目的 测定种植体周围炎患者龈沟液(GCF)中白细胞介素-8(IL-8)水平,探讨IL-8与种植体周围炎的关 系。方法 选择健康种植体和种植体周围炎患者各35例,测定种植体周GCF量并运用双抗体夹心酶联免疫吸附 法检测GCF中IL-8总量及浓度;同时检测种植体周临床指标,包括菌斑指数、探诊深度、牙龈指数和松动度。结果  种植体周围炎GCF量、IL-8总量和浓度均高于健康种植体(P<0·01);GCF量、IL-8总量与临床指标之间有正相关 关系(P<0·01)。结论 种植体周围GCF量及GCF中IL-8水平的变化与种植体周围炎的发生有关,临床检测GCF 中IL-8水平可作为诊断种植体周围炎的客观指标之一。  相似文献

目的:检测种植体周围炎患者龈沟液(gingival crevicular fluid,GCF)中高迁移率族蛋白B-1(high mobility groupbox l,HMGB-1)的水平,以探讨HMGB-1水平与种植体周围炎间的关系。方法 :经患者知情同意,选取行种植手术1年以上的健康种植体39颗和种植体周围炎40颗,共79颗种植体,收集种植体周围GCF,并运用ELISA法检测其中HMGB-1、IL-1β、IL-8、TNF-α的浓度,同时检测种植体周围的临床指标。结果 :轻度种植体周围炎组的HMGB-1、IL-1β、IL-8及TNF-α浓度高于健康种植体组,重度种植体周围炎的浓度水平又高于轻度种植体周围炎组,HMGB-1与IL-1β、TNF-α的浓度之间亦存在正相关关系(P<0.01)。结论:种植体周围GCF中HMGB-1水平变化与种植体周围炎的病变程度有关,且与经典炎症因子间存在显著相关性,临床检测GCF中HMGB-1水平可作为诊断种植体周围炎的客观指标。  相似文献

目的:比较慢性重度牙周炎病人和健康人龈沟液(gingival crevicular fluid,GCF)中可溶性细胞间黏附分子1(soluble intercellular adhesion molecule-1,sICAM-1)的浓度.研究牙周炎是否会引起GCF中的sICAM-1含量改变.方法:纳入慢性重度牙周炎病人和健康人各30例共182个牙,记录临床牙周检查指标,用滤纸条法收集GCF,GCF测量仪Periotron 8000测量GCF总量,用ELISA法测定GCF中sICAM-1浓度.结果:牙周炎病人的GCF总量较健康人高(P<0.01),牙周炎病人的sICAM-1浓度较健康人高(P<0.01).结论:GCF中的sICAM-1浓度的升高可能是由牙周炎症反应引起的,其浓度与GCF总量无明显相关性.  相似文献

目的:探讨龈沟液(GCF)中白细胞介素8(IL -8)在慢性牙周炎病程中的变化、IL- 8与牙周临床检测指标以及同龈下菌斑中细菌含量的相关关系。方法: 5名非牙周炎患者的10颗牙周健康牙和30名慢性牙周炎(CP)患者的70颗牙(其中10颗为健康牙, 60颗为牙周炎患牙)纳入本研究。采集观察牙的GCF样本、龈下菌斑样本,同时记录所有牙的牙龈指数(GI)、牙周袋探诊深度(PPD)、临床附着丧失水平(CAL)。用ELISA法检测样本中IL -8的水平,用厌氧菌培养技术培养菌斑标本,用PCR法检测菌斑中的牙龈卟啉单胞菌(Pg),结果:慢性牙周炎组的GCF中IL -8的总量高于临床牙周健康组,而IL -8的质量浓度在各组中无差异。GCF中IL- 8浓度与GI、PPD、CAL无相关关系,而IL -8的总量与GI、PPD、CAL呈正相关关系。龈下菌斑中Pg的检出量与GCF中IL- 8的含量间未发现有相关关系。结论:慢性牙周炎患者龈沟液中IL- -8量增加与龈沟液分泌增加有关而与牙龈卟啉单胞菌检出CFU无关。  相似文献

