摘    要:目的:探究长链非编码RNA-FENDRR在膀胱癌组织中的表达情况及其与膀胱癌患者临床预后的关系。方法:收集50例(对)膀胱癌组织,用qRT-PCR检测FENDRR的表达量;同样在T24、5637、EJ和 253J-BV 等4种膀胱癌细胞系中验证其表达水平。用χ2检验分析FENDRR和患者临床特征的相关性,用Kaplan-Meier生存曲线分析不同表达组患者的生存状况,用Log-rank检验两组患者的肿瘤特异性生存 率差异。结果:LncRNA-FENDRR在膀胱癌组织中明显下调(t=7.222,P<0.000 1),细胞系中验证结果一致,其中膀胱癌细胞系5637中表达量最低(P<0.000 1)。相关性分析发现FENDRR表达水平与患 者浸润深度有关(χ2=4.612,P<0.05)。生存分析发现,lncRNA-FENDRR低表达组患者的肿瘤特异性生存率显著低于高表达组(HR=3.17,95%CI:1.37~7.32,P<0.05)。结论:LncRNA-FENDRR在膀胱 癌组织中显著下调,其低表达与膀胱癌患者的不良预后有关。

Abstract:Objective: To investigate the expression of long non-coding RNA-FENDRR(FOXF1 adjacent non-coding developmental regulatory RNA) in bladder cancer(BCa) tissues and its relationship with prognosis of patients with BCa. Methods: A total of 50 bladder cancer tissues were collected and qRT-PCR assay was performed to detected the expression of lncRNA-FENDRR, and its expression levels were also verified in four bladder cancer cell lines including T24, 5637, EJ and 253J-BV. Chi-square test was performed to evaluate the correlation between FENDRR expression and clinicopathological features of patients with BCa. Kaplan-Meier method and Log-rank test were performed to determine the correlation between FENDRR expression and prognosis of BCa patients. Results: LncRNA-FENDRR was significantly down-regulated in bladder cancer tissues(t=7.222,P<0.000 1).The results were consistent in bladder cancer cell lines, among which the expression of lncRNA-FENDRR in cell line 5637 was the lowest(P<0.000 1). Correlation analysis showed that expression levels of lncRNA-FENDRR were associated with the depth of invasion in patients(χ2=4.612, P<0.05). Survival analysis revealed that the tumor-specific survival rate of patients with low expression of lncRNA-FENDRR was significantly lower than that with high expression of lncRNA-FENDRR(HR=3.17,95% CI:1.37-7.32, P<0.05). Conclusion: LncRNA- FENDRR is significantly down-regulated in bladder cancer tissues and is associated with poor prognosis of bladder cancer patients.
