁,合并腰椎、骨盆或下肢外伤的病例占所有血栓栓塞病例的56.31%(11人).结论 年龄大于50岁,肥胖病合并腰椎、骨盆或下肢外伤,合并心血管病患,长期制动的患者,引发血栓栓塞发病率明显增高.病人可因下床,排便,咳嗽等而突然导致肺动脉栓塞死亡.诊治和预防血栓栓塞应引起骨科医生的高度重视.  相似文献

乙肝病毒血清标记物与血清HBV-DNA的关系   总被引:1,自引:0,他引:1  
目的:探讨不同乙肝感染模式的患者与HBV-DNA定量结果进行对比分析.方法:采用ELISA方法检测乙肝病毒血清标记物, 荧光定量PCR (FQ-PCR) 检测HBV-DNA, 比较二者之间的关系.结果:HBsAg、 HBeAg、 HBcAb阳性组和HBsAg、 HBeAg阳性组 HBV-DNA检出率较高, 分别为96.61%和100%.HBsAg、 HBeAg、 HBcAb阳性组HBV-DNA含量平均水平为3.86×108, HBsAg、 HBeAg阳性组HBV-DNA含量平均水平为1.73×108;HBsAg、 HBeAb、 HBcAb阳性组和HBsAg、 HBcAb阳性组检出率也较高, 达45% 以上, HBV-DNA含量平均水平分别为1.30×107和1.31×106.结论:FQ-PCR检测HBVDNA能准确地反映体内的HBV真实感染和复制情况, 在乙肝的诊断和治疗中, 不能单凭两对半的检测, 同时应做HBV-DNA的定量测定.  相似文献

Cysteinyl leukotrienes (CysLTs) are a family of inflammatory lipid mediators synthesized from arachidonic acid by a variety of cells, including mast cells, eosinophils, basophils, and macrophages. This article reviews the data for the role of CysLTs as multi-functional mediators in allergic rhinitis (AR). We review the evidence that: (1) CysLTs are released from inflammatory cells that participate in AR, (2) receptors for CysLTs are located in nasal tissue, (3) CysLTs are increased in patients with AR and are released following allergen exposure, (4) administration of CysLTs reproduces the symptoms of AR, (5) CysLTs play roles in the maturation, as well as tissue recruitment, of inflammatory cells, and (6) a complex inter-regulation between CysLTs and a variety of other inflammatory mediators exists.  相似文献

To explore whether the proinflammatory products of the 5-lipoxygenase (5-LOX) pathway are involved in microglia-mediated toxicity toward neuronal cells, we evaluated the effects of 5-LOX inhibitors using an in vitro assay system where human neuronal SH-SY5Y cells are exposed to toxic secretions from THP-1 monocytic cells or human microglia. The specific 5-LOX inhibitors, REV 5901, zileuton, and 5-hydroxyeicosatetraenoic acid lactone; the nonselective LOX inhibitors, phenidone and dapsone; the dual 5-LOX/cyclooxygenase inhibitor, tepoxalin; and the selective inhibitor of the 5-LOX-activating protein (FLAP), MK-886, inhibited such toxicity. The toxicity was enhanced by the 5-LOX product leukotriene (LT)D(4) and reduced by the selective cysteinyl LT receptor (CysLT(1)) antagonist MK-571. The mRNAs for 5-LOX and FLAP were detected in THP-1 cells and human microglia but not in SH-SY5Y cells. The data suggest that inhibition of proinflammatory LT production by 5-LOX inhibition could selectively reduce toxicity of microglial cells and thus be beneficial in neuroinflammatory diseases.  相似文献

Relationship between lipoxygenase and human testicular cancer   总被引:7,自引:0,他引:7  
The metabolism of arachidonic acid by either the cyclooxygenase (COX) or lipoxygenase (LOX) pathway generates eicosanoids, which have been implicated in the pathogenesis of a variety of human diseases, including cancer. They are now believed to play important roles in tumor promotion, progression, and metastasis, and the involvement of LOX expression and function in tumor growth and metastasis has been reported in human tumor cell lines. Expressions of 5-LOX and 12-LOX in human testicular cancer (TC), and normal testis (NT) tissues were examined, as well as effects of their inhibitors on cell proliferation in TC cell line. Expressions of 5-LOX and 12-LOX were detected by immunohistochemistry. Effects of LOX inhibitors on TC cell growth were examined by MTT assay. While 5-LOX and 12-LOX expressions were slightly detected in NT tissues, expressions of 5-LOX and 12-LOX were significant detected in TC tissues by immunohistochemistry. The LOX inhibitors inhibited the growth of TC cells. LOX is induced in TC, and results may suggest that LOXs are essential for cell growth of TC cells.  相似文献