目的 探讨牙周基础治疗对牙周炎患牙龈沟液(gingival crevicular fluid,GCF)中白细胞介素(interleukin,IL)-35水平的影响.方法 选取30例慢性重度牙周炎患者的患牙30颗,收集龈上洁治和龈下刮治治疗前后患牙的GCF并记录相关的临床指标.酶联免疫吸附测定法检测GCF中IL-35含量,Pearson相关分析GCF中IL-35与牙龈指数(gingival index,GI)、平均探诊深度(probing depth,PD)、附着丧失(clinical attachment loss,CAL)平均值的相关性.结果 牙周洁、刮治后患牙牙龈GCF中IL-35含量显著低于治疗前,两组比较差异有统计学意义(t=4.043,P<0.01).牙周洁、刮治后患牙GI值和PD值均低于治疗前,治疗前后比较差异有统计学意义(P<0.05).治疗前GCF中IL-35含量与GI值呈正相关(r=0.557,P<0.01),而与患牙牙周袋的PD值和CAL值均无相关性(P>0.05).结论 牙周基础治疗可明显影响患牙GCF中IL-35水平,IL-35可能在牙周炎的发生发展过程中具有重要作用.  相似文献

目的:探讨慢性牙周炎龈沟液(gingival crevicular fluid,GCF)中白细胞介素-8(interleukin-8,IL-8)与硫化物(suleus sulphide level,SUL)的关系。方法:采用双抗体夹心ELISA法、金刚牙周诊断仪对龈沟液中IL-8和硫化物的含量和临床指标进行测定。23个牙周健康牙作正常对照组(C),12个慢性牙周炎健康牙作实验组1(T1),30个慢性牙周炎患牙作实验组2(T2)。用标准化滤纸条采集观察牙位GCF样本,记录相应位点30’硫化物浓度,同时记录龈沟出血指数(SBI)、牙周袋探诊深度(PPD)、牙周附着丧失水平(CAL)。结果:①受检位点龈沟液中IL-8总量存在显著性差别,其中慢性牙周炎炎症牙位组IL-8总量高于健康牙位组、正常对照组(p〈0.05),但龈沟液中IL-8浓度无显著性差别。龈沟液中IL-8总量与SBI、PPD、CAL之间有明显相关关系(P〈0.05),但IL-8的浓度与临床指标间相关关系不明显。②慢性牙周炎炎症牙位组的硫化物浓度与健康牙位组、正常牙位组之间均有显著性差别(P〈0.05),慢性牙周炎健康牙位组与正常牙位组之间无显著性差别。③慢性牙周炎炎症牙位组硫化物浓度与临床指标间具正相关关系(P〈0.05),而健康牙位组和正常对照组硫化物浓度与临床指标间无相关关系。GCF中慢性牙周炎炎症牙位组的硫化物浓度与IL-8总量具负相关关系(P〈0.05),而慢性牙周炎健康牙位组和正常对照组的硫化物浓度与IL-8总量之间无相关关系。结论:慢性牙周炎患牙龈沟液中IL-8和硫化物的含量与临床指标之间有相关性。龈沟液中细菌代谢产物所产生的硫化物对IL-8的含量存在一定抑制作用。  相似文献

目的探讨龈沟液中白介素-18(IL-18)和基质金属蛋白酶-13(MMP-13)表达与种植体周围炎的关系,评价其作为种植体周围炎诊断客观指标的意义。方法选择2011年5月至2013年5月在佳木斯大学口腔医学院口腔种植科行ITI种植体种植的患者30例作为研究对象(种植体40颗),根据种植体周围情况将其分为健康种植体组(28颗)和炎症种植体组(12颗):将对侧同名健康天然牙作为对照(健康天然牙组,40颗)。分别检测各组的牙周探诊深度(PD)、出血指数(SBI)、龈沟液(GCF)量及GCF中的IL-18、MMP-13含量并进行分析。结果炎症种植体组的PD、SBI值显著高于健康种植体组和健康天然牙组,差异有统计学意义(P〈0.05)。炎症种植体组的GCF量以及GCF中IL-18和MMP-13含量均高于健康种植体组和健康天然牙组,差异有统计学意义(P〈0.05)。健康种植体组与健康天然牙组比较,两组的PD、SBI、GCF量以及GCF中IL—18和MMP-13含量的差异均无统计学意义(P〉0.05)。结论IL-18和MMP-13与种植体周围炎有密切关系,可作为种植体周围炎早期诊断的有效检测指标。  相似文献

