ROLE OF BIVALENT CATIONS AND CHOLINE IN ATP-INDUCED APOPTOSIS OF HUMAN LYMPHOCYTESWITH P2Z RECEPTORS

The role of bivalent cations and choline in ATP induced apoptosis via P2Z purinceeptor was investigated in human leukemic lymphocytes. In vitro exposure of leukemic lymphocytes with P2Z receptors to 1mmol/L ATP or 0.1 mmol/L benzoylbenzoic ATP(13zATP) for 8 h in the presence of choline, 1 mmol/L Mg^2 or other bivalent cations, and ATP induced DNA breaks, associated with apoptosis were quantifie dby TdT assay. We observed that (l) Extracelhalar Mg^2 or Ca^2- stimulated ATP-indueed DNA fragmenration in a dose-depandent manner, and the compatihle evidence was provided by the inhibition of ATP-induced DNA fragmentation in the present of EGTA or EDTA; (2) ATP induced DNA fragmentation was completely inhibited by 1 mmol/L Zn^2 ; (3)ATP-induced DNA breaks were not afl[ected by Ba^2 , Sr^2 ,Co^2 when they were substituted for cxtracellular Mg^2 or Ca^2 ; (4)Chohne, an inhibitor of phospholipase D(PLD) stimulated by ATP through P2Z receptor in human lymphoeytes, was also a partial inhibitor of ATP-induced DNA fragmentation, and the results were coniqrmed by flow eytometrie analysis (FCA);(5)ATP induced DNA fragmentation was completely obliterated when the temperature was lower than 10℃. These results suggest that the endonuclease and PLD may be involved in ATP-induced apoptosis in human lymphocytes via P2Z receptor.

C-TYPE NATRIURETIC PEPTIDE INHIBITS UPREGULATION OF αl-ADRENOCEPTOR AND INOSITOL 1,4,5-TRISPHOSPHATE RECEPTOR IN RAT VASCULAR SMOOTH MUSCLE AFTER VASCULAR ENDOTHELIAL INJURY

Objective. In a model of balloon injury of rat aortic endothelium, the effects of C-type natriuretic peptide(CNP) on al-adrenoreceptar and inositol 1,4,5-triphosphate (IP3) receptor were studied. Methods. Aortic injuries were produced by vascular endothelium-denudation, αl- adrenoreceptor in smooth muscle sarcolemma and IP3 receptor in smooth muscle sarcoplasmic reticulum in the rat aorta were assayed by radioactive analysis method. Results. It was found that neoinfma was formed and the coraents of DNA, collagen and elastin of each intimamedia were significantly increased in 7 days and 21 days after balloon injury of rat aorta, α1-adrenoreceptor in smooth muscle sarcolemma and IP3 receptor in sarcoplasmic reticulum were also upwodated. Results also showed that the administration of CNP i.p significantly decreased the contents of DNA, collagen and elaslin of each iraima-media, and inhibited the up-regulation of α1-adrenoreceptor and IP3 receptor. Conelusion. The inhibition of the up-regulation of α1-adrenoreceptor and IP3 receptor by CNP might be one of the mechanisms of its suppressive action on intimal proliferation.

EFFECTS OF β-ENDORPHIN ON PHYTOHEMAGGLUTININ -INDUCED LYMPHOCYTE PROLIFERATION AND MOUSE PLAQUE-FORMINGCELL RESPONSE VIA AN OPIOID RECEPTOR MECHANISM

The effects of opioid peptides on immune responses were investigated.It was found that β-endorphin(β-END) can depress proliferative responses to PHA in rat splenocytes but enhance those in mice,and it could also inibit the plaque-forming cell(PFC) response to sheep red blood cells when mouse splenocytes immunized in vivo were cultured in vitro with the peptide.The peptide antagonist naloxone was able to reverse β-END suppression of the PFC response.The data indicate that β-END suppresses antibody production or secretion via a specific opioidreceptor-mediated mechanism.

EFFECTS OF INTEGRIN ALPHA Ⅱb^R995A MUTATION ON RECEPTOR AFFINITY AND pp125 （FAK） PHOSPHORYLATION

Objective To investigate the role of cytoplasmic domain of integrin alpha lib in platelet signal transduction. Methods Binding capacity of integrin alpha Ⅱb^R995A to antibody platelet activation complex-1 (PAC-1) and pp125 focal adhesion kinase (FAK) phosphorylation of cells were detected by flow cytometry, immune precipitation, and Western blotting. Without activation, wild-type alpha Ⅱ bbeta3 Chinese hamster ovary (CHO) cells failed to bind to PAC-1, but mutant chimera alpha Ⅱb^R995A beta3 CHO cells were able to bind with PAC-1. Furthermore, phosphorylation ofppl25 (FAK) in wild-type alpha Ⅱ bbeta3 CHO cells occured only when cells were adhered to fibrinogen, but could not be detected in bovine serum albumin suspension. However in the mutant chimera group, it could be detected in both conditions. Conclusion The mutation in integrin alpha Ⅱb^R995A alters its affinity state as a receptor, thus also mediating cytoplasmic signal transduction leading to the phosphorylation of pp125 (FAK) without ligand binding.     

性激素受体在人脑膜瘤中的表达

目的 :了解雌激素受体 (ER)、孕激素受体 (PR)、雄激素受体 (AR)在脑膜瘤中的表达情况以及在脑膜瘤中 3种受体表达间的相互关系。方法 :采用链霉菌抗生素蛋白 -过氧化酶 (SP)免疫组化法对 4 5例脑膜瘤石蜡切片进行 ER、PR和 AR的检测。采用χ2 检验、spearman等级相关分析法对 3种性激素受体间的相关关系 ,以及 3种受体在实验组与对照组中表达的差异性进行统计学分析。结果 :(1)脑膜瘤中存在性激素受体的表达。AR阳性率为 5 3.33% (2 4 / 4 5 ) ,ER为 2 4 .4 4 % (11/ 4 5 ) ,PR为 6 2 .2 2 % (2 8/ 4 5 )。 (2 ) AR与 PR间存在相关性 (r=0 .316 5 ,P<0 .0 5 ) ,AR与 ER、ER与 PR间无相关性 (P>0 .0 5 )。(3) 3种性激素受体在男女患者中的表达差异无显著性 (P均 >0 .0 5 )。结论 :(1) AR、PR在脑膜瘤中有较高的表达率 ,而 ER的表达率较低。 (2 ) AR、PR间存在相关性 ,PR的表达可能受雄激素系统调节。