BACKGROUND: Evidence suggests that small airways contribute to clinically significant processes in asthma. Cysteinyl leukotrienes (CysLTs) are considered to be pivotal mediators in the pathogenesis of asthma. Montelukast (MK), a specific CysLT1 receptor antagonist, is metabolized in two main hydroxylated metabolites (termed M5 and M6, respectively). OBJECTIVES: The aims of this study were to compare the responsiveness of small and large human bronchi to the three CysLTs, to evaluate the antagonist activity of MK, M5 and M6 in these preparations of human bronchi, and to characterize the CysLT receptors involved in the contractile response. METHODS AND RESULTS: In isolated small bronchus (i.d. 0.5-2 mm), the potencies (-log molar EC50) of LTC4, LTD4 and LTE4 were 9.3 (n=11), 9.1 (n=30) and 8.4 (n=14), respectively. The three CysLTs were about 30-fold more potent in small bronchi than in larger bronchi (i.d. 4-6 mm). In small bronchi, MK significantly shifted to the right the CysLT concentration-effect curves with pA2 values against LTC4, LTD4 and LTE4 of 9.1 (n=3), 9.0 (n=11) and 8.7 (n=5), respectively. The antagonist potencies of M6 and M5 were similar to MK and fivefold lower, respectively. A similar activity of MK against the three CysLTs suggested that CysLT1 receptors are involved in the contraction of human bronchus. Analysis by RT-PCR also indicated that human bronchus mainly expressed CysLT1 receptors. CONCLUSION: MK exerts a potent antagonist activity against the particularly potent constricting effects of CysLTs in isolated human small bronchi, which only expressed the CysLT1 receptor subtype. The metabolites of MK are also potent in vitro antagonists, but may not participate in the therapeutic activity of MK due to their low plasma concentrations in patients treated with the recommended dose of MK.  相似文献

It has been suggested that the enzymatic pathway of 5-lipoxygenase (5-LOX) influences brain functioning and pathobiology. The mRNAs for both the enzyme 5-LOX and its activating protein FLAP have been found in the cerebellum. In this work, we investigated the cellular expression of 5-LOX in the adult mouse cerebellar cortex. We used the in situ mRNA hybridization assay, immunocytochemistry, laser capture microdissection, and our previously developed method for assaying the DNA methylation status of a putative mouse 5-LOX promoter. Since both 5-LOX mRNA in situ hybridization signal and FLAP immunoreactivity co-localize with calbindin 28 kD immunoreactivity (a Purkinje cell marker) but not with S-100β immunoreactivity (a Bergmann glia marker), the suggestion is that the 5-LOX pathway is expressed in cerebellar Purkinje cells. We found that methylation in the sites targeted by methylation-sensitive restriction endonucleases AciI and HinP1I but not BstUI and HpaII was greater in DNA samples obtained from a high-5-LOX-expressing cerebellar region (Purkinje cells) versus a low-5-LOX-expressing region (the molecular cell layer), suggesting a possible epigenetic contribution to the cell-specific 5-LOX expression in the cerebellum. We propose that Purkinje cell-localized 5-LOX and FLAP expression may be involved in the cerebellar synthesis of leukotrienes and/or could influence the Dicer-mediated microRNA formation and processes of neuroplasticity.  相似文献

There is evidence of an association between depression and anxiety and cardio- cerebro-vascular conditions, but the mechanisms of this association are unknown. Here we review a possible role for the 5-lipoxygenase (5-LOX) pathway. 5-LOX is an enzyme that, in association with 5-LOX-activating protein (FLAP), leads to the synthesis of leukotrienes from omega-6 arachidonic acid. Production of active leukotrienes can be reduced by dietary omega-3 fatty acids, which also are beneficial in cardiac and psychiatric (e.g., depression) pathologies. Human 5-LOX and FLAP gene polymorphisms are a risk factor in atherosclerosis and cardio-cerebro-vascular pathologies; an overactive 5-LOX pathway is found in these diseases. Studies with 5-LOX-deficient transgenic mice suggest that 5-LOX activity may contribute to anxiety- and depression-like behaviors. Future research should characterize the role of the 5-LOX pathway in comorbid cardio-cerebro-vascular and psychiatric disorders and in the therapeutic actions of dietary omega-3 fatty acids.  相似文献

The role of 5-lipoxygenase (5-LOX) in the pathophysiology of the organ injury/dysfunction caused by endotoxin is not known. Here, we investigate the effects of treatment with 5-LOX inhibitor zileuton in rats and targeted disruption of the 5-LOX gene in mice (5-LOX(-/-)) on multiple organ injury/dysfunction caused by severe endotoxemia. We also investigate the expression of beta2-integrins CD11a/CD18 and CD11b/CD18 on rat leukocytes by flow cytometry. Zileuton [3 mg/kg intravenously (i.v.)] or vehicle (10% dimethyl sulfoxide) was administered to rats 15 min prior to lipopolysaccharide (LPS; Escherichia coli, 6 mg/kg i.v.) or vehicle (saline). 5-LOX(-/-) mice and wild-type littermate controls were treated with LPS (E. coli, 20 mg/kg intraperitoneally) or vehicle (saline). Endotoxemia for 6 h in rats or 16 h in mice resulted in liver injury/dysfunction (increase in the serum levels of aspartate aminotransferase, alanine aminotransferase, gamma-glutamyl transferase, alkaline phosphatase, bilirubin), renal dysfunction (creatinine), and pancreatic injury (lipase, amylase). Absence of functional 5-LOX (zileuton treatment or targeted disruption of the 5-LOX gene) reduced the multiple organ injury/dysfunction caused by endotoxemia. Polymorphonuclear leukocyte infiltration (myeloperoxidase activity) in the lung and ileum as well as pulmonary injury (histology) were markedly reduced in 5-LOX(-/-) mice. Zileuton also reduced the LPS-induced expression of CD11b/CD18 on rat leukocytes. We propose that endogenous 5-LOX metabolites enhance the degree of multiple organ injury/dysfunction caused by severe endotoxemia by promoting the expression of the adhesion molecule CD11b/CD18 and that inhibitors of 5-LOX may be useful in the therapy of the organ injury/dysfunction associated with endotoxic shock.  相似文献