目的研究慢性牙周炎患者牙周基础治疗前后龈沟液(gingival crevicular fluid,GCF)中IL-17、IL-23表达水平的变化,探讨IL-23/IL-17轴在慢性牙周炎发生、发展过程中的作用。方法选取来我科就诊的慢性牙周炎患者50例为研究对象。于牙周基础治疗前后,采集龈沟液并测量体积,运用夹心酶联免疫吸附试验测定IL-17和IL-23的质量浓度以及测量两者之间的关系。结果牙周基础治疗后龈沟液中的IL-17、IL-23表达水平均较治疗前显著下降,差异均有统计学意义(P<0.05)。牙周基础治疗前后龈沟液中的IL-17、IL-23表达水平与牙周临床指标呈正相关。结论 IL-17、IL-23在慢性牙周炎发生发展过程中起重要作用。  相似文献

目的:探讨维吾尔族绝经早期的牙周炎患者龈沟液IL-6及血清雌二醇(E2)水平与牙周炎的关系。方法:共79例绝经年限均≤5a的妇女纳入本研究。采集30颗牙周健康牙和49颗牙周炎患牙的龈沟液(GCF),记录牙周临床观察指标。采集慢性牙周炎患者的血样本。应用放射免疫分析法检测GCF中IL-6和血清中E2的浓度。结果:牙周健康组GCF中IL-6浓度为(1088.10±102.33)pg/ml;慢性牙周炎组GCF中IL-6浓度明显高于牙周健康牙组(P<0.005);慢性牙周炎患牙GCF中IL-6浓度与GI、PPD、CAL均呈正相关(r=0.564,P<0.005;r=0.335,P<0.05;r=0.324,P<0.05)。血清E2≤30pg/ml组的妇女牙周炎患牙牙周临床指标、GCFIL-6浓度与E2>30pg/ml组无显著差异(P>0.05)。结论:健康牙GCF中含有微量IL-6。牙周炎患牙GCF中IL-6的水平反映了牙周炎症的严重程度,可以尝试作为判断维族妇女牙周病变程度的一个指标。绝经早期患慢性牙周炎的维族妇女,血清E2水平与牙周炎患牙的病变程度及GCF中IL-6的水平无关。  相似文献

目的:检测侵袭性牙周炎(AgP)龈沟液中白细胞介素(IL-8)的总量和浓度并探讨其与牙周临床指标的关系。方法:采用常规滤纸条法收集侵袭性牙周炎实验组患牙(T1)和健康牙(T2)各位点及正常对照组(C)各位点的龈沟液(GCF)样本,用ELISA法检测各样本中IL-8的总量和浓度。结果:3组受检牙龈沟液中IL-8的总量和浓度不同。侵袭性牙周炎患牙组GCF中IL-8总量高于健康牙位组(P〈0.05)及正常对照组;而3组中IL-8浓度差异也有显著性,侵袭性牙周炎患牙组GCF中IL-8的浓度高于健康牙位组(P〈0.05)和正常对照组;虽然GCF中IL-8浓度与牙周探诊深度(PD)、牙龈出血指数(BOP)、附着丧失(AL)无相关关系;但IL-8总量与以上牙周临床指标相关。结论:在侵袭性牙周炎患者龈沟液中IL-8是参与牙周炎症反应的重要调节因子。  相似文献

The aim of the present study was to analyze the levels of osteocalcin, deoxypyridinoline (Dpd) and interleukin-1beta as markers of bone metabolism in peri-implant crevicular fluid (PICF) from peri-implantitis patients. PICF was sampled from a total of 34 endosseous titanium implants from 16 patients; nine females (mean age 52.8, range 40-62 years) and seven males (mean age 56.0, range 36-66 years). The implants had been in place for a period of 9-112 months (mean; 35.8 months) since the loading. These sites were categorized as six peri-implantitis, eight peri-implant mucositis and 20 healthy implant. PICF volume from peri-implantitis sites was significantly higher than mucositis and healthy implant sites (P < 0.01). Osteocalcin levels in PICF from mucositis sites were significantly higher than healthy implants (P < 0.05), whereas peri-implantitis sites were not significantly different from either mucositis or healthy implant sites. Dpd could not be detected in any of the samples examined. IL-1beta levels in PICF from peri-implantitis sites were significantly higher than levels from peri-implant mucositis (P < 0.05) and healthy implant sites (P < 0.01). In conclusion, osteocalcin in PICF may reflect increased local bone turnover around implants. Further, IL-1beta should be a useful marker for peri-implant inflammation.  相似文献

目的:研究IL-8在牙周炎病程中的变化及与临床指标的关系,方法:采用双抗夹心ABC-ELISA法测定慢性牙周炎(CP)患者,健康对照者以及CP治疗前后患者的龈沟液中IL-8含量,IL-8总量,同时检测临床指标并作相关性检验。结果:CP患者龈沟液中IL-8检出率显著高于健康对照者(P<0.025),在CP患者和健康者之间以及CP治疗前后患者的龈沟液中IL-8总量,龈沟液(GCF)量均存在统计学差异(分别为P<0.01,P<0.05),而IL-8含量无统计学差异(P>0.05),IL-8总量,GCF量与临床指标存在正相关性(P<0.01),结论:IL-8总量在牙周炎病程中呈动态性改变,检测GCF中IL-8的水平对评价牙周炎的程度及指导临床治疗有一定价值。  相似文献

The gingival crevicular fluid (GCF) contains various biomarkers, such as interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor-α (TNF-α), and IL-10, among others. These cytokines have been reported to correlate with gingival inflammation and periodontal status. Therefore, the analysis of GCF may be useful for the diagnosis of periodontal status. Pentraxin 3 (PTX3) is the first identified long pentraxin, and is released by several cell types in response to proinflammatory signals. The aim of this study was to determine the levels of IL-1β, IL-6, IL-8, TNF-α, IL-10 and PTX3 in GCF from diseased and healthy sites in patients with chronic periodontitis. Cross-sectional clinical data were obtained from 50 patients with chronic periodontitis. GCF samples were collected with paper strips from one periodontal diseased site and one periodontally healthy site per subject. The levels of IL-1β, IL-6, IL-8, IL-10 and TNF-α were determined using a multiplexed bead immunoassay, and the PTX3 level was measured using an enzyme-linked immunosorbent assay. Mean clinical parameters were significantly higher at diseased sites (P < 0.01) as compared to healthy sites, and the mean levels of PTX3, IL-1β, IL-6, IL-8, IL-10 and TNF-α were higher in diseased sites (P < 0.01) than in healthy sites. There were strong correlations between PTX3 or IL-1β and periodontal status. These results suggest that GCF PTX3 levels might be useful as a diagnostic marker for periodontal disease.  相似文献

ObjectiveComposition of the restorative materials may cause inflammatory responses by monocyte activation and changes in the levels of cytokine released from different cells. Interleukin-6 (IL-6), interleukin-8 (IL-8) and Tumor necrosis factor alpha (TNF-α) are important cytokine for evaluating of the inflammatory process. The aim of this study was to evaluate the different restorative materials used in class V cavities effect on gingival crevicular fluid inflammatory cytokine levels.Design60 individuals having Class V carious cavities participated in the study. Cavities were restored with FiltekZ250, DyractXP, Fuji IX, Cavex avalloy restorative materials. Changes in clinical and biochemical parameters were evaluated before restorations, seven and 21 days after restorations. Contralateral tooth intact enamel surface was determined as control side. Periotron8000 device was used for detection of GCF volume. Cytokine level of GCF was evaluated by Human ELISA kits. Data were analyzed using Mann-Whitney U test and Wilcoxon signed ranks test. The correlations between clinical parameters and biochemical parameters were examined by Spearman's rank correlation analysis.ResultsAfter restorative treatments PI and GI scores were decreased compared with baseline evaluations. There was a significant difference in GCF levels between experimental and control sites in all groups. GCF IL-6 levels in all groups except Filtek Z250, GCF IL-8 levels in all groups except Fuji IX, GCF TNF-α level in only Fuji IX showed significant differences between experimental and control sites.ConclusionsThe obtained data supported that all of the tested materials caused changes in GCF cytokine levels.  相似文献

BACKGROUND: Apoptosis plays an important role in the maintenance of tissue homeostasis. Considering that apoptosis mediators may play a role in the pathogenesis of drug-induced gingival overgrowth, this study was conducted to evaluate p53, bcl-2, and interleukin-15 (IL-15) levels in gingival crevicular fluid (GCF) of cyclosporin A (CsA)-treated patients. METHODS: Twenty renal transplant patients exhibiting CsA-induced gingival overgrowth and 15 systemically healthy gingivitis patients were included in the study; 15 systemically and periodontally healthy volunteer subjects served as the healthy control group. GCF samples were obtained from one interdental site with gingival overgrowth (GO+) and one site without (GO-) from each CsA-treated patient; hyperplasia index, probing depth, papilla bleeding index, and plaque presence were recorded. One site from each gingivitis patient and healthy control was selected, GCF samples were obtained, and the same clinical parameters were recorded. GCF p53, bcl-2, and IL-15 levels were analyzed by enzyme-linked immunosorbent assay. The results were tested statistically. RESULTS: p53 and bcl-2 levels were below the minimum detectable level in all GCF samples analyzed. CsA GO+ and CsA GO- sites, as well as gingivitis sites, exhibited significantly higher GCF levels of IL-15 compared to healthy controls (P<0.05). The difference between CsA GO+ sites and gingivitis sites was not statistically significant, although the total amount of IL-15 in CsA GO+ sites was lower than gingivitis sites (P>0.05). The total amount of IL-15 in CsA GO- sites was significantly lower than gingivitis sites (P<0.05). No significant correlation was found between the clinical parameters and GCF IL-15 levels (P>0.05). CONCLUSIONS: The pathogenesis of CsA-induced gingival overgrowth is multifactorial. The findings of the present study indicate that IL-15 may play a role in the pathogenesis of CsA-induced gingival overgrowth due to its interactions with CsA and its role in apoptosis and inflammation.  相似文献

BACKGROUND: The protein components of GCF can be separated by reverse-phase microbore HPLC on a C18 column with detection on the basis of 214 nm absorbance. A single major symmetrical protein peak eluting with a retention time of 26 min (50% acetonitrile) was evident in gingival crevicular fluid (GCF) from periodontitis patients but not in healthy GCF. This protein was identified as human MRP-8 by N-terminal amino acid sequencing and liquid chromatography quadropole mass spectrometry. AIMS: To quantify the amount of MRP-8 detectable in GCF from individual healthy, gingivitis and periodontitis affected sites and to study the relationship, if any, between the levels of this responsive protein and periodontal health and disease. METHODS: GCF was sampled (30 s) from healthy, gingivitis, and periodontitis sites in peridontitis subjects (n=15) and from controls (n=5) with clinically healthy gingiva and no periodontitis. Purified MRP-8 was sequenced by Edmann degradation and the phenylthiohydantoin (PTH) amino acid yield determined (by comparison of peak area with external PTH amino acid standards). This value was subsequently used to calculate the relative amount of protein in the peak eluting with a retention time of 26.0 min (MRP-8) in individual GCF chromatograms. RESULTS: Higher levels of MRP-8 were detected in inflammatory sites: periodontitis 457.0 (281.0) ng; gingivitis 413.5 (394.5) ng compared with periodontally healthy sites in diseased subjects 14.6 (14.3) ng and in controls 18.6 (18.5) ng, p=0.003. There was at least 20-fold more MRP-8 in the inflammatory compared with the healthy sites studied. CONCLUSIONS: The preliminary data indicate that MRP-8 is present in GCF, with significantly greater amounts present at diseased than healthy sites. A systematic study of the relationship of this protein to periodontal disease could prove useful in further clarifying whether MRP-8 could be a reliable GCF biomarker of gingivitis and periodontitis.  相似文献
